A contribution to the inhibition of the proprotein convertase furin
Aufgrund der vielfältigen Funktionen in zahlreichen pathologischen Prozessen ist Furin ein
attraktives Zielmolekül zur Wirkstoffentwicklung. Obwohl eine dauerhafte Furinhemmung
möglicherweise zu Nebenwirkungen führt, ist eine kurzzeitige oder mitelfristige Therapie akuter
furinabhängiger Infektionen durch spezifische Hemmstoffe durchaus denkbar. Die antivirale oder
antibakterielle Wirkung vieler Furininhibitoren wurde in Zellkulturen belegt.
Die antivirale Aktivität ausgewählter Furininhibitoren wurde in unterschiedlichen Zellkulturtests
untersucht. Bei den verwendeten Konzentrationen zeigten die Inhibitoren eine vernachlässigbare
Zytotoxizität (Zellviabilität > 80 %).
Furin ist eine Proproteinkonvertase, deren physiologische Aufgabe in der Prozessierung
lebensnotwendiger Vorläuferproteine (Proproteine) besteht. Andererseits ist die Furinaktivität auch
mit unterschiedlichen pathologischen Prozessen assoziiert. Dazu zählen einige Krebsarten, diverse
Stoffwechselerkrankungen sowie verschiedene bakterielle und virale Infektionen. Daher ist Furin
ein interessantes Zielmolekül für die Entwicklung entsprechender Arzneistoffe.The serine protease furin belongs to the family of proprotein convertases and fulfills important physiological functions in the processing of numerous vital precursor proteins (proproteins). On the other hand, furin is also associated with diverse pathological processes, e.g., various bacterial and viral infections. Therefore, furin emerged as an attractive target for drug development.
Based on the consensus furin cleavage site comprising the multibasic sequence -Arg-Xaa-Arg/Lys-Arg- (Xaa: any amino acid), new fluorogenic furin substrates and substrate-analogue furin inhibitors were synthesized within this work. All compounds were obtained by a combination of solid phase and solution synthesis.
The new substrates are more efficiently cleaved and possess improved kcat/KM values compared to already known derivatives. The improved specificity constants enable more sensitive kinetic measurements at reduced substrate and enzyme concentrations. Compared to the previously used compound Phac-Arg-Val-Arg-Arg-AMC, the new substrate acetyl-Arg-Arg-Tle-Arg-Arg-AMC exhibits a 30 times higher specificity constant and enables a more convenient Ki value determination of highly potent furin inhibitors (which were previously characterized as tight-binding inhibitors) under classical, non-tight-binding conditions.
Several cyclic furin inhibitors were developed by selective modifications of a previously published structure with pronounced cell penetrating properties. All these oligoarginine-containing compounds with the benzamidine moiety 4-Amba (4-amidinobenzylamide) in P1 position exhibit a high inhibitory potency towards furin. Moreover, the potency of previously developed linear oligoarginine derivatives with a C-terminal 4-Amba residue could be improved by converting their N-terminal α-amino group into a more basic guanidine.
The previously published compound 4-guanidinomethyl-Arg-tert-Leu-Arg-4-Amba (MI-1148, Ki = 5.5 pM), developed in our group, is the most efficient furin inhibitor known so far. In a first approach, its backbone has been C-terminally elongated by replacement of the P1 4-Amba with various stable or cleavable P1-P1’ segments. However, the most potent of these derivatives possesses only a moderate Ki value of 20 nM. In another attempt, a series of stepwise C-terminally truncated derivatives was developed. The loss of the P1 benzamidine led to a
dramatic drop in the inhibitory potency. Interestingly, the truncated derivatives showed an unexpected behaviour in the kinetic test, deviating from the usually observed classical competitive inhibition mechanism. As the binding mode on furin was not known, only IC50 values could be determined for these compounds.
All effective synthetic furin inhibitors are multibasic compounds. The results of previous work also revealed that a basic P1 residue is of prime importance for a strong furin inhibition by peptidomimetic inhibitors. However, numerous adverse effects have been described in literature for benzamidine derivatives. Therefore, the 4-Amba residue in inhibitor MI-1148 was replaced by various other groups, most of which are less basic. Interestingly, some of these derivatives showed the same aberrant enzyme kinetic behaviour as observed for the truncated peptides. According to mathematical and graphical analysis, it could be classified as a parabolic competitive inhibition. This kinetic model is characterized by the simultaneous binding of two inhibitor molecules on two different sites in one enzyme molecule. The P1 benzylamine and aminopyridine analogues exhibit a relatively high inhibitory potency. Even more active is a fluoro-substituted benzamidine compound (Ki = 22.3 pM), which also effectively inhibits the propagation of Dengue and West Nile viruses and the cleavage of the hemagglutinine precursor of the highly pathogenic avian influenza virus A H7N1 in cell culture.
A first study in mice revealed a suboptimal toxicological profile for inhibitor MI-1148. By contrast, its tribasic analogues possessing a neutral P5 phenylacetyl residue (still a strong furin inhibitor) or P1 benzylamide moiety (a poor inhibitor) were better tolerated. The similar toxicity of these two compounds suggests that the low tolerability in vivo does not correlate with the strength of furin inhibition and is rather caused by unspecific off-target effects.
The results of the study also suggest that the toxicity depends at least to some extent on the overall multibasic character of the inhibitor MI-1148. However, no suitable alternative for the P1-Amba residue could be identified thus far. Besides, benzamidine groups are found in some marketed drugs. This suggests to retain the benzamidine in P1 position and replace other residues instead. Since the basic P5 and P4 residues are also essential for furin inhibition, only the P2 arginine was replaced by the slightly less basic lysine; the resulting inhibitor is nearly as effective as the P2 arginine derivative in cell culture and shows a better tolerability in vivo
Synthesis and characterization of substrate-analogous inhibitors of the serine proteases Matriptase and Kallikrein 7
Die Serinproteasen Matriptase und Kallikrein 7 sind in einer Vielzahl physiologischer Prozesse involviert. Eine gestörte Aktivität dieser Proteasen ist für eine Reihe von Erkrankungen verantwortlich.
Daher ist es von therapeutischem Interesse, die Aktivität der Proteasen regulieren zu können.
Zu diesem zwecke wurde im Rahmen der vorliegenden Arbeit eine Reihe von substratanalogen Hemmstoffen für Matriptase und Kallikrein 7 entwickelt und auf ihre Hemmwirkung hin untersucht.
Für beide Protease konnten wirksame und selektive Inhibitoren dargestellt werden.The serine proteases matriptase and kallikrein 7 are involved in a variety of physiological processes. A disturbed activity of these proteases is responsible for a number of diseases.
Therefore, it is of therapeutic interest to be able to regulate the activity of these proteases.
For this purpose, a series of substrate-analogue inhibitors for matriptase and kallikrein 7 have been developed and investigated for their inhibitory potency.

Entwicklung und Charakterisierung neuer peptidomimetischer Inhibitoren des Westnil Virus NS2B-NS3 Protease
Potent inhibitors of the West Nile virus NS2B-NS3 protease could be useful drugs for the treatment of WNV infections. Therefore, in the present work, new substrate analogue inhibitors against the WNV protease have been developed.
In this present work, new decarboxylated arginine memtics inhibitors against the WNV NS2B-NS3 protease were developed. In comparison to the known agmatine derivatives, the P1 trans-4-Guanidinocyclohexylmethylamid containing inhibitors have higher potency and selectivity against the WNV. Moreover, the first crystal structure of the WNV protease in complex with small noncovalently-binding inhibitor was solved.Wirksame Hemmstoffe der WNV NS2B-NS3-Protease könnten nützliche Wirkstoffe für die Behandlung von Erkrankungen nach WNV-Infektionen sein. Daher wurden in der vorliegenden Arbeit neue, substratanaloge Inhibitoren gegen die WNV-Protease entwickelt und charakterisiert.
