Functional properties of active site variants of yeast cytochrome C
Mitochondrial cytochromes c are small, soluble respiratory proteins which exhibit a high degree of amino acid sequence and tertiary structural homologies across the entire eukaryotic kingdom. Theinfluence of mutations within two conserved regions of yeast-iso-1-cytochrome c on the physicochemical
properties of this protein were examined. i) Invariant residue Phe82, located at the exposed heme edge
on the protein surface, was replaced by Tyr, Leu, lie, Ala, Ser and Gly. ii) Internal residues Asn52, Tyr67, Thr78 and 1le75 form the environment about WAT166, a buried water molecule near the heme.
This region undergoes relatively large oxidation-state linked conformational changes and these residues
are collectively referred to as ‘water switch’ residues. The mutants Asn52Ala, Tyr67Phe, Ile75Met and Thr78Gly were studied. The effects of variation at these conserved positions on i) the oxidation-reduction
equilibrium, ii) the electron transfer kinetics with inorganic electron transfer partners, and iii) the
equilibrium and kinetics of heme axial ligand Met8O displacement by added azide and alkaline pH of yeast
iso-1-cytochrome c, were determined.
The reduction potentials (pH 6, 25 °C, μ=0.1M) of position 82 mutants decreased by up to
45 mV with decreasing size of the side chain. Small differences in the ΔH° and ΔS° of reduction were responsible for the observed reduction potentials. Electron transfer rates (corrected for driving force and electrostatics) with the inorganic electron transfer partners Fe(edta)^2- (as reductant) and Co(phen)₃ ³⁺ (as
oxidant) increased up to 17-fold as the size of the mutant side chain decreased (pH 6, 25 °C, μ= 0.1M). Electron transfer reactivity relative to wild type was dependent on the identity of the inorganic electron transfer partner, indicating that mutations at position 82 change that portion of the surface of the
eytochrome that contacts electron transfer partners. Eyring plots of the rate data showed small

Politicising stardom: Jane Fonda, IPC Films and Hollywood, 1977-1982
PhDThis thesis is an empirical analysis of Jane Fonda’s films, stardom, and political activism during the most commercially successful period of her career. At the outset, Fonda’s early stardom is situated in relation to contemporaneous moral and political ideologies in the United States and how she functioned as both an agent and symbol of these ideologies. Her anti-war activism in the early-1970s constituted the apex of Fonda’s radicalisation and the nadir of her popular appeal; a central question of this thesis, therefore, is how her stardom was rehabilitated for the American mainstream to the point of becoming Hollywood’s most bankable actress.
As the star and producer of IPC Films, Fonda developed political projects using commercial formats, namely Coming Home (1978), The China Syndrome (1979), Nine to Five (1980), and Rollover (1981). The final IPC film, On Golden Pond (1981), signalled an ideological breach in this political strategy by favouring a familial spectacle, and duly outperformed its predecessors significantly. The first and last chapters of this work provide historical parameters for IPC in Fonda’s career, while the remaining chapters are structured by the conceptual and political aspects of each IPC project. Julia (1977) is discussed as an IPC prototype through its dramatisation of political consciousness. Coming Home, The China Syndrome, Nine to Five, and Rollover all exhibit this motif whereas On Golden Pond employs melodramatic nostalgia. Often discussed reductively as a star symbolising change, this thesis instead uses archival and published sources to analyse Fonda’s individual agency in historical context, as well as the cultural and political impact of her stardom. The IPC enterprise provided cinematic apparatus for Fonda’s political recuperation within the American mainstream, which, more broadly, harboured significance for the nation’s conservative resurgence at the end of the 1970s

