Entwicklung neuer massenspektrometrischer Methoden für die Suche, Strukturaufklärung und biologische Untersuchung von peptidischen Toxinen
Natürliche Toxine sind von großer Bedeutung für den Menschen. Ihre biochemischen Eigenschaften resultieren in einem „Ying und Yang“ an tödlichem und zugleich lebensrettenden Potenzial für uns. Etliche pharmazeutische Wirkstoffe basieren auf natürlichen Giften, insbesondere auf peptidischen Toxinen, welche jedes Jahr hunderttausende Menschenleben retten. Im gleichen Moment sind einige dieser Toxine verantwortlich für hunderttausende Todesfälle und fatale gesundheitliche Einschränkungen.
Durch die konstante Verbesserung bio-analytischer und speziell massenspektrometrischer Methoden sind wir auf dem Weg zu einem „kompletten Bild“ an Informationen bezüglich Identität, Konzentration und Konformation bzw. räumliche Interaktionstelle von Naturstoffen oder Toxinen in verschiedenen biologischen Systemen. Diese neuen technologischen Möglichkeiten werden uns zwangsläufig zu einem umfassenden Verständnis von Toxinen und anderen Naturstoffen und ihrer Interaktion mit anderen biologisch relevanten Molekülen führen. Hierdurch werden wir das enorme Reservoir an solchen Verbindungen besser aufdecken und ihr Potential als Modellverbindungen, diagnostische Werkzeuge und schlussendlich als neue therapeutische Wirkstoffe nutzen.
In dieser Arbeit sind die Ergebnisse bezüglich der Suche, Strukturaufklärung, der biochemischen und pharmazeutischen Untersuchung zweier verschiedener Toxin-Klassen, unter Zuhilfenahme neuer massenspektrometrischer Analysemethoden, gezeigt und diskutiert.
Das erste Beispiel handelt von dem nicht-ribosomal synthetisierten Peptid-Polyketid-Hybrid Albicidin, welches von dem Zuckerrohr-Pathogen Xanthomonas albilineans synthetisiert wird. Durch die Sequenzierung des Genoms von Xanthomonas albilineans war es uns möglich die einzigartige biologische Nische und den besonderen Genotyp, innerhalb der Xanthomonaden, welcher etliche ungewöhnliche Biosynthese Gencluster enthält, zu beschreiben. Einer dieser Gencluster ist für die Biosynthese des Phytotoxins Albicidin, einem starken DNA-Gyrase Inhibitor, welcher auch hochwirksam gegen Gram-positive und Gram-negative Bakterien ist, verantwortlich. Die Strukturaufklärung von Albicidin deckte ein bisher unbekanntes chemisches Gerüst auf, welche bis auf Cyano-Alanin und Zimtsäure ausschließlich aus para-Aminobenzoesäure-Derivaten besteht. Die heterologe Produktion und darauf aufbauende in vitro Experimente verschiedener Module der Nicht-Ribosomalen-Peptidsynthetase gab uns eine detaillierte Einsicht in die Biosynthesemaschinerie, was uns half ein umfassendes Biosynthesemodell für Albicidin aufzustellen.
Durch non-targeted Tandem-Massenspektrometrie (MS/MS) konnten wir mehrere Albicidin Derivate, wie z.B. N-terminal carbamoyliertes Albicidin identifizieren. Die Struktur von Carbamoyl-Albicidin konnte durch hochauflösende MS/MS aufgeklärt und die Biosynthese ebenfalls durch heterologe Produktion der Carbamoyltransferase und in vitro Assays sowie einer Geninaktivierungsmutante verifiziert werden.
Die Totalsynthese von Albicidin sowie Carbamoyl-Albicidin ermöglichte uns schließlich die detaillierte Testierung der biologischen Aktivitäten durch in vitro Gyrase-inhibierung und Testierung der antibakteriellen Eigenschaften. Carbamoyl-Albicidin zeigte hierbei stärkere inhibitorische Effekte gegenüber Gyrase als Albicidin, was dafür spricht, dass die Carbamoylierung ein Teil der biosynthetischen Reifung von Albicidin ist.
Der zweite Teil dieser Arbeit handelt von der Einführung und ersten Anwendung von Top-down Massenspektrometrie für die Analyse von Schlangengiften. In einer Machbarkeitsstudie haben wir Gift der Königskobra, Ophiophagus hannah, mit Hilfe einer Top-down Methode analysiert und etliche Toxine verschiedener Proteinfamilien nachgewiesen. Unter diesen Toxinen waren hauptsächlich Drei-Finger-Toxine, ein Kunitz-artiger Proteaseinhibitor, Ohanin, eine bisher unbekannte A2-Phospholipase sowie ein Cystein-reiches-Sekretionsprotein (Ophanin). Eine komplementäre bottom-up Analyse führte zu einem kompletten, Lokus-aufgelösten Venom-Repertoire der Königskobra - der größten bekannten Giftschlange.
Als erstes Beispiel für eine Otter-Spezies haben wir das Venom der Anatolischen Wiesenotter, Vipera anatolica, durch eine Kombination aus Top-down und Bottom-up Venomics analysiert. Durch diese Analyse konnten wir etliche Metalloproteasen, Cystein-reiche-Sekretionsproteine, einen Metalloproteaseinhibitor, A2-Phospholipasen, Disintegrine, eine Serinprotease, Typ-C Lektine sowie einen Kunitz-artigen Proteaseinhibitor identifizieren. Des Weiteren konnten wir etliche Isoformen der oben beschriebenen sowie unbekannten Toxine detektieren, was für eine große, bisher nicht beschriebene Komplexität von Schlangengiften spricht, die mit herkömmlichen bottom-up Techniken unentdeckt geblieben wäre. Schlussendlich konnten wir hierdurch eine bioaktive Komponente des Venoms als ein heterodimeres Disintegrin identifizieren, welches vielversprechende Aktivität gegen Krebszellen zeigte.Natural toxins play an important role for human health. Their biochemical properties result in a “ying and yang” of deadly and lifesaving potential. Several drugs are based on natural and especially peptidic toxins, saving hundred thousands of human lives every year. At the same time, some of these toxins are responsible for hundreds of thousands of deaths and permanent health issues.
With the constant improvement of bioanalytical and especially mass spectrometric techniques, progress has been made towards the accomplishment of “complete pictures” of identities, concentration and spatial location of biomolecules, and in particular toxins, in different biological systems. These technological improvements will enable us to gain a more complete understanding of toxins and all other natural products and their biological interactions. Furthermore these improvements will enable us to better uncover and utilize their enormous potential as biomedical models, diagnostic tools and ultimately novel therapeutics.
In this work, the discovery, structural elucidation, biochemical investigation and finally assessment as potential drugs of two classes of natural toxins are shown and discussed.
The first part is dedicated to the non-ribosomally synthesized peptide-polyketide-hybrid albicidin, which is produced by a sugar cane pathogenic bacterium, Xanthomonas albilineans. By means of the sequencing of the genome of Xanthomonas albilineans we were able to describe its unique biological niche and special genotype among other xanthomonads, containing a number of unusual biosynthetic gene clusters. One of these gene clusters is responsible for the biosynthesis of the phytotoxin albicidin, a potent DNA gyrase inhibitor, which is also highly active against Gram positive and Gram negative bacteria. The structure elucidation revealed the hitherto unknown structure of albicidin, revealing a unique polyaromatic oligopeptide mainly composed of p-amino benzoic acids and cyano-alanine. Heterologous production and in-vitro studies of several domains of the non-ribosomal peptide synthetase rendered detailed insights into the biosynthetic machinery, revealing a comprehensive biosynthetic model for albicidin. Besides the main metabolite, we found several derivatives by non-targeted tandem mass spectrometry (MS/MS) experiments, for example an N-terminally carbamoylated version of albicidin. Carbamoyl-albicidin was characterized by high resolution MS/MS from cultures of X. albilineans and the putative biosynthesis confirmed by gene inactivation of the carbamoyltransferase, heterologous production of the carbamoyltransferase and in vitro reconstitution of the carbamoylation reaction. The chemical synthesis of both albicidin and carbamoyl-albicidin finally enabled us to assess their bioactivities by means of in vitro gyrase inhibition and antibacterial assays. Carbamoyl-albicidin showed stronger inhibitory effects against the gyrase compared to albicidin, which identifies the carbamoylation as an important biosynthetic step of albicidin maturation.