Im Rahmen dieser Arbeit ist es durch Einbau besonders geeigneter, decarboxylierter Argininmimetika gelungen, Hemmstoffe der WNV NS2B-NS3-Protease mit Hemmkonstanten um 0,15 μM zu erhalten. Im Vergleich zu den bereits bekannten Verbindungen mit Agmatin in P1-Position besitzen die Derivate mit einem trans-4-Guanidinocyclohexylmethylamid eine erhöhte Selektivität gegenüber einigen trypsinartigen Serinproteasen, wie Matriptase. Mit einem der neu entwickelten Inhibitoren konnte erstmals eine Kristallstruktur der WNV-Protease mit einem nichtkovalent-bindenden niedermolekularen Inhibitor gelöst werden
Development of host serine protease inhibitors for antiviral therapy
Die Entwicklung neuer Strategien zur antiviralen Therapie ist von entscheidender Bedeutung, da viele Viren eine erhebliche Bedrohung für die globale Gesundheit darstellen. Nach dem Eindringen in ihren Wirt sind die meisten Viren auf die Aktivität von Wirtsproteasen angewiesen, um sich zu replizieren. Daher kann die Hemmung solcher Proteasen die Virusausbreitung verringern. Der Fokus dieser Arbeit lag auf dem Design, der Synthese und der Charakterisierung von Inhibitoren für die Wirtsproteasen Matriptase und S1P im Kontext potenzieller neuer antiviraler Wirkstoffe.
Projekt A: Matriptase-Inhibitoren:
Die membrangebundene trypsinartige Serinprotease Matriptase kann die Hämagglutinine bestimmter Influenzaviren spalten und damit potenziell zur Virusreplikation beitragen. In unserer Arbeitsgruppe wurde in den letzten Jahren eine Vielzahl von Matriptase-Inhibitoren vom 3-Amidinophenylalanin-Typ (Phe(3-Am)) entwickelt. Ziel dieser Doktorarbeit war es, hauptsächlich monobasische Matriptase-Inhibitoren des Phe(3-Am)-Typs mit hoher Selektivität gegenüber den Gerinnungsproteasen Thrombin und Faktor Xa zu entwickeln und zu synthetisieren, da die Hemmung dieser Enzyme potenziell zu Blutungskomplikationen führen kann. Dies sollte durch eine gründliche Analyse der aktiven Zentren aller drei Enzyme und die Einbeziehung von Docking-Studien erreicht werden. Frühere Hemmstoffserien lieferten zwei monobasische Matriptase-Inhibitoren (5 und 28) mit einer hohen Selektivität gegenüber Thrombin (SF 126 bzw. 410), die mit einer mutmaßlichen Kollision des Thrombin-spezifischen 60er Insertionsloops mit der Cyclohexylureidogruppe der Hemmstoffe begründet wurde. In dieser Arbeit wurden sowohl das C-terminale Piperidid als auch die Harnstoffgruppe derivatisiert, wobei letztendlich ein Piperidyl-4-tert-butylharnstoff als optimale C-terminale Gruppe zum Erreichen einer hohen Selektivität gegenüber Thrombin identifiziert wurde. In Kombination mit geeigneten Substituenten am N-terminalen Biphenylsystem wurden mehrere neue monobasische Derivate mit solcher Selektivität entwickelt. Die höchste Selektivität wurde für die monobasische Verbindung I5 (SF 1533) und den dibasischen Inhibitor I20 (SF 3643) erreicht.
Während der C-Terminus modifiziert wurde, um die Affinität für Thrombin zu verringern, sollten die Substituenten am N-terminalen Biphenylsystem eine erhöhte Selektivität gegenüber FXa gewährleisten. Diese Gerinnungsprotease weist einen Phe174-Rest in ihrer S3/4-Binderegion auf, an analoger Position befindet sich in Matriptase ein Glutamin (Gln175). Basierend auf der Kristallstruktur eines verwandten Phe(3-Am)-Inhibitors im Komplex mit Matriptase (PDB: 2GV6) wurden Docking-Studien durchgeführt, um N-terminale Substituenten zu identifizieren, die möglicherweise in der Lage sind, Gln175 der Matriptase zu adressieren, wodurch nur die Affinität für Matriptase und nicht für FXa verbessert werden sollte. Diese Studien schlugen 3- und 4-Hydroxymethylgruppen als geeignete Substituenten für diese Wechselwirkung mit Gln175 vor, welche somit die Grundlage für alle im Rahmen dieser Arbeit durchgeführten N-terminalen Variationen bildeten. Die erhaltenen Inhibitoren zeigten jedoch nicht die gewünschte Selektivität gegenüber FXa. Von allen 24 neuen Derivaten zeigt nur die dibasische Verbindung I21 (SF 105) eine adäquate Selektivität. Es wurden vier neue Kristallstrukturen von drei verschiedenen Inhibitoren (I5, I12 und I20) im Komplex mit Matriptase, Thrombin und Trypsin erhalten, die das Vorliegen der vorhergesagten Wechselwirkung zwischen Gln175 und der 4-Hydroxymethylgruppe von Inhibitor I5 belegen konnten. Darüber hinaus ermöglichten die Strukturinformationen dieser Komplexe die Etablierung detaillierterer Struktur-Wirkungsbeziehungen, welche die Weiterentwicklung dieses Inhibitortyps unterstützen könnten. Ausgewählte Hemmstoffe zeigten eine starke inhibitorische Wirkung auf die Replikation eines Matriptase-abhängigen H9N2-Influenzavirusstamms.
Projekt B: S1P Inhibitoren:
Die site-1 protease (S1P) ist ein Mitglied der Proproteinconvertasen und an der Regulierung des intrazellulären Cholesterin- und Fettsäurehaushalts beteiligt. Darüber hinaus spaltet es die Glykoprotein-Vorläufer aller bekannten humanpathogenen Arenaviren und stellt damit einen interessanten Angriffspunkt für eine gezielte antivirale Therapie dar. Bisher wurde nur ein einziger Artikel über potente, reversible S1P-Inhibitoren mit in vitro-Aktivität veröffentlicht und alle folgenden Studien zur Wirkung der S1P-Hemmung auf verschiedene biologische Systeme wurden mit der in diesem Artikel beschriebenen Verbindung PF-429242 durchgeführt. In dieser Arbeit wurden peptidische Boronsäuren basierend auf der Substratsequenz RRLL↓ und deborylierte Analoga als neue S1P-Inhibitorklasse etabliert. Des Weiteren wurden weitere Derivate der Verbindung PF-429242 synthetisiert. Die Synthese peptidischer Boronsäure-Inhibitoren wurde optimiert und umfangreiche Studien zur Etablierung von Boronsäure-SPPS durchgeführt. Die enzymkinetische Charakterisierung zeigte, dass peptidische Boronsäuren hochwirksame S1P-Inhibitoren sind und lässt vermuten, dass die Boronsäurefunktion kovalent mit dem Serin im aktiven Zentrum interagiert. Verbindung I34, die ein Borocyclopentylglycin in P1-Position trägt, bindet an S1P mit einem IC50-Wert von 73 nM und damit fünfmal stärker als der Referenzinhibitor PF-429242 (IC50 = 374 nM).
