    Dimer-dimer stacking interactions are important for nucleic acid binding by the archaeal chromatin protein Alba

    Archaea use a variety of small basic proteins to package their DNA. One of the most widespread and highly conserved is the Alba (Sso10b) protein. Alba interacts with both DNA and RNA in vitro, and we show in the present study that it binds more tightly to dsDNA (double-stranded DNA) than to either ssDNA (single-stranded DNA) or RNA. The Alba protein is dimeric in solution, and forms distinct ordered complexes with DNA that have been visualized by electron microscopy studies; these studies suggest that, on binding dsDNA, the protein forms extended helical protein fibres. An end-to-end association of consecutive Alba dimers is suggested by the presence of a dimer-dimer interface in crystal structures of Alba from several species, and by the strong conservation of the interface residues, centred on Are and Phe(60). In the present study we map perturbation of the polypeptide backbone of Alba upon binding to DNA and RNA by NMR, and demonstrate the central role of Phe(60) in forming the dimer dimer interface. Site-directed spin labelling and pulsed ESR are used to confirm that an end-to-end, dimer dimer interaction forms in the presence of dsDNA.Peer reviewe

    Estudo tecnológico, fitoquímico e biológico de Lippia alba (Miller) N. E. Brown Ex Britt. & Wils. (Falsa-Melissa) Verbenaceae

Programa de Pós-Graduação em Farmácia.O desenvolvimento de soluções extrativas visando a elaboração de fitoterápico exige um estudo multidisciplinar caracterizado pela pesquisa no campo da botânica, etnofarmacologia, fitoquímica, tecnologia e biologia. A espécie Lippia alba (Mill.) N. E. Brown ex Britt. & Wils. da família Verbenaceae, popularmente conhecida como erva-cidreira, falsa melissa e outros, é muito utilizada em todo o Brasil, principalmente como antiespasmódica, sedativa e calmante. Num extensivo levantamento bibliográfico sobre o gênero Lippia, buscou-se informações que forneçam subsídios para o planejamento dos estudos tecnológicos, fitoquímicos e biológicos, analisando-se 95 citações bibliográficas referentes a 58 binômios científicos. Os dados sobre nomenclatura botânica (e sinonímias científicas) e sobre os constituíntes químicos de espécies do gênero foram sistematizados, verificando-se a predominância de trabalhos sobre os constituintes voláteis das plantas, além daqueles sobre flavonóides que aparecem com relativa freqüência em espécies do gênero. Informações sobre iridóides, verbascosídeos, fenilpropanóides, quinonas e outras substâncias menos freqüentes também foram encontradas. Constatou-se confusão quanto à denominação científica das espécies de Lippia devido a falta de uniformidade no uso de binômios válidos, gerando um grande número de sinonímias, que podem dificultar estudos posteriores. O estudo tecnológico de soluções extrativas (SEs) hidroalcoólicas a 70, 80 e 90% obtidas por maceração (SEM) e percolação (SEP) incluiu a caracterização da matéria-prima e soluções extrativas, com o objetivo de contribuir para a otimização do processo extrativo. Os resultados confirmam as observações de trabalhos anteriores, onde o teor de resíduo seco (RS) e o teor de flavonóides totais (FT) apresentaram uma relação inversa. Os FTs para as SEs a 90% são os mais altos nas soluções analisadas. Estas soluções foram testadas frente à ação anticonvulsivante, nos modelos da convulsão induzida por pentilenotetrazol (PTZ) e eletrochoque máximo (ECM). Na análise fitoquímica de L. alba, obteve-se frações purificadas a partir do fracionamento de uma SEM80%, com o auxílio do monitoramento farmacológico. O grupo de substâncias que pode servir de marcador químico (SBN4-C) é composto de uma mistura de 4 substâncias, separadas por CLAE, necessitando ainda serem caracterizadas e identificadas. Resultados promissores foram obtidos, na análise da atividade antimicrobiana sobre S. aureus, com a SEM80% e suas frações acetato de etila e n-butanol. SEM40% e SEM80% foram testadas quanto à atividade antinociceptiva, sendo que a de 40% demonstrou melhor atividade. As preparações testadas quanto a ação anticonvulsivante foram a SEM80%, sua fração n-butanólica, subfrações originadas desta (precipitado e sobrenadante), e subfrações separadas do sobrenadante através de uma coluna de Sephadex LH-20 (SBN4-C e SBN4-D). Também foram testadas a SEM90% e SEP90%, resultantes do desenvolvimento tecnológico. A dose de 0,05 mg/Kg para a fração SBN4-C apresentou o melhor resultado no modelo de convulsão induzida por PTZ. Neste mesmo modelo, a SEM90% e a SEP90% mostraram os melhores resultados a partir da dose de 200 mg/Kg, para a maioria dos parâmetros avaliados. O modelo do PTZ foi considerado o método mais adequado para a avaliação da atividade anticonvulsivante de preparações extrativas a partir de folhas de L. alba e suas frações

    Female barn owls (Tyto alba) advertise good genes

    The good genes hypothesis of sexual selection postulates that ornamentation signals superior genetic quality to potential mates. Support for this hypothesis comes from studies on male ornamentation only, while it remains to be shown that female ornamentation may signal genetic quality as well. Female barn owls (Tyto alba) display more black spots on their plumage than males. The expression of this plumage trait has a genetic basis and it has been suggested that males prefer to mate with females displaying more black spots. Given the role of parasites in the evolution of sexually selected traits and of the immune system in parasite resistance, we hypothesize that the extent of female plumage `spottiness’ reflects immunological defence. We assessed the genetic variation in specific antibody production against a non-pathogenic antigen among cross-fostered nestlings and studied its covariation with the plumage spottiness of genetic parents. The magnitude of the antibody response was positively correlated with the plumage spottiness of the genetic mother but not of the genetic father. Our study thereby provides the first experimental support, to our knowledge, for the hypothesis that female ornamentation signals genetic quality.

