Abstract LB-164: The association between p53, MDM2, and MDM2 isoform C protein expression and survival in a multiethnic population of breast cancer patients
Abstract
Breast cancer is the most common cancer among women in the United States. Despite improvements in survival rates, the disease remains the second leading cause of cancer death for women in the U.S. Breast cancer incidence and survival dramatically vary by race and ethnicity. These disparities may be attributed to racial/ethnic differences in the factors influencing breast cancer development and progression. These include factors such as personal (eg. body size and reproductive history), clinical (eg. stage, grade, histology), treatment course, and genetic susceptibility.
Inactivation of the tumor suppressing, p53 pathway is a common event in breast cancer. Depending on the molecular subtype, p53 mutation is observed in 12-84% of breast cancers and is correlated with poor clinical outcome. In addition to p53 mutation events, p53 pathway inactivation in tumors can also result from an increase in the levels of MDM2, an ubiquitin ligase that functions as a negative regulator of p53. The mdm2 gene produces at least 40 differentially spliced transcripts. The MDM2 isoforms A, B, and C have been observed to be frequently overexpressed in many tumor types and correlated with poor prognosis, however, the underlying mechanism is poorly understood.
To examine the protein expression profiles for p53, MDM2, and MDM2 isoform C (MDM2-C) in breast cancer from a multiethnic population, we conducted immunohistochemical (IHC) analyses utilizing commercially available antibodies to p53 and MDM2, and a newly developed monoclonal antibody specific for MDM2-C (Okoro DR et al. 2013. PLoS ONE) on a total of 791 invasive breast tumors included in a clinically-annotated population-based tissue microarray (TMA). Based on Cox proportional hazards regression analysis and adjusting for age, stage, and estrogen and progesterone receptor status, we did not observe significant associations between expression of p53, MDM2, or MDM2-C and mortality across all cases. However when we examined associations within major racial/ethnic groups included in our TMA (Caucasian, Japanese, and Native Hawaiian), we observed a significant positive association of p53 expression (HR=1.63, 95% CI: 1.02-2.59) and a marginally significant association of MDM2-C (HR=1.69, 95% CI: 0.94-3.01) with all-cause mortality for Japanese women. We also observed associations between MDM2-C expression and all-cause and breast cancer-specific mortality for Caucasian women (HR=0.60, 95% CI: 0.33-1.08 and HR=0.28, 95% CI: 0.12-0.67, respectively). However, in contrast to Japanese women, expression of MDM2-C was associated with lower risk of mortality for Caucasian women. No significant associations for Native Hawaiian women were observed. Recent reports indicate that the functional effects of MDM2 isoforms (A, B, and C) are dependent on the presence of wild-type or mutant p53, and our preliminary results suggest that the association between breast cancer outcome and MDM2-C expression is context dependent on p53 status.

Morpho-functional changes of pituitary-adrenal system in the long-term course of experimental diabetes mellitus

Summary
Different neurohumoral systems are involved in the development and advance of diabetes mellitus as well as its complications, including hypothalamic-pituitary-adrenal system. The increase of functional activity of the latter contributes to the worsening of the disease course and leads to the development of diabetic macroangiopathies (retinopathy, nephropathy). That’s why our goal was to discover common regularities of morpho-functional changes in pituitary-adrenal system in the long-term course of experimental diabetes mellitus (EDM).
The investigation involved 20 mature male Wistar rats, which were equally divided into two groups: control group and experimental one. Diabetes mellitus was induced by single dose intraperitoneal administration of streptozotocin (diluted in 0.1 М of citrate buffer with the pH of 4.5) in a dose of 6 mg per 100 grams of body mass. The research work involved the use of histological (hematoxylin-eosin staining, Heidenhain's Azan staining technique) and electron-microscopic investigation methods. We have discovered that on the 56th day of EDM hyperglycemia leads to the increase of functional activity of pituitary-adrenal system that is morphologically manifested in: corticotrophs – as increase of their number, hypertrophy and hyperplasia of structural components of Golgi complex and granular endoplasmic reticulum with the increase in volume ratio of secretory granules, including marginated ones, causing significant increase of ACTH in blood; adrenal cortex – as marked thickening of zona fasciculata and zona reticularis, increase of endocrinocyte area in these zones and lipid droplets in them as well as the increase of blood cortisol level.
On the increase of experimental period (day 70) constant hyperfunctioning of pituitary-adrenal system results in its exhaustion. These changes in pituitary are morphologically characterized by ballooning degeneration of corticotrophs, decrease of volume density of secretory granules within them, including the marginated ones, decrease of ACTH in blood; while in adrenal cortex these changes are marked by hydropic degeneration of endocrinocytes of all zones, partial necrosis of endocrinocytes in zona fasciculata, decrease of blood cortisol level.

