    Hb Vila Real [beta36(C2)Pro®His] in Italy: characterization of the amino acid substitution and the DNA mutation

    A rare high oxygen affinity hemoglobin variant was identified in a 22-year-old male patient from Napoli (Naples, Italy) affected by erythrocytosis. A detailed structural characterization of the variant hemoglobin was carried out, both at the protein and DNA levels essentially by mass spectrometric procedures and allele-specific amplification techniques. The amino acid substitution was determined by liquid chromatography tandem mass spectrometric analysis of the tryptic digest as β36(C2)Pro → His; the corresponding DNA mutation was identified as C → A at the second position of codon 36 of the β chain (CCT → CAT). These variations identified the presence of Hb Vila Real, described only once before in a Portuguese woman. Haplotype analysis of DNA polymorphisms showed that the β-globin gene of Hb Vila Real was associated with haplotype I

    Comparative proteomics and immunoproteomics of Helicobacter pylori related to different gastric pathologies

    Helicobacter pylori is a Gram-negative bacterium which causes ulcer, atrophic gastritis, adenocarcinoma, or mucosa-associated lymphoid tissue lymphoma. A comparative proteomic and immunoproteomic analysis was chosen to identify the antigenic patterns of three different H. pylori strains. These strains were probed against single sera from H. pylori-positive patients affected by gastric adenocarcinoma or duodenal ulcer. We found a quite heterogeneous antigenic pattern, both from strain and sera points of view, thus underlying both a strain- and a host-specificity. The different antigenic repertoires introduced the importance of the strain to be used for immunoblotting as a diagnostic test

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
