    Circadian clocks responsible for daily time keeping in a wide range of organisms synchronize to daily temperature cycles via pathways that remain poorly understood. To address this problem from the perspective of the molecular oscillator, we monitored temperature-dependent resetting of four of its core components in the fruitfly Drosophila melanogaster: the transcripts and proteins for the clock genes period (per) and timeless (tim). The molecular circadian cycle in adult heads exhibited parallel responses to temperature-mediated resetting at the levels of per transcript, tim transcript and TIM protein. Early phase adjustment specific to per transcript rhythms was explained by clock-independent temperature-driven transcription of per. The cold-induced expression of Drosophila per contrasts with the previously reported heat-induced regulation of mammalian Period 2. An altered and more readily re-entrainable temperature-synchronized circadian oscillator that featured temperature-driven per transcript rhythms and phase-shifted TIM and PER protein rhythms was found for flies of the &#39;Tim 4&#39; genotype, which lacked daily tim transcript oscillations but maintained post-transcriptional temperature entrainment of tim expression. The accelerated molecular and behavioural temperature entrainment observed for Tim 4 flies indicates that clock-controlled tim expression constrains the rate of temperature cycle-mediated circadian resetting.</span

    A new promoter element associated with daily time keeping in <em>Drosophila</em>

    Circadian clocks are autonomous daily timekeeping mechanisms that allow organisms to adapt to environmental rhythms as well as temporally organize biological functions. Clock-controlled timekeeping involves extensive regulation of rhythmic gene expression. To date, relatively few clock-associated promoter elements have been identified and characterized. In an unbiased search of core clock gene promoters from 12 species of Drosophila, we discovered a 29-bp consensus sequence that we designated as the Clock-Associated Transcriptional Activation Cassette or ‘CATAC’. To experimentally address the spatiotemporal expression information associated with this element, we generated constructs with four separate native CATAC elements upstream of a basal promoter driving expression of either the yeast Gal4 or firefly luciferase reporter genes. Reporter assays showed that presence of wild-type, but not mutated CATAC elements, imparted increased expression levels as well as rhythmic regulation. Part of the CATAC consensus sequence resembles the E-box binding site for the core circadian transcription factor CLOCK/CYCLE (CLK/CYC), and CATAC-mediated expression rhythms are lost in the presence of null mutations in either cyc or the gene encoding the CLK/CYC inhibitor, period (per). Nevertheless, our results indicate that CATAC's enhancer function persists in the absence of CLK/CYC. Thus, CATAC represents a novel cis-regulatory element encoding clock-controlled regulation

    One man's war: a case for systemic change in military mental health

    In the following pages, I will use the story of my patient Lee, who I have been treating in my role as his therapist for a period of three and half years. This case study will be used to illustrate the difficulties that service members face when transitioning back into the community. I will also challenge some of the current leading paradigms that are used for treating individuals with symptoms related to their combat experiences. This case will highlight the importance of individualized, culturally competent treatment within the context of a strong therapeutic alliance and the absence of possible secondary gain. Through a person-in-environment and with a historical appreciation, this case will explore Lee's journey in hopes of providing some generalizable knowledge and offer a different way to conceptualize traumatic experiences and individuals' responses to them.DSWIncludes bibliographical referencesby Brandi M.     "Love, luck, and lollipops": children's television programming in Philadelphia, 1948-1969

    This project explores children's television programming in Philadelphia from the 1940s through the 1960s. Though Philadelphia was the third-largest television market during this period, very little scholarship on its shows exists. The city's broadcasting infrastructure allowed stations to provide programming to air nationally, and many children's shows that began as local Philadelphia fare became national hits. Personalities such as Dick Clark and Ed McMahon went on to national fame after appearing on Philadelphia television. Philadelphia stations created television series for children that were both entertaining and educational. The city was important to the history of the medium, and it was also at the forefront of the attempts to teach through television. The first efforts at in-school television were pioneered in Philadelphia, but the commercial stations had a commitment to education as well. Many shows had explicit educational goals, while others taught by example and encouraged social education. Though critics lamented the state of children's shows as frivolous entertainment as early as the 1950s, Philadelphia stations rose above the criticism and met the criteria that child experts put forth for programming. By examining in detail the children's shows that Philadelphia produced during this time period, it becomes evident that Philadelphia programming elevated what local television was expected to achieve. Shows were just as wholesome and enlightening, if not more so, than their nationally-known counterparts. Philadelphia programming deserves to be recognized as important in the history of children's television and should be remembered as imaginative, creative, and full of passionate people willing to try new approaches to children's entertainment.M.A.Includes abstractIncludes bibliographical referencesby Brandi Scardill

