    Modulation of 3-hydroxy-3-methylglutaryl-CoA reductase gene expression by CuZn superoxide dismutase in human fibroblasts and HepG2 cells

    The homeostasis of intracellular cholesterol in animal cells is highly regulated by a complex system in which the microsomal rate-limiting enzyme 3-hydroxy-3-methylglutaryl CoA (HMG-CoA) reductase plays a key role in cholesterol synthesis. Substantial evidence has demonstrated that the cytosolic antioxidant enzyme CuZn superoxide dismutase (SOD1) inhibits the HMG-CoA reductase activity in rat hepatocytes and in human fibroblasts by decreasing cholesterol synthesis. Although these data that suggest that SOD1 exerts a physiological role in cholesterol metabolism, it is still unclear whether the decrease of HMG-CoA reductase activity is mediated by transcriptional or by posttranscriptional events. The results of the present study, obtained by one-step RT-PCR assay, demonstrated that both SOD1 and the metal-free form of enzyme (Apo SOD1) inhibit HMG-CoA reductase gene expression in hepatocarcinoma HepG2 cells, in normal human fibroblasts, and in fibroblasts of subjects affected by familiar hypercholesterolemia.     Presented at the Student Experience in the Research University (SERU) symposium, Berkeley, CA, May 4, 2012.Presented at the 6th annual SERU/UCUES Research Symposium. In this research paper, we utilize data from the Community and Civic Engagement Module of the multi-institutional Student Experience in the Research University (SERU) survey in order to estimate potential direct andindirect effects of community engagement experiences on undergraduate students' academic and social gains. A latent variable structural equation model of student achievement was developed to explore the potential relationship between participation in community-based learning activities (including enrollment in a course with a service-learning component and involvement in community based research) and students' perceptions of academic and cultural development. Specifcally, our empirical investigation addresses two questions: Does participation in community-based learning opportunities contribute, either directly or indirectly, to participating students' improved academic learning and skills; and does participation in community-based learning opportunities contribute, either directly or indirectly, to participating students' prosocial skills development?     Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Kajian dalam skripsi ini dilatarbelakangi oleh praktik cloning account yang ada pada progam serbu seru Bukalapak. Dalam praktik cloning account tersebut, pengguna membuat atau menggunakan lebih dari satu akun untuk mengikuti progam dengan tujuan memperbesar peluang untuk menjadi penyerbu terpilih, hal ini berdampak dirasa merugikan untuk pengguna lain yakni pengguna satu akun dimana akan memperkecil peluang menjadi penyerbu terpilih. kemudian hingga pembatalan pengiriman barang oleh tim Bukalapak karena alasan kesamaan data atau indikasi kecurangan antara pengguna satu akun dengan pengguna cloning account karena, data yang digunakan oleh pengguna cloning account bukan hanya miliknya pribadi melainkan milik orang lain yang mana akan terdapat kesamaan data seperti alamat penduduk didesa untuk selanjutnya, dilakukan pemilihan ulang penyerbu terpilih oleh tim Bukalapak. Dari latar belakang diatas, peneliti menggunakan rumusan masalah (1). Bagaimana tinjauan etika bisnis Islam terhadap cloning account serbu seru pada aplikasi Bukalapak? (2). Bagaimana tinjauan etika bisnis Islam terhadap dampak cloning account serbu seru pada aplikasi Bukalapak? Jenis penelitian yang digunakan dalam penelitian ini merupakan penelitian lapangan (field research) yang menggunakan pendekatan kualitatif. Sedangkan dalam pengumpulan data penulis menggunakan teknik pengumpulan data wawancara, observasi dan dokumentasi. Selanjutnya dilakukan pengolahan data yang meliputi editing, organizing, dan penemuan hasil riset. Teknik analisa data yang digunakan dalam penelitian ini adalah metode induktif. Hasil dari penelitian ini menyimpulkan bahwa, cloning account serbu seru Bukalapak kurang sesuai dengan beberapa nilai etika bisnis Islam seperti nilai S}iddiq yang mana di sini berarti mampu melaksanakan taransaksi dengan jujur dengan melandaskan tindakan dan ucapan sesuai dengan ajaran Islam. Juga dengan nilai Ama>nah di sini berarti mampu dapat dipercaya dalam melakukan transaksi. Dan untuk Dampak yang ditimbulkan oleh pengguna cloning account jika ditinjau dengan Etika Bisnis Islam kurang sesuai dengan nilai keadilan yang mana di dalamnya ada konsep tidak terdapatnya ghara>r, ghara>r dalam pengertian ada kezaliman terhadap pelaku ekonomi lainnya

