Young and unknown artist – Ieva Grigienė is keen on making her identity visible for her old and potencial clients. Also, she has the urge to tell everyone about jewelry particularity. After the theory part, which examined brand identity, social media, luxury brands, jewelry creation exceptions, a communication plan was made. 2019 on December, Ieva created her Facebook page according to communication plan. Quality analysis showed if artists aim to create her identity as young, creative and friendly person, has succeeded. During this social experiment communication experts had open questions, which required their opinion about Griga juvelyrika brand and young artist – Ieva Grigienė. Hypothesis that social media users can misunderstand or read completely different brand and jewellrer identity, wasn’t fully denied. All research participats thought that Ieva is creative and warmly communicating person but no one answered that Griga juvelyrika is a luxury brand. Though a lot luxury features were assigned to Grigienė personal brand. Analysis showed that Ieva created good impression and her personal brand provokes good emotions. Respondents gave few ideas and reccomendations how to make this brand and artists identity better, how to expand clients and followers circle. Production of biologically active proteins in plants
6. SOUHRN 1) Byly zavedeny tři rostlinné expresní systém umožňující produkci biologicky aktivních proteinů. První využívá dočasné exprese z vektoru dopraveného do buňky Agrobacteriem po infiltraci listů tabáku bakteriální suspenzí. Pomocí tohoto systému jsme schopni vyprodukovat množství rekombinantního proteinu dosahujícího 5% CRP během 3 dnů. Druhý expresní systém využívá suspenzní kultury stabilně transformovaného mechu Physcomitrella patens. Za kontrolovaných podmínek pěstování v bioreaktoru je výtěžek rekombinantního proteinu srovnatelný s výtěžkem z agroinfiltrace. Expresní systém využívající kombinace virové expresní kazety, založené na TRV, a jejího přenosu do rostlinné buňky pomocí Agrobacteria je pro produkční účely vzhledem k nízkým výtěžkům méně vhodný, nežli předchozí dva systémy. 2) Ve všech třech systémech lze produkovat heterologní proteiny, které se svými vlastnostmi neliší od proteinů produkovaných v původním organismu. Pro dosažení požadované aktivity heterologního proteinu produkovaného rostlinou, stejně jako pro dosažení vysokého výtěžku, je nutné provést řadu optimalizací. Významné je zjištění, že v případě virových antigenů (HPV16L1 protein) i jednořetězcových fragmentů protilátek (scFv MEM97) je pro rostlinu výhodnější exprimovat sekvence genů rekombinantních proteinů s vyšším..
MEASUREMENT OF THE TURBOCOMPRESSOR
katedra: KEZ; přílohy: CD ROM; rozsah: 44 s , přílohy 20 sThe purpose of this Bachelor Thesis is to measure the characteristics of a turbocharger and to recalculate them in respect of the guaranteed values declared by the manufacturer. The turbocharger was manufactured in 1965 by ČKD and now it is located in the laboratory of the Technical University of Liberec. The Thesis is divided into several chapters. The first chapter deals with the theory of radial turbochargers and marginally also with the theory of axial turbochargers. The second chapter describes the entire measuring line with all the measuring devices and instruments. The third chapter defines the procedure of the measurements performed on the turbocharger. The fourth chapter contains a graphical evaluation of the measured and calculated values. The subject matter of the fifth chapter is a proposal for a laboratory exercise titled Turbocharger Measurement, and the sixth chapter contains the conclusion of the whole measurement. The values that have been measured and calculated are shown in the charts in Appendix II and their sample form is attached as Appendix I.Cílem této bakalářské práce je naměření charakteristik turbokompresoru a jejich přepočítání na zaručené stavy deklarované výrobcem. Turbokompresor byl vyroben v roce 1965 ve firmě ČKD a dnes je umístěn v laboratoři Technické univerzity v Liberci. Práce je rozčleněna do několika kapitol. V první kapitole je zpracována teorie radiálních turbokompresorů a okrajově axiálních turbokompresorů. Ve druhé kapitole je popsána celá měřící trať se všemi měřícími přístroji a zařízeními. Ve třetí kapitole je uveden postup měření na turbokompresoru. Ve čtvrté kapitole je grafické vyhodnocení naměřených a vypočítaných hodnot. V páté kapitole je návrh laboratorní úlohy Měření na turbokompresoru a v poslední šesté kapitole je závěr celého měření. The outburst of dammed lake Maashey (North-Chuya ridge, Central Altai)
The dammed lakes are widespread in mountainous areas and usually occur when river flow is blocked by landslides, rock glaciers, etc. Among such lakes, the most dangerous are those located in the periglacial zone and blocked by rock glaciers. Continued deglaciation of mountainous areas under changing climate conditions contributes to accumulation of large volumes of melt water in lakes, which may increase pressure on the dam, cause its failure and subsequent outburst flood. In this article we describe the development of such a lake before its outburst and the process of its outburst. The object of study was Maashei Lake (North Chuya Ridge, Central Altai) located in the zone of mountain glaciation and dammed by a rock glacier, where the lake outburst occurred in July 2012. The lake area before the outburst was 259 × 103 m2 and water volume 1.21 × 106 m3. As a result of the outburst, the lake was completely drained. We analyzed the published works on Lake Maashei, materials of our own field studies in the lake basin combined with remote sensing data. We hypothesized that the mechanism of the outburst occurred in 2012 was caused by the water erosion of the filtration channel in the dam body. The mechanism of this outburst was numerically simulated using the method presented in this article. The modeling allowed to reproduce the outburst flood hydrograph, to estimate such characteristics as maximum water discharge, volume of the outburst flood, water flow velocities and the size of the formed breach. Estimated maximum discharge was 694 m3s−1, flow velocities varied from 0.2 to 5-7 m s−1, and the outburst flood period was about 5.5 hours. The breach was formed to the full height of the dam (10 m). Its calculated morphometric characteristics were as follows: average width 47.5 m (measured 41.5m), скоss-section area 476 m2 (measured 415 m2). Association of extraintestinal manifestations of inflammatory bowel disease in a province of western Hungary with disease phenotype: Results of a 25-year follow-up study
AIM: IBD is a systemic disease associated with a large number of extraintestinal manifestations (EIMs). Our aim was to determine the prevalence of EIMs in a large IBD cohort in Veszprem Province in a 25-year follow-up study.