Abstract
The purpose of the paper is to reveal the peculiarities of the formation of value professional orientation of college students in specialty “Accountant”. The study was conducted to confirm the authors’ concept intended to facilitate a more effective assimilation of the system of human and professional values by accountant students from the experimental group. Based on this concept, the students from the experimental group were supposed to demonstrate better learning results than students in the regular group. For this purpose, the authors’ courses "Philosophical and Historical Context for the Development of Axiology" and a special course "Ethics Code for Professional Accountants" were used to distinguish the training program. By applying the authors’ methodology, special pedagogical conditions and models, we succeeded in raising the level of value professional orientation of future accountants and in achieving better learning results. The scientific approach, which is based on the comparison of techniques for the development of students' value sphere to enhance the educational attainment of college students, had not previously been applied.

Summary. Recently, there has been a significant increase in interest in research on hypertension (HT), primarily due to its high prevalence. The interest in studying this problem is also exacerbated by the often insufficient effectiveness of existing treatments. The effect of concomitant pathologies on HT, in particular non-alcoholic fatty liver disease (NAFLD), remains poorly understood.
The aim of the study – to evaluate the changes in the serum albumin binding function (SABF) and its relationship with the biochemical parameters of the blood when HT and HT combined with NAFLD and to suggest ways of medical correction of the detected disorders.
Material and methods. 76 individuals with stage 2 HT with degree 2–3 arterial hypertension were examined. They were divided into two groups. Group 1 included 28 patients with HT without concomitant diseases who received basic hypertension therapy, and group 2 included patients with concomitant NAFLD. The latter in turn was divided into two subgroups: 2a – 27 patients who in addition to basic HT therapy received additional Antral hepatoprotector 200 mg three times a day for 2 months, and 2b – 21 patients who received only basic HT therapy. All of them underwent a standard clinical examination, as well as SABF, protein fractions, and liver function indicators. The comparison group consisted of 25 healthy individuals, comparable in age and sex.
Results and Discussion. Patients in group 1 showed moderate changes in the functional state of the liver, but they did not exceed the norm, patients in group 2 – a significant decrease in SABF, as well as protein metabolism (decrease in total protein, albumin, albumin-globulin ratio and increase globulins) and liver function (increased activity of aspartate aminotransferase, alanine aminotransferase, gamma-glutamyltranspeptidase, thymol levels, alkaline phosphatase and total bilirubin). After treatment, the majority of patients in subgroup 2 had a statistically significant increase in SABF and a quantitative improvement in protein fractions and functional state of the liver. In subgroup 2-b, where hepatoprotective treatment was not performed, significant changes in most indicators did not occur. The results may be related to the positive effect of the drug on the liver, which leads to improved functional status of hepatocytes and their protein-synthesizing ability. Conclusions. Changes of the functional state of the liver are observed when HT without concomitant pathology. HT with NAFLD is accompanied by a significant decrease in SABF, changes in protein metabolism and the functional state of the liver. Prescribing Antral to such patients helps to increase SABF, normalize protein metabolism and improve the functional state of the liver.