In the second part of this work, the first application of top-down mass spectrometry in snake venomics is shown and discussed. In a case study with venom from king cobra, the application of top-down mass spectrometry enabled us to identify several proteins from different toxin families. Among those toxins are eleven three-finger toxins (7-7.9 kDa), a Kunitz-type inhibitor (6.3 kDa), ohanin (11.9 kDa), a novel phospholipase A2 molecule (13.8 kDa) and a cysteine-rich secretory protein (CRISP) ophanin (25 kDa). Complementary bottom-up MS/MS analyses contributed to complete a locus-resolved venom phenotypic map for Ophiophagus hannah, the world's largest venomous snake.
As a first example for a viperid species we characterized the venom proteome Vipera anatolica, the Anatolian Meadow Viper by a combination of top-down and bottom-up venomics. From this analysis we identified snake venom metalloproteases, cysteine-rich secretory protein isoforms, a metalloprotease inhibitor, several type A2 phospholipases, disintegrins, a snake venom serine protease, a C-type lectin and a Kunitz-type protease inhibitor. Furthermore, we detected several isoforms of above mentioned proteins as well as previously unknown proteins, indicating an extensive complexity of the venom which would have remained undetected with conventional venomic approaches. With these results we could identify one of the bioactive fractions of the venom to be a hetero-dimeric disintegrin, which showed remarkable activity against cancer cells
Synthese der Bacteriocine Amylocyclicin A und Plantazolicin in Bacillus amyloliquefaciens FZB42
Bacillus amyloliquefaciens FZB42 ist ein grampositives Bodenbakterium. Es kann in der Rhizosphäre das Wachstum von Pflanzen fördern und durch die Produktion von Sekundärmetaboliten phytopathogene Organismen hemmen. Aus der Genomanalyse und den dazugehörigen Arbeiten war bekannt, dass Bacillus amyloliquefaciens FZB42 nicht-ribosomal je drei antimikrobielle Polyketide und Lipopeptide herstellt, sowie zwei Siderophore und das Dipeptid Bacilysin. Für Bacillus typische Lantibiotika oder große Bacteriocine wurden nicht gefunden. In dieser Arbeit wird erstmalig gezeigt, dass Bacillus amyloliquefaciens FZB42 auf ribosomale Weise antibakterielle Peptide herstellt. Zwei bisher unbekannte Bacteriocine, Amylocyclicin A und Plantazolicin, und deren dazugehörigen Gencluster konnten identifiziert und charakterisiert werden. Amylocyclicin A ist ein unmodifiziertes Peptid, dessen N- und C-Terminus kovalent verbunden sind. Es wurde der Gruppe I der zirkulären Bacteriocine zugeordnet, dessen Mitglieder sich durch schwache Homologie untereinander, aber durch wahrscheinlich ähnliche 3D-Strukturen auszeichnen. Die Masse beträgt 6381 Da und die Substanz ist stark aktiv gegen grampositive Bakterien. Das Biosynthesecluster umfasst sechs Gene für die Synthese, den Export, die Zyklisierung und die Immunität. Plantazolicin ist ein hydrophobes, stark modifiziertes Peptid aus der TOMM-Gruppe, einer Gruppe aus Microcin B17-ähnlichen Peptiden, die nach neueren Erkenntnissen verbreiteter ist, als bisher bekannt. Plantazolicin ist schwach aktiv gegen grampositive Bakterien und besitzt die Masse 1335 Da. Das Biosynthesecluster umfasst zwölf Gene, mit allen nötigen Genen für Synthese, Modifikation, Regulation, Immunität und Export.Bacillus amyloliquefaciens FZB42 is a Gram-positive, plant-associated bacterium, which stimulates plant growth and produces secondary metabolites that suppress soil-borne plant pathogens. Five gene clusters direct the non-ribosomal synthesis of the cyclic lipopeptides surfactin, bacillomycin, fengycin, an unknown peptide and the iron-siderophore bacillibactin. Three gene clusters direct the non-ribosomal synthesis of the antibacterial acting polyketides macrolactin, bacillaene and difficidin; in addition to the non-ribosomal synthesis of the antibacterial dipeptide bacilysin. Genes involved in ribosome-dependent synthesis of lantibiotics and other peptides are scarce. Only two incomplete gene clusters directing immunity against mersacidin and subtilin were found. In this work two ribosomally synthesized antibacterial peptides, amylocyclicin A and plantazolicin, and their corresponding gene clusters were identified. Amylocyclicin A is a circular peptide with a mass of 6381 Da and strong activity against Gram-positive bacteria. Six genes are responsible for the synthesis, maturation, export and immunity of this peptide belonging to group I of circular bacteriocins. Plantazolicin is a strongly modified hydrophobic peptide bearing a molecular mass of 1,335 Da and displaying antibacterial activity toward closely related Gram-positive bacteria. Essential modification contains the incorporation of azole heterocycles, which derive from Cys, Ser, and Thr residues of the precursor peptide and addition of two methyl groups. Twelve genes are responsible for synthesis, modification, export and immunity of this peptide belonging to the TOMM group of thiazol/oxazol modified microcins
MAP-Kinase-abhängige Opioid-Rezeptor-Expression in sensorischen Neuronen und PC2-abhängige Opioidpeptid Procezierung und Freisetzung von Immunzellen
Opioide sind für ihre zentrale analgetische Wirkung bekannt. Jedoch immer mehr Hinweise zeigen, dass Opioide auf peripheren sensorischen Neuronen analgetische Wirkung nach der Aktivierung ihrer Rezeptoren entlocken.Während der lokalen entzündlichen Schmerzen, Opioidrezeptoren zunehmend akkumulieren auf peripheren sensorischen Nervenenden und Opioid-Peptide werden durch einwandernde Immunzellen in das subkutane entzündetes Gewebe freigesetzt. Um schließlich eine starke Antinocizeption nach Freisetzung zu herstellen, müssen sie die Perineuralen Barriere von peripheren sensorischen Nerven eindringen. Diese Doktorarbeit untersucht i) ob Nervenwachstumfaktor (NGF) abhängige Signalwege verantwortlich für die erhöhte sensorische Neurone mu-Opioid-Rezeptor-Expression bei entzündlichen Schmerzen sind; ii) ob die Immunzellen endogene Opioid-Peptide Endorphin (END), enkephaline (ENK) und Dynorphin (DYN) ergeben sich aus der korrekten Prozessierung ihrer Vorläufern durch spezifische Prohormonkonvertasen und anschließlich diese Opioide freisetzen können; iii) ob exogen applizierten oder endogen freigesetzte Opioidpeptide die perineuralen Barriere von sensorischen Neuronen durchdringen und anschließend in der Lage sind, Antinozizeption unter normalen und pathologischen Bedingungen zu entlocken.In einem Tiermodell der einseitigen Hinterpfote Entzündungen nach vollständigem Freund-Adjuvans (FCA) Inokulation, i.pl. Einspritzung des vollen Opioid-Agonist Fentanyl führte zu verbesserten antinociceptive Wirkung. Lokale immunoneutralization von NGF in FCA behandelten Tieren mit einem spezifischen Antiserum oder i.pl. NGF Behandlung von naiven Tieren führte zu einer Abnahme oder Zunahme, beziehungsweise der Antinozizeption. Radioaktiv markierte Ligandenbindung mit [3H] DAMGO, Western Blot und Immunhistochemie mit einem mu-Opioid-Rezeptor (MOR) spezifische Antikörper identifiziert signifikanten Anstieg in der Anzahl der MOR-Bindungsstellen, MOR-Protein sowie MOR-immunorreactive (MOR-IR) neuronalen Zellen innerhalb Spinalganglien (DRG). Parallel dazu phosphorylierten p38-MAPK (p-p38-MAPK)-Protein und die Anzahl der p-p38-MAPK-IR Neuronen die MOR exprimieren in DRG sowie die peripher geleiteten axonale Transport von MOR deutlich erhöht.Schließlich wurden NGF-induzierte Effekte in DRG, dem axonalen Transport und der Potenzierung oder verbesserte Wirkung von Opioiden Antinozizeption durch die intrathekale Hemmung der p38-MAPK, aber nicht ERK-1/2 aufgehoben, vermutlich eine entscheidende Rolle für NGF abhängige Signalwege wie p38 MAP-Kinase. Endogene Opioid-Peptide (END, ENK und DYN) sind die Liganden