    Hyperfine splitting of [Al VI] 3.66 mu m and the Al isotopic ratio in NGC 6302

    The core of planetary nebula NGC 6302 is filled with high-excitation photoionized gas at low expansion velocities. It represents a unique astrophysical situation in which to search for hyperfine structure (HFS) in coronal emission lines from highly ionized species. HFS is otherwise blended by thermal or velocity broadening. Spectra containing [Al vr] 3.66 mu m P-3(2) <- P-3(1), obtained with Phoenix on Gemini South at resolving powers of up to 75000, resolve the line into five hyperfine components separated by 20-60 km s(-1) as a result of the coupling of the I = 5/2 nuclear spin of Al-27 with the total electronic angular momentum J. The isotope Al-26 has a different nuclear spin of I = 5, and a different HFS, which allows us to place a 3 sigma upper limit on the Al-26/Al-27 abundance ratio of 1/33. We measure the HFS magnetic dipole coupling constants for [Al vr], and provide the first estimates of the electric quadrupole HFS coupling constants obtained through astronomical observations of an atomic transition

    Stearoyl-CoA desaturase (SCD) is a key enzyme that determines the composition and metabolic fate of ingested fatty acids, in particular the conversion of trans-vaccenic acid (TVA) to conjugated linoleic acid (CLA). The present study addressed the hypothesis that intestinal TVA absorption and biotransformation into CLA can be modulated by EPA and 3,10-dithia stearic acid (DSA) via altered SCD mRNA levels and desaturation indices (cis-9, trans-11-CLA:TVA and oleic acid:stearic acid ratios) in Caco-2 and T84 cells, two well-established in vitro models of the human intestinal epithelium. The study determined the effect of acute (3 h with 0.3 mm-EPA or 0.3 mm-DSA) and acute-on-chronic (1 week with 0.03 mm-EPA or -DSA, followed by respectively, 0.3 mm-EPA or -DSA for 3 h) treatments. In both cell lines, acute EPA treatment did not alter SCD desaturation indices, whereas the acute-on-chronic treatment affected these surrogate markers of SCD activity. This was associated with reduced sterol regulatory-element binding protein-1c and SCD mRNA levels. In contrast, acute and acute-on-chronic DSA treatments significantly reduced SCD desaturation indices without affecting SCD mRNA levels in Caco-2 cells. The present study on intestinal cells shows that the conversion rate of TVA to c9, t11-CLA is affected by other fatty acids present in the diet such as EPA, confirming previous observations in hepatic and mammary cell models

    Los itinerarios ecológicos suelen centrarse en la observación de monumentos, de componentes del patrimonio cultural o elementos de interés del espacio natural, sin integrar las características del medio físico del espacio. Esto no implica que el diseño del itinerario sea incorrecto pero una forma de mejorarlo o complementarlo es integrar todos los aspectos que configuran el ambiente, es decir, el medio biológico, paisajístico, histórico, patrimonial… Estos itinerarios también deben ayudar a los visitantes, independientemente de sus características, a percibir todos los elementos que integran el medio natural, así como las relaciones entre ellos. Además, lo ideal es que el visitante llegue a conocer la evolución y los procesos que se dan dentro del espacio, así como los programas de conservación y elementos incorporados y desarrollados por la mano humana dentro del espacio. Los itinerarios pueden dirigirse a varios tipos de visitantes, ya sean grupos de jóvenes o mayores o individuos aislados, como los excursionistas.     Pure-rotational 1D-CARS spatiotemporal thermometry with a single regenerative amplifier system

    We report spatiotemporal pure-rotational coherent anti-Stokes Raman spectroscopy (CARS) in a one-dimensional imaging arrangement obtained with a single ultrafast regenerative amplifier system. The femtosecond pump/Stokes photon pairs, used for impulsive excitation, are delivered by an external compressor operating on a ∼35% beam split of the uncompressed amplifier output (2.5 mJ/pulse). The picosecond 1.2 mJ probe pulse is produced via the second-harmonic bandwidth compression (SHBC) of the ∼65% remainder of the amplifier output (4.5 mJ/pulse), which originates from the internal compressor. The two pump/Stokes and probe pulses are spatially, temporally, and repetition-wise correlated at the measurement, and the signal generation plane is relayed by a wide-field coherent imaging spectrometer onto the detector plane, which is refreshed at the same repetition rate as the ultrafast regenerative amplifier system.     peer-reviewedBackground Currently, a number of bioinformatics methods are available to generate appropriate lists of genes from a microarray experiment. While these lists represent an accurate primary analysis of the data, fewer options exist to contextualise those lists. The development and validation of such methods is crucial to the wider application of microarray technology in the clinical setting. Two key challenges in clinical bioinformatics involve appropriate statistical modelling of dynamic transcriptomic changes, and extraction of clinically relevant meaning from very large datasets. Results Here, we apply an approach to gene set enrichment analysis that allows for detection of bi-directional enrichment within a gene set. Furthermore, we apply canonical correlation analysis and Fisher's exact test, using plasma marker data with known clinical relevance to aid identification of the most important gene and pathway changes in our transcriptomic dataset. After a 28-day dietary intervention with high-CLA beef, a range of plasma markers indicated a marked improvement in the metabolic health of genetically obese mice. Tissue transcriptomic profiles indicated that the effects were most dramatic in liver (1270 genes significantly changed; p < 0.05), followed by muscle (601 genes) and adipose (16 genes). Results from modified GSEA showed that the high-CLA beef diet affected diverse biological processes across the three tissues, and that the majority of pathway changes reached significance only with the bi-directional test. Combining the liver tissue microarray results with plasma marker data revealed 110 CLA-sensitive genes showing strong canonical correlation with one or more plasma markers of metabolic health, and 9 significantly overrepresented pathways among this set; each of these pathways was also significantly changed by the high-CLA diet. Closer inspection of two of these pathways - selenoamino acid metabolism and steroid biosynthesis - illustrated clear diet-sensitive changes in constituent genes, as well as strong correlations between gene expression and plasma markers of metabolic syndrome independent of the dietary effect. Conclusion Bi-directional gene set enrichment analysis more accurately reflects dynamic regulatory behaviour in biochemical pathways, and as such highlighted biologically relevant changes that were not detected using a traditional approach. In such cases where transcriptomic response to treatment is exceptionally large, canonical correlation analysis in conjunction with Fisher's exact test highlights the subset of pathways showing strongest correlation with the clinical markers of interest. In this case, we have identified selenoamino acid metabolism and steroid biosynthesis as key pathways mediating the observed relationship between metabolic health and high-CLA beef.
