    Chemopreventive effect of modified zeng-sheng-ping on oral squamous cell carcinoma by regulating tumor associated macrophages through targeting tnf alpha induced protein 6

    Abstract Background Oral squamous cell carcinoma (OSCC) is the most common malignancy of the head and neck. Zeng-Sheng-Ping, composed of Sophora tonkinensis Gagnep., Bistorta officinalis Delarbre, Sonchus arvensis L., Prunella vulgaris L., Dioscorea bulbifera L., and Dictamnus dasycarpus Turcz., was regarded as an anti-cancer drug with significant clinical efficacy, but was discontinued due to liver toxicity. Our research group developed a modified Zeng-Sheng-Ping (ZSP-M) based on original Zeng-Sheng-Ping that exhibited high efficiency and low toxicity in preliminary investigations, although its pharmacodynamic mechanism is still unclear. Here, we aimed to elucidate the pharmacodynamic material basis of ZSP-M and investigate its chemopreventive effect on OSCC by modulating tumor associated macrophages (TAMs). Methods Components of ZSP-M were characterized using ultra-performance liquid chromatography-mass spectrometry. Chemopreventive effect induced by ZSP-M against experimental oral cancer was investigated using the 4-nitroquinoline N-oxide precancerous lesion mouse model. RNA sequencing analysis was used to gain a global transcriptional view of the effect of ZSP-M treatment. A cell co-culture model was used to study the targeted effect of ZSP-M on TAMs and the biological properties of OSCC cells and to detect changes in TAM phenotypes. The binding of ZSP-M active compounds to TNF alpha induced protein 6 (TNFAIP6) protein was analyzed by molecular docking and dynamic simulation. Results Forty main components of ZSP-M were identified, the most abundant of which were flavonoids. ZSP-M inhibited the degree of epithelial dysplasia in precancerous lesions by inhibiting the expression of the TNFAIP6 and CD163 proteins in the precancerous lesions of the tongue. ZSP-M inhibited proliferation, colony formation, migration and invasion of SCC7 cells by targeting TAMs. ZSP-M reduced the expression of CD163+ cells, inhibited the expression of TNFAIP6 protein, Arg1 mRNA and Il10 mRNA in TAMs, and reduced IL-10 cytokine release in the co-culture environment. This effect was maintained after the addition of recombinant TNFAIP6 protein. Computer simulations showed that trifolirhizin and maackiain are well-connected to TNFAIP6. Conclusions ZSP-M counteracts the immunosuppressive action of TAMs by specific targeting of TNFAIP6, thereby exerting chemopreventive activity of OSCC

    Generation and manipulation of “smart” droplets

    We report the generation and manipulation of electrorheological (ER) droplets that exhibit the giant ER effect. The experiments were carried out on specially designed microfluidic chips, in which the ER droplets were generated by using the microfluidic flow-focusing approach. Both the size and formation rate of these droplets can be controlled through digitally applied electrical signals. The principle of droplet manipulation is based on the electrical responsiveness of ER droplets and hence the denotation of “smart” when the electrical signals can be triggered by sensing/control devices. Due to the unique characteristics of the GER effect, the smart droplets can deform and even stop the microfluidic channel flow under an applied electric field. The pressure difference induced by the smart droplets inside the micro-channel is controllable by varying the field strengths, droplet sizes and particle concentrations in the GER suspension. By trapping and timed release of smart droplets in different micro-branch channels, we demonstrate that the smart droplets generated upstream cannot only be stored or displayed in the desired downstream channel(s) and thereby offer the potential of micro-droplet display, but also be useful in counting, flow directing and sorting the desired number of passive droplets sandwiched between two smart droplets. Such capabilities of smart droplets will enable the programmable control of discrete processes in bio-analysis, chemical reactions, digital microfluidics, and digital droplet display

    Manipulations of microfluidic droplets using electrorheological carrier fluid

    Electrorheological (ER) fluids are a type of ‘‘smart’’ colloid capable of reversible viscosity variations, or even solidification, in response to an applied electric field. The response time can be as short as a few milliseconds. By using the ER fluid as the carrier fluid in microfluidic chips, we report the generation and manipulation of microdroplets and bubbles via integrated, digitally controlled micro-electrodes equipped with a feedback system. By utilizing the strong electric response of the ER fluid, the flow rate can be easily controlled digitally, thereby making tunable the size of the droplets generated and their separations. In particular, ordering change in a chain of droplets is demonstrated. The maneuverability presented in this paper may have potential applications in a variety of lab chips for chemical reactions, bioassays, as well as microfluidic logic computation

    Hybrid approach to high-frequency microfluidic mixing

    We report the experimental verification of the predicted chaotic mixing characteristics for a polydimethylsioxane microfluidic chip, based on the mechanism of multistage cross-channel flows. While chaotic mixing can be achieved within short passage distances, there is an optimal side channel flow pulsation frequency beyond which the mixing becomes ineffective. Based on the physical understanding of a Poincaré section analysis, we propose the installation of passive flow baffles in the main microfluidic channel to facilitate high-frequency mixing. The combined hybrid approach enables chaotic mixing at enhanced frequency and reduced passage distance in two-dimensional flows.<br/

    在本篇論文中,我們在SO(4)群6維的均項性(homogenous)空間,建立一個均項性但各向異性(anisotropic)的度規(metric),使得這個6維空間分開成3個大的空間維度,以及另外3個極小的額外維度。再利用建立的度規計算在 空間中的曲率,並推導在6+1維度下的愛因斯坦方程式,討論額外維度收縮的過程。   接著我們讓此模型做演化,並分別與古典宇宙學中宇宙演化的各個階段:暴漲(Inflation)、輻射主控時期(Radiation dominated era)、物質主控時期(Matter dominated era)分別做對照。並且計算在此模型中一些重要的數值。In this thesis, we use SO(4) group as a 6 dimensional homogenous space, we can build up a homogenous but anisotropic metric such that the 6 dimensional space become 3 large ordinary dimension and 3 compact extra-dimension. We will calculate the space curvature on the metric. Finally, we will obtain an Einstein’s equation for such a space describing the evolution of space compactification and topology. Then we will evolve this model with every phase of universe: inflation, radiation dominated era and matter dominated era. Fit this model with cosmology. Finally we will give some estimation and evaluate important quantities in this model

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