der Opioid-Rezeptoren auf sensorischen Neuronen. Sie sind während stressigen Reizes ausgelöst und binden an die peripheren Opioidrezeptoren um Schmerzen zu hemmen. Doch die subzelluläre Wege der POMC, PENK und PDYN prozessierung zu END, ENK und DYN wurde noch nicht beschrieben in Entzündungszellen. Doppelte Immunfluoreszenz konfokale Mikroskopie zeigt der Kolokalisation Vorläufer Opioiden POMC, PENK oder PDYN mit ihren wichtigsten prozessierungsenzym (PC2) innerhalb subkutaner Gewebe von entzündeter Pfote. Immunhistochemie und Western Blot haben Akkumulation des Vorläufer-Proteinen POMC, PENK oder PDYN innerhalb zirkulierenden und lokalen Immunzellen von PC2-Knockout-Mäusen nachgewiesen. Entschprechend, Radioimmunoassay hat eine Abnahme der Gesamtgehalt und Formyl-Methionin-leucyl-Phenylalanin (fMLP)-induzierte Freisetzung von Opioid-Peptiden Endprodukte END, ENK und DYN von Immunzellen gemessen. Entzündliches Schmerz kann durch intraplantar Injektion oder durch endogene Sekretion Opioid-Peptide von Ratten Leukozyten im entzündeten Pfoten kontrolliert werden. Antinociception benötigt Bindung von Opioidpeptiden an Opioidrezeptoren auf peripheren sensorischen Nervenenden. In Abwesenheit von Entzündung, dringen hydrophilen Opioidpeptide die perineuralen Barriere nicht leicht durch und somit entlocken sie keine Antinozizeption. Daher war das Ziel zu untersuchen, die Bedingungen unter denen endogenen hydrophilen Opioidpeptide (dh Met-Enkephalin und Β-Endorphin) aus Neutrophilen, nozizeptive Schwellen in nichtentzündetete Gewebe bei Ratten erhöhen können. Nach intraplantar Behandlung mit hypertoner Kochsalzlösung wurde die perineuralen Barriere reversibel durchlässig für Stunden und intraplantaren Injektion von Opioid-Peptiden erhöhte mechanische nozizeptive Schwellen. Darüber hinaus führte MIP2-induzierte Rekrutierung von Leukozyten-Produzierte Opioid-Peptide in das subkutane Gewebe, ohne die Integrität des perineuralen Barriere zu beeinflussen, zu Formyl-Met-Leu-Phe (fMLP)-ausgelöst Opioid-Peptid-Freisetzung und die anschließende Schmerz-Hemmung. Zusammen genommen, zusätzlich zu dem Opioid-System in das zentrale Nervensystem zunehmende Beweise vermuten eine Rolle für die Opioid-System in das periphere Nervensystem. Unter der entzündlichen Schmerzen die Anzahl der Opioidrezeptoren auf peripheren sensorischen Nerven ist hochreguliert in einer NGF p38 MAPK-Kinase-Weg. In parallel Opioidpeptid-produzierende Immunzellen migrieren aus dem Blutkreislauf in unmittelbarer Nähe dieser Nerven um Opioidpeptide auf korrekte Verarbeitung aus ihren Vorläufern, um biologisch aktive Endprodukte freizusetzen. In PC2-Knockout-Mäusen dieses Opioid-Verarbeitung wurde behindert, was zu einer Akkumulation von Vorläufer und ein reduzierte Menge an Opioidpeptid Endprodukten führte. Die perineuralen Barriere, die normalerweise, dass Opioid-Peptide Zugang zu peripheren Opioidrezeptoren verhindert, ist unter entzündeten Bedingungen zerstört. Unter nicht-entzündlichen Erkrankungen des Zugangs von Opioid-Peptiden kann durch hypertone Lösungen erleichtert werden. Antinozizeption vermittelt durch endogen freigesetzte Opioidpeptide hat von daher drei wichtige Anforderungen zuerfüllen: 1) eine kritische Anzahl von Opioid-Rezeptoren im peripheren sensorischen Neuronen. 2) Opioidpeptid Expression, korrekte Prozessierung und freisetzung von Leukozyten und 3) die Eröffnung des perineuralen Barriere für Opioidpeptid Zugriff zu Opioid-Rezeptoren auf sensorische Neuronen . Dies kann für intrinsische Mechanismen der Schmerzkontrolle wichtig sein wie sich im Rahmen von Stress-vermittelte Analgesie gezeigt worden hat.Opioids are well known for their central analgesic effects. However, growing evidence shows that opioids also elicit analgesic effects following activation of their receptors on peripheral sensory neurons. During local inflammatory pain opioid receptors increasingly accumulate on peripheral sensory nerve endings and opioid peptides are delivered by immigrating immune cells into subcutaneous inflamed tissue. To finally result in potent antinociception upon their release they have to penetrate the perineural barrier of peripheral sensory nerves. The work of this thesis examined i) whether nerve growth factor (NGF) dependent signaling pathways are responsible for the increased sensory neuron mu-opioid receptor expression during inflammatory pain; ii) whether the immune cell derived endogenous opioid peptides endorphine (END), enkephaline (ENK) and dynorphin (DYN) result from the correct processing of their precursors by specific prohormone convertases and a subsequent release; iii) whether exogenously applied or endogenously released opioid peptides penetrate the perineural barrier of sensory neurons and subsequently are able to elicit antinociception under normal and pathological conditions. In an animal model of unilateral hindpaw inflammation following Freund’s complete adjuvant (FCA) inoculation, i.pl. administration of the full opioid agonist fentanyl led to enhanced antinociceptive efficacy. Local immunoneutralization of NGF in FCA treated animals by a specific antiserum or i.pl. NGF treatment of naive animals resulted in a decrease or increase of antinociception, respectively. Radiolabeled ligand binding with [3H]DAMGO, Western blot and immunohistochemistry using a mu-opioid receptor (MOR) specific antibody identified significant increases in the number of MOR binding sites, MOR protein as well as MOR-immunorreactive (MOR-IR) neuronal cells within dorsal root ganglia (DRG). In parallel, phosphorylated p38-MAPK (p-p38-MAPK) protein and the number of p-p38-MAPK-IR neurons expressing MOR in DRG as well as the peripherally directed axonal transport of MOR significantly increased. Finally, NGF-induced effects occurring in DRG, on axonal transport and on the potentiation or enhanced efficacy of opioid antinociception were abrogated by the intrathecal inhibition of p38- but not ERK-1/2- MAPK suggesting a crucial role for NGF dependent signal pathways such as p38 MAP kinase. Endogenous opioid peptides (END, ENK and DYN) are the ligands of such opioid receptors on sensory neurons. They are released during stressful stimuli and interact with peripheral opioid receptors to inhibit pain. However, the subcellular pathways of POMC, PENK and PDYN processing into END, ENK and DYN have not yet been delineated in inflammatory cells. Double immunofluorescence confocal microscopy showed colocalization of the opioid precursors POMC, PENK or PDYN with their key processing enzyme (PC2) within subcutaneous inflamed paw tissue. Immunohistochemistry and Western blot detected accumulation of the precursor proteins POMC, PENK or PDYN within circulating and resident immunocytes of PC2 knockout mice. Consistently, radioimmunoassay measured significant decreases in the total content and formyl-methionyl-leucyl-phenylalanine (fMLP)-induced release of opioid peptide endproducts END, ENK and DYN from immune cells. Inflammatory pain can be controlled by intraplantar opioid injection or by secretion of endogenous opioid peptides from leukocytes in inflamed rat paws. Antinociception requires binding of opioid peptides to opioid receptors on peripheral sensory nerve terminals. In the absence of inflammation, hydrophilic opioid peptides do not penetrate the perineurial barrier easily and, thus, do not elicit antinociception. Therefore, the aim was to examine the conditions under which endogenous, neutrophil-derived hydrophilic opioid peptides (i.e. Met-Enkephalin and β-endorphin) can raise nociceptive thresholds in noninflamed tissue in rats. Following intraplantar treatment with hypertonic saline, the perineurial barrier was reversibly permeable for hours and intraplantar injection of opioid peptides increased mechanical nociceptive