ARF-GAP-mediated interaction between the ER-Golgi v-SNAREs and the COPI coat
No full textIn eukaryotic cells, secretion is achieved by vesicular transport. Fusion of such vesicles with the correct target compartment relies on SNARE proteins on both vesicle (v-SNARE) and the target membranes (t-SNARE). At present it is not clear how v-SNAREs are incorporated into transport vesicles. Here, we show that binding of ADP-ribosylation factor (ARF)-GTPase-activating protein (GAP) to ER-Golgi v-SNAREs is an essential step for recruitment of Arf1p and coatomer, proteins that together form the COPI coat. ARF-GAP acts catalytically to recruit COPI components. Inclusion of v-SNAREs into COPI vesicles could be mediated by direct interaction with the coat. The mechanisms by which v-SNAREs interact with COPI and COPII coat proteins seem to be different and may play a key role in determining specificity in vesicle budding

No full textRecent advances in membrane contact site (MCS) biology have revealed key roles for MCSs in inter-organellar exchange, the importance of which is becoming increasingly apparent. Roles for MCSs in many essential physiological processes including lipid transfer, calcium exchange, receptor tyrosine kinase signalling, lipid droplet formation, autophagosome formation, organelle dynamics and neurite outgrowth have been reported. The ER forms an extensive and dynamic network of MCSs with a diverse range of functionally distinct organelles. MCSs between the ER and endocytic pathway are particularly abundant, suggesting important physiological roles. Here, our current knowledge of the formation and function of ER contact sites with endocytic organelles from studies in mammalian systems is reviewed. Their relatively poorly defined molecular composition and recently identified functions are discussed. In addition, likely, but yet to be established, roles for these contacts in lipid transfer and calcium signalling are considered. Denne oppgaven er en lesning av Dag Solstads roman T. Singer (1999), hvor jeg særlig legger vekt på temaene skam, identitet og intime relasjoner, og forsøker å tolke dem i lys av moderniteten. I utviklingen av analysen benytter jeg meg av Anthony Giddens teorier om individets identitet i (sen)modernitetens æra, samt skamteori slik den er utformet av Helen Merrell Lynd og Finn Skårderud. Jeg forholder meg til Giddens overordende oppfatning om at selvet er gjennomsyret av modernitetens abstrakte systemer, og kontinuerlig formes av og selv former disse systemene i en bevegelse som Giddens kaller modernitetens refleksivitet. Slik er menneskets irrganger og dannelsen av identiteten avhengig av å betraktes på bakgrunn av samfunnets grunnleggende strukturer for å kunne begripes.
Foruten å konsentrere meg om identitetens refleksive vilkår i (sen)moderniteten, fokuserer jeg på skammens betingelser i samme epoke, og finner grunnlag for dette valget i T. Singers inngående skildring av denne emosjonen. Jeg kobler videre skam og identitet til hverandre, og undersøker hva romanens hovedperson og hans skamfølelse kan fortelle leseren om Singers opplevelse av sin egen identitet.
I T. Singer knyttes skamfullheten til isolasjon og tilbaketrekning fra den sosiale sfære, og jeg forfølger derfor også Singers regressive bevegelse fra begynnelse til slutt, og analyserer på nært hold hans tre eneste relasjoner og den langsomme avviklingen av dem. Oppgaven munner ut i en diskusjon rundt hovedpersonens dødsbevissthet.
Det overordnede målet med lesningen er å utforske forholdet mellom romanen T. Singer og samfunnet den er skapt i, for slik å komme dypere inn i den litterære teksten, som på alle måter er mitt hovedanliggende. Av den grunn leser jeg T. Singer som et frittstående verk, uten å trekke inn Dag Solstads berømmede forfatterskap forøvrig. Det er min vri og min mulighet til å lese T. Singer med et åpent blikk. Forhåpentligvis vil en slik lesning gi mening

The Emp24 complex recruits a specific cargo molecule into endoplasmic reticulum-derived vesicles
No full textMembers of the yeast p24 family, including Emp24p and Erv25p, form a heteromeric complex required for the efficient transport of selected proteins from the endoplasmic reticulum (ER) to the Golgi apparatus. The specific functions and sites of action of this complex are unknown. We show that Emp24p is directly required for efficient packaging of a lumenal cargo protein, Gas1p, into ER-derived vesicles. Emp24p and Erv25p can be directly cross-linked to Gas1p in ER-derived vesicles. Gap1p, which was not affected by emp24 mutation, was not cross-linked. These results suggest that the Emp24 complex acts as a cargo receptor in vesicle biogenesis from the ER
Embedding research (ER) led by nurses, midwives and allied health professionals (NMAHPs): the NMAHP-ER model.
