These datasets are contain Beta gal assay images, DAPI stained nuclei and 3D FISH images from oncogene and replicative senescent cells. The Betagal assay was performed to demonstrate the senescent condition. The DAPI stained nuclei images are from different siRNA treated OIS samples. The FISH images display the presence of SHADs in SAHFs.
The Summary of work:
While transcriptional and epigenetic changes associated with senescence are well studied, the role of the extensive senescence-associated 3D genome reorganization remains elusive. Here, we have generated genome wide chromatin interaction maps, epigenetic, replication-timing, whole genome bisulfite sequencing and gene expression profiles from cells entering replicative senescence (RS) or upon oncogene induced senescence (OIS). We identify Senescence Associated Heterochromatin Domains (SAHDs). Differential intra vs inter SAHD interactions lead to the formation of senescence associated heterochromatin foci (SAHFs) in OIS but not in RS. This OIS-specific configuration brings active genes located in genomic regions adjacent to SAHDs in close spatial proximity and favours their expression. Finally, screening of factors for SAHF induction revealed DNMT1 as a novel component that induces SAHFs by promoting HMGA2 expression. Upon DNMT1 depletion, OIS cells transition to a 3D genome conformation akin to that of cells in replicative senescence. These data show how multi-omics and imaging can identify critical features of RS and OIS and discover new determinants of acute senescence and SAHF formation

Sati
This chapter examines the history of the traditional Hindu practice of widow self-immolation, commonly known as sati, which is one of the mostly widely known and discussed forms of ritual suicide in world religions. The chapter begins by briefly placing sati within the context of other historically practiced forms of “following into death” (in German, Totenfolge), and discussing those features of sati that make it unique among these practices. Then, in three separate sections, it provides an account of the earliest surviving sources on sati, which likely date as far back as the fourth century BCE; outlines an important medieval debate on the validity of the practice that took place within the orthodox Hindu legal tradition known as Dharmaśāstra; and, lastly, notes some major later developments regarding sati, including especially its legal prohibition by the British colonial government in India in 1829.</p

Drama tari ini mengisahkan tentang kesetiaan Sati Devi pada suaminya, Dewa SIwa serta kekecewaan Sati terhadap ayahnya Prajapati Daksa yang telah menghina dan mengutuk Dewa Siwa. Atas kekecewaan hatinya terlahir dari ayah yang sombong dan arogan dan menghina suaminya di hadapan para dewa dan brahmana, Sati melepas raganya. Sati practice in india: historical analysis.
This research work is devoted to the analysis of Sati - the practice of widow self-immolation in Hindu society - from a historical perspective. The topic of Sati is interesting in that it covers many different aspects (religion, tradition, politics, women’s rights, feminism) and has been, as well as still is, surrounded by conflicting evaluations. The main goal of this work is to explore the phenomenon of Sati from the perspective of different historical periods and different actors, revealing the conflicting evaluations of the practice of Sati. The main objectives of this research work are: to investigate the practice of Sati in pre-colonial India; to review the evaluations of Sati by the first Westerners in India; to analyse the transformation of the practice during colonial period; to explore the significance of Sati during the period of Indian nationalism; to analyse the practice of Sati in the post-colonial period and discuss the case of Roop Kanwar. Methodology: this research is based on a historical analysis of the Sati phenomenon. The primary sources are taken from Andrea Major's “Sati a Historical Anthology” (2007), which contains stories, memoirs and discussions on the Sati practice by scholars, travellers, political figures and reformers in the period of 16th-21st centuries. The analysis from the perspective of different historical periods of India reveals that the Sati practice has always been viewed with controversy and as a serious moral dilemma

Always exceptional and effecting only a tiny minority of Hindu widows, it has remained close to the surface of social and political life and has played a disproportionately prominent role within Indian history and culture. The importance given to this rite in 'Western' accounts of India since the fifteenth century, as well as the significance of its 'ethos', if not its actual practice, within Indian culture, has meant that sati has remained in the public eye for several centuries and has taken on a variety of different meanings at different times, and for different observers.