thresholds. In addition, MIP2-induced recruitment of opioid peptide containing leukocytes into subcutaneous tissue without affecting the integrity of the perineural barrier resulted in the establishment of formyl-Met-Leu-Phe (fMLP)-triggered opioid peptide release and subsequent pain inhibition. Taken together, in addition to the opioid system within the central nervous system increasing evidence suggests a role for the opioid system in the peripheral nervous system. Under conditions of inflammatory pain the number of opioid receptors on peripheral sensory nerves is up-regulated in an NGF p38 MAPK kinase pathway. In parallel opioid peptide expressing immune cells migrate from the circulation into direct vicinity of these nerves to release opioid peptides upon correct processing from their precursors to biologically active end products. In PC2 knockout mice this opioid processing was hindered leading to an accumulation of precursors and a lack of opioid peptide endproducts to be released. The perineural barrier which normally prevents opioid peptides from their access to peripheral opioid receptors is destroyed under inflamed conditions. Under non-inflammatory conditions the access of opioid peptides can be facilitated by hypertonic solutions. Antinociception mediated by endogenously released opioid peptides has, therefore, three important requirements: 1) a critical number of opioid receptors at the peripheral sensory neurons; 2) opioid peptide expression, correct processing and release from leukocytes and 3) opening of the perineurial barrier for opioid peptide access to sensory neuron opioid receptors. This may be important for intrinsic mechanisms of pain control as has been shown in the context of stress-induced analgesia
Studies towards the biosynthesis of Omphalotin
Originally discovered in 1987, Omphalotin is an undodecapeptide (which is) synthesized by the basidiomycete Omphalotus olearius. The fungal secondary metabolite shows a strong nematicide and selective activity against the plant parasite Meloidogyne incognita. Structurally, it bears various remarkable characteristics, as for example nine methylations of the α-nitrogen atoms in the peptide backbone. Despite the nematicide peptide and its structure being discovered almost 30 years ago, the biosynthetic mechanism still remains unknown.
In this study, a gene cluster was identified which is potentially responsible for the biosynthesis of Omphalotin. It contains genes encoding for a prolyl oligopeptidase (OphP) and a fusion protein OphMA, the latter consisting of a methyl transferase (OphM) fused to the Omphalotin precursor peptide (OphA). In consequence, a ribosomal biosynthesis pathway involving the necessary and sufficient participation of the enzymes OphMA and OphP could be assumed. To further investigate this hypothesis, genes which are potentially involved in the biosynthetic pathway of Omphalotin were heterologously expressed in different host organisms. Particularly, Pichia pastoris could be identified as the most efficient expression system. Corresponding in vitro studies revealed that OphMA forms a homodimer which capable to self-methylate its C-terminal region consisting of the Omphalotin core sequence. The ability of self-methylation and the homodimeric mode of interaction could be confirmed via protein crystallization experiments. In heterologous co-expression studies of ophMA and ophP in Pichia pastoris, the production of Omphalotin could be observed and proven by detailed mass spectrometric analyses.
Altogether, Omphalotin was proven to be the first ribosomally synthesized and post-translationally modified peptide (RiPP) of fungal origin which could be autonomously produced in a heterologous expression system. Furthermore, it is so far the only known RiPP synthesized as a precursor peptide (OphA) translationally fused to its associated modification enzyme (OphM).Das von dem Basidiomyceten Omphalotus olearius synthetisierte Undodecapeptid Omphalotin zeigt eine starke und selektive, nematizide Aktivität gegen den Pflanzenparasiten Meloidogyne incognita. In der Struktur zeigt das Peptid außergewöhnliche Modifikationen. Dabei sollen hier nur die neun Methylierungen an den α-Stickstoffatomen im Peptidrückgrat genannt werden. Auch wenn das Peptid seit nunmehr 30 Jahren entdeckt und dessen Struktur fast ebenso lange aufgeklärt ist, blieb der Biosynthesemechanismus bislang verborgen.
In dieser Arbeit wurde die Biosynthese des pilzlichen Sekundärmetaboliten untersucht. Im Genom von O. olearius wurde ein Gencluster identifiziert, der potentiell für die Biosynthese von Omphalotin verantwortlich ist. Unter anderem beinhaltet dieser die Gene für eine Prolyl-Oligopeptidase (OphP) und das Fusionsprotein OphMA, bestehend aus einer Methyltransferase (OphM) und dem Vorläuferpeptid von Omphalotin (OphA). Daraus ergibt sich die Hypothese einer ribosomalen Biosynthese, bei der die Enzyme OphMA und OphP notwendig und hinreichend sind, um Omphalotin A zu produzieren.
Die potentiell an der Biosynthese beteiligten Gene wurden heterolog in verschiedenen Wirtsorganismen exprimiert und die Hefe Pichia pastoris als effizientestes Expressionssystem gewählt. In in vitro Studien wurde gezeigt, dass OphMA als Homodimer interagiert und sich am eigenen C Terminus im Bereich der Omphalotin-Sequenz selbst methyliert. Durch die Kristallisation des Enzyms wurde ebenfalls die intermolekulare Selbst-Methylierung, sowie die Interaktion von OphMA als Dimer bestätigt. Im nächsten Schritt wurde ophMA mit ophP koexprimiert. Es wurde nachgewiesen, dass die entsprechenden Pichia-Zellen durch die parallele Synthese beider Enzyme Omphalotin A produzieren, wodurch die ursprüngliche Hypothese bestätigt wird.
Damit ist Omphalotin das erste RiPP aus einem Pilz, das vollständig heterolog produziert werden konnte und weiterhin das einzige RiPP, dessen Vorläuferpeptid (OphA) zusammen mit seinem modifizierenden Enzym (OphM) transkribiert und translatiert wird
Integrative Schlangengiftproteomik: Massenspektrometrie-gestützte Einblicke in die Toxinzusammensetzungen, ihre Quantifizierung und weiterführende Bildgebung
The venoms of snakes are a cocktail of biochemically active substances that can have devastating effects, but also serve as template for novel drugs. Every year snakebites claim hundreds of thousands of victims, either fatally injured or maimed. Therefore, their toxins are in scientific focus and deciphering venoms is a subfield of toxinology, known as ‘venomics’. Over the centuries, venom research has evolved into an interdisciplinary branch spanning topics such as evolution, biochemistry and pharmaceutics, but also structural biology and analytics have become key elements in the modern venomics. Especially, mass spectrometry (MS) has established itself as one of the most important methods due to its continuous improvement and high versatility.
This thesis deals with the composition of snake venoms and the adaptation as well as further development of modern MS techniques for toxin analysis. In the first part, the investigation of the entire Old World vipers (Viperinae) subfamily showed that a large part of their venoms and variations have hardly been described by proteomics in the literature. The toxin families found in viper venoms had a qualitative consensus and can be classified according to their relative abundance. However, compositions at the genus and species level show remarkable quantitative differences and most venoms are either protease or lipase dominated. The lack of metadata and a high variance in used quantification methods were one of the major problems for direct comparisons. Nevertheless, this chapter presents the first holistic database of all studied Old World viper venom proteomes until the beginning of 2021.
