HDAC2-dependent miRNA signature in acute myeloid leukemia
Acute myeloid leukemia (AML) arises from a complex sequence of biological and finely orchestrated events that are still poorly understood. Increasingly, epigenetic studies are providing exciting findings that may be exploited in promising and personalized cutting-edge therapies. A more appropriate and broader screening of possible players in cancer could identify a master molecular mechanism in AML. Here, we build on our previously published study by evaluating a histone deacetylase (HDAC)2-mediated miRNA regulatory network in U937 leukemic cells. Following a comparative miRNA profiling analysis in genetically and enzymatically HDAC2-downregulated AML cells, we identified miR-96-5p and miR-92a-3p as potential regulators in AML etiopathology by targeting defined genes. Leo&Giulia standing for public health : an animated series to promote the values of public health among school-aged children. Best practices and field-trial protocol
Background: The "Leo&Giulia standing for public health" project is an innovative digital health education model targeting primary school children. The project, developed during the COVID-19 pandemic, aims to educate primary school-aged children about public health issues through an animated cartoon series. It highlights the importance of early-life health promotion and the potential role of educational settings in shaping health behaviours. Study design: A 2-year school-based cluster-randomized controlled community trial will be conducted among 8-10-year-old pupils in the province of Pavia, Northern Italy. Methods: The intervention group will receive an educational programme via a new episode of "Leo&Giulia" animated series, focusing on smoking prevention. The study will assess changes in knowledge, attitudes towards smoking, and communication about smoking risks among peers and parents. The trial involves baseline and follow-up assessments through questionnaires targeting both children and parents. Results (expected): We assume that children in the intervention group will demonstrate increased knowledge and awareness of smoking-related health risks and develop negative attitudes towards tobacco use compared to the control group. Enhanced communication about tobacco harms among peers and between children and parents, as well as increased parental involvement in anti-smoking socialization practices, are expected secondary outcomes. Discussion and conclusions: "Leo&Giulia" integrates health education into the school curriculum, leveraging the appeal of animated content to engage children in public health topics. The project is expected to contribute to the field of health education by demonstrating the effectiveness of digital health interventions in childhood, foreseeing potential long-term impacts on health behaviors and in shaping future public health strategies

Effect of vascular risk factors on increase in carotid and femoral intima-media thickness. Identification of a risk scale
Background: The well established correlation between intima-media thickness (IMT) and the risk of cardiovascular and cerebrovascular events and death is usually measured in subjects with multiple vascular risk factors, which makes it difficult, after application of the usual analysis-of-variance linear combination of effects model, to establish whether each cardiovascular risk factor has, per se, an effect on IMT. Method and results: In this study we investigated five "pure" groups of patients (865), i.e. each presenting only one of the following risk factors: hypertension, obesity, overweight, smoking, hypercholesterolaemia and a control group of 37 healthy subjects. We measured, both as discrete and as continuous variables, the following indices: intima-media thickening of the common carotid artery (IMT(C)) and of the common femoral artery (IMT(F)) and the ankle-brachial index (ABI). Descriptive statistics were used to analyse the prevalence of pathological values for the three indices in the different groups. Subsequently the entire group of 902 subjects was included in a correlation analysis in which the Pearson correlation coefficient for each pair of variables was computed. In order to assign the risk factors a continuous ranking, and obtain a more general idea of the correlation structure, principal component analysis (PCA) was used. The scores obtained from PCA made it possible to build a scale of severity of the vascular risk factors considered. All the risk factors considered were demonstrated to strongly affect the studied indices. Overweight was shown to be the least important risk factor with regard to intima-media thickening, followed by smoking, hypercholesterolaemia, hypertension and finally obesity, which emerged as the greatest risk factor. Conclusions: The strong correlation between the indices made it possible to compute a composite general score, which provides an univocal risk estimation at single-patient level. IMT(F) was demonstrated to be the most sensitive descriptor. The construction of this risk scale has implications for preventive treatment and the frequency of instrumental examinations, allowing clear quantitative definition of the extent of the damage. : Exosomes are extracellular vesicles secreted by cells with a key role in a wide range of biological processes including cancer. These vesicles are involved in intercellular communication and deliver diverse cargo molecules, including miRNAs (exo-miRNAs), to recipient cells affecting their physiology. Exo-miRNAs have a role in promoting tumor, progression, metastatization, and remodeling of tumor microenvironment, therefore making them interesting biomarkers to study.Here we provide a detailed technical protocol for exosome isolation (which can be applied to cell culture as well as physiological fluids), validation of their vesicular identity, miRNA extraction, and quantitative and qualitative analysis to evaluate the sample purity and concentration
Restauro, ripristino e invenzione della policromia pittorica nell'architettura medievale. Alcuni esempi tra Napoli e Puglia nel XIX secolo
Sulle orme della Francia di Viollet-le-Duc, l’Italia della metà dell’Ottocento intraprese il restauro in stile dei suoi edifici medievali. Il fervore e la gloria nazionale all’alba dell’Unità si riversarono nelle singole aree regionali che scelsero i monumenti più rappresentativi della propria storia da ripristinare nelle presunte forme originali. Nel Meridione, Napoli, ancora capitale del Regno delle Due Sicilie, fu tra i centri più ricettivi degli sviluppi storico-critici dell’Oltralpe, complici i suoi circuiti accademici. Nella città partenopea si aprì quindi una fervida stagione di recupero dell’Evo Medio, a discapito delle stratificazioni moderne delle architetture e con un orientamento retrospettivo in cui il colore divenne un’inderogabile presenza. Dagli interventi nel Duomo al lavoro di Federico Travaglini nel S. Domenico Maggiore, architetto poi attivo nei cantieri pugliesi di Altamura e Troia, la policromia, struttiva e dipinta, confermò un nuovo modo di guardare al Medioevo, di certo non filologico ma nemmeno fatto di un “candido manto di chiese” così come lo aveva descritto Rodolfo il Glabro. A questa tendenza maggioritaria di uso ridondante delle cromie come cifra distintiva dell’Età di Mezzo si sarebbero poi opposte alcune voci meno conosciute. Tra queste quella di Sante Simone, architetto a cavallo tra Puglia e Napoli e contemporaneo di Travaglini, che, nonostante dovette piegarsi alle volontà delle committenze e quindi impiegare la policromia, fu uno strenuo difensore della semplicità della nuda pietra
Notizie da Venezia e dall'Oriente negli inediti dispacci sforzeschi di Leonardo Botta
I rapporti diplomatici tra il ducato di Milano e la repubblica di Venezia furono particolarmente vivaci nella seconda metà del Quattrocento. Viste vicinanza geografica e rilevanza politica della Serenissima, il duca milanese Galeazzo Maria Sforza decise di inviare nel 1473 un ambasciatore ufficiale in Laguna, Leonardo Botta. Questa ambasceria, durata 7 anni, si distinse per essere stata una delle più lunghe e fruttuose, sia per Milano che per la scomoda, ma potente vicina Venezia. Durante questi anni presso la Serenissima, l’oratore inviò numerosissime lettere al suo signore, con l’intento di informarlo riguardo tutto quello che avveniva a Venezia, allora la porta verso l’Oriente. Il mio lavoro, partito dalla trascrizione di 121 missive inedite, inviate da Botta al duca, e conservate nell’Archivio di Stato di Milano, vuole fornire uno spaccato della realtà e della vita veneziana viste con gli occhi e descritte con le parole di un perspicace ambasciatore lombardo del tempo. Oltre ai contenuti, estremamente vari, lo studio si sofferma anche sulla cifra, sulla grafia e sulla lingua usate da Botta, con lo scopo di presentare un quadro completo della fonte esaminata. Gli argomenti trattati sono, si diceva, molti: ecco comparire nelle lettere i Turchi, pericolosi invasori da est che assediano la cittadella di Scutari, l’Ungheria e la Persia, potenziali alleate per Venezia, ma anche le altre realtà italiane, Firenze, Roma, Napoli, Ferrara, con le quali la Serenissima e Milano volevano, ognuna per ragioni distinte, giungere ad un accordo generale, e infine i dissidi personali dello Sforza col celeberrimo condottiero Bartolomeo Colleoni, suddito di San Marco.