    The Author/Translator Interactional Process. A Case Study

    See Naples and Kill (1988) is a lively and colourful novel by the con-temporary English writer, Gregory Dowling, translated into Italian in 2015. Following the tradition of translation studies (Venuti 2000, Bass-nett 2002, Cronin 2006), this paper analyses the rewriting process of literary translation, considering in particular the fruitful but sometimes tense and even conflictual relationship between writer and translator. The translation of the novel See Naples and Kill was an ongoing rewriting process entailing a constant dialogue between the writer and the translator. Therefore, the study aims at answering two main ques-tions: what happens if the rewriting process of translation is constant-ly questioned by the author? What happens if the author has a good mastery of the target language and s/he is her/himself a translator? By exploring the relationship between translation and re-creation, the research focuses on the differences and similarities between the primary creation (source text) and the secondary creation (target text), and aims to verify in which way the dialogic encounter of two different personalities and cultures does not make them merge but, by retaining their own uniqueness, leads eventually to their mutually en-riching each other. A comparative analysis of the source text and the different drafts of the translated version accompanied by the author’s comments will shed light on the tense author-translator relationship in the specific case under investigation and how both actors handle this tension in order to create a new work resulting from the (dis)agreement of the two parties

    L’objectiu d’aquesta recerca és estudiar l’àcid glucònic com a indicador de l’estat sanitari dels raïms de vinificació. La quantificació d’aquest àcid permet saber si raïms aparentment sans estan afectats per la Botrytis cinerea i en quin grau. És un indicador útil tant per a les  cooperatives com per als productors, ja que permet controlar la qualitat del raïm en recepció, establir-ne el preu i determinar quin s’utilitzarà per elaborar vi i quin es destinarà a altres finalitats. Per últim, aquesta recerca és una aproximació científica al món de la vinya i el vi, activitat econòmica amb més de 2.000 anys de tradició a la meva comarca. L’Alt Empordà, situat al nord de la Costa Brava, amb una gran riquesa cultural i paisatgística, és una zona fortament marcada pels contrasts entre mar i muntanya. A aquestes peculiaritats del terreny, s’hi suma un altre tret distintiu: la presència regular de la Tramuntana. Aquest vent del nord té efectes molt beneficiosos en l’estat sanitari de la vinya, ja que crea un clima més sec i redueix la proliferació de bacteris.This research aims to study gluconic acid as an indicator of the health conditions of grape used for winemaking. Determining the level of gluconic acid helps to find out whether similar grapes are affected or not by Botrytis cinerea and to what extent. It is a key indicator for both cooperatives and producers to monitor the quality of grapes upon reception at their facilities, allowing to fix the right price and to sort out those grapes addressed to wine-making while diverting the remaining ones for other industrial purposes. The research intends to be a scientific approach both to the world of vine and wine, an outstanding economic activity in our region: the Alt Empordà, located in the North of the Costa Brava and enjoying a rich cultural heritage as well as a remarkable landscape. The area’s main climate feature is the Tramuntana, a strong northern wind blowing regularly and keeping humidity at its lowest. It becomes a natural way of preserving vineyards free from bacterial proliferation

Com que és un autor encara força desconegut, aquest treball pretén acostar la seva figura i obra a un públic actual. En primer lloc, s'ha recollit i sintetitzat la bibliografia que parla de l'autor per fer-ne un estat de la qüestió que ha de ser útil tant per introduir el personatge com per senyalar les llacunes biogràfiques i contextuals que encara caldria omplir. En segon lloc, s'han seleccionat les cinc esparses amoroses del poeta com a tast representatiu de la seva poètica i se n'han elaborat diferents productes editorials: una edició diplomàtica, una de crítico-interpretativa i, finalment, una d'actualitzada. D'aquesta manera, es pretén abraçar un públic molt divers que podria estar interessat per la poesia medieval.[eng] Joan Basset is one of the most productive and original poets of the Vega-Aguiló compilation. As he still is an unknown author, the aim of this work is to approach his profile and lyrical work to an actual reader. In the first place, it is assembled a status of the issue based on the literary works about the poet and his poetical context. This compilation pretends to be an introduction into the world of the poet and, what is more, a base for a future more deep study about his figure. In the second place, five love esparses have been selected as a representation of the poetic frame of the poet. Different editorial products have been elaborated of the same five poems: a diplomatic edition, an interpretative and critic edition and, finally, a contemporary version. The diversity of the offer wants to be an attraction for the different possible public of Middle Age literature