Im Falle der Derivate von PF-429242 wurden die Reaktionsbedingungen für beide reduktiven Aminierungen, die zur Synthese solcher Inhibitoren erforderlich sind, optimiert. Keines der neuen Derivate der veröffentlichten Verbindung PF-429242 zeigte jedoch eine höhere S1P-Affinität als die Referenz selbst.The development of novel strategies for antiviral therapy is of crucial importance, as many viruses represent a significant threat to global health. After entering their host, most viruses require the action of host proteases to replicate. Hence, the inhibition of such proteases can decrease the viral spread. This work focused on the design, synthesis, and characterization of inhibitors for the host proteases matriptase and S1P in the context of potential novel antiviral agents.
Project A: Matriptase inhibitors
The membrane-bound trypsin-like serine protease matriptase can cleave the hemagglutinins of certain influenza viruses, thereby potentially contributing to viral replication. In our working group, a multitude of matriptase inhibitors of the 3-amidinophenylalanine (Phe(3-Am)) type have been developed over the past years. In this doctoral thesis, the aim was to design and synthesize mainly monobasic matriptase inhibitors of the Phe(3-Am)-type with a high selectivity against the clotting proteases thrombin and factor Xa, as inhibiting the latter enzymes can potentially cause bleeding complications. This was supposed to be achieved by thorough analysis of the active sites of all three enzymes and the incorporation of docking studies. Previous compound series had provided two monobasic matriptase inhibitors (5 and 28) with a high selectivity against thrombin (SF 126 and 410, respectively) that was linked to a putative steric clash between the 60 insertion loop of thrombin with the inhibitors’ C-terminal cyclohexyl ureido group. In this work, the C-terminal piperidide as well as the ureido moiety were derivatized, ultimately revealing a piperidyl-4-tert-butyl urea as the optimal C-terminal group to achieve a high selectivity against thrombin. In combination with appropriate substituents on the N-terminal biphenyl system, several new monobasic derivatives with such selectivity were developed. The highest selectivity was achieved for monobasic compound I5 (SF 1533) and the dibasic inhibitor I20 (SF 3643).
While the C-terminus was modified to decrease affinity for thrombin, the substituents on the N-terminal biphenyl system were intended to provide an enhanced selectivity against fXa. This clotting protease features a Phe174 residue in its S3/4 binding region, where matriptase carries a Gln in the analogous position (Gln175). Based on the crystal structure of a related Phe(3-Am) inhibitor in complex with matriptase (PDB: 2GV6), docking studies were performed to identify N-terminal substituents that are potentially able to address Gln175 of matriptase, thereby improving only the affinity for matriptase and not for fXa. These studies suggested 3- and 4-hydroxymethyl groups as appropriate substituents for this interaction with Gln175, providing the basis for all N-terminal variations performed in the context of this work. However, the obtained inhibitors did not exert the desired selectivity against fXa. Of all 24 new derivatives, only the dibasic compound I21 (SF 105) shows a decent selectivity. Nevertheless, four new crystal structures of three different inhibitors (I5, I12 and I20) in complex with matriptase, thrombin and trypsin were obtained, proving the presence of the predicted interaction between Gln175 and the 4-hydroxymethyl group of inhibitor I5. Moreover, the structural information provided by these complexes allowed the establishment of more detailed structure-activity-relationships that could aid the further development of this inhibitor type. Selected inhibitors demonstrated a strong inhibitory effect on the replication of a matriptase-dependent H9N2 influenza virus strain.
Project B: S1P inhibitors
The site-1 protease (S1P) is a member of the proprotein convertases and involved in the regulation of intracellular cholesterol and fatty acid levels. Furthermore, it cleaves the glycoprotein precursor of all known arenaviruses pathogenic to humans and therefore represents an interesting target for antiviral therapy. So far, only a single article about potent, reversible S1P inhibitors with in vitro activity has been published and all following studies concerning the effect of S1P inhibition on various biological systems have been performed with compound PF-429242 that was described in said article. In this work, peptidic boronic acids based on the substrate sequence RRLL↓ and deborylated analogues were established as a new S1P inhibitor class and further derivatives of compound PF-429242 were synthesized. The synthesis of peptidic boronic acid inhibitors was optimized and extensive studies were performed regarding the establishment of boronic acid SPPS. The enzyme kinetic characterization revealed peptidic boronic acids as highly potent S1P inhibitors and strongly suggests that the boronic acid moiety covalently interacts with the active-site serine. Compound I34 bearing a boro-cyclopentylglycine in P1 position binds to S1P with an IC50 value of 73 nM and therefore fivefold stronger than the reference inhibitor PF-429242 (IC50 = 374 nM).
In case of PF-429242 derivatives, the reaction conditions for both reductive aminations required to synthesize such inhibitors were optimized. However, none of the new derivatives of the published compound PF-429242 exhibited a higher S1P affinity than the reference itself
Charakterisierung der hämagglutininspaltenden Transmembranserinproteasen Matriptase und TMPRSS2
Influenza is one of the commonest infectious diseases affecting millions of people every year including 290,000 – 650,000 heavy casualties. Influenza viruses undergo constant genetic changes and every 10 – 50 years new influenza virus strains emerge that potentially cause a severe pandemic. In this modern interconnected world, experts believe the next influenza pandemic will be a “devastating global health event with far-reaching consequences” [1]. Novel effective anti-influenza drugs are in need. One strategy of influenza research is to focus on host-specific proteases that are essential for virus activation and spread. Trypsin-like serine proteases are crucial for influenza activation by mediating the cleavage of the viral surface glycoprotein HA and hence promoting the fusion potential of the virus. Therefore, their inhibition provides a promising therapeutic approach. The present work focused on the characterization of two relevant HA cleaving type-II transmembrane serine proteases matriptase and TMPRSS2.
Chapter 3 and chapter 4 of this thesis engaged with the recombinant production of matriptase (chapter 3) in order to obtain a pure functional enzyme of high quality for a SAR study with novel monobasic (hence potentially bioavailable) matriptase inhibitors of the 3-amidinophenylalanine type (chapter 4). Adequate amounts of high-quality matriptase enzymes were isolated using a new expression system and in total 5 matriptase crystals were available at the end of this thesis for structural analysis. The matriptase inhibitor design in this thesis focused on matriptase-affine compounds with a fair selectivity profile against the blood coagulation enzymes thrombin and fXa. In total, 18 new monobasic and potentially bioavailable, as well as four new dibasic compounds of the 3-amidinophenylalanine types were tested. Based on the last published crystal structure of this inhibitor type in complex with matriptase from 2006 (PDB code 2GV6) docking was used as a structure-based virtual screening method for lead optimization of the compounds N-terminus. Selected compounds were suggested to interact with the carbonyl side chain of Gln175 of matriptase to achieve a higher affinity of matriptase compared to fXa.
The 4-tert-butylureido-piperidine could be identified as suitable C-terminus in combination with 3-fluoro-4-hydroxymethyl biphenylsulphonyl N-terminally in order to obtain excellent selectivity over thrombin. The binding mode of this compound (compound 55) was crystallographically determined in complex with matriptase as well as trypsin. Trypsin proved as a suitable alternative to matriptase for detailed binding mode analysis of the compounds N-terminus. However, different preferences were detected for the C-terminus.
Dibasic compounds showed higher matriptase affinity and selectivity in comparison with the monobasic analogues. However, the tested monobasic compounds were still decent matriptase inhibitors that are additionally suitable for cell culture and animal studies in their benzamidine prodrug forms, which are well established from related inhibitors of thrombin. In addition, selected monobasic as well as dibasic compounds demonstrated strong suppression of the replication of certain H9N2 influenza viruses in a matriptase-expressing MDCK II cell model. These matriptase inhibitors could be potential lead structures for the development of new drugs against H9 strains for influenza.