In the second part of the thesis, four elapid and viper venoms were characterized for the first time in detail by proteotranscriptomics and served in the three corresponding chapters as models to investigate problems and open questions in the field of venomics, respectively. The venom of the Anatolian meadow viper (Vipera anatolica senliki), like most vipers, contains toxins of high molecular weight that cannot be detected intact until now. Current top-down approaches to measure proteins directly from the crude venom are limited to about 30 kDa and thus only partially useful for viper venoms. With an alternative in-source decay (ISD) using 1,5-diaminonaphthalene (DAN), this limit could be increased to 70 kDa. The applied matrix-assisted laser desorption/ionization (MALDI) made it also possible to de novo annotate and identify various toxin families and proteoforms even in parallel in a sample. The following chapter highlights that current venomics approaches only allow relative quantifications, as different toxins would require dozens of different standards for absolute values according to classical methods, which is practically impossible. However, many snake toxins contain sulfur, mostly in structurally relevant disulfide bridges, which can be quantified in comparison to a single sulfur standard of known concentration. The venom atomization by an inductively coupled plasma (ICP) allows the elemental sulfur content to be accurately measured by MS. With a known sequence, the concentrations of the individual toxins could be exactly determined. In combination with three parallel bottom-up and top-down protocols, this hybrid approach of molecular and elemental MS enabled the absolute quantification and detailed characterization of venom proteomes using of desert cobras (Walterinnesia) as example.
The last chapter deals with a more biological aspect of the venom apparatus, as little is known about the tissue-internal processes of venom production and storage. For this purpose, fixed venom gland tissue of the Egyptian cobra (Naja haje) was scanned using MALDI. The high two-dimensional resolution in tenth micrometer range allows the detection of spatially resolved spectra of toxin-specific m/z masses. This MS imaging (MSI) of in situ toxin mapping by spatial intensity maps across a venom gland showed that the different families and individual toxins are produced only in specific areas and are not ubiquitous in the gland.Die Gifte von Schlangen bestehen aus einem Pool biochemisch aktiver Stoffe, die verheerende Wirkungen entfalten, aber auch als Grundlage neuer Medikamente dienen können. Jedes Jahr fordern Schlangenbisse hunderttausende Opfer, entweder tödlich verletzt oder durch die Folgen der Vergiftung lebenslang eingeschränkt. Daher stehen ihre Toxine im Fokus und die Entschlüsselung der Gifte sind ein Unterbereich der Toxinologie, der als Venomics bezeichnet wird. Über die Jahrhunderte hat sich die Giftforschung in einen interdisziplinären Zweig der Wissenschaft entwickelt, der Themen wie Evolution, Biochemie und Pharmazie umspannt, aber auch die Strukturbiologie und Analytik sind mittlerweile zu Kernelementen geworden. Vor allem aber hat sich die Massenspektrometrie (MS) durch ihre stete Weiterentwicklung und hohe Vielseitigkeit als eine der wichtigsten Methoden etabliert.
Diese Arbeit beschäftigt sich mit den Zusammensetzungen von Schlangengiften sowie der Adaption und Weiterentwicklung von modernen MS Techniken für die Analyse von Toxinen. Im ersten Teil zeigte die Untersuchung der gesamten Unterfamilie der Echten Vipern (Viperinae), dass ein Großteil ihrer Gifte noch kaum proteomisch in der Literatur beschrieben worden ist. Die in Viperngift vorkommenden Toxinfamilien zeigen einen qualitativen Konsens und lassen sich entsprechenden ihrer relativen Häufigkeit einteilen. Dennoch weisen Gattungen und Arten markante Unterschiede in ihren quantitativen Zusammensetzungen auf und so sind die meisten Gifte entweder Protease oder Lipase dominiert. Fehlende Metadaten und eine hohe Varianz an Quantifizierungsmethoden stellten hierbei eines der größten Probleme für direkte Vergleiche dar. Dennoch konnte hier die erste vollständige proteomische Datenbank der Viperinae Gifte bis zum Jahr 2021 beschrieben werden.
Im zweiten Teil der Arbeit wurden die Gifte von vier Elapiden- und Vipernarten erstmals proteotranskriptomisch detailliert charakterisiert und dienten in den drei entsprechenden Kapiteln als Modell um Probleme und offene Fragestellungen im Bereich der Venomics zu untersuchen. Das Gift der Anatolischen Wiesenotter (Vipera anatolica senliki) enthält, wie die meisten Vipern, Toxine mit hohem Molekulargewicht, die nicht intakt detektiert werden können. Aktuelle Top-Down-Verfahren, um Proteine direkt aus dem Gift zu vermessen, sind meistens auf etwa 30 kDa begrenzt und damit nur zum Teil für Viperngifte nützlich. Durch die Verwendung einer alternativen Fragmentierung in der Ionisierungsquelle (engl. in-source decay, ISD) mittels 1,5-Diaminonaphthalin (DAN) konnte diese Grenze auf 70 kDa erhöht werden. Diese Matrix-unterstützte Laser-Desorption/Ionisation (engl. matrix-assisted laser desorption/ionization, MALDI) ermöglichte es auch Toxinfamilien und Proteoformen parallel in einer Probe de novo zu annotieren. Das folgende Kapitel beschäftigt sich mit der Tatsache, dass heutige Venomics-Ansätze nur eine relative Quantifizierung zulassen, da die verschiedenen Toxine nach klassischen Verfahren für absolute Werte auch dutzende verschiedener Standards benötigen würden, was in der Praxis unmöglich umzusetzen ist. Viele Schlangentoxine enthalten jedoch Schwefel, meistens in strukturrelevanten Disulfidbrücken, der im Vergleich zu einem Schwefelstandard bekannter Konzentration quantifiziert wenden kann. Die Atomisierung des Giftes durch ein induktiv gekoppeltes Plasma (engl. inductively coupled plasma, ICP) erlaubt es massenspektrometrisch den elementaren Schwefelanteil genau zu vermessen. Bei bekannter Sequenz ließen sich so die Konzentrationen der einzelnen Toxine exakt bestimmen. In Kombination mit drei parallelen Bottom-Up- und Top-Down-Protokollen ermöglichte dieser hybride Ansatz von molekularer und elementarer MS die absolute Quantifizierung und detaillierte Charakterisierung vom Giftproteomen, gezeigt am Beispiel der Wüstenkobras (Walterinnesia).
