Sistema de acciones para estimular las potencialidades físicas en niños con Síndrome de Prader Willi
This research responds to the need to introduce modifications to the Therapeutic Physical Culture process, when working with children with Prader Willi Syndrome. Theoretical-methodological shortcomings were found that limit the stimulation of physical potentialities in them. The objective is to propose a system of actions to stimulate these potentialities, containing stages, orientations and general and specific methodological indications, with procedures that make this process feasible. The essential relationships between the affective-motivational components and the stimulation of physical potentialities are revealed.Esta investigación responde a la necesidad de introducir modificaciones al proceso de la Cultura Física Terapéutica, cuando se trabaja con niños portadores del Síndrome de Prader Willi. Se constataron insuficiencias teórico-metodológicas que limitan la estimulación de las potencialidades físicas en estos. El objetivo consiste en proponer un sistema de acciones para estimular dichas potencialidades, contentivo de etapas, orientaciones e indicaciones metodológicas generales y específicas, con procedimientos que viabilizan este proceso. Modelling and analysis of large-scale models of signalling networks
Cells rely on a system of signal processing and transmission networks to perform and coordinate their basic activities. The transmission process is governed through chemical signals and reactions which can be mediated by proteins or other smaller molecules. This process is collectively named cell signalling. Errors in cell signalling can lead to the development of severe diseases such as cancer. As such, an understanding at a system level of cell signalling can help us identify aberrant processes which can be therapeutically targeted. Considering the huge variety of proteins and their modifications, these kinds of systems can be quite large, complex and dynamic. This makes the modelling of cell signalling systems particularly challenging. The aim of the PhD project consists of developing new and existing methods used for the modelling cell signalling networks for the understanding of processes involved in it and the mechanisms of cell function. This was achieved through the development of various modelling tools (CARNIVAL, PHONEMeS, CellNOptR and Dynamic-Feeder). At the core of these tools stands the integration of optimisation techniques with simulation analysis in order to provide information about the behaviour of the biological system representing our signalling networks. Such an approach allows the prediction of perturbation outcomes (i.e. treating cells with drugs) and can give help in the planning of prospective treatment strategies. Model-based design of cell signalling systems, especially optimisation-based, is a major focus of this work, and here I will be mostly focusing on understanding, exploiting and designing methods of integer linear and non-linear dynamic optimisation techniques for structural and parametric identification of large signalling networks based on experimental data. A machine learning and optimization approach to uncover TP53 regulatory patterns
TP53, the Guardian of the Genome, is the most frequently mutated gene in human cancers and the functional characterization of its regulation is fundamental. To address this we employ two strategies: machine learning to predict the mutation status of TP53 from transcriptomic data, and directed regulatory networks to reconstruct the effect of mutations on the transcipt levels of TP53 targets. Using data from established databases (Cancer Cell Line Encyclopedia, The Cancer Genome Atlas), machine learning could predict the mutation status, but not resolve different mutations. On the contrary, directed network optimization allowed to infer the TP53 regulatory profile across: (1) mutations, (2) irradiation in lung cancer, and (3) hypoxia in breast cancer, and we could observe differential regulatory profiles dictated by (1) mutation type, (2) deleterious consequences of the mutation, (3) known hotspots, (4) protein changes, (5) stress condition (irradiation/hypoxia). This is an important first step toward using regulatory networks for the characterization of the functional consequences of mutations, and could be extended to other perturbations, with implications for drug design and precision medicine
De delegado a romancista : representações da violência e figura policial nos romances de Joaquim Nogueira
Este trabalho se debruça sobre o tema das representações sociais da violência e em torno da figura policial, com o objetivo de compreender como é formulada a representação da violência nos romances de Joaquim Nogueira, ex-delegado de polícia, de modo a identificar os nexos entre o modelo ficcional de policial e as representações acerca da violência por parte do autor. Os principais conceitos teóricos mobilizados foram representações da violência, identidade policial e mimese e, na composição do modelo teórico metodológico, que combinou análise dialética e análise interpretativa, foram utilizadas também as noções de figuração, para Auerbach, e de poder, para Foucault. O corpus de investigação foram os romances Informações sobre a Vítima (2002) e Vida Pregressa (2003), ambos analisados enquanto romances da violência, a partir dos elementos textuais presentes nos textos relacionando-os com a representação da realidade, da violência e a figura policial desenvolvidas pelo autor. Considerando a análise textual, bem como a análise das relações de violência (e de poder) apresentadas nos textos, juntamente com a construção individual e profissional de Venício, o narrador protagonista, percebeu-se que a figura policial estabelecida pelo autor é multifacetada, que não é nem a vivida pelo autor enquanto delegado de polícia, nem mera ficção. A figura policial desenvolvida por Nogueira, especialmente em Venício, é dual, posto que ora representa a instituição policial e, por isso, pode ser vista como universal; ora figura a instituição policial, pois transcende a realidade e motiva a reflexão do leitor para pensar a situação da instituição a partir de um cenário imaginário passível de veracidade.This paper focuses on the theme of social representations of violence and the police figure, with the aim of understanding how the representation of violence is formulated in the novels of Joaquim Nogueira, a former police chief, to identify the links between the fictional model of the police officer and the representations of violence proposed by the author. The main theoretical concepts mobilized were representations of violence, police identity and mimesis. In the composition of the theoretical methodological model, which combined dialectical analysis and interpretative analysis, also used the notions of figuration, for Auerbach, and power, for Foucault. The research corpus was the novels Informações sobre a Vítima (2002) and Vida Pregressa (2003), both analyzed as novels of violence, based on the textual elements present in the texts relating them to the representation of reality, representation of violence and the police figure developed by the author. Considering the textual analysis, as well as the analysis of the relations of violence (and power) presented in the texts, together with the individual and professional construction of Venício, the protagonist narrator, it was noticed that the police figure established by the author is multifaceted, it’s neither the one experienced by the author as a police delegate, nor mere fiction. The police figure developed by Nogueira, especially in Venício, is dual, sometimes it represents the police institution and, therefore, can be seen as universal; sometimes it figures the police institution, since it transcends reality and motivates the reader to reflect about the institution based on an imaginary scenario that can be seen as true