We examined the transmission of human papillomavirus (HPV) in 25 heterosexual, monogamous couples (25 men, 25 women), followed up over an average of 7.5 months. A total of 53 heterosexual transmission events were observed among 16 couples (14 male-to-female and 39 female-to male). Sexual transmission involved 13 different oncogenic and nononcogenic HPV types; 8% were vaccine-covered types transmitted between partners. The overall rate of HPV transmission from the penis to the cervix was 4.9/100 person-months, which was substantially lower than that from the cervix to the penis (17.4/100 person-months). Transmission between the hands and genitals, as well as apparent self-inoculation events (primarily in men), were also observed. Couples who transmitted HPV were more sexually active and used condoms less frequently. These results have implications for HPV prevention and control strategies, including the targeting of prophylactic vaccines

Abstract
Topicality. Sports and technical skills, as an element of a holistic process of sports training, occupy one of the priority places in the overall structure of sports skills. The study, formation, improvement and development of the most rational models of sport technique are closely related to the use of modern video recording systems.
The task of the study is to analyse the knee strike technique, applied by the athletes of different qualifications with the help of "Qualisys" optoelectronic system.
Results of the research. Analysis of highly skilled athletes’ competitive activities shows that a knee strike is mainly used in close combat. Much less often such a strike can be put in a jump from a long or average distance. In our work, we have analysed the technique of right knee strike, performed from the starting position characteristic of close combat.
Conclusions. When studying the biokinematic structure of the knee strike technique, used by athletes of different qualification, differences in the dynamics of changes in the speed of the lower extremity joints on athlete’ impact side of were found, which indicates higher coordination of movements, performed by highly skilled athletes. Thus, regarding highly qualified athletes the peaks of the maximum speed of their ankle and knee joints coincide in time, contrary to the skilled athletes. The speed of the skilled athletes’ ankle and hip joints at certain times is higher than those of highly skilled. However, the maximum value of speed of the highly skilled athletes’ strike link, as a rule, is higher on average by 0.5 m ∙ s-1. The general patterns include the reduction of the resulting knee speed for athletes of different qualification at the time of strike performance in reference to the maximum values that precede the strike interaction.

Safety and Mobility Impacts of Winter Weather – Phase I, August 2011
Highway agencies spend millions of dollars to ensure safe and efficient winter travel. However, the effectiveness of winter weather maintenance practices on safety and mobility are somewhat difficult to quantify.
Phase I of this project investigated opportunities for improving traffic safety on state-maintained roads in Iowa during winter weather conditions.
The primary objective was to develop several preliminary means for the Iowa Department of Transportation (DOT) to identify locations of possible interest systematically with respect to winter weather-related safety performance based on crash history.
Specifically, metrics were developed to assist in identifying possible habitual, winter weather-related crash sites on state-maintained rural highways in Iowa. In addition, the current state of practice, for both domestic and international highway agency practices, regarding integration of traffic safety- and mobility-related data in winter maintenance activities and performance measures were investigated. This investigation also included previous research efforts.

PATHOPHYSIOLOGICAL ANALYSIS OF SYSTEMIC AND RENAL DISREGULATION OF PATHOLOGICAL PROCESSES IN THE DAMAGE OF PROXIMAL TUBULE IN THE Nephrolithiasis

Abstract
The pathogenesis of urolithiasis in the modern view lies disregulating the pathological process of primary surplus mobilization reactions of protection, which leads to their further transformation in the reaction of damage that is the underlying basis for the formation of vicious circles. 299 patients with nephrolithiasis were examined, 10 patients were in the control group. Found that systemic and renal dysregulation pathological processes of stone the size of stones of 0.6 – 1.0 cm, top, middle cup the upper third of the ureter led to the formation of large and small “vicious circles”. Renal and extrarenal dysregulation pathological processes contributed to the increase in damage to the proximal tubule, since the primary damage to the said nephron. Department is not able to mobilize protective adaptive reactions aimed at increasing the excretion of acids and protein reabsorption, since they require additional energy costs. The use of the drug blemaren in nephrolithiasis reveals protective properties to reduce the manifestations of systemic and renal dysregulation pathological processes and promotes the rupture of formed large and small “vicious circles" with a decrease in proteinuria, the level of tumor necrosis factor-α, improving the transport of sodium ions, fibrinolytic activity of urine and it is advisable to develop guidelines for further use in the clinic of the drug blemaren as a promising method of treatment of urolithiasis.