Im letzten Kapitel geht es um grundlegende biologische Aspekte des Giftapparates, da noch wenig über die gewebsinternen Abläufe der Giftproduktion und -lagerung bekannt ist. Hierfür wurde fixiertes Giftdrüsengewebe der Uräusschlange (Naja haje) mittels MALDI abgerastert. Die hohe zweidimensionale Auflösung im Mikrometerbereich erlaubt es ortsaufgelöste Spektren der toxinspezifischen m/z-Massen zu detektieren. Durch dieses MS-basierende Bildgebungsverfahren (engl. MS imaging, MSI) einer in situ Kartierung der räumlichen Ionenintensitäten konnte gezeigt werden, dass die unterschiedlichen Toxinfamilien und auch einzelne Toxine jeweils nur in bestimmten Bereichen der Drüse produziert werden und nicht ubiquitär auftreten
Characterization of the Labyrinthopeptin-Synthetase LabKC in vitro
Der erstmals 1988 beschriebene Actinomyceten-Stamm Actinomadura namibiensis produziert in seinem Kulturüberstand die drei Sekundärmetabolite LabA1, A2 und A3. Dabei handelt es sich um ribosomal synthetisierte Peptide, welche aufgrund ihrer posttranslational eingeführten Lanthionin-Verbrückungen in die Gruppe der Lantibiotika eingeordnet wurden. Die Aufklärung der Röntgenkristallstruktur von Labyrinthopeptin A2 erbrachte weitere detaillierte Einblicke in die bis dato noch relativ unbekannte Klasse III Lantibiotika. Eine zentrale Erkenntnis war vor allem die Entdeckung eines neuartigen Labionin-Motives. Mit der Identifizierung des Genclusters der Labyrinthopeptine wurde zudem der Grundstein für die weitere Untersuchung der Biosynthese gelegt. Neben den Sequenzen für die Vorläuferpeptide von LabA1 und LabA2 codieren weiterhin zwei Gene für den Export der Peptide, sowie ein Gen für ein Enzym LabKC, welches für die Einführung der Labionin-Verbrückungen verantwortlich ist. Durch Vergleich mit anderen bekannten Enzymen konnten in der Sequenz von LabKC eine N-terminale Lyase- und eine zentrale Ser/Thr-Kinase-Domäne sowie eine C-terminale Domäne mit Ähnlichkeit zu Lantibiotika-Zyklasen identifiziert werden. Der vorgeschlagene Mechanismus für die Labionin-Bildung sieht zunächst die Phosphorylierung von zwei Serin-Resten im LabA2 Präpropeptid vor, gefolgt von der anschließenden Zyklisierung über ein C-terminales Cystein durch eine doppelte Michael-Addition. Ausgehend von diesen Vorkenntnissen war das Hauptziel der vorliegenden Arbeit zunächst die Etablierung eines in vitro Enzym-Assays für die Untersuchung der Labyrinthopeptin-Biosynthese. Damit sollte eine detaillierte Charakterisierung der Funktionsweise des LabA2-prozessierenden Enzyms LabKC ermöglicht werden. Hierzu wurde dieses zunächst erfolgreich kloniert, in E. coli exprimiert und gereinigt. Die Bereitstellung geeigneter Peptidsubstrate für die enzymatische Umsetzung erfolgte mittels chemischer Peptidsynthese an fester Phase. Nach Optimierung der Assay-Bedingungen sowie unter Verwendung des Nukleotids GTP als Co-Substrat für die Phosphorylierungsreaktion konnte schließlich die komplette Dehydratisierung und Zyklisierung eines synthetischen LabA2-Präpropeptids erreicht werden. Für die Analytik der enzymatischen Umsetzungen wurden massenspektrometrische Methoden angewandt. Nach der Optimierung und Charakterisierung wichtiger Parameter für die in vitro Umsetzung mit LabKC stand die Untersuchung des katalytischen Mechanismus im Mittelpunkt der weiteren Arbeiten. Unter Anwendung von Mutagenese-Methoden konnten katalytisch wichtige Aminosäureseitenketten in der Lyase- und Ser/Thr-Kinase Domäne von LabKC identifiziert und ein entsprechender Mechanismus postuliert werden. Der Reaktionsablauf der Labyrinthopeptin-Biosynthese wurde zudem durch Umsetzungen synthetischer Labyrinthopeptin-Derivate mit LabKC untersucht. Ein weiteres Ziel war die detaillierte Untersuchung der Rolle des Leaderpeptids für die Biosynthese der Labyrinthopeptine. Im Rahmen dieser Arbeit konnte durch Testierung zahlreicher LabA2-Leaderpeptid-Varianten die absolute Notwendigkeit dieser Aminosäuresequenz für die erfolgreiche Prozessierung durch LabKC gezeigt werden. Weiterhin wurde ein Erkennungsmotiv identifiziert und charakterisiert. Im Fokus zukünftiger Arbeiten steht die Aufklärung des Mechanismus für die Labionin-Zyklisierung sowie die Strukturaufklärung von LabKC oder homologen Enzymen mittels Röntgenkristallographie.The actinomycete Actinomadura namibiensis, which was firstly described in 1988, produces in its culture supernatant the three secondary metabolites LabA1, A2 and A3. These ribosomally synthesized peptides were classified as lantibiotics due to their posttranslationally introduced lanthionine bridges. Furthermore the successful elucidation of the crystal structure of Labyrinthopeptin A2 gave new insights in the so far unknown class III lantibiotics including the discovery of the novel Labionin motif. With the overall identification and sequencing of the Labyrinthopeptin gene cluster, further experiments yielded in the elucidation of the biosynthesis. The gene cluster consists of two structural genes coding for the precursor peptides of Labyrinthopeptin A1 andA2, two genes with sequence similarity to ATP dependent ABC transporters for the peptide export (labT1 and labT2), and one gene (labKC) coding for a putative modifying enzyme involved in the formation of the Labionin bridges. An N-terminal lyase, a central Ser/Thr Kinase domain as well as a C-terminal Domain with similarity to lantibiotics cyclases could be identified by sequence alignment with homologue enzymes. The proposed mechanism for formation of the labionin motif starts with the phosphorylation of two serine residues in the LabA2 prepropeptide, followed by a cyclization reaction including a C-terminal Cysteine residue by a twofold Michael-type addition. With this knowledge the first goal of the thesis was the establishment of an in vitro enzyme assay for the elucidation of the Labyrinthopeptin biosynthesis and the detailed enzymatic characterization of the LabKC enzyme. Therefore the coding sequence of LabKC was cloned in E. coli, expressed and further purified. The generation of suitable peptide substrates was accomplished by solid phase chemical synthesis. After further optimization of the assay conditions including GTP as co-substrate for the phosphorylation reaction, the complete dehydration and cyclisation of the LabA2 prepropeptide could be observed, by LC-ESI-MS methods. After optimization and characterization of the in vitro conditions, further work was focused on the evaluation of the catalytic mechanism. Usage of common mutagenesis techniques, succeeded in the identification of important catalytic amino acid side chains in the Lyase and Ser/Thr-Kinase domain. With these results a catalytic mechanism for LabKC was postulated. The catalytic reaction of the Labyrinthopeptin biosynthesis was further investigated by usage of synthetic Labyrinthopeptin derivatives. The investigation of the leader peptide in respect to the biosynthesis mechanism was a further goal in this work. By usage of numerous leader peptide variants, it could be shown that this sequence is obsolete for the successful processing by LabKC. Furthermore, a recognition motif in the leader peptide could be identified and further characterized. Future work will focus on the elucidation of the Labionin-cyclization reaction as well on the structure elucidation of LabKC or its homologues by X-ray crystallography
Synthesis of derivatives of the antiviral peptide antibiotic Feglymycin and studies towards the mode of action
