    Die dreidimensionale Darstellung biologischer Gewebestrukturen spielt eine zentrale Rolle in der modernen biomedizinischen Forschung. Insbesondere die Blockflächen-Bildgebung in Kombination mit episkopischer Fluoreszenz-Bildgebung (EFIC) ermöglicht eine serielle Aufnahme von Schnittflächen und die anschließende Rekonstruktion hochaufgelöster 3D-Gewebemodelle. Ein zentrales technisches Hindernis bei dieser Methode stellt jedoch die hohe Lichtdurchlässigkeit des Paraffins dar, welches zur Einbettung von Gewebeproben verwendet wird. Aufgrund dieser optischen Transparenz kommt es zu einem störenden „Durchscheinen“ von Signalanteilen aus tieferliegenden Schichten. Dies führt zu Bildüberlagerungen, reduziert den Kontrast und erschwert eine präzise räumliche Zuordnung von Strukturen innerhalb der Probe (Rosenthal et al., 2004). Herkömmliche Ansätze wie die Verwendung von Carbon Black konnten dieses Problem bislang nur unzureichend lösen, da sie entweder zu inhomogener Färbung oder zu starker Signalabsorption führten. Ziel dieser Arbeit war es, eine Methode zur effektiven Kontrastverbesserung bei EFIC durch Einfärbung des Paraffins mit Sudan IV zu entwickeln und zu evaluieren. Dabei wurden verschiedene Konzentrationen von Sudan IV (0,1 %–0,8 %) in Kombination mit 0 %–10 % Stearinsäure getestet, um eine möglichst homogene, lichtabsorbierende Paraffinmatrix zu erzeugen, ohne dabei die natürliche Autofluoreszenz des Gewebes oder die Schnittqualität zu beeinträchtigen. Die bildgebenden Untersuchungen zeigten, dass mit zunehmender Sudan-IV-Konzentration eine progressive Reduktion des Durchscheinens erreicht werden konnte. Die Paraffinmischung mit 0,4 % Sudan IV und 5 % Stearinsäure erwies sich als optimal: Sie ermöglichte eine deutliche Kontraststeigerung an der Schnittfläche, eine klare Abgrenzung zu tieferliegenden Gewebeschichten sowie eine sehr gute Erhaltung der Autofluoreszenzstruktur innerhalb des Lymphknotens. Höhere Konzentrationen führten zu keiner weiteren Verbesserung, sondern verstärkten stattdessen Inhomogenitäten und beeinträchtigten die Handhabung. Ein Vergleich mit klassischen histologischen Färbungen (Hämatoxylin-Eosin und Azan) zeigte, dass die durch Autofluoreszenz gewonnenen Bilder grundlegende morphologische Strukturen wie Kapsel, Trabekel und Kompartimentierung zuverlässig abbilden können. Es bestand eine klare strukturelle Übereinstimmung zu den gefärbten Referenzschnitten. Gleichzeitig wurde jedoch deutlich, dass die Auflösung und der Kontrast der Autofluoreszenzbildgebung begrenzt sind. Für detaillierte zelluläre Differenzierungen und die Identifikation feingeweblicher Merkmale bleiben klassische histologische Färbungen unverzichtbar. Die Autofluoreszenz bietet somit eine nützliche Ergänzung, aber keine vollwertige Alternative zur differenzierten Gewebeanalyse. Insgesamt zeigt diese Arbeit, dass Sudan IV – insbesondere in Kombination mit Stearinsäure – ein wirkungsvolles Mittel zur Kontrastierung von Paraffinblöcken für die EFIC darstellt. Die entwickelten Protokolle verbessern die Bildqualität signifikant und bilden die Grundlage für eine standardisierbare volumetrische Histologie zur präzisen Analyse biologischer Gewebe.The three-dimensional visualization of biological tissue structures plays a central role in modern biomedical research. In particular, block-face imaging combined with episcopic fluorescence imaging (EFIC) enables the serial acquisition of tissue sections and the subsequent reconstruction of high-resolution 3D tissue models. However, a major technical challenge of this method is the high optical transparency of paraffin, which is commonly used for tissue embedding. This transparency results in unwanted signal contributions from deeper layers “shining through,” leading to image superimpositions, reduced contrast, and difficulties in the precise spatial localization of structures within the sample. Conventional approaches, such as the use of carbon black, have thus far been insufficient to overcome this issue, as they typically result in either inhomogeneous staining or excessive signal absorption. The aim of this study was to develop and evaluate a method for effective contrast enhancement in EFIC by staining paraffin with Sudan IV. Different concentrations of Sudan IV (0.1%–0.8%) in combination with 0%–10% stearin-acid were tested in order to generate a homogeneous, light-absorbing paraffin matrix without compromising tissue autofluorescence or sectioning quality. Imaging experiments demonstrated that increasing concentrations of Sudan IV progressively reduced signal bleed-through. A paraffin mixture containing 0.4% Sudan IV and 5% stearin-acid proved optimal: it provided a marked improvement in contrast at the block surface, a clear separation from deeper tissue layers, and excellent preservation of autofluorescence structures within the lymph node. Higher concentrations, in contrast, did not further improve image quality but instead caused inhomogeneities and impaired handling. Comparison with conventional histological stains (hematoxylin–eosin and Azan) showed that autofluorescence-based images reliably depicted key morphological features such as capsule, trabeculae, and compartmentalization, with clear structural correspondence to stained reference sections. However, it also became evident that the resolution and contrast of autofluorescence imaging are limited. For detailed cellular differentiation and the identification of fine tissue features, classical histological stains remain indispensable. Autofluorescence imaging therefore represents a valuable complement, but not a full substitute, for differentiated tissue analysis. In summary, this study demonstrates that Sudan IV—particularly in combination with stearin-acid —is a powerful tool for enhancing the contrast of paraffin blocks in EFIC. The developed protocols significantly improve image quality and provide a foundation for standardized volumetric histology, enabling more precise analyses of biological tissues

