    NF-κB in anti-Inflammatory activity of probiotics: an update

    The NF-κB (nuclear factor κappa B) belongs to a family of transcription factors that regulate a number of key cellular pathways and control many physiological processes, including the immune and inflammatory responses. The beneficial effect of probiotics is presently recognized in Both the intestinal and extra-intestinal development. The growing literature on the subject suggests an important role of specific probiotic strains, mixtures of strains, and secreted products, all of which have a role in either the preventive or therapeutic treatment of certain human disorders. During the last two decades, a number of studies have shown that probiotic strains and their extracellular products can join the immune system and activate receptors in different key pro-inflammatory pathways such as the NF-κB route, either by increasing or suppressing the signaling pathways. This brief review does not include an exhaustive bibliography of the subject, but focuses instead on an update to the latest research of the past three years on the role of the NF-κB signaling pathway in the anti-inflammatory effect of probiotics.Fil: Rodriguez, Ana Virginia.     Antibacterial activity of lactobacillus plantarum CRL 759 against methicillin-resistant staphylococcus aureus and pseudomonas aeruginosa

    This study investigates the antibacterial properties of potential probiotic Lactobacillus plantarum CRL 759 against Pseudomonas aeruginosa and methicillin-resistant Staphylococcus aureus (MRSA) isolated from samples of human diabetic foot, by different methods. The results demonstrated that L. plantarum CRL 759 cell-free supernatant (SLp759) inhibited both P. aeruginosa and MRSA growth. In addition, SLp759 inhibited the adhesion of pathogenic microbes. The characterization of SLp759 showed that after neutralization of acidic SLp759, no activity against pathogens strains was observed. Moreover, treatment with proteolytic enzymes did not modify antibacterial activity, indicating that no bacteriocin was present in the supernatant. In addition, results obtained by HPLC analysis indicated that the inhibitory effect was the result of production of two organic acids: lactic and acetic. From the results reported here, L. plantarum CRL 759 has potential applications as a probiotic by preventing or breaking multidrug-resistant microorganism carriage.Fil: Layús, Barbara Ivana.     Role of simvastatin and methyl-β-cyclodextin on inhibition of poliovirus infection

    Cells exposed to simvastatin or to methyl-β-cyclodextrin show reduced poliovirus infection, without alteration in virus binding or on the kinetics of genome entry, suggesting that the steps which are altered are those post uncoating and genome entry. Reduction of infection by cyclodextrin is reversed by increasing MOI whereas that produced by simvastatin treatment is not, suggesting that the effects on infection are not due to a reduction in cholesterol. The differences in the characteristics of inhibition can be explained by the differential effects of the compounds. Cyclodextrin inhibits the store-operated calcium channels, suggesting that reduction in infection is through translational inhibition. Simvastatin produces vesicles from internal membranes which cannot sustain viral RNA synthesis, reducing infection through reduced transcription. The results indicate that the impact on viral infection by the cholesterol-modifying agents is due to the cellular changes produced rather than due to disruption of the cholesterol-rich domains.Fil: Liu, Shumei.     Disruption of lipid rafts enhances the effect of lactobacilli on the production of tumor necrosis factor-alpha in mononuclear blood cells

    We investigated, to determine whether disruption of lipid rafts would influence the effect of three selected strains of Lactobacillus on TNF-alpha production by peripheral blood mononuclear cells. Two strains increased TNF-alpha production; and the third one reduced cytokine levels. Disruption of rafts changed the immunomodulatory effect of the three strains.     Phosphatidylinositol-specific phospholipase C activity in Lactobacillus rhamnosus with capacity to translocate

    Phosphatidylinositol-specific phospholipase C (PI-PLC) activity was investigated in 25 different lactic acid bacteria (LAB) strains belonging to the genera Lactobacillus, Weisella, and Enterococcus. PI-PLC activity was detected in 44% of the strains studied in culture medium without carbon source. From the PI-PLC positive strains, Lactobacillus rhamnosus ATCC 7469 was selected for translocation studies. Healthy mice were orally administered with a daily dose of 2.0U109 of viable L. rhamnosus suspension. Viable bacteria were detected in liver and spleen of mice fed with LAB for 7 days. Bacterial colonies isolated from liver were biochemically characterized, and further subjected to randomly amplified polymorphic DNA. Amplification patterns of five strains displayed identical profiles to L. rhamnosus. PI-PLC activity was determined in the strains recovered from liver.Fil: Rodriguez, Ana Virginia.     A Postbiotic Formulation of Lactiplantibacillus plantarum CRL 759 Attenuates Endotoxin Induced Uveitis

    Purpose: To evaluate the anti-inflammatory activity of a cell-free supernatant from Lactiplantibacillus plantarum CRL 759, in phosphate buffer modified according to Sorensen called POF-759.Methods: The activity of POF-759 administered by means of eye drops was evaluated on animals subcutaneously injected with the lipopolysaccharide animals in which uveitis was induced by a subcutaneous injection of lipopolysaccharide (EIU). Clinical signs of ocular inflammation, cytokines and proteins were examined in the aqueous humor. Additionally, cellular infiltration was evaluated by histopathological analysis.Results: The new postbiotic administered locally decreases signs of ocular damage, the number of infiltrating cells in the anterior and posterior chambers, the proinflammatory mediators and the proteins in the aqueous humor on mice with EIU.Conclusions: Our results provide an impetus to relieve ocular inflammation and to identify and develop preventive and therapeutic approaches, to avoid deterioration and to maintain healthy eyes on inflammatory processes.Fil: Layús, Barbara Ivana.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
