    Heterotrimeric G-proteins have been implicated in having a role in many plant signalling pathways. To understand further the role of G-proteins, a preliminary experiment was performed to assess the impact of the G? subunit loss-of-function mutation gpa1-1 on the Arabidopsis transcriptome. The analysis indicated that the G? subunit may play a role in response to jasmonic acid (JA). Consistent with this, G? mutants showed a reduced response to JA in inhibition of chlorophyll accumulation and root growth, whilst G? gain-of-function plants overexpressing G? showed the opposite phenotype. The levels of JA and related compounds were unaffected in the gpa1-1 mutant, as was autoregulation of the Allene Oxide Synthase (AOS) gene that encodes a key enzyme for JA biosynthesis. In contrast, further analyses using G? loss- and gain-of-function Arabidopsis lines indicated that G? positively modulates the expression of the Vegetative Storage Protein (VSP) gene. This indicates that the G? subunit regulates a subset of JA-regulated genes defining a branch point in this signalling pathway in Arabidopsis. Further analysis of the impact of G? loss of function upon the JA-regulated transcriptome using Arabidopsis full genome arrays indicated that up to 29% of genes that are >2-fold regulated by JA in the wild type are misregulated in the G? mutant. This supports the observation that a significant proportion of, but not all, JA-regulated gene expression is mediated by G

    In vivo Expression von XIAP und Smac Protein in Gliomen und die Korrelation mit der Prognose

    Recent studies demonstrate that defects in apoptotic pathways are involved in both the development of astrocytomas and in their intrinsic resistance to genotoxic therapies. A major role in apoptosis regulation has been demonstrated for XIAP. Animal and in vitro glioma studies encourage therapeutic attempts at depleting XIAP with its endogenous antagonist Smac or mimics of its function. In this study the protein expressions of XIAP and Smac in tissue micro arrays of 194 astrocytomas of different WHO grade (I-IV) were immunohistochemically analyzed. XIAP expression was found to increase significantly with the grade of malignancy, while Smac was expressed at the lowest levels in grade III tumors, whereas the highest expression was observed in glioblastoma. XIAP protein was generally not observed in neuronal cells, while Smac was strongly expressed in neurons of the normal brain and the infiltration zone of glioblastoma. Further, a significant correlation of patient age and XIAP expression was identified exclusively in glioblastoma. The two proteins exhibited a differing distribution in the tumor tissue. While the expression of XIAP was highly correlated in the tumor center and the periphery, Smac showed no correlation. This was interpreted as an evidence for the differing regulation of the two proteins, XIAP expected to be an immanent characteristic of the tumor while Smac seems to react to regional stimuli. A correlation of both proteins with cleaved caspase-8 and -9 as well as the rate of apoptosis as detected by the TUNEL assay could not assess any direct associations. Anti-apoptotic Smac revealed a significant association with survival in WHO grade II with high levels linked to a worse prognosis. XIAP displayed no association with survival in WHO grade-dependent Kaplan-Meier analysis. In conclusion, it is shown that XIAP is significantly upregulated in high grade glioma, confirming the feasibility of an anti-XIAP therapy. The Smac expression profile indicates that differences of apoptosis escape might exist between anaplastic astrocytoma and primary glioblastoma.Mehrere Studien konnten zeigen, dass Schäden in den Signalwegen der Apoptose zum einen an der Entstehung von Astrozytomen eine Rolle spielen und zum anderen an der intrinsischen Resistenz dieser Tumoren gegenüber genotoxischen Therapien beteiligt sind. Für XIAP konnte eine bedeutsame Funktion bei der Regulation der Apoptose nachgewiesen werden. Tierstudien und in vitro Daten legen als therapeutischen Ansatz für astrozytären Tumoren eine Depletierung von XIAP durch seinen endogenen Antagonisten Smac oder andere Moