    Family relationships in childhood and common psychiatric disorders in later life : systematic review of prospective studies

    Background Most evidence for associations between childhood adversity and adult mental illness is retrospective. Aims To evaluate prospective evidence of associations between poor parent-child relationships and common psychiatric disorders in later life. Method Systematic review of studies published between 1970 and 2008 including: (a) more than 100 participants, (b) measures of relationships in the home during childhood, (c) at least 10 years between assessment of exposures, and (d) measures of anxiety, depression, suicide, suicidal ideation or post-traumatic stress disorder (PTSD). Analysis was by narrative synthesis. Results Twenty-three papers were identified reporting data from 16 cohorts. Abusive relationships predicted depression, anxiety and PTSD. Maternal emotional unavailability in early life predicted suicide attempts in adolescence. Results of studies investigating less severe relationship problems were Suggestive but not conclusive of causal association, due partly to methodological heterogeneity. Conclusions Given the prevalence and disabling nature of common psychiatric problems, these studies highlight the need to minimise harm associated with dysfunctional parent-child relationships

    © 2010 Dr. Scott John PattersonHepatitis B is a major global health problem with an estimated 2 billion people having been infected with the hepatitis B virus (HBV) and an estimated 350-400 million people suffering chronic infection. Hepatitis B is the 10th leading cause of death worldwide and hepatocellular cancer, a complication of chronic hepatitis B infection, is the 5th most common cancer worldwide. In this thesis I review the virology, epidemiology, natural history and therapy of hepatitis B infection. The mainstay of medical management of chronic hepatitis B are oral nucleos(t)ide analogues which have the capacity to profoundly suppress viral replication and prevent disease progression. The major shortcoming of nucleos(t)ide therapy is the development of resistance. Therapy for these patients is increasingly difficult in the setting of multi-drug resistance. In this thesis I investigate the use a newer nucleotide, tenofovir disoproxil fumarate, in patients with previous multi-drug (lamivudine and adefovir dipivoxil) resistant hepatitis B. I show tenofovir is safe and effective in such patients, although probably less so than in nucleos(t)ide naïve patients. Post-liver transplant recurrence of hepatitis B may be severe and life threatening. Standard prophylaxis regimes utilise hepatitis B immune globulin (HBIG) and lamivudine indefinitely post-transplant. In this thesis I investigate the use of combination lamivudine and adefovir dipivoxil prophylaxis, either post-transplant (after a period of HBIG and lamivudine) or pre-transplant (avoiding the use of prolonged HBIG entirely). I show combination adefovir and lamivudine (both pre- and post-transplant) achieves and/or maintains suppression of hepatitis B viral replication and prevents the development of recurrence. This has the benefit of significant cost-savings and is less inconvenient for liver transplant recipients

    Effects of Two Aromatic Hydrocarbon Derivatives on Mammalian Cells: Cytogenetic, LHtrastructural and Electron Spin Resonance Analyses Advisor: Dr. Rosalyn Patterson Master of Science degree conferred May 21, 1979 Thesis dated May, 1979 A cytogenetic analysis of Muntiacus muntjac fibroblast cells exposed to various concentrations of Rhodamine B (DSC Red No. IS) and amaranth (FDSC Red No. 2) was examined. The dye of particular interest in this study was Rhodamine B which is used to color many consumer products. The dye has been found to cause carcinogenic effects in animals injected subcutaneously with Rhodamine B. Cytogenetic data revealed a significant increase in chromosomal aberrations of the experimental cells. A higher percentage of chromosomal aberrations was observed in cells treated with Rhodamine B as compared to those cells treated with amaranth. An increase in the concentration of Rhodamine B resulted in an increase in the types and amounts of aberrations observed. Chromosomes that were particularly susceptible to becoming aberrant were chromosome pairs 1 and an electron spin resonance analysis was performed to investigate the action of Rhodamine B as a possible free radical solvator. Data obtained revealed a possible 1-6 addition product of Rhodamine B and TEMPONE in which TEMPONE appears to add reversibly to Rhodamine B and thereby complete quenching of the signal is not achieved. Conclusive evidence has not been obtained on radical solvation of Rhodamine B and TEMPONE, but such studies are forthcoming and will be performed at much lower temperatures. An ultrastructural analysis was performed to observe the effects of Rhodamine B at the subcellular level.