Das aus Streptomyces DSM 11171 isolierte lineare 13mer-Peptid Feglymycin reiht sich aufgrund des hohen Anteils an Hydroxyphenylglycinen in der Aminosäuresequenz in die Klasse der Peptidantibiotika mit Vertretern wie Vancomycin oder Ramoplanin A2 ein. Es besitzt außerdem eine nahezu alternierende Anordnung an L- und D-konfigurierten Aminosäuren, sowie eine helikale Sekundärstruktur im Kristall. Feglymycin ist jedoch nicht nur strukturell äußerst interessant, es besitzt auch eine vielseitige biologische Aktivität, denn das Peptid inhibiert sowohl die Replikation von HI-Viren im Zellkulturtest, als auch das Wachstum von S. aureus-Stämmen in vitro. Neueste Experimente zeigten, dass Feglymycin auch die zytoplasmatischen Enzyme MurA und MurC der bakteriellen Zellwandbiosynthese inhibiert. Die Realisierung der ersten Totalsynthese für Feglymycin ebnete schließlich den Weg zur genaueren Untersuchung dieses Peptidwirkstoffes. Die vorliegende Arbeit beschreibt die Synthese und Charakterisierung von Feglymycin-Derivaten mit dem Ziel einer genaueren Untersuchung der Wirkmechanismen sowie der Identifizierung einer pharmakophoren Region des Peptids. Die Darstellung des Feglymycin-Enantiomers zeigte, dass die biologischen Aktivitäten unabhängig von der absoluten Konfiguration des Moleküls sind. Dies weißt darauf hin, dass Feglymycin wahrscheinlich weder mit einem spezifischen Rezeptor interagiert, noch im aktiven Zentrum eines Enzyms bindet. Ein Alanin-Scan als eine erste Studie zur Struktur-Aktivitäts-Wirkungsbeziehung ergab, dass lediglich die Deletion der aciden Aminosäureseitenkette der C-terminalen Asparaginsäure zu einem leichten Abfall der antiretroviralen Aktivität sowie der Inhibitionswirkung auf die Enzyme MurA und MurC führt. Auch in Bezug auf die antibakterielle Aktivität gegen S. aureus in vitro konnten für die Wachstumshemmung essenzielle Aminosäurebausteine in der Sequenz von Feglymycin identifiziert werden. Erstaunlich war dabei, dass der jeweilige Austausch der 3,5-Dihydroxy-D-phenylglycine gegen D-Alanin relativ gut toleriert wurde. Des Weiteren bestätigten Testierungen von Peptidfragmenten frühere Annahmen, nach denen sich eine mögliche pharmakophore Region des Peptids im N-terminalen Teil des Naturstoffes befindet. Weiterhin scheint die Möglichkeit zur Ausbildung von mehreren Wasserstoffbrückenbindungen durch die C-terminale Aminosäure, sowohl im Naturstoff, als auch bei entsprechenden Peptidfragmenten, wichtig für die biologischen Aktivitäten zu sein. Diese Erkenntnisse stellen wertvolle Bausteine auf dem Weg zur Generierung eines Peptidomimetikums für Feglymycin mit verringerter struktureller Komplexität und verbesserten pharmakologischen Eigenschaften dar. Mit einem fluorgelabelten Peptidderivat konnten erste 19F-Festkörper-NMR-Experimente durchgeführt werden, welche Hinweise auf eine mögliche Wechselwirkung von Feglymycin mit Lipidmembranen liefern sollen. Durch die Annelierung eines weiteren Aromaten an die Aminosäureseitenkette von Phenylalanin wurde ein fluoreszierendes Feglymycin-Analogon erhalten, welches zur Lokalisierung des Peptids direkt an seinem Wirkort verwendet werden könnte. Mit der Synthese der beschriebenen Feglymycin-Derivate wurde gezeigt, dass die hochkonvergente Totalsynthesestrategie für den Naturstoff durch Kondensation von Peptidfragmenten in Lösung für den Aufbau von kleinen, zielgerichteten Substanzbibliotheken ausgezeichnet geeignet ist.Due to its high amount of hydroxyphenylglycines the 13mer peptide Feglymycin can be assigned to the group of peptide antibiotics together with Vancomycin and Ramoplanin A2. In addition, it contains a nearly alternating sequence of L- and D-amino acids and exhibits a helical crystal structure. But Feglymycin is not only structurally interesting, it shows also miscellaneous biological activities like inhibition of the replication of HIV in cell culture and growth inhibition of S. aureus strains in vitro. Latest experiments also revealed that Feglymycin inhibits the cytoplasmatic enzymes MurA and MurC of the bacterial cell wall biosynthesis. Finally, the realization of the totals synthesis for Feglymycin paved the way for a detailed investigation of this peptide. This work describes the synthesis and characterization of Feglymycin derivatives to further investigate the mode of action of the biological activities as well as to identify a pharmacophoric region of this peptide. The synthesis of the Feglymycin enantiomer showed that the biological activities are independent of the absolute configuration of the molecule. This indicates that Feglymycin probably neither interact with a specific receptor, nor binds in the active center of an enzyme. An alanine scan as a first structure-activity-relationship study showed that only the deletion of the acidic side chain of the C-terminal aspartic acid causes a weak decrease of the antiretrovirale activity and the inhibition effect on the enzymes MurA and MurC. Also regarding the antibacterial activity against S. aureus in vitro essential parts of the amino acid sequence could be indentified. Testing of peptide fragments confirmed the assumption that a possible pharmacophoric region is located in the N-terminal part of this natural product. These results are important steps on the way towards the generation of a peptidomimetic for Feglymycin with reduced structural complexity and improved pharmacological properties. Using a Fluor-labeled peptide derivative first 19F-solid-state-NMR measurements were performed to investigate a potential interaction of Feglymycin with lipide membranes. In addition, a fluorescent peptide analogue was synthesized by annealing a second aromatic ring to the side chain of phenylalanine which could be used to locate the peptide directly at its target side. With the generation of the described Feglymycin derivatives it was shown that the highly convergent strategy for the total synthesis of the natural product through condensation of peptide fragments in solution is exellently suitable to create small, target-oriented peptide libraries
First Total Synthesis of the Antiviral Antibiotic Feglymycin" and "Studies for the Synthesis of Complestatin-Analogues on Solid Support
Im Rahmen dieser Arbeit sollte zum einen ein Syntheseprotokoll für die Darstellung von Complestatinanaloga an fester Phase entwickelt werden. Zum anderen wurde eine erste Totalsynthese des Naturstoffs Feglymycin in Lösung anvisiert, welcher zusätzlich auf eine antiretrovirale bzw. antibakterielle Wirkung untersucht werden sollte. Die teilweise essentiellen basischen Reaktionsbedingungen im Zuge der Festphasensynthese von Complestatinanaloga führten zur Epimerisierung von empfindlichen Aminosäuren während Kupplungsreaktionen und hierdurch zur Ausbildung diastereomerer Produktgemische. Weitere Nebenprodukte entstanden während der Abspaltung temporärer Schutzgruppen, wodurch eine chromatographische Aufreinigung der linearen Zielstrukturen nicht möglich war. Die ausgearbeitete Strategie zur Synthese von Complestatinanaloga konnte hierdurch nicht erfolgreich abgeschlossen werden. Das antivirale Peptidantibiotikum Feglymycin konnte mittels Fragmentkondensationen in Lösung erstmals synthetisiert werden, wodurch ein Strukturbeweis des Naturstoffs gelang. Der konsequente Verzicht auf zu basische Reaktionsbedingungen im Laufe der gesamten Synthese ermöglichte es, Epimerisierungen von empfindlichen Arylglycinen zu unterdrücken. Hierzu trug zudem maßgeblich der Einsatz des Kupplungsreagenzes DEPBT bei. CD-spektroskopische Messungen der Verbindung weisen auf die Ausbildung von beta-Faltblattstrukturen bzw. beta-Helices in Lösung hin. Entgegen ursprünglicher Angaben zeigte Feglymycin in durchgeführten Reihenverdünnungstests beachtliche antibakterielle Aktivität gegenüber Staphylococcus aureus Stämmen. Eine Mögliche Ursache hiefür könnte die erstmals im Ionenkanalexperiment nachgewiesene Membranaktivität des Peptids sein. Der Naturstoff wurde zusammen mit synthetischen Intermediaten und dargestellten enantiomeren Verbindungen im Zellkulturtest auf eine antiretrovirale Wirkung hin untersucht. Feglymycin und die enantiomere Verbindung, sowie einige synthetische Vorläufer besitzen hiernach beachtliche anti-HIV-Aktivität. Die Ergebnisse dieser Studien lassen zudem vermuten, dass eine pharmakophore Region von Feglymycin vorzugsweise in der N-terminalen Region lokalisiert ist, unabhängig von der absoluten Konfiguration der einzelnen Aminosäuren.Aim of this work was the establishment of a protocol for the synthesis of complestatin analogues on solid support and for a first total synthesis of the natural product feglymycin in solution. Additionally antibacterial and antiretroviral activities of feglymycin should be investigated. During the solid phase peptide synthesis of linear complestatin analogues the strongly basic reaction conditions, which were partly essential for acceptable conversions, resulted in epimerisation of sensitive amino acids during coupling sequences. Furthermore, partial incomplete removal of temporary protecting groups and side reactions during Alloc-deprotection led to the formation of undesired by-products which could, together with diasteriomeric impurities, not be purified by chromatography. The elaborated synthetic strategy therefore could not be completed successfully. The antiviral peptide antibiotic feglymycin was synthesized successfully by fragment condensation in solution, leading to the evidence of structure of this compound. The application of weakly basic conditions throughout the whole synthesis and the use of DEPBT as coupling reagent suppressed epimerisation of numerous sensitive aryl glycines. CD-spectroscopic analysis of feglymycin indicates the formation of beta-sheet or beta-helical structures. Conducted antibacterial testings have successfully shown that the activity of feglymycin against Staphylococcus aureus is exceptional, contrary to previous statements. A possible reason for that could be the detection of a membrane spanning activity of the natural product during conducted ion channel experiments. For investigation of antiretroviral activity a selection of synthetic intermediates and enantiomeric compounds, as well as the natural product, were chosen for anti-HIV testing in cell culture. Feglymycin, its enantiomer and some synthetic fragments exhibit strong anti-HIV activity. After comparison of all results it may be assumed, that the potential pharmacophore of feglymycin is located in the N-terminal region, independent of absolute configuration of the substrates
