    Duni a Parma. Tracce d’archivio tra opera italiana e teatro francese

    La permanenza di Egidio Romualdo Duni a Parma fu un episodio cruciale sia nella biografia del compositore, sia nella storia dell'opéra-comique francese. Fu infatti a Parma che Duni entrò in contatto con la incipiente tradizione francese e che decise di trasferirsi a Parigi dove contribuì significativamente a consolidare e trasformare il nuovo genere di teatro musicale. Grazie allo studio dei documenti conservati nell'Archivio di Stato di Parma e al loro incrocio con la recente bibliografia musicologica, ho ricostruito il ruolo effettivo del compositore nell'organico di corte, gl incarichi ricevuti e soprattutto i contatti che ha potuto stabilire nel ducato Borbonico. Il quadro che emerge rende conto dell'attività di un artista inconsueto nella tradizione italiana, per livello culturale, riflessione critica ed artistica, capacità progettuale trasversale tra teatro musicale e teatro di parola; tutte doti che Duni avrebbe saputo poi spendere ai massimi livelli nell'ambiente intellettuale parigino. E' stato inoltre possibile datare diverse pagine musicali di Duni, circostanziare con precisione i suoi primi passi nel dramma francese, individuare il repertorio italiano e francese in cui era stato coinvolto, prima della celebre stagione riformatrice del teatro parmigiano. Lo studio delle carte di Archivio ha reso anche possibile ricostruire la graduale espansone della orchestra ducale di corte fino alla morte del duca Filippo I e l'organizzazione della musica in uno dei più dinamici e cosmopoliti ducati italiani del pieno Settecento

    Meiosis progression and donor age affect expression profile of DNA repair genes in bovine oocytes

    Several genetic and physiological factors increase the risk of DNA damage in mammalian oocytes. Two critical events are meiosis progression, from maturation to fertilization, due to the extensive chromatin remodelling during genome decondensation and aging which is associated to a progressive oxidative stress. In this work, we studied the transcriptional patterns of three genes, RAD51, APEX-1 and MLH1, involved in DNA repair mechanisms. The analyses were performed by Real-Time quantitative PCR (RT-qPCR) in immature and in vitro matured oocytes collected from 17 ± 3 mo old heifers and 94 ± 20 mo old cows. Batches of 30-50 oocytes for each group (three replicates) were collected from ovarian follicles of slaughtered animals. The oocytes were freed from cumulus cells at the time of follicle removal, or after IVM carried out in M199 supplemented with 10% foetal calf serum, 10 IU LH /ml, 0.1 IU FSH /ml and 1 μg 17β-oestradiol/ml. Total RNA was extracted by Trizol method. The expression of bovine GAPDH gene was used as internal standard, while primers for bovine RAD51, APEX-1 and MLH1 genes were designed from DNA sequences retrieved from GeneBank. Results obtained indicate a clear up-regulation of RAD51, APEX-1 and MLH1 genes after IVM ranging between 2- and 4-fold compared to GV oocytes. However, only RAD51 showed a significant transcript increase between the immature oocytes collected from young and old individuals. This finding candidates RAD51 as gene marker for discriminating bovine immature oocytes in relation to the donor age

    Marine invertebrates provide a series of natural products with different biological activities. Several of these compounds and their derivatives showed a potent anticancer effect. Tunicates represent an important source of bioactive agents, leading to the isolation of ecteinascidin-743 (ET-743), a compound isolated from the Caribbean sea squirt Ecteinascidia turbinata with a potent cytotoxic activity against a variety of tumours in vitro and in vivo. Current phase II clinical trials against soft tissue sarcomas in Europe and the United States indicate that ET-743 represents a highly promising anticancer agent. Another example is aplidine from the Mediterranean tunicate Aplidium albicans, with a broad spectrum activity against various types of cancers, such as colorectal, lymphoma, thyroid and renal cancers. In the present work, we reported, for the first time, that a partially purified methanolic extract prepared from the ascidian Ciona intestinalis inhibited cell proliferation in human cell lines of different origin, including Caco2, HPB-ALL, U-937 and HL-60 and induced early apoptotic events, such as caspase-3 activation and internucleosomal DNA degradation. We suggest the presence in the Ciona intestinalis extract of bioactive compounds possessing anticancer activity

    Vitamin E in early stages of sea bass (Dicentrarchus labrax) development.

    This study reports titration of vitamin E levels in the sea bass (Dicentrarchus labrax) using high-pressure liquid chromatography. The first part of the work is devoted to vitamin E detection in: (1) plasma of maturing females and males characterized by different body sizes; (2) seminal fluid and eggs; and (3) developing embryos of sea bass fed with vitamin E. In the second part of the study, variations of vitamin E levels during larval development are analyzed. The results show a direct correlation between plasma vitamin E content and body size for both adult male and female sea bass. High vitamin E levels were found in seminal fluid, in eggs before and after fertilization, and in embryos during development and at hatching, whereas vitamin E level was low in dead embryos and in embryos with limited survival. During larval development, the vitamin E content decreased slowly but steadily during the first four days of larval growth; subsequently, it progressively increased from day 9 to day 40. In teratogenic larvae, vitamin E content was significantly higher than in normal larvae. This study provides evidence on how vitamin E exerts an antioxidant defense in sea bass reproduction