TMPRSS2 is widely discussed for its role in influenza activation. With a TMPRSS2 dependancy of HA-activation of certain subtypes, the characterization of this protease is an important prerequisite for being available as a target for influenza drug design. However, only little is known about the physiological function of TMPRSS2 and no experimental structure data are available at the moment to enable a structure-based drug development. Therefore, chapter 5 of this thesis focused on the characterization of TMPRSS2 in order to develop a strategy for the isolation of proteolytically active TMPRSS2 from cell culture. Even though, no functional TMPRSS2 could be recovered at the end of this work some new structural characteristics of TMPRSS2 were identified as crucial for functionality insight the cell. In general, TMPRSS2 without the cytosolic part, the transmembrane domain and the LDLRA domain is able to undergo autocatalytically activation if an artificial signal peptide was added N-terminal to enable entry into the endoplasmic reticulum. The presence of the cysteine-rich SRCR domain and the presence of the disulfide chain that connects the SPD and the stem region after activation cleavage have been identified as crucial for activity. N-terminal truncation of TMPRSS2 did not result in obvious dislocation within the cell: as the full-length positive control truncated TMPRSS2 was exclusively found in cell compartments surrounding the nucleus in immunofluorescence experiments. However, a reduced proteolytic cleavage activity towards H3-HA in co-expression experiments has been observed and might be a result of dislocation, since truncated TMPRSS2 is not bound to the biomembrane anymore. In addition, TMPRSS2 has been identified as a potential substrate of matriptase in vitro, which suggests possible participation in several zymogen cascades.Matriptase und TMPRSS2 sind humane Transmembran Serinproteasen, die in der Lage sind, verschiedene Subtypen des Influenza-A Oberflächenproteins Hemagglutinin proteolytisch zu spalten und somit die Vermehrung von Influenzaviren in der Wirtszelle ermöglichen. Die Charakterisierung dieser Wirtsproteasen ist eine wichtige Voraussetzung zur Entwicklung wirksamer und selektiver Inhbibitoren als potentielle Wirkstoffe zur Therapie von Influenzainfektionen. In der vorliegenden Arbeit wurde ein Expressionssystem zur Herstellung rekombinanter C122S mutierter Matriptase etabliert und das erhaltene Protein im Folgenden für die enzymkinetische Charakterisierung verschiedener Inhibitoren sowie zur Strukturanalyse eingesetzt. Des Weiteren wurden verschiedene verkürzte und/oder mutierte TMPRSS2 Formen hergestellt und auf ihre proteolytische Aktivität mit dem Ziel untersucht, eine isolierbare TMPRSS2 Variante für enzymkinetische Messungen bereit zu stellen.
In dem Wirtsorganismus E. coli wurden zwei verschiedene Matriptasevarianten produziert und miteinander enzymkinetisch verglichen. Die Serinproteasedomänen der Wildtyp-Matriptase und der mutierten C122S-Matriptase wurden jeweils mit 6 verschiedenen AMC-Substraten vermessen und zeigten dasselbe enzymkinetische Profil. Die etwas schlechteren Ausbeuten bei der Produktion der C122S-Matriptasevariante wurde durch die deutlich kostengünstigere Herstellung kompensiert. Im Folgenden wurden größere Mengen der C122S-Matriptase produziert und für Kristallisationationsexperimente eingesetzt. Durch den Einsatz der Hochdurchsatz Kristallisationsrobotik konnte ein geeigneter Kristall zur Sturkturanalyse gefunden werden. Mittels „Micoseed Matrix Screening“ konnten in einem zweiten Ansatz 4 weitere gut geformte Matriptase Proteinkristalle erhalten werden, indem zwei für die Röntgenstrukturanalyse qualitativ nicht geeignete und zerkleinerte Proteinkristalle dem neuen Kristallisationsansatz beigefügt wurden.
Im Fokus der Entwicklung neuer monobasischer 3-Amidinophenylalanin Derivate im Rahmen dieser Arbeit standen potente Matriptase Verbindungen die eine hohe Selektivität gegenüber den Serinproteasen der Blutgerinnungskaskade Faktor Xa und Thrombin aufweisen. Insgesamt wurden 18 monobasische und somit potentiell bioverfügbare, als auch 4 dibasische Verbindungen getestet. Auf Basis einer Kristallstruktur der Matriptase in Komplex mit einem Inhibitor des 3-Amidinophenylalanin-Typs (PDB code: 2GV6, Verbindung 20) wurde Docking
genutzt um mögliche Optimierungen des N-Terminus zu identifizieren. Es wurden Verbindungen mit einer 3-Hydroxymethylgruppe ausgesucht, da eine Interaktion mit dem Gln175 der Matriptase vorhergesagt wurde, die zu einer erhöhten Matriptase Affinität, aber nicht fXa Affinität, beitragen soll. Die monobasische Verbindung 55 mit einem 4-tert-Butylureido Piperidin am C- und 3-Fluor-4-Hydroxymethyl Biphenylsulphonyl am N-Terminus zeigte eine hervorragende Selektivität gegenüber Thrombin (1533-fach). Der kristallographische Bindemodus dieser Verbindung in Komplex mit Matriptase zeigte eine Interaktion der 4-Hydroxymethylgruppe mit dem Gln175, was zu einer moderaten Selektivität gegenüber fXa führte (12-fach). Der Bindemodus von Verbindung 55 (als auch Verbindung 56) wurde zudem in Komplex mit Trypsin bestimmt. Das gut verfügbare Trypsin erwies sich als geeignete Alternative zu Matriptase, wenn eine Interaktion des Inhibitors mit der S3/4 Bindetasche im Fokus steht. Der Inhibitor C-Terminus zeigte dagegen eine abweichende Orientierung in beiden Proteasen. Die Trypsin/Verbindung 56-Kristallstruktur zeigte außerdem, dass ein N-terminales 3-Hydroxymethyl theoretisch mit dem Gln175 in Matriptase interagieren würde. Verbindungen mit einem 3-Hydroxymethyl zeigten allerdings eine verminderte Matriptase Affinität (als auch Selektivität) in enzymkinetischen Messungen.
Aufgrund erfolgloser Isolierversuche aktiver TMPRSS2 aus E.coli Bakterien wurden verschiedene Strategien entwickelt um die anspruchsvolle Transmembran Serinprotease aus Säugerzellen zu isolieren. Dazu wurden zunächst verkürzte als auch mutierte TMPRSS2-Varianten eingesetzt um strukturelle Eigenschaften zu identifizieren, die essentiell für eine Proteaseaktivität sind. Die Hauptstrategie basierte auf dem Entfernen von mindestens der Transmembrandomäne um löslisches und somit leichter isolierbares Enzym zu erhalten. Obwohl im Rahmen dieser Thesis keine aktive TMPRSS2 isoliert wurde, konnten durch diese Versuche neue Erkenntnisse zur TMPRSS2 Funktionalität gewonnen werden. Es wurde gezeigt, dass der zytosolische Teil, die Transmembrandomäne als auch die LDLRA Domäne keine Voraussetzung zur autokatalytischen Aktivierung von TMPRSS2 sind. Die SRCR Domäne als auch die Disulfidbrücke, welche die Proteasendomäne mit der Stammregion nach der Aktivierungsspaltung verbindet, sind hingegen essentiell für TMPRSS2 Aktivität in der Zelle. Trotz Entfernung der in der Zellmembran verankernden Transmembrandomäne konnte in Immunofluoreszens-Versuchen keine offensichtliche Dislokalisation der verkürzten Varianten im Vergleich zum Wildtyp festgestellt werden. Des Weiteren wurde TMPRSS2 in vitro als ein Substrat der Matriptase identifiziert. Dies deutet auf die Involvierung von TMPRSS2 in mehreren Zymogenkaskaden hin
Synthesis and characterization of the haemagglutinin-cleaving proteases furin and HAT
In der vorliegenden Arbeit wurden Inhibitoren der hämagglutininspaltenden Proteasen Furin und HAT synthetisiert und charakterisiert. Während die Proproteinkonvertase Furin das Hämagglutinin (HA) hochpathogener Influenzaviren vom Subtyp H5 und H7 aktiviert, bewirkt die trypsinartige Serinprotease HAT die Spaltung des HA saisonaler Influenzaviren der Subtypen H1, H2 und H3. Die entwickelten Hemmstoffe dieser Proteasen können somit als Ausgangsstoffe für die Entwicklung neuer antiviraler Wirkstoffe zur Therapie der Influenza dienen.