    Free fatty acids modulate LDL receptor activity in BHK-21 cells

    It has been shown that dietary fatty acids affect serum low density lipoprotein (LDL) levels, but the mechanism responsible for this effect is still under debate. Here we investigate the effect of different free fatty acids on LDL receptor activity in BHK-21 cells. These cells possess a classical LDL receptor strongly regulated by substances like 25-OH-cholesterol or lovastatin. Preincubation of cells for 24 h with both oleic (cis 18:1) and its trans counterpart, elaidic acid, enhanced I-125-LDL binding, internalization and degradation, being oleic acid more effective than elaidic acid. Among polyunsaturated fatty acids (PUFA) of the n-6 series arachidonic acid (20:4) enhanced LDL receptor activity more than linoleic acid (18:2), and among PUFA of the n-3 series docosahexaenoic (22:6) and eicosapentaenoic acids (20:5) were more effective compared to a-linolenic acid (18:3). Conversely, preincubation of cells with saturated fatty acids, palmitic (16:0) and stearic (18:0) acids, decreased binding, internalization and degradation of I-125-LDL. Scatchard analysis of binding data obtained with palmitic and oleic acids showed that these two fatty acids affect LDL receptor number without altering receptor affinity. The regulatory effect of free fatty acids on LDL receptor activity in BHK-21 cells is consistent with the hypothesis that the ability of fatty acids to modulate LDL-cholesterol levels in vivo is mediated, at least in part, by an action on receptor-dependent uptake of LDL.     "The Shift from Belt Conveyor Line to Work-cell Based Assembly Systems to Cope with Increasing Demand Variation and Fluctuation in The Japanese Electronics Industries"

    As consumption patterns become increasingly sophisticated and manufacturers strive to improve their competitiveness, not only offering higher quality at competitive costs, but also by providing broader mix of products, and keeping it attractive by launching successively new products, the turbulence in the markets has intensified. This has impelled leading manufacturers to search the development of alternative production systems supposed to enable them operate more responsively. This paper discusses the trend of abandoning the strategy of relying on factory automation technologies and conveyor-based assembly lines, and shifting towards more human-centered production systems based on autonomous work-cells, observed in some industries in Japan (e.g. consumer electronics, computers, printers) since mid-1990s. The purpose of this study is to investigate this trend which is seemingly uneconomic to manufacturers established in a country where labor costs are among the highest in the world, so as to contribute in the elucidation of its background and rationality. This work starts with a theoretical review linking the need to cope with nowadays' market turbulence with the issue of nurturing more agile organizations. Then, a general view of the diffusion trend of work-cell based assembly systems in Japanese electronics industries is presented, and some empirical facts gathered in field studies conducted in Japan are discussed. It is worthy mentioning that the abandonment of short cycle-time tasks performed along conveyor lines and the organization of workforce around work-cells do not imply a rejection of the lean production paradigm and its distinctive process improvement approach. High man-hour productivity is realized as a key goal to justify the implementation of work-cells usually devised to run in longer cycle-time, and the moves towards this direction has been strikingly influenced by the kaizen philosophy and techniques that underline typical initiatives of lean production system implementation. Finally, it speculates that even though the subject trend is finding wide diffusion in the considered industries, it should not be regarded as a panacea. In industries such as manufacturing of autoparts, despite the notable product diversification observed in the automobile market, its circumstances have still allowed the firms to rely on capital-intensive process, and this has sustained the development of advanced manufacturing technologies that enable the agile implementation and re-configuration of highly automated assembly lines.