    In my study I analyze a significant late-medieval memoir, known as „The Memoires of Helene Kottanner (1439–1440), written by Helene, or Elena Kottanner, an Austrian woman, daughter of Peter Wolfram from Ödenburg (Sopron), acting in the service of Queen Elizabeth of Hungary. The source, which may be considered the oldest German memoir written by a secular woman, depicts the events of an interesting period of the medieval Hungarian history with “vividness and poignancy” (Maya Bijvoet Williamson). After the death of King Albert (1437–1439), his ambitious wife, Elizabeth – while a large part of the Hungarian nobles wanted Władysław III of Poland to be the king of Hungary – tried to maintain his own authority in Hungary (at that time she was already pregnant and hoped that her new-born will be a boy), therefore she ordered her servant, Elena Kottanner to steal the Holy Crown of Saint Stephen from the royal stronghold, Plintenburg (Visegrád). The woman and an unnamed Hungarian collaborator managed to remove the Crown secretly, rushing to the Queen with it, who within an hour of the crown’s arrival at her castle of Komorn (Komárom), bore a son, Ladislaus Posthumous (1440–1457). Three months later, the little boy was crowned King of Hungary in Stuhlweissenburg (Székesfehérvár). In her memoir, Helene Kottanner – as an eye-witness author – gives a unique, detailed and remarkable picture about these events. My main goal is to analyze the rhetorical methods, the historiographical practice presented by Helene, which can be detected in her text. I demonstrate that the servant intended to emphasize her own role in the mentioned events, and tried to legitimate the Hungarian kingship of Ladislaus Posthumous. Besides, she presented certain events as symbolic of the fate of the future king: according to Helene, God protects her and Elizabeth, and the whole undertaking, while the Devil is on the side of their enemies (Władysław III of Poland and the Hungarian nobles). Furthermore, I also intend to demonstrate what possible goals could Helene have had with her memoir. It is quite possible that the servant wrote her opus in order to get her reward for her services provided to the queen and the future king. I demonstrate this problem in context of the Hungarian political situation in the 1440s and the 1450s

    Thesis (Master's)--University of Washington, 2016-06Helene is a cross-genre text through the lens of a young female woman, Helene, that both ruminates on the grief caused by having a female body, specifically one that fails to meet the expectations of others, and pays homage to the possibility of life without a body. The title character, grappling with the death of her grandfather and its impact on her sense of self and body, processes her grief through various forms: letters written to her grandfather's spirit, fragmented prose and poetry, an episode of an imaginary television sitcom of Helene's life where well-known figures stand in for her family members, and magazine articles, advertisements, and self-guided exercise that execute Helene's desire to exist in the spiritual realm. Writing Abuse, Shame, and Death: A Poetics of Spirit within the Failing Body studies the creation of Helene as writing in spite of body through the practice of mediumship. It discusses the text as a response to the death of the author's own grandfather, a death that helped her realize the acts of repression in her life and her writing. Helene and Writing Abuse, Shame, and Death: A Poetics of Spirit within the Failing Body are thus a process, not product, of grieving that moves through the internal conflict the author has felt as a woman mourning both the loss of a loved one and the temporary loss of identity and agency as a result of abuse

    Chick Lit in Historical Settings by Frida Skybäck

    Chick lit is mostly contemporary portrayals of single women in cities. The Swedish author Frida Skybäck writes “chick lit in corsets”, that is, chick lit in a historical setting, and she writes primarily for teenage girls. Her two novels Charlotte Hassel (2011) and Den vita frun (2012) balance between chick lit jr. and romance. They can also be read as historical novels, and in my article I highlight how Skybäck has consciously played with the different genres to convey a feminist message and to strengthen young readers

    I have written this to myself as much as for you. As you’re about to, I have seen what these characters saw and heard in the trees and the waves of the ocean. The cries of pain and joy in their cellular memory will surely speak to yours. Like you, I came here to find words for what I’ve experienced and to give life to those experiences on the page. What is known to me without words wanted to be spoken through the paper, intuitively transferred with textures felt softly, coarsely, deeply. Inside this collection of poetry, translations, and prose including both fiction and non-fiction, you’ll receive a doorway into those intuitions. Some of your questions won’t have answers until you feel them real, but they will invite you in to yourself to find what mysterious qualities you have. You might lose time while underground, but rest assured, you will find your fire in the voices of the wind and sea if you dare to look further and aren’t afraid to let go of expectations. You may find pieces of you here and wonder about your own motives and purposes. Clues and details of what really happened keenly seen by the poet and author will help you see into the unexplored aspects of your own heart channels and thought fields. I sincerely hope my journey as expressed herein takes you to the parts of you that aren’t easy to articulate or even acknowledge, and to the parts of you that have always been there, waiting to be disclosed; waiting to be seen from unseen angles.
