    Lo spirito dello haiku

    No full text

N-acetyl-l-cysteine fosters inactivation and transfer to endolysosomes of c-Src

The non-receptor-protein tyrosine kinase c-Src is overexpressed and activated in a large number of human cancers, in which it is associated with tumor development and progression. Canonical regulation takes place by means of an alternative phosphorylation of tyrosine residues Tyr419 for activation and Tyr530 for inactivation. An independent redox regulation mechanism, involving cysteine residues, has also been proposed, in which oxidation activates the enzyme. Here we present a kinetic analysis of the effect of N-acetyl-l-cysteine (NAC) on c-Src, demonstrating that reduction reverts the oxidation-driven activation. In cancer cells, we show that NAC treatment produces an increase in specifically labeled reduced thiols of c-Src cysteines, thus confirming a redox transition. In addition to a decrease in Tyr419 phosphorylation, this leads to a massive shift of c-Src from plasma membranes where its active form is located to endolysosomal compartments. With the objective of deciphering the complex issue of c-Src regulation and of devising new strategies to revert its activation in cancers, redox regulation thus emerges as a promising area for study

CpG oligodeoxinucleotides promote phospholipase D despendent phagolysosome maturation and intracellular mycobacterial killing in M. tubercolosis infected type II alveolar epithelial cells

CpG oligodeoxynucleotides have been previously shown to enhance antimycobacterial response in human monocytes/macrophages. The present study reports evidences showing the capability of CpG oligodeoxynucleotides to induce (i) host phospholipase D (PLD) activation, (ii) PLD dependent reactive oxygen intermediate production, (iii) PLD dependent phagolysosome maturation and (iv) PLD dependent intracellular mycobacterial killing in type II alveolar epithelial cells. These are the first evidences showing that alveolar epithelial cells may represent efficient effecter cells during primary innate antimycobacterial immune response

Topological properties of semigroup primes of a commutative ring

A semigroup prime of a commutative ring R is a prime ideal of the semigroup (R, ·). One of the purposes of this paper is to study, from a topological point of view, the space S(R) of prime semigroups of R. We show that, under a natural topology introduced by B. Olberding in 2010, S(R) is a spectral space (after Hochster), spectral extension of Spec(R) , and that the assignment R⦠S(R) induces a contravariant functor. We then relateâin the case R is an integral domainâthe topology on S(R) with the Zariski topology on the set of overrings of R. Furthermore, we investigate the relationship between S(R) and the space X(R) consisting of all nonempty inverse-closed subspaces of Spec(R) , which has been introduced and studied in Finocchiaro et al. (submitted). In this context, we show that S(R) is a spectral retract of X(R) and we characterize when S(R) is canonically homeomorphic to X(R) , both in general and when Spec(R) is a Noetherian space. In particular, we obtain that, when R is a Bézout domain, S(R) is canonically homeomorphic both to X(R) and to the space Overr(R) of the overrings of R (endowed with the Zariski topology). Finally, we compare the space X(R) with the space S(R(T)) of semigroup primes of the Nagata ring R(T), providing a canonical spectral embedding X(R) âa S(R(T)) which makes X(R) a spectral retract of S(R(T))

Physical Properties of the Zona Pellucida

The zona pellucida (ZP) is the extracellular coat that surrounds the mammalian oocyte. It forms a spherical shell of remarkably uniform thickness (5-10 mm in eutherian mammals) composed of three glycosylated proteins (ZP1, ZP2 and ZP3). Penetration of this shell by spermatozoa plays a crucial role in mammalian fertilization and any inability of spermatozoa to penetrate the ZP inevitably leads to infertility. The purpose of this work is to shed light to the three-dimensional structure of the ZP, its construction and its properties as a polymer. By means of the Atomic Force Microscopy (force-distance curves) we have determined for the first time several ZP structural properties (elasticity, plasticity, adhesion, etc.) during the ovulatory, periovulatory and fertilized phase. Moreover information about single polymers interactions has been obtained by means detachment experiments (pull-off curves)

    No full text