Ein Bis-Amidinohydrazonderivat wurde in einem screening als Startpunkt für die Entwicklung neuer niedermolekularer Furininhibitoren identifiziert. In vier Syntheseserien wurden Furininhibitoren dieses Typs dargestellt. In der ersten Serie wurden kommerziell erhältliche Carbonylverbindungen als Ausgangsmaterial verwendet. Die wirksamste Verbindung dieser Serie mit einem Ki-Wert von 1,5 µM ist der vom Terephthalaldehyd abgeleitete Inhibitor. In zwei weiteren Serien wurden Derivate mit drei oder vier basischen Funktionen synthetisiert. Die Kopplung von 4 Guanidinobuttersäure an die Leitstruktur MI-0007 in der zweiten Serie und die Darstellung tetrabasischer Verbindungen unter Verwendung von in der dritten Serie führten zu Inhibitoren mit Hemmkonstanten von 0,5 - 0,6 µM. Ein aus der Literatur bekanntes, vom Salicylaldehydbenzylether abgeleitetes, monovalentes Amidinohydrazon mit einem Ki-Wert von 11,8 µM wurde als Leitstruktur einer vierten Serie von Furininhibitoren genutzt. Jedoch waren die Verbindungen dieser Serie weniger wirksam als die entsprechende aus der Literatur bekannte Ausgangsverbindung.
Mit Ausnahme einer Verbindung, weisen alle im Rahmen dieser Arbeit synthetisierten Amidinohydrazonderivate eine hohe Selektivität gegenüber den untersuchten trypsinartigen Serinproteasen Thrombin, Faktor Xa und Plasmin auf (Ki-Werte > 30 µM).
Für die trypsinartige Serinprotease HAT wurden bisher nur wenige Inhibitoren in der Literatur beschrieben. Deshalb begannen die Arbeiten zur Entwicklung der HAT-Inhibitoren mit einer Testung von im Arbeitskreis verfügbaren Verbindungen. Neben substratanalogen Inhibitoren mit einem C terminalen 4-Amidinobenzylamid konnten Verbindungen mit einem zentralen 3-Amidinophenylalanin des 3-TAPAP-Typs als Startpunkte für die Entwicklung von HAT-Hemmstoffen identifiziert werden.
In sämtliche substratanaloge Inhibitoren wurde in P4- ein Benzylsulfonylrest und in P1-Position ein 4-Amidinobenzylamid eingebaut. In einer ersten Serie mit einem Prolin als P2-Rest wurden in P3 Position verschiedene D-Aminosäurereste akzeptiert, besonders jedoch das basische D-Arginin oder D-homo-Arginin. Akzeptiert werden auch hydrophobe Aminosäurederivate, wie ein tert.-butyl-geschütztes D-Asp oder D-Glu (Ki < 40 nM). Aufgrund der Präferenz für basische und hydrophobe Aminosäurereste in P3-Position, wurden in einer zweiten Serie die Seitenketten von D-Asp und D-Glu mit verschiedenen zyklischen Aminen modifiziert. Die wirksamste Verbindung wurde durch Kopplung von 1-(2-Pyrimidyl)piperazin an die Seitenkette von D-Glu erhalten und hemmt HAT mit einem Ki-Wert von 17 nM.
Da substratanaloge Inhibitoren mit einem P2-Prolin häufig auch andere trypsinartige Serinproteasen hemmen, wurde der Einfluss anderer Aminosäuren in P2-Position in Kombination mit D-Arginin als P3-Rest untersucht. Die stärkste Hemmwirkung innerhalb dieser Serie mit Ki Werten < 30 nM wurde für kleine hydrophobe Aminosäurereste, wie 2-Aminobuttersäure (Abu), Norvalin (Nva), Valin oder Alanin gefunden. Ein Vorteil der Derivate mit Abu oder Nva in P2-Position ist deren verbesserte Selektivität gegenüber den Proteasen Thrombin, Faktor Xa und Plasmin. Die Verbindung Bzls-D-Arg-Nva-4-Amidinobenzylamid ist mit einer Hemmkonstante von 15 nM der erste Inhibitor, der die anderen getesteten Proteasen schwächer hemmt und somit die höchste Selektivität für HAT aufweist.
Ausgewählte substratanaloge Inhibitoren wurden in Zellkulturversuchen eingesetzt, um die Hemmung der Vermehrung von Influenzaviren zu untersuchen. Eine besonders starke Hemmwirkung auf Influenzaviren der Subtypen H1 und H3 wurde durch Inhibitoren mit Norvalin oder 2-Aminobuttersäure in P2-Position bestimmt. Die Inhibitoren weisen darüber hinaus keine signifikante Zytotoxizität auf, so dass die reduzierte Virusausbreitung auf die HAT-Hemmung zurückzuführen ist.
In der vorliegenden Arbeit wurden zahlreiche substratanaloge Inhibitoren identifiziert, die HAT mit Ki-Werten < 30 nM hemmen. Im Gegensatz zu den bereits etablierten antiviralen Wirkstoffen hemmen alle im Rahmen dieser Arbeit entwickelten Furin- und HAT-Inhibitoren Zielstrukturen des Wirts. Deshalb sollten auch bei längerer Verwendung dieser Verbindungen keine Resistenzen auftreten.Inhibitors of the haemagglutinin cleaving proteases furin and HAT were synthesized and characterized in this thesis. While the proproteinconvertase furin is activating the haemagglutinin (HA) of highly pathogenic influenza viruses of the subtype H5 and H7, the trypsin-like serine protease HAT activates the HA of seasonal influenzaviruses of the subtypes H1, H2 and H3. Therefore, the described inhibitors may be useful tools for the development of new antiviral drugs for the treatment of the flu.
In a first screening a bis-amidinohydrazone-derivate was identified as a lead structure for new low molecular weight furin inhibitors, which was modified in four subsequent synthesis cycles. Commercially available carbonyl compounds were used in a first series. The most potent derivative was derived from terephthalaldehyde and possesses an inhibitory constant of 1,5 µM. Inhibitors with three or four basic residues were obtained from second and third series. The coupling of 4-guanidionbutyric acid to the lead structure in the second series and the dimerization of the initial screening hit with chloroaceticacidchloride led to compounds with a inhibitory potency of 0,5 to 0,6 µM. A known salicylaldehyde benzylether-derived compound was used as a lead structure of the fourth series. However, all inhibitors from this attempt were less potent than the lead structure.
With one exception all amidinohydrazone derived inhibitors from this thesis feature a high selectivity against the related trypsin-like serineproteases thrombin, factor Xa and plasmin (Ki > 30 µM).
