The interaction between endopolygalacturonase from Fusarium moniliforme and PGIP from Phaseolus vulgaris studied by surface plasmon resonance and mass spectrometry
A combination of surface plasmon resonance (SPR) and matrix-assisted laser-desorption-ionization-time-of-flight mass spectrometry (MALDI-TOF-MS) was used to study the interaction between endopolygalacturonase (PG) from Fusarium moniliforme and a polygalacturonase-inhibiting protein (PGIP) from Phaseolus vulgaris. PG hydrolyses the homogalacturonan of the plant cell wall and is considered an important pathogenicity factor of many fungi. PGIP is a specific inhibitor of fungal PGs and is thought to be involved in plant defence against phytopathogenic fungi. SPR was used either to study the effect of the PG glycosylation on the formation of the complex with PGIP, and as a sensitive affinity capture of an interacting peptide from a mixture of PG fragments obtained by limited proteolysis. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
Proteômica abrangente de alta resolução na análise de neutrófilos humanos ativados pelo peptideo formyl Methyl Leucyl Phenylalanine (fMLP)
Tese (doutorado)—Universidade de Brasília, Faculdade de Medicina, Programa de Pós-Graduação em Patologia Molecular, 2017.Os neutrófilos são granulócitos polimorfonucleares que utilizam mecanismos intra e extracelulares para eliminar patógenos. A ativação de neutrófilos pela N-formil-Metionil-Leucyl-Phenylalanine (fMLP), uma molécula quimiotáctica liberada de bactérias Gram-positivas, disparadores de explosão respiratória, modificações morfológicas, degranulação e liberação de NETs. No entanto, a desregulamentação de neutrófilos está envolvida com várias patologias, em parte, devido à exacerbação da descarga de espécies reativas de oxigênio e enzimas citolíticas Embora haja um número considerável de estudos sobre condições, as questões sobre como essas desregulações permanecem sem resposta; Para os caminhos exatos que são percorridos por esses caminhos intracelulares ainda estão preenchidos com lacunas. Atualmente, a comunidade científica vem usando uma ferramenta de análise conhecida como espectrometria de massa (MS) para desempenhar melhor o papel dos neutrófilos na resposta imune; a realização de estudos relacionados aos neutrófilos de proteômica comparativa são ferramentas essenciais para a compreensão dos mecanismos intracelulares que regem a resposta inflamatória. Usando ferramentas proteômicas de alta resolução, o presente trabalho identificou 8362 proteínas no total, na ativação de neutrófilos por fmp de 100 nmL / L. Em conclusão este trabalho representa a eficiência dos métodos de análise proteômica proporcionaram a identificação de 8362 proteínas, sendo 301 proteínas reguladas diante do estímulo com fMLP. Pois, proteínas importantes para a resposta inflamatória dos neutrófilos foram identificadas, mostrando que a análise comparativa dos mapas proteômicos utilizando diferentes métodos de aquisição e extração proteicas atuaram de forma complementar na robustez da identificação mais abrangente de proteínas de neutrófilos, tanto quiescentes quanto ativados com fMLP. Ademais, E que existe uma forte conexão entre a família das pequenas GTPases com a maioria dos processo desencadeados pela ativação de vias por fMLP, como por exemplo as evidências de alterações estruturais em MEV. Com as análises conjugadas será possível sugerir pontos de modulação da resposta inflamatória promovidas por agentes ativadores relacionados a receptores ligados a proteína G. A identificação de regulações por subunidades do proteassoma 26S tendo extensa participação nas vias reguladas, o que pode ser um indicativo para um mecanismo de regulação por esse complexo. Outrossim, a proteômica se mostra uma ferramenta essencial para a análise de vias, pois proporciona a identificação de proteínas que estão em pequenas quantidades como citocinas liberadas pela ativação. No mais, os mecanismos de morte celular programada com divergência entre vias de ativação e bloqueio da apoptose, podendo acarretar desregulação pois os dados tentem a sugerir um fino balanço entre as vias de vida e morte. O presente trabalho apresenta um conjunto de proteínas com diversas funções relacionadas aos processos da resposta inflamatória, que incluem a atividade oxidativa, motilidade, metabolismo energético e sinalização intra e intercelular. Este estudo adiciona dados relacionados à ativação de neutrófilos por fMLP, também revelando proteínas que não foram previamente identificadas em neutrófilos, ampliando as possibilidades de modulação da resposta inflamatória. Nosso estudo também comtempla o maior número de proteínas nos neutrófilos ativados por fMLP, ampliando assim as possibilidades de sugerir uma modulação da resposta inflamatória em neutrófilos.Neutrophils are polymorphonuclear granulocytes that use intra- and extracellular mechanisms to eliminate pathogens. Activation of neutrophils by N-formyl-Methionyl-Leucyl-Phenylalanine (fMLP), a chemotactic molecule released from gram-positive bacteria, triggers respiratory burst, morphological modifications, degranulation and NETs´ release. However, neutrophils´ deregulation are involved with several pathologies, in part, due to the exacerbated discharge of reactive oxygen species and cytolytic enzymes; in other words, prolonged lifespan of activated neutrophils and their migration to non-injured tissues may result in severe clinical conditions. Although there are a considerable number of studies on conditions, questions about how such deregulations still remain unanswered; For the exact pathways that are traversed by these intracellular pathways are still filled with gaps. Currently, the scientific community has been using an analysis tool known as mass spectrometry (MS) to better play the role of neutrophils in the immune response; studies related to comparative proteomics neutrophils are essential tools for understanding the intracellular mechanisms governing the inflammatory response. In conclusion, this work represents the efficiency of the methods of proteomic analysis, which allowed the identification of 8362 proteins, 301 of which were regulated proteins before the stimulation with fMLP. Because proteins important for the inflammatory response of neutrophils were identified, showing that the comparative analysis of proteomic maps using different methods of protein acquisition and extraction acted in a complementary way in the robustness of the more comprehensive identification of neutrophil proteins, both quiescent and activated with fMLP . In addition, there is a strong connection between the family of small GTPases with most of the processes triggered by the activation of fMLP pathways, such as the evidence of structural alterations in SEM. With the conjugated analyzes it will be possible to suggest modulation points of the inflammatory response promoted by activating agents related to G protein-linked receptors. The identification of 26S proteasome subunit regimens having extensive participation in the regulated pathways, which may be indicative of a mechanism of regulation by this complex. In addition, the mechanisms of programmed cell death with divergence between activation pathways and apoptosis blockade, may lead to deregulation as the data try to suggest a fine balance between the life and death pathways. The present work presents a set of proteins with diverse functions related to the processes of the inflammatory response, which include oxidative activity, motility, energetic metabolism and intra and intercellular signaling. This study adds data related to the activation of neutrophils by fMLP, also revealing proteins that were not previously identified in neutrophils, increasing the possibilities of modulation of the inflammatory response.
