THE ACTIVE SITE OF THE CYTOCHROME c OXIDASE PROTEIN AS PROBED BY CARBON MONOXIDE PHOTOLYSIS AND FTIR SPECTROSCOPY

Altschuld (1981) PNAS 78, 234.Author Institution:Cytochrome c Oxidase contains two heme irons, a and , and two coppers, A and B. The protein catalyzes the reduction of dioxygen to water within the heme pocket. Carbon monoxide, like , binds to the iron, and it is photolizable with visible light. After photolysis the CO relaxes back to the Fe following a power law time dependence: (l + t/to). This relationship is expected from a distribution in activation enthalpy caused by a distribution in protein conformation (1), The peak of this enthalpy distribution, Hp, can be obtained by fitting the CO kinetics, simultaneously at all temperatures, to the function and is found to be 40kJ/mole. This value, much larger than for simpler heme proteins, has been explained by FTIR which has been used to identify the photolyzed CO associating with the B Cu (2). Since the CO cannot escape the heme pocket at these low temperatures (210K), we infer that the Fe and the Cu are separated by a clear pathway, 4 to 6 {\AA} long, within this pocket. That the two metals can both bind CO and that they are in close proximity should be helpful in elucidating the mechanism of reduction. A passable intermediate would be Fe-0-0-Cu. The transmission spectrum of the Fe bound CO vibration has been fit very well to a Gaussian bandshape convoluted with a sin x/x instrument function at all temperatures between 10 and 275K. The Gaussian shape also reflects the distribution in conformational states frozen into the protein at low temperature. (Contributed Paper) INFRARED SPECTROSCOPY OF METAL PROTEINS
Author Institution: Department of Physiological Chemistry, The Ohio State University College of MedicineBiological macromolecules are critically dependent upon a particular manner of folding for their biological activity. This, in turn, depends upon solvent composition, temperature, and other factors. Thus classical infrared sampling techniques which employ dried films, KBr pellets, or ``good’’ infrared solvents, destroy biologically native structures, and are of limited value in structure analysis. We have therefore studied a variety of metal-protein complexes in concentrated aqueous solution by infrared spectroscopy. Studies which illustrate the use of quantitative infrared spectroscopy for analysis of biological structures include determination of the temperature dependence of the equilibrium for azide bound ionically or covalently to metmyoglobin, and the use of isotope shift data in the study of carbon monoxide complexes with oxygen-carrying proteins from vertebrates and lower animal forms
Compliance with community mitigation and interventions in Pandemic Influenza : a community policing strategy
CHDS State/LocalA number of response plans and strategies have been published concerning preparation for an oncoming Pandemic Influenza. The majority of federal guidance and state planning with respect to pandemic preparation focuses excessively on the availability and distribution of effective vaccine and antiviral remedies; pharmaceutical solutions. Effective vaccines, presently unavailable, will not be in production and available for application for at least eight months after the onset of an identified pandemic. Community mitigations and interventions such as school closures, event cancellations, limited travel, quarantine and work at home plans are traditional responses to slowing the spread of a virus. In order to effectively implement these time tested strategies, voluntary community compliance with interventions becomes exceedingly important. The recent global experience with SARS and current mathematical modeling of virus spread characteristics support community mitigation efforts. The community policing model, having evolved over the last twenty years, provides a pre-existing framework to engage the public in grass roots pandemic education, awareness, planning and problem solving partnerships. The Incident Command System provides a structure for collaborative, multi-agency approach to successfully implementing a community awareness and compliance initiative. INFRARED STUDIES OF PORPHYRINS. VALENCE AND LIGAND FIELD EFFECTS IN METAL COMPLEXES

Proc., 21, 46 (1962).Author Institution: Department of Physiological chemistry, Johns Hopkins School of Medicine‘‘Previous studies of metal-porphyrin have demonstrated the manner in which electronic effects of peripheral substituents on the porphyrin ring are transmitted through centrally coordinated divalent iron to cis-coordinated carbon monoxide, and how they affect the strength of pi-bonding to the iron. In the present study, the effects of the centrally coordinated metal on vibrational modes of 2,4-diacetyldeuteroporphyrin IX dimethyl ester have been examined, when the metal is Mn(III), Fe(III),Co(II), Fe(II), Co(II), Ni(II) Cu(U), Zn(II), Pd(II), or Ag(II). All of these compounds except the Co(III) complex have been purified and characterized by ultraviolet, visible, and infrared spectroscopy, and by elemental analyses. The Co(III) complex bus been purified and identified by visible and infrared spectroscopy. The major vibrational bands, for which assignments have been at least partially determined, have been examined in terms of shifts in frequency with each of several types of metal properties. In addition to requiring the separation of these compounds into complexes of trivalent, and first and second row transition metals, the frequencies of several vibrational modes such as the methane CH out-of-place deformation exhibit the non-linearity with number of d electrons which is characteristic of ligand field stabilization effects. Other modes, such as the corresponding in-plane deformation are affected by the metal to a negligible extent. The frequency of the acetyl Co stretching mode of the trivalent metal complexes is about higher than that of the divalent metal complexes, but is otherwise negligibly affected by the metal. However, the relative intensities of these vibrations vary in a manner which is consistent with ligand field stabilization.’