Until now only few inhibitors for the trypsin-like serinportease HAT were described. Thus, the first attempt in this field was the screening of available compounds for HAT inhibition. Besides, substrate analogue compounds with a 4-amidinobenyzlamide moiety on the C-terminal part and inhibitors with a 3-amidinophenylalanine of the TAPAP-type were identified as possible starting points for the development of HAT inhibitors.
For all substrate analogue inhibitors a P4-benzylsulfonyl residue and a P1-Amidinobenzylamidine residue were used. In a first series with a proline in P2-position, several d-configurated aminoacids were well-accepted in P3-position. Especially d-Arg and d-homo-Arg were favored as a P3-residue. Nevertheless, hydrophobic derivatives, like a tert.-butyl protected d-Asp or d-Glu, were also well-suited for incorporation. Due to the preference of HAT for basic or hydrophobic amino acids in P3, cyclized amines were coupled to the side chains of d-Asp or d-Glu in a second series. Most potent compound from this attempt was obtained via coupling of 1-(2-pyrimidyl)piperazin to the free side chain of d-Glu (Ki = 17 nM).
Substrate analogue inhibitors with a P2-Proline are common inhibitors of several trypsine-like proteases. Hence, the influence of other amino acid residues replacing the proline was examined keeping d-Arg in P3-position. Small hydrophobic residues like 2-aminobutyric acid (Abu), norvaline (Nva), Ala or Val are a good replacement for proline and the compounds inhibit HAT with Ki-values below 30 nM. An advantage of these inhibitors is their enhanced selectivity against the other tested proteases thrombin, factor Xa and plasmin.
Selected compounds were also tested in cell based assays to examine their ability to suppress viral replication. In these experiments the inhibitors with an Abu or Nva residue in P2-position effectively inhibit the replication of influenza viruses of the subtypes H1 and H3. It should be noted that this effect is not associated with any toxic effects to the cells.
Several substrate analogue compounds which inhibit HAT with inhibition constants < 30 nM are presented in this thesis. Contrary to until now known antiviral drugs, the synthesized compounds are targeting host cell proteases. Therefore, there will be no rising of resistances against these drugs
Entwicklung neuer Hemmstoffe der Typ II transmembranären Serinprotease Matriptase.
Several new inhibitors were prepared and tested as potential matriptase inhibitors. The most potent compounds were found among the tertiary amides of arylsulfonylated-3-amidinophenylalanines. The incorporation of suitable bis-substituted biphenylsulfonyl groups in combination with appropriate C-terminal urea structures provided the final strong potent monobasic matriptase inhibitors with acceptable selectivity against related trypsin-like serine proteases. Such compounds are suitable candidates for their further evaluation in cell culture experiments to elucidate the potential role of matriptase in pathophysiological processesNeben dem Screening vorhandener Verbindungen wurden zahlreiche neue Proteasehemmstoffe synthetisiert und als Matriptaseinhibitoren untersucht. Die wirksamsten Verbindungen sind tertiäre Amide arylsulfonylierter 3-Amidinophenyl-alanin-Derivate. Der Einbau geeigneter disubstituierter Biphenylsulfonyl-Reste in Kombination mit substituierten C-terminalen Harnstoffen an der Piperididgruppe führte zu hochaffinen Matriptaseinhibitoren mit einem relativ günstigen Selektivitätsprofil gegenüber anderen trypsinartigen Serinproteasen. Diese Inhibitoren sind geeignete Kandidaten für weiterführende Experimente in Zellkultur oder zur Anwendung in Tiermodellen, um pathophysiologische Prozesse zu hemmen, die durch Matriptase vermittelt werden
Development, Synthesis and Characterization of novel Furin Inhibitors
In der vorliegenden Arbeit wurden neuartige, effektive, peptidomimetische Inhibitoren der Proproteinkonvertase Furin synthetisiert und charakterisiert. Ausgehend von der Struktur des irreversiblen Referenzinhibitors Decanoyl-RVKR-chlormethylketon wurden Verbindungen entwickelt, in die anstelle des P1 Arg-chlormethylketon-Segmentes decarboxylierte Arginin- und Lysinmimetika sowie weitere zyklische basische Reste eingebaut wurden. Der Decanoyl-Rest wurde zunächst durch einen Phenylacetyl- oder Acetylrest sowie das Lysin in der P2-Position durch Arginin ersetzt. Die auf die Synthese folgende Bestimmung der Inhibitorkonstanten ergab, dass als wirksamster Inhibitor Phenylacetyl-RVR-4-(amidino)benzylamid (17) Furin mit einem Ki-Wert von 0,81 nM hemmt.
Basierend auf dieser Leitstruktur wurden zur weiteren Optimierung verschiedene Inhibitorserien synthetisiert, bei denen nacheinander in allen Positionen der Struktur (P2-P5) verschiedene natürliche und unnatürliche Aminosäurereste oder Aminosäurederivate eingebaut wurden. So konnte gezeigt werden, dass der Austausch des P4-Arginins durch ein Lysin oder die Eliminierung der basischen Gruppe in der P2-Position zu einer drastischen Reduktion der inhibitorischen Effektivität führt. Der systematische Einbau aller proteinogenen Aminosäuren in die P3-Position zeigte, dass fast alle Derivate dieser Serie Furin effektiv hemmen, dabei waren neben Valin auch Isoleucin, Phenylalanin oder Tyrosin besonders geeignet. Der Einbau saurer Aminosäuren an dieser Stelle führte jedoch zu weniger wirksamen Inhibitoren, wahrscheinlich bedingt durch elektrostatische Abstoßung aufgrund des stark negativ geladenen, aktiven Zentrums des Furins.
In der P5-Position wurden verschiedene Fettsäurereste eingebaut. Die Bestimmung der Inhibitorkonstanten ergab, dass sie bis zu einer Länge von 10 Kohlenstoffatomen annähernd im Bereich der Leitstruktur (17) liegt. Die weitere Verlängerung der Kohlenstoffkette führte allerdings zu einem sukzessiven Abfall der inhibitorischen Wirkung. Die Synthese von Inhibitoren mit basischen P5-Resten führte zu sehr potenten Wirkstoffen, von denen sich 3-(Guanidinomethyl)phenylacetyl-RVR-4-(amidino)benzylamid (38) mit einem Ki-Wert von 5,6 pM als der effektivste Inhibitor für Furin herausstellte.
Untersuchungen zur Stabilität einiger Verbindungen mittels HPLC zeigten, dass unter den gewählten Bedingungen kein nennenswerter Zerfall oder Abbau nachzuweisen war. Als Beispiel dient Inhibitor 17, der in wässriger Lösung über 10 Tage bei Raumtemperatur stabil ist.
In Zusammenarbeit mit der AG Garten (Institut für Virologie der Universität Marburg) wurden ausgesuchte Inhibitoren auf ihre Zytotoxizität in Zellkultur untersucht. Die wenig zytotoxisch wirkenden Verbindungen wurden anschließend in virologischen Untersuchungen zur Hemmung der Vermehrung und Ausbreitung hochpathogener Viren der klassischen Geflügelpest (H7N1) eingesetzt. Dabei wurde nachgewiesen, dass unter anderen die Inhibitoren 17 und 4-(Guanidinomethyl)phenylacetyl-RVR-4-(amidino)benzylamid (40) die Virusvermehrung effektiv hemmen. Die quantitative Bestimmung infektiöser Viren in Zellkulturen über einen Zeitraum von mehreren Tagen zeigte, dass die Zugabe des Inhibitors 40 zu einer signifikanten Verlangsamung der Virusausbreitung führt. So betrug der Virustiter nach 45 h nur 1,7 % der Kontrollkultur ohne Inhibitor.
