A Rediscovery of the First-Generation Female Architects in Taiwan: Xiu Zelan and Wang Chiu-Hwa

YB-1 expression promotes epithelial-to-mesenchymal transition in prostate cancer that is inhibited by a small molecule fisetin
Epithelial-to-mesenchymal transition (EMT) plays an important role in prostate cancer (PCa) metastasis. The transcription/translation regulatory Y-box binding protein-1 (YB-1) is known to be associated with cancer metastasis. We observed that YB-1 expression increased with tumor grade and showed an inverse relationship with E-cadherin in a human PCa tissue array. Forced YB-1 expression induced a mesenchymal morphology that was associated with down regulation of epithelial markers. Silencing of YB-1 reversed mesenchymal features and decreased cell proliferation, migration and invasion in PCa cells. YB-1 is activated directly via Akt mediated phosphorylation at Ser102within the cold shock domain (CSD). We next identified fisetin as an inhibitor of YB-1 activation. Computational docking and molecular dynamics suggested that fisetin binds on the residues from β1 - β4 strands of CSD, hindering Akt’s interaction with YB-1. Calculated free binding energy ranged from -11.9845 to -9.6273 kcal/mol. Plasmon Surface Resonance studies showed that fisetin binds to YB-1 with an affinity of approximately 35 μM, with both slow association and dissociation. Fisetin also inhibited EGF induced YB-1 phosphorylation and markers of EMT both in vitro andin vivo. Collectively our data suggest that YB-1 induces EMT in PCa and identify fisetin as an inhibitor of its activation
The Theory Of The Ideal King In “Chun Chiu Fan LU”(春秋繁露)
[[abstract]]The substance of this paper is the “Theory of the King/Kingdom in Chun Chiu Fan LU(春秋繁露)”. First, I think the theory of an “ideal king” could be the problematic idea in “Chun Chiu Fan LU”. The background of the politics and the society in pre-Han dynasty, many intellects are always concerned with the morality of king and all kinds of problems in the kingdom. So, they had a great deal of discussions about king and kingdom. Chun Chiu Fan Lu continues with the political tradition; the articles in this book are written in the same way which combines the philosophical theory and king’s practice. For this reason, we can determine the problematic idea in Chun Chiu Fan LU is “ideal king”.
Then, I need to build up the theory of ideal king in Chun Chiu Fan LU. According to the articles in this writings, there are three horizons about the ideal king theory. The first is the theory of cosmology and ontology. In this horizon, we can understand the theory combines the almighty lord possessing flawless morality with the lawof air which includes Yin Yan (陰陽) and Five Elements(五行). The ideal king needs to learn the flawless morality of lord. The second is the theory about human-nature. The ideal king needs to develop his moral character through the Five Classics. The last horizon is the theory about ideal king’s moral practice. The ideal king should build up a peace and wealthy land so that the almighty lord will bless the king and kingdom. Finally, the kingdom will become a paradise where has no war and no famine.
