The efficiency of nitrogen fixation of the model legume Medicago truncatula (Jemalong A17) is low compared to Medicago sativa
Medicago truncatula (Gaertn.) (barrel medic) serves as a model legume in plant biology. Numerous studies have addressed molecular aspects of the biology of M. truncatula, while comparatively little is known about the efficiency of N-2 fixation at the whole plant level. The objective of the present study was to compare the efficiency of N-2 fixation of M. truncatula to the genetically closely related Medicago sativa (L) (alfalfa). The relative growth of both species relying exclusively on N-2 fixation versus nitrate nutrition, H-2 evolution, nitrogen assimilation, the concentration of amino acids and organic acids in nodules, and N-15(2) uptake and distribution were studied. M. truncatula showed much lower efficiency of N-2 fixation. Nodule-specific activity was several-fold lower when compared to M. sativa, partially as a result of a lower electron allocation to N-2 versus W. M. truncatula or M. sativa plants grown solely on N-2 fixation as a nitrogen source reached about 30% or 80% of growth, respectively, when compared to plants supplied with sufficient nitrate. Moreover, M. truncatula had low %N in shoots and a lower allocation of N-15 to shoots during 1 h N-15(2) labeling period. Amino acid concentration was about 20% higher in M. sativa nodules, largely as a result of more asparagine, while the organic acid concentration was about double in M. sativa, coinciding with a six-fold higher concentration of malate. Total soluble protein in nodules was about three times lower in M. truncatula and the pattern of enzyme activity in that fraction was strongly different. Sucrose cleaving enzymes displayed higher activity in M. truncatula nodules, while the activity of phosphoenolpyruvate carboxylase (PEPC) was much lower. It is concluded that the low efficiency of the M. truncatula symbiotic system is related to a low capacity of organic acid formation and limited nitrogen export from nodules. (C) 2010 Elsevier GmbH. The periods 1958-61 and 1974-77, when Srinivas was 38-41 and 58-61 years old. were his most productive periods with highest publication activity
Asparagine as a major factor in the N-feedback regulation of N-2 fixation in Medicago truncatula
The objective of this study was to assess whether a whole plant N-feedback regulation impact on nitrogen fixation in Medicago truncatula would manifest itself in shifts of the composition of the amino acid flow from shoots to nodules. Detected shifts in the phloem amino acid composition were supposed to be mimicked through artificial phloem feeding and concomitant measurement of nodule activity. The amino acid composition of the phloem exudates was analyzed from plants grown under the influence of treatments (limiting P supply or application of combined nitrogen) known to reduce nodule nitrogen fixation activity. Plants in nutrient solution were supplied with sufficient (9 μM) control, limiting (1 μM) phosphorus or 3 mM NHNO (downregulated nodule activity). Low phosphorus and the application of NHNO reduced per plant and specific nitrogenase activity (H evolution). At day 64 of growth, phloem exudates were collected from cuts of the shoot base. The amount of amino acids was strongly increased in both phloem exudates and nodules of the treatments with downregulated nodule activity. The increase in the downregulated treatments was almost exclusively the result of a higher proportion of asparagine in both phloem exudates and nodules. Leaf labeling with N showed that nitrogen from the leaves is retranslocated to nodules. An artificial phloem feeding with asparagine resulted in an increased concentration of asparagine in nodules and a decreased nodule activity. A possible role of asparagine in an N-feedback regulation of nitrogen fixation in M. truncatula is discussed
Measuring Digital Twin Entanglement in Industrial Internet of Things
Digital Twins (DTs) have recently emerged as a valuable approach for modeling, monitoring, and controlling physical objects in Industrial Internet of Things applications. Measuring the quality of entanglement between the digital and physical counterparts plays a crucial role in the adoption of DTs. In this paper, we propose a concise yet expressive metric for representing the quality of entanglement, namely Overall Digital Twin Entanglement (ODTE), based on two key factors: timeliness and completeness. Furthermore, the paper presents the development of our industrial testbed implemented on top of Kubernetes, where we show practical applications of the proposed ODTE metric by highlighting and discussing its benefits in realistic use cases
Chiral discrimination in the formation reaction and at equilibrium for N,N,N ',N '-tetramethyl-1,2-diaminocyclohexane-PtG(2) complexes
The effect of an extremely bulky carrier ligand, such as N,N,N,N-tetramethyl-1,2-diaminocyclohexane (Me4DACH, R,R and S,S configurations of the asymmetric carbon atoms of the chelate ring), upon the rate of formation and stability of Me4DACH–PtG2 derivatives has been investigated (G = 9-EtG, 3-GMP, and 5-GMP). Of the three possible rotamers, two head-to-tail (ΔHT and ΛHT) and one head-to-head (HH), only the former two are found in solution. The very small difference in H8 chemical shifts between ΔHT and ΛHT rotamers indicates that the relationship between the two guanines is very similar in the two conformers which are canted to the same degree but in opposite directions. A smaller interligand steric clash appears to favour the HT rotamer in which the H8 atoms of the guanines are on the same side of pseudo-axial N–Me’s with respect to the platinum coordination plane. In
the case of 5-GMP, because of the absence of aminic protons on the carrier ligand, the 5-phosphate directs its H-bonding potential towards the second ligand in cis-position (the coordinated H2O molecule in the mono adduct, [Me4DACH–Pt(H2O)(5-GMP)], and the cis-G in the bis-adduct). As a consequence the rotamer in which such an interaction is possible, with the nucleotide keeping its favoured anti conformation, is more stable than expected. In
contrast with the behaviour of 5-GMP, 3-GMP is found to increase the reactivity of the mono adduct in which the phosphate is directed towards the cis-water molecule and to give a negligible contribution to the stability of the HT rotamer in which the 3-phosphate is directed towards the cis-nucleotide