No full textPrevious embedded researcher models have focused predominantly on an individual being a temporary team member and embedded for a project-limited short-term placement. To develop an innovative research capacity building model to address the challenges of developing, embedding and sustaining, research led by Nurses, Midwives, and Allied Health Professionals (NMAHPs) in complex clinical environments. This healthcare and academic research partnership model offers an opportunity to support the 'how' of enabling NMAHP research capacity building from within the researchers' clinical area of expertise. Collaboration between three healthcare and academic organisations and the iterative process of cocreation, development and refinement took place over 6 months during 2021. The collaboration relied on virtual meetings, emails, telephone calls and document review. A codesigned NMAHP embedded research (ER) model is ready for trialling with the individual being an existing clinician working collaboratively within the healthcare setting and with academia to develop the skills to become the ER. This model supports NMAHP-led research activity in clinical organisations in a visible and manageable way. As a shared, long-term vision, the model will contribute to research capacity and capability of the wider healthcare workforce. It will lead, facilitate and support research in and across clinical organisations in collaboration with higher education institutions. [Abstract copyright: © Author(s) (or their employer(s)) 2022. Long-term Patterns in Australia’s Terms of Trade
No full textWe examine two important aspects of Australia’s terms of trade using 135 years of annual data up to 2003/04. Since Australia predominantly exports commodities and imports manufactures, the Prebisch-Singer hypothesis suggests that there should be a negative trend in the terms of trade. But the trend is no more than –0.1 per cent per annum, less than the trend decline in world commodity prices relative to manufactured goods prices. The weaker trend appears to be the result of Australia exporting, and importantly diversifying toward, commodities with faster price growth. Extending the sample using projections for the terms of trade for the two years to 2005/06 based on commodity price movements to date, the apparent downward trend disappears. Indeed, based on these projections, the terms of trade will have increased by around 50 per cent over the period 1987–2006, unwinding the decline over the preceding 30 years. We also investigate the volatility of the terms of trade and demonstrate that it was significantly higher between 1923 and 1952. This is attributable to substantially higher volatility in the export prices of a few key commodity exports. Volatility declined after 1952 due to smaller shocks to the prices of these goods. The diversification in Australia’s export base since then means that the terms of trade are less susceptible to shocks to prices of individual commodity exports.terms of trade; commodity prices; Prebisch-Singer
Überlegungen zur hybriden Form des vermeintlich ersten Tonfilms THE JAZZ SINGER (USA 1927, Alan Crosland)
No full textAlan Croslands Film THE JAZZ SINGER wird in zahlreichen wissenschaftlichen Abhandlungen zum Übergang vom Stumm- zum Tonfilm als der Film beschrieben, der dem Tonfilm endgültig zum Durchbruch verhalf (vgl. Dibbets 1998, 197; Nowell-Smith 1998, 193; Henzel 2006, 47). Doch obwohl THE JAZZ SINGER maßgeblich dazu beigetragen hat, das neue Medium Tonfilm am Markt zu etablieren, handelt es sich bei diesem Film nicht um den ersten Tonfilm, wie gelegentlich behauptet wird (vgl. Ferrari 2004, 70), sondern lediglich um einen „Stummfilm mit einigen vertonten Einschüben“ (Dibbets 1998, 197). THE JAZZ SINGER ist ein so genannter part-talkie, ein Film also, der nur zum Teil vertont wurde. An ausgewählten Stellen enthält er „lippensynchrone Lieder und Dialog“ (Dibbets 1998, 197). Das expressive Spiel der Figuren und der Einsatz von Zwischentiteln hingegen erinnern an den Stummfilm. Doch nicht nur die Tatsache, dass THE JAZZ SINGER kein hundertprozentiger Tonfilm ist, steht laut Christoph Henzel einer Definition als erstem Tonfilm entgegen. Darüber hinaus ist er auch nicht der erste Film, der technisch in der Lage war, Bild und Ton zu synchronisieren. Vielmehr positioniert er sich als ein Ereignis unter vielen, als ein Glied innerhalb einer ganzen Reihe technischer Entwicklungen vom Stumm- zum Tonfilm (vgl. Henzel 2006, 48/49)