Catalytic Differences between Flavohemoglobins of <i>Giardia intestinalis</i> and <i>E. coli</i>
The sole known heme enzyme of the parasitic protist Giardia intestinalis is a flavohemoglobin (gFlHb) that acts as a nitric oxide dioxygenase (NOD) and protects the organism from the free radical nitric oxide. To learn more about the properties of this enzyme, we measured its nitric oxide dioxygenase, NADH oxidase, and cytochrome c reductase activities and compared these to the activities of the E. coli flavohemoglobin (Hmp). The turnover number for the NOD activity of gFlHb (23 s−1) is about two-thirds of that of Hmp (34 s−1) at pH 6.5 and 37 °C. The two enzymes differ in their sensitivity towards molecules that act as heme ligands. For both gFlHb and Hmp, inhibition with miconazole, a large imidazole ligand, is adequately described by simple competitive inhibition, with KI = 10 μM and 0.27 μM for gFlHb and Hmp, respectively. Inhibition plots with the small ligand imidazole were biphasic, which is consistent with previous experiments with carbon monoxide as a probe that show that the active site of flavohemoglobins exists in two conformations. Interestingly, the largest difference is observed with nitrite, which, like imidazole, also shows a biphasic inhibition plot; however, nitrite inhibits gFlHb at sub-millimolar concentrations while Hmp is not significantly affected. NADH oxidase activity measured under aerobic conditions in the absence of nitric oxide for Hmp was more than twice the activity of gFlHb. The addition of 1 mM hydrogen peroxide in these assays stimulated the NADH oxidase activity of gFlHb but not Hmp. Both enzymes had nearly identical cytochrome c reductase activities but the extent of the contribution of indirect reduction by flavohemoglobin-generated superoxide was much lower with gFlHb (4% SOD-inhibited) than with Hmp (17% SOD-inhibited). Although the active sites of the two enzymes share the same highly conserved residues that are important for catalysis, differences in the distal ligand binding site may account for these differences in activity and sensitivity towards NOD inhibitors. The differences observed in the NADH oxidase and cytochrome c reductase assays suggest that gFlHb may have evolved to protect the protist, which lacks both superoxide dismutase and catalase, from the damaging effects of superoxide by minimizing its production and from peroxide by actively reducing it
Irish Nationalist Organisations in the North East of England, 1890 – 1925
This thesis is the first major study of organised Irish nationalism in the North East of England, set against the wider context of events in Britain and Ireland, from the division that followed Parnell’s fall in 1890 until shortly after the foundation of the Irish Free State and the Irish Civil War. It is a significant contribution to our understanding of the history of the largest ethnic group in Britain before the Second World War – the Irish. It is also an important regional study, revealing the vitality and diversity of the North East’s expression of Irish nationalism that was probably not equalled anywhere else in England and Wales, other than in London. That vitality was manifested in the raising of the Tyneside Irish Brigade for the British Army in 1914. The Tyneside Irish was the crowning achievement of the pre-1918 Irish nationalist organisations in the North East, and arguably in Britain, demonstrating the organisations’ commitment both to John Redmond and to the region, where so many Irish migrants had settled. Irish nationalism’s diversity in the North East was embodied in the Irish Labour Party, which, alone in England, took root on Tyneside, and sought to blend class and ethnic issues at a time of national crisis in Ireland. This organisation casts light on the complex issue of the transference of working-class Irish Catholic allegiance from nationalism to the labour movement in Britain, and, therefore, in the assimilation of that community into the wider British community. Though none of these nationalist organisations has left any extensive archive, this thesis utilises Irish and English manuscript sources, and a wide array of Catholic, labour, and regional newspapers, to demonstrate that these organisations were not only an important part of the history of the Irish in the North East, but also of the North East itself
Current problems of educational administration in the state of Kuwait
This research is an inquiry into the current problems and difficulties of educational administration in Kuwait. The study comprises eight chapters. The introduction defines the scope and purpose of the study and the methodology employed. Chapter Two examines the past and present development of education in the country. Chapter three describes the stages of development of the organisation of the Ministry of Education and its current problems. Chapter Four analyses the trends towards decentralization in the educational system of Kuwait. Chapter Five deals specifically with the individual schools, their administrative structure and function in the Kuwaiti educational system. Chapter Six examines particularly the administrative difficulties in post-secondary education. Chapter Seven discusses the recent academic and administrative problems of university education in Kuwait. The conclusion summarises the current problems of educational administration in Kuwait and Includes suggestions for strategies for change and improvement

UND graduates shared commencement spotlight with honored alumnae Karen Nyberg and Linda Pancratz
On Friday, Aug. 8, in the Chester Fritz Auditorium, the University of North Dakota held one of its largest summer commencement ceremonies on record.
At the ceremony, honorary degrees were bestowed upon NASA Astronaut Karen Nyberg and successful businesswoman Linda Pancratz, alongside about 515 total graduates.
Nyberg graduated from UND with a degree in mechanical engineering and has gone on to become the 50th woman in space. Nyberg delivered last summer\u27s commencement address from the International Space Station. This year she will present to accept her degree.
Pancratz graduated from UND with a bachelor\u27s degree in business administration. She has since become the chairman and chief executive officer of the investment company Mountain Capital LLC.
UND President Robert Kelley presided over the ceremony, his seventh summer commencement. Each year, UND graduates more than 2,700 students in ceremonies in May, August and December.
The commencement will be rebroadcast on Grand Forks Cable Channel 3 on Aug. 12-15 at noon and 8 p.m.

The holiday: Britishness and British film
The representation of the supposed free space of the holiday by a medium of mass entertainment offers a highly condensed image that demands analysis. In my thesis I question the ways in which the holiday film constructs a sense of Britishness based around the idea of community that is shaped and pressured by forces at different historical moments. Modern capitalist society offers us a structure where the holiday is presented to us as the ultimate contrast from work. It is commodified, and we choose to enter into this ideology, take our break, and return to work, refreshed. The holiday also offers a particular type of freedom, which distinguishes it from other forms of leisure. It can be considered as more of an ‘event’ than a weekend break from work, for instance. The emergence of the holiday as a form of mass entertainment for the working class appears to coincide with the birth of cinema in the same respect. By studying the holiday film I try to reveal what it tells us about British culture, the nation and British life, and how cinema audiences may have engaged with and responded to these texts