METHODS: Eight hundred and seventy-three IBD patients were enrolled (ulcerative colitis/UC/: 619, m/f: 317/302, mean age at presentation: 38.3 years, average disease duration: 11.2 years; Crohn's disease/CD/: 254, m/f: 125/129, mean age at presentation: 32.5 years, average disease duration: 9.2 years). Intestinal, extraintestinal signs and laboratory tests were monitored regularly. Any alteration suggesting an EIMs was investigated by a specialist.
RESULTS: A total of 21.3 % of patients with IBD had EIM (UC: 15.0 %, CD: 36.6 %). Age at presentation did not affect the likelihood of EIM. Prevalence of EIMs was higher in women and in CD, ocular complications and primary sclerosing cholangitis (PSC) were more frequent in UC. In UC there was an increased tendency of EIM in patients with a more extensive disease. Joint complications were more frequent in CD (22.4 % vs UC 10.2 %, P<0.01). In UC positive family history increased the risk of joint complications (OR:3.63). In CD the frequency of type-1 peripheral arthritis was increased in patients with penetrating disease (P=0.028). PSC was present in 1.6 % in UC and 0.8 % in CD. Dermatological complications were present in 3.8 % in UC and 10.2 % in CD, the rate of ocular complications was around 3 % in both diseases. Rare complications were glomerulonephritis, autoimmune hemolytic anaemia and celiac disease.
CONCLUSION: Prevalence of EIM in Hungarian IBD patients is in concordance with data from Western countries. The high number of EIM supports a role for complex follow-up in these patients
Specific Effects of the 1988 Earthquake on Topography and Glaciation of the Tsambagarav Ridge (Mongolian Altai) Based on Remote Sensing and Field Data
Strong earthquakes could serve as a trigger for glacier detachment and associated ice–rock avalanches. The 1988 Tsambagarav earthquake (M = 6.4) initiated collapse of part of the glacier tongue and a further ice–rock avalanche with an abnormal 5 km long path in Zuslan valley, Tsambagarav ridge (Mongolian Altai). Early documentation of surface effects in 1988, remote sensing and field data gathered 16 and 30 years after this event allowed for the assessment of the seismic impact on a reduction of “damaged” glacier under conditions of global warming as well as estimating topography changes in this arid and seismically active area. Because of the earthquake, the glacier immediately lost 10.4 % of its area (0.1 km2 of tongue surface). Additionally, 56% of its area was lost during 1988–2015, shrinking much faster than neighboring glaciers of similar size and exposition. Collapse of snow–ice cornice in the accumulation zone could play a key role in rapid acceleration of the detached ice block and abnormally long path of the ice–rock avalanche. A large amount of debris material provided more than 16 years of ice melting. Downstream, the valley avalanche debris cover repeats the topography of underlying Pleistocene moraines, which should be considered in regional paleogeographical reconstructions. © 2022 by the authors. Novel genetic risk markers for ulcerative colitis in the IL2/IL21 region are in epistasis with IL23R and suggest a common genetic background for ulcerative colitis and celiac disease
OBJECTIVES: Recently, a genome-wide association study showed that single-nucleotide polymorphisms (SNPs) in the chromosome 4q27 region containing IL2 and IL21 are associated with celiac disease. Given the increased prevalence of inflammatory bowel disease (IBD) among celiac disease patients, we investigated the possible involvement of these SNPs in IBD. METHODS: Five SNPs strongly associated with celiac disease within the KIAA1109/TENR/IL2/IL21 linkage disequilibrium block on chromosome 4q27 and one coding SNP within the IL21 gene were analyzed in a large German IBD cohort. The study population comprised a total of 2,948 Caucasian individuals, including 1,461 IBD patients (ulcerative colitis (UC): n=514, Crohn's disease (CD): n=947) and 1,487 healthy unrelated controls. RESULTS: Three of the five celiac disease risk markers had a protective effect on UC susceptibility, and this effect remained significant after correcting for multiple testing: rs6840978: P=0.0082, P(corr)=0.049, odds ratio (OR) 0.77, 95% confidence interval (CI) 0.63-0.93; rs6822844: P=0.0028, P(corr)=0.017, OR 0.73, 95% CI 0.59-0.90; rs13119723: P=0.0058, P(corr)=0.035, OR 0.75, 95% CI 0.61-0.92. A haplotype consisting of the six SNPs tested was markedly associated with UC susceptibility (P=0.0025, P(corr)=0.015, OR 0.72, 95% CI 0.58-0.89). Moreover, in UC, epistasis was observed between the IL23R SNP rs1004819 and three SNPs in the KIAA1109/TENR/IL2/IL21 block (rs13151961, rs13119723, and rs6822844). CONCLUSIONS: Similar to other autoimmune diseases such as celiac disease, rheumatoid arthritis, type 1 diabetes, Graves' disease, and psoriatic arthritis, genetic variation in the chromosome 4q27 region predisposes to UC, suggesting a common genetic background for these diseases
