    Cardiomyopathy in farmed Norwegian salmon

    This paper describes the gross findings, histopathology, and ultrastructural and scanning electron microscopical (SEM) appearance of farmed Norwegian Atlantic salmon dying from a disease known locally as 'acute heart failure'. Pathological findings were mainly cardiac, and some fish showed haemopericardium due to rupture of the atrial wall. Lesions were largely restricted to the spongy portion of ventricle and atrium, and comprised myocardial degeneration and necrosis, with variable degrees of endocardial-associated hypercellularity, plus macrophage and lymphocyte infiltration; epicarditis was also present in most fish. Transmission electron microscope findings confirmed the myofibrillar disruption, while the SEM showed crater-like discontinuities in the endocardial plasmalemma. The cause and pathogenesis of these severe lesions are unknown; whether they are primarily degenerative or inflammatory requires more detailed study. The definitive version was published in Parasitology 128 (2004): 577-584, doi:10.1017/S0031182004005025.Human serum high-density lipoprotein (HDL) is necessary and sufficient for the short-term maintenance of Plasmodium falciparum in in vitro culture. However, at high concentrations it is toxic to the parasite. A heat-labile component is apparently responsible for the stage-specific toxicity to parasites within infected erythrocytes 12–42 h after invasion, i.e. during trophozoite maturation. The effects of HDL on parasite metabolism (as determined by nucleic acid synthesis) are evident at about 30 h after invasion. Parasites treated with HDL show gross abnormalities by light and electron microscopy.Professor Hajduk was supported by NIH.     Haemorrhagic kidney syndrome of Atlantic salmon, Salmo salar L.

    This report describes a new syndrome affecting farmed Atlantic salmon on the Canadian east coast that has resulted in increased morbidity and mortality in affected stocks. The major pathological findings are apparent only microscopically and include renal interstitial haemorrhage and acute tubular necrosis and tubular casting. As a result, the disease has become known as haemorrhagic kidney syndrome (HKS). Affected fish are lethargic and anorectic, and lark external lesions. Clinically, HKS fish are anaemic, hypoproteinaemic and hyperosmolalic, with increased serum concentrations of sodium and chloride. At necropsy, internal changes ranged from apparently normal to include one or several of the following: swelling and/or patchy reddening of the kidney, pale gills, exophthalmos, serosanguinous ascites, darkening of the posterior intestine and splenomegaly. Ultrastructurally, viral inclusions were found in the cytoplasm of erythrocytes of HKS fish, and there were unusual electron-dense inclusions within the tips of renal tubular microvilli of HKS fish. The significance and relevance of the ultrastructural findings to HKS are unknown. Virus isolation was attempted using CHSE, RTG-2, FH-10, BE and EPC cell lines; no virus was isolated. Bacteriological analysis failed to reveal significant pathogens. Analysis of tissues for heavy metals and pesticides was negative. Assays for clostridial toxins, lipopolysaccharide and verotoxins were negative.     Investigating the mechanism of FOXG1 in glioblastoma stem cells

    Glioblastoma multiforme (GBM) is the most common and aggressive primary brain cancer in adults. Relapse after conventional surgery and chemo/radiotherapy is thought to be driven by glioblastoma stem cells (GSCs). GSCs have phenotypic sumularities to normal neural stem cells (NSCs) and frequently overexpress many key neurodevelopmental transcription factors, including FOXG1, a key forebrain transcription factor with reporgramming activity. Evidence points to an important funtional role of FOXG1 in driving the NSC-like identity of GSCs. Here we explore the mechanisms by which FOXG1 acts, by characterising its key downstream transcritional targets and protein-protein interactions. Analysis of the transcriptional changes induced by FOXG1 overexpression identified key targets, including cell cycle and epigenetic regulators, such as Fox06 and Chd3. Fox06 was identified as a clear downstream target with a functional role in enabling FOXG1 to drive exit from quiescence. This also revealed the FOXG1, a transcriptional repressor, can lead to gene activation. The H3K4 demethylase JARID1B (KDM5B/PLU-1), a negative regulator of promoter activation, is thought to be a protein partner of FOXG1. We hypothesised that FOXG1 may activate gene expression by sequestration of this repressor. However, our studies suggest JARID1B is not essential for FOXG1's ability to drive a proliferative NS cell-like state, nor transcriptional activiation of Fox06 or Chd3. FOXG1 immunoprecipitation studies highlighted difficulties in obtaining good quality, protein-specific antibodies for characterising protein partners. we therefore exploited CRISPR/Cas9 technology to epitope-tag endogenous FOXG1 in patient-derived glioblastoma NS (GNS) cells. These cellular models can now be used in future studies to define FOXG1's key protein partners and aid the identification of ways to therapeutically target FOXG1 in GBM