Zur Optimierung der Reinigung von rekombinant hergestelltem Furin für Kristallisationsexperimente wurden basierend auf der Leitstruktur N-terminal biotinylierte Inhibitoren für die Affinitätschromatographie synthetisiert. Diese Verbindungen wurden in Zusammenarbeit mit der AG Than (Institut für Altersforschung, Fritz Lipmann-Institut, Jena) für die Reinigung von Maus-Furin eingesetzt, indem kommerziell erhältliche Streptavidinsäulen mit den biotinylierten Inhibitoren beladen wurden. Mit diesen Affinitätssäulen gelang eine effektive Reinigung des Furins im Vergleich zu den bisher verwendeten, klassischen Reinigungsmethoden.
Die im Rahmen dieser Arbeit synthetisierten, niedermolekularen Verbindungen sind die wirksamsten, reversibel bindenden Furinhemmstoffe, die bisher weltweit entwickelt wurden. Sie sind daher für weiterführende Untersuchungen in Zellkulturen oder gegebenenfalls sogar in Tiermodellen geeignet.In the present work, novel effective peptidomimetic Inhibitors for the proprotein convertase furin were synthesized and characterized. Several compounds were designed based on the structure of the irreversible inhibitor Decanoyl-RVKR-chloromethyl ketone, which contain in place of the P1 Arg-chloromethyl ketone moiety decarboxylated arginin- or lysin derivatives as well as some other basic groups. Initially, the decanoyl residue was replaced with an acetyl or phenylacetyl group and the lysin in P2-position was replaced with and arginin. Following the synthesis, the determination of the inhibitor constants showed that the compound Phenylacetyl-RVR-4-(amidino)benzylamide (17) was a highly effective Inhibitor (0.81 nM).
For optimization, different series of inhibitors were synthesized based on this lead structure 17. Therefore, natural and unnatural amino acids and amino acid derivatives were inserted successively in all other positions (P2-P5). It could be shown that the exchange of the P4-arginin as well as the elimination of the basic group in P2 position led to a drastic reduction of the inhibitory efficiency. The systematic incorporation of all proteinogenic amino acids in P3 position led to compounds which could inhibit furin comparable to inhibitor 17. Besides valin, isoleucin, phenylalanin or tyrosin are also well suited in this position. In contrast, the determination of the inhibitory constants of compounds with acidic amino acids in P3-position showed a reduced inhibitory potency, probably due to electrostatic repulsion from the negatively charged active site of furin.
Different fatty acids were introduced in P5 position in course of the following synthesis. The determination of the inhibitory constants showed that up to a length of 10 carbon atoms the inhibitory activity was comparable to the lead structure. The further prolongation of the carbon chain led to a gradual loss of the inhibitory efficiency. The introduction of basic residues in P5 position yielded highly potent Inhibitors of which 3-(Guanidinomethyl)phenylacetyl-RVR-4-(amidino)benzylamide (38) inhibits Furin most efficient with a Ki-value of 5.6 pM.
In further course of the work, the stability of some synthesized compounds was analyzed via HPLC. Under the applied conditions no degradation or decay of the chosen compounds could be observed. For example, inhibitor 17 was stable in aqueous solution over 10 days at room temperature.
In collaboration with the Garten laboratory (Institute of Virology, University of Marburg) some compounds were tested in cell culture for cytotoxicity. The inhibitors with negligible cytotoxic effects were further used in virological assays for the determination of the inhibition of virus spread and proliferation of highly pathogenic avian influenza (H7N1). Among others, inhibitor 17 and 4-(Guanidinomethyl)phenylacetyl-RVR-4-(amidino)benzylamide (40) were able to reduce efficiently the virus proliferation. The quantitative determination of infectious viruses in cell cultures over a period of several days showed a significant reduction in virus spread. After 45 h, the virus titer reached a value of only 1.7 % in comparison to the control without inhibitor.
To purify recombinant synthesized furin for utilization in crystallization experiments, some N-terminally biotinylated inhibitors based on the lead structure were synthesized for the application in affinity chromatography. These derivatives were used in collaboration with the workgroup of Mr. Than (Institute for Age Research, Fritz Lipmann-Institut, Jena) for the purification of mouse furin. Therefore, commercially available streptavidin columns were loaded with the biotinylated compounds. In comparison to the classical purification methods, these affinity columns were well suited for an effective purification of furin.
The small molecular inhibitors synthesized during this work are the most efficient reversible furin inhibitors worldwide until today. Therefore, they are well suited for further tests in cell cultures or even in animal models
Development of enhanced furin inhibitors with reduced toxicity as potential broad spectrum antiviral drugs
At the beginning of the twentieth century, the focus of peptide research was largely on human signalling hormones. A breakthrough in the field of peptide therapeutics was the first medicinal application of insulin isolated from animal pancreas, which revolutionized the treatment of type 1 diabetes. Since then, more than 80 peptide drugs have been used for a variety of diseases, including diabetes, cancer, osteoporosis, multiple sclerosis, HIV infection, and chronic pain. The production of synthetic therapeutic peptides has become possible with the development of solid-phase peptide synthesis by Merrifield in the year 1963. This milestone has provided access to pure and sufficient amounts of peptides enabling countless research projects to determine the functional and structural bioactive properties of peptides.
In the context of drug discovery, this thesis focused on the development of synthetic inhibitors of the Ca2+-dependent type-I transmembrane protein furin, which belongs to the proprotein convertase (PC) family of serine endoproteases. It is ubiquitously expressed in vertebrates and invertebrates and activates a large number of proprotein substrates in the secretory pathway, including prohormones, proenzymes, and proforms of receptors or extracellular matrix proteins. Furin also plays a crucial role in tumorigenesis, neurodegenerative disorders, and diabetes or arteriosclerosis as well as in many bacterial and viral diseases. It cleaves numerous precursors of bacterial toxins such as Pseudomonas exotoxin A, Shiga-, and diphtheria toxin. Some enveloped viruses possess surface proteins that contribute to the fusion between viral and host cell membranes and have to be activated by furin or related PCs. Examples of such glycoproteins include those from highly pathogenic avian Influenza viruses, Measles, Ebola, and Flaviviruses such as Dengue-, West Nile-, and Zika-virus. Recent studies revealed that furin is implicated in the activation of the spike (S) protein of the new SARS-CoV-2. Given its implications in a variety of biological processes and diseases, furin emerged as a target of paramount importance in drug discovery. To suppress the propagation cycle of furin-dependent viruses, novel peptidomimetic furin inhibitors were developed in this work. Based on the preferred multibasic cleavage motif of furin (–Arg–X–Lys/Arg–Arg↓–) and on previous work from our group, novel inhibitors of furin were synthesized by a combination of solid-phase peptide synthesis and solution synthesis.
In a first manuscript, several series of new macrocyclic furin inhibitors have been designed and synthesized. The development of cyclic peptides is a widely used strategy to improve the stability and bioavailability of peptide drugs. The cyclization was performed between different positions of these substrate-analogue inhibitors and by incorporating different linker segments. The inhibitory potency of these compounds was determined in enzyme kinetic assays with soluble human furin. In cooperation with other groups the binding mode of selected cyclic inhibitors in complex with furin was determined and their antiviral potency was tested in cells infected with Respiratory syncytial virus (RSV).