Construction of a biosynthetic mutant of Amycolatopsis balhimycina via module insertion in the non-ribosomal peptide synthetase BpsB using a combinatorial biosynthetic approach and its mass spectrometric characterization
Herstellung einer Biosynthesemutante von Amycolatopsis balhimycina durch Modulinsertion in die nicht-ribosomale Peptidsynthetase BpsB mittels eines kombinatorisch-biosynthetischen Ansatzes und deren massenspektro-metrische Charakterisierung Die vorliegende Arbeit beschäftigt sich mit der kombinatorischen Biosynthese bei nicht-ribosomalen Peptidsynthetasen (NRPS), die aufgrund der Komplexität der Strukturen von nicht-ribosomal synthetisierten Peptiden eine ernstzunehmende Alternative zur chemischen Synthese darstellt, um Strukturderivate zu erzeugen. Ziel dieser Arbeit war die Komplettierung der bisher bei NRPS literaturbekannten Modul-manipulationen „Modulaustausch“ und „Moduldeletion“ durch das noch ausstehende Experiment der „Modulinsertion“. Als Modellverbindung diente das von A. balhimycina synthetisierte Vancomycin-Typ Glykopeptidantibiotikum Balhimycin, welches durch sein stark modifiziertes Peptidgrundgerüst (Chlorierung, oxidative Seitenkettenzyklisierung, N-Methylierung und Glykosylierung) ein sehr komplexes Beispiel einer nicht-ribosomalen Peptidsynthese darstellt. In der vorliegenden Arbeit ist es gelungen, ein zusätzliches Modul in die NRPS BpsB von A. balhimycina über eine double crossover-Strategie einzuführen, was sowohl auf genetischer Ebene mittels PCR und Southern Blot-Hybridisierung als auch auf Proteinebene mittels Disk-SDS-PAGE und anschließende massenspektrometrische Identifizierung der Gelbande nachgewiesen werden konnte. Daran schloss sich eine eingehende Charakterisierung des Produktionsprofils dieser kombinatorischen Biosynthese-Mutante mittels HPLC-ESI-MS und -MS/MS an. Diese Analyse erbrachte den Beweis, dass das eingeführte Modul auch enzymatische Aktivität besitzt und somit das Peptidrückgrat des Balhimycins um eine Aminosäure erweitert. Neben dem Beweis der Machbarkeit einer Modulinsertion bei NRPS gewährte dieses kombinatorische Biosynthese-Experiment auch Einblicke in die Flexibilität des gesamten Biosyntheseapparates. Beispielsweise war die Chlorierung der Produkte durch die Halogenase BhaA nicht beeinträchtigt, während N-Methylierung und Glykosylierung in allen 17 nachgewiesenen Biosyntheseprodukten fehlten. Bezüglich der oxidativen Seitenkettenzyklisierungen brachte die erste P450-abhängige Monooxygenase OxyB noch eine gewisse Toleranz für das neue Substrat bzw. die veränderte NRPS auf, während OxyA und OxyC keine nachweisbare Aktivität mehr zeigten.The topic of the thesis is combinatorial biosynthesis with non-ribosomal peptide synthetases (NRPSs).Whereas module exchanges and module deletions are manipulations already known in literature, no example of a successful module insertion in a NRPS has been published. The model system for such an experiment was Amycolatopsis balhimycina producing the vancomycin-type glycopeptide antibiotic balhimycin which is characterized by diverse modifications of the peptidic backbone (chlorination, oxidative side-chain cyclization, N-methylation, glycosylation). The insertion of an additional module in the NRPS BpsB of Amycolatopsis balhimycina was obtained via a double crossover strategy and could be verified on the genetic and proteinogenic level. The subsequent mass spectrometric characterization of the production profile of this mutant showed the expected amino acid insertion in the peptidic backbone of balhimycin. This was the proof that the new module bears enzymatic activity
Synthesis of the A´-ring of Labyrinthopeptin A2 and a reference compound for the verification of the unique amino acid labionin
Die vorliegende Arbeit beschäftigt sich mit zwei Problemstellungen. Zum einen beschreibt die Arbeit die Synthese des A´-Rings von Labyrinthopeptin A2. Dieses pentazyklische 18-mer Lantibiotikum wurde 1988 im Rahmen eines von der Firma Sanofi-Aventis durchgeführten Screenningprogramms aus dem Aktinomyzeten Actinomadura namibiensis DM 6313 isoliert. Die Strukturaufklärung dieses Naturstoffs gelang erst 2009, wobei eine strukturelle Besonderheit entdeckt wurde. Dabei handelt es sich um die Triaminortrisäure Labionin, die aus einem zentralen quartären Kohlenstoffatom mit einem Lanthioninmotiv und einer ungewöhnlichen Methylenbrücke besteht, die eine kovalente Verknüpfung zu einer weiteren Aminosäure bildet. Bioaktivitätsstudien von Labyrinthopeptin A2 in der Maus zeigten eine ausgezeichnete Wirksamkeit gegen neuropathische Schmerzen. Die Synthese des aa- disubstituierten Vorläufers von Labionin, der quartären Aminosäure (2R,4S)-5-tert-Butyl-1-methyl-2-azido-4-(bis-Boc-amino)-2-(hydroxymethyl)pentandioat, war erst nach mehrmaliger Änderung der Synthesestrategie möglich. Nach der erfolgreichen Synthese dieser Aminosäure wurde eine Route zur Synthese des A´-Rings entwickelt. Im zweiten Teil der Arbeit wurde eine Syntheseroute zur Darstellung einer diastereomerenreiner 2,4-Diamino-2-methylglutarsäure entwickelt. Diese Verbindung ist ein Abbauprodukt der ungewöhnlichen Aminosäure Labionin und wird zum Nachweis dieser Aminosäure verwendet. Bisher wurde Labionin nur im Labyrinthopeptin A2 nachgewiesen, doch ein Vergleich des Labyrinthopeptin-Biosyntheseclusters mit Biosyntheseclustern andere Aktinomyzeten, lässt auf die Anwesenheit der Aminosäure Labionin in anderen Lantibiotika schließen. Die Synthese von 2,4-Diamino-2-methylglutarsäure erfolgte ausgehend von Cbz-Glycin-OAll. durch eine chelatverbrückte Claisen-Umlagerung und anschließenden Aufbau einer Amino- und Carboxygruppe aus der Doppelbindung des Umlagerungsproduktes. Die Synthese führte jedoch aufgrund der geringen Stereoselektivität der verwendeten asymmetrischen Dihydoxylierung nach Sharpless zu einem razemischen Gemisch, bei dem die Diastereomrenpaare voneinander getrennt werden konnten.The current work deals with two problems. In the first part of the theses the synthesis of a labioninprecursor and of the A´-ring of labyrinthopeptin A2 is described. The second part concentrates on the synthesis of diaminomethylglutaricacid as a reference compound for the verification of labionin in peptides. Labyrinthopeptin A2 is a pentacyclic lantibiotic that consists of 18 amino acids. This peptide was isolated from Actinomadura namibiensis DM 6313 in 1988. The structure was elucidated in 2009 and shows a unique amino acid, termed labionin. This triaminotriacid consists of a central quaternary carbon atom which forms a lanthioninmotif and a unique methylenbridge. For the synthesis of the A´-ring an aa-disubstituted amino acid was needed. This building was synthesized in 10 reaction steps starting from glycine in a total yield of 15%. It served as a precursor of labionin and successfully led to the development of an innovative synthesis of the A´-ring. The second part of the theses deals with the synthesis of diaminomethylglutaricacid for the verification of labionin in peptides. Diaminomethylglutaricacid is a degradation product of labionin that results of the desulfuration of labionin containing peptides. It was synthesized from Cbz-protected alanineallylester using a Claisen-type rearrangement to generate the quaternary carbon atom. The double bond in the resulting allylalanine was transformed to an aminoacidmotif. The resulting diaminomethylglutaricacid provides the first method for the verification of the amino acid labionin in peptides