    Traces and shards of self-injury: Strange accounting with “Author X”

    In this strange account autoethnography, three or four authors explore their lived experiences with self-injury. Strange accounting is both a post-modern style of text, and a method for keeping identities concealed when risks and secrets are in play. Author X, a post-modern place-keeper for an anonymous author who may or may not have contributed to this manuscript, introduces a new dimension and layer of concealment. With Author X in-play and under erasure, the reader will never be sure if there were three or four authors on this manuscript. Through strange accounting, a post-structuralist/postmodernist frame will be applied to understanding the self-injury experience. We frame self-injury as a social practice and, for some, an everyday norm, while remaining acutely aware of the stigma surrounding the topic of self-injury. Each of us, coupled with Author X, provide the others cover to trace stories of self-injury through the literature, our flesh, and our lives

Le due città sono confrontate con i provvedimenti emanati anche in altre città dell'Emilia-Romagna attraverso gli statuti cittadini. L'analisi delle fonti dimostra che i problemi di igiene e decoro nella maggior parte delle città permasero anche nei secoli successivi, documentati anche da suppliche da parte dei cittadini.The author reviews waste disposal management in Rimini and Bologna in the 13th and 14th centuries. These cities are compared to other cities in the Emilia-Romagna region, by examining the statuti, in order to assess what laws were applied and the effectiveness thereof. The statuti were intents, political proposals aimed at finding solutions to complex issues, most of which were never solved and reappeared in subsequent centuries

    In the last few years we have witnessed the upsurge of publications in the area of conservation and restoration defending the idea that Cesare Brandi´s “Teoria del restauro” has become antiquated and is even impossible to comprehend, due to the identification of inherent contradictions and the difficult reading of its complex form of Italian language. This article aims to discuss some of the points presented by the Spanish author Salvador Muñoz Viñas as contradictions found in Brandi´s theory. Are they really contradictions? Reductionist interpretations end up leading to an polarized debate, that tends to fall into criticism for the sake of criticism. Here we would like to emphasize the need to get over this unproductive dichotomy between “brandians par excellence” and “postmodernists” of conservation. To the development of our argument, we will discuss the concept of restoration, the themes of integrity, authenticity and the brandian concept of potential unity.Nos últimos anos, testemunha-se o surgimento de publicações na área de conservação e restauração, defendendo a ideia de que a “Teoria da Restauração” de Cesare Brandi se tornou antiquada e até mesmo impossível de compreender, devido à identificação de contradições e à leitura difícil decorrente de uma pretensa erudição de sua escrita na língua italiana. Este artigo tem como objetivo discutir alguns dos pontos apresentados pelo autor espanhol Salvador Muñoz Viñas como contradições encontradas na teoria de Brandi. Eles são realmente contradições? Interpretações reducionistas acabam levando a um debate polarizado, que tende a cair na crítica pela crítica. Este artigo defende a necessidade de superar essa dicotomia improdutiva entre "brandianos por excelência" e "pós-modernistas" na conservação do patrimônio cultural. Para o desenvolvimento desse argumento, será discutido o conceito de restauração, os temas de integridade, autenticidade e o conceito brandiano de unidade potencial da obra de arte

    A multinuclear magnetic resonance study of nitrile oxides

    The 13C NMR signal of the CNO carbon atom in nitrile oxides is in general so broad at room temperature that it is often undetectable. This result has been related to the quadrupolar relaxation mechanism of the adjacent 14N nucleus. The unusual behavior of trimethylsilanecarbonitrile oxide, exhibiting a sharp 13C triplet and a narrow 14N singlet, indicates a dramatic increase in the 14N relaxation time, ascribed to the reduction of the electric field gradient around the 14N nucleus arising from a higher electron density on the adjacent carbon atom. © 1982