A second paper describes the optimization of a non-cyclic inhibitor starting from a previously designed linear inhibitor MI-1148 that inhibits furin with a Ki value of 5 pM but suffers from a considerably toxicity in mice and rats. Additional studies with structurally related compounds suggested that the severe toxicity of these compounds is caused by the multibasic character of this inhibitor type and not by the inhibition of the host protease furin. By replacing the arginine residues (pKa of side chain 13.5) with the noncanonical amino acid canavanine (pKa 7.0), a new series of potent furin inhibitors was prepared. The best compound MI-1851 inhibits furin with a Ki value of 10.1 pM. Furthermore, the toxicity of this compound was significantly reduced. In cell culture studies, the novel canavanine-containing compounds also showed a significant antiviral activity against furin-dependent viruses, such as RSV, WNV, and Dengue-2 virus, and most importantly, the current SARS-CoV-2 virus.
In a third co-authored paper, this inhibitor MI-1851 was also used to demonstrate an antiviral effect on the new SARS-CoV-2. Moreover, through a series of newly synthesized FRET-substrates derived from the sequence of the potential S1/S2 cleavage site of the SARS-CoV-2 spike protein, it was possible to prove the SARS-CoV-2 S protein activation at this sequence by furin.
In a fourth paper, the synthesis and characterization of a small series of 11 new fluorescence substrates are described, which were tested with five basic PCs including furin, PC1, PC2, PC5A, and PC7. For four of the tested PCs, improved substrates have been identified, which are more efficiently cleaved than the known reference substrates. The improved furin substrates enabled kinetic measurements at significantly reduced enzyme concentrations, thereby avoiding tight-binding conditions. At these reduced furin concentrations, a slow-binding behaviour was observed for the first time with inhibitor MI-1148, which enabled the determination of its individual kon and koff rate constants. The new inhibitors and substrates may help further elucidate the role of furin and related PCs in physiology and disease.Zu Beginn des 20. Jahrhunderts konzentrierte sich die Peptidforschung weitestgehend auf menschliche Signalhormone. Der erste Durchbruch gelang mit der medizinischen Anwendung des Insulins aus tierischen Bauchspeicheldrüsen, das die Behandlung von Typ-1-Diabetes revolutionierte. Seitdem werden mehr als 80 therapeutisch wirksame Peptide bei unterschiedlichsten Krankheitsbildern wie Diabetes, Krebs, Multiple Sklerose, HIV-Infektion und chronische Schmerzen eingesetzt. Die Synthese therapeutischer Peptide wurde mit der Entwicklung der Festphasenpeptidsynthese durch Merrifield im Jahr 1963 möglich. Dieser Meilenstein ermöglichte den Zugang zu reinen und ausreichenden Mengen an Peptiden und ermöglichte unzählige Forschungsprojekte zur Bestimmung ihrer Funktion und Struktur.
Der Fokus dieser Arbeit lag auf der Entwicklung synthetischer Inhibitoren der calciumabhängigen subtilisinartigen Serinprotease Furin, die zur Familie der Proproteinkonvertasen (PCs) gehört. Es wird ubiquitär in Vertebraten und Invertebraten exprimiert und aktiviert zahlreiche Proproteinsubstrate wie Prohormone, Proenzyme und Proformen von Rezeptoren oder extrazellulärer Matrixproteine. Furin ist auch an der Tumorigenese, neurodegenerativen Erkrankungen, Diabetes oder Arteriosklerose beteiligt, sowie an zahlreichen bakteriellen und viralen Erkrankungen. Beispielsweise spaltet es Vorstufen bakterieller Toxine wie Pseudomonas Exotoxin A, Shiga-Toxin und Diphtherietoxin. Einige Viren besitzen Oberflächenproteine, die zur Fusion viraler und Wirtszellmembranen beitragen und durch Furin oder verwandte PCs aktiviert werden. Beispiele sind die Glykoproteine hochpathogener Vogelgrippeviren, von Masern-, Ebola- oder Flaviviren wie Dengue-, West-Nil- und Zika-Viren. Interessanterweise ist Furin auch an der Aktivierung des Spike-Oberflächenproteins des neuen SARS-CoV-2 beteiligt. Im Rahmen dieser Arbeit wurden neuartige Furininhibitoren entwickelt, synthetisiert, enzymkinetisch und strukturell charakterisiert, sowie ihre antivirale Wirksamkeit bestimmt.
Im ersten Manuskript wurden mehrere Serien makrozyklischer Furininhibitoren synthetisiert. Die Entwicklung zyklischer Peptide ist eine weit verbreitete Strategie zur Verbesserung der Stabilität und Bioverfügbarkeit therapeutischer Peptide. Die Zyklisierung erfolgte zwischen verschiedenen Resten an unterschiedlichen Positionen dieser substratanalogen Inhibitoren mittels verschiedener Linkersegmente. Die inhibitorische Wirksamkeit dieser Verbindungen wurde durch enzymkinetische Untersuchungen mit löslichem humanen Furin bestimmt. In Zusammenarbeit mit Kooperationspartnern wurde der Bindungsmodus ausgewählter zyklischer Inhibitoren im Komplex mit Furin und ihre antivirale Wirksamkeit in mit dem Respiratorischen Synzytial-Virus (RSV) infizierten Zellen ermittelt.
Die zweite Arbeit beschreibt die Optimierung des bereits bekannten linearen Inhibitors MI-1148, der Furin mit einem Ki-Wert von 5 pM hemmt, sich jedoch an Mäusen und Ratten als relativ toxisch erwiesen hat. Zusätzliche Studien mit strukturell verwandten Verbindungen legten nahe, dass die Toxizität dieser Verbindungen durch den stark multibasischen Charakter dieses Verbindungstyps und nicht durch die eigentliche Hemmung der Wirtsprotease Furin verursacht wird. Durch Austausch der Argininreste (pKa der Seitenkette 13.5) durch die nicht-kanonische Aminosäure Canavanin (pKa 7.0) wurde eine neue Serie wirksamer Furinhemmstoffe hergestellt. Die effektivste Verbindung MI-1851 hemmt Furin mit einem Ki-Wert von 10.1 pM. Wie erwartet, hat sich diese Verbindung in Mäusen als deutlich weniger toxisch erwiesen. In Zellkulturstudien zeigten die neuartigen canavaninhaltigen Verbindungen eine signifikante antivirale Aktivität gegen furinabhängige Viren, wie RSV, West-Nil- und Dengue-2-Virus.
In einer dritten Arbeit aus dem Arbeitskreis von Prof. Friebertshäuser wurde für den Inhibitor MI-1851 auch eine antivirale Wirkung auf das neue SARS-CoV-2 nachgewiesen. Darüber hinaus konnte durch eine Reihe neu synthetisierter FRET-Substrate, die von der Sequenz der potenziellen S1/S2-Spaltstelle im Spike-Protein verschiedener Coronaviren abgeleitet wurden, die Aktivierung der SARS-CoV-2 Sequenz durch Furin nachgewiesen werden.
In einer vierten Arbeit wird die Synthese und Charakterisierung einer kleinen Serie von 11 neuen Fluoreszenzsubstraten beschrieben, die mit den fünf basischen PCs Furin, PC1, PC2, PC5A und PC7 vermessen wurden. Für vier der getesteten PCs (außer PC5A) wurden verbesserte Substrate identifiziert, die effizienter als die bekannten Referenzsubstrate gespalten werden. Die verbesserten Furinsubstrate gestatteten Messungen bei deutlich reduzierten Enzymkonzentrationen, bei denen erstmals ein sogenanntes slow-binding Verhalten für den Inhibitor MI-1148 festgestellt wurde. Unter diesen Bedingungen konnten die individuellen Assoziations- und Dissoziations-Geschwindigkeitskonstanten kon und koff-bestimmt werden. Die neuen Inhibitoren und Substrate können zukünftig helfen, die Rolle des Furins und verwandter PCs bei physiologischen Prozessen und Erkrankungen weiter aufzuklären
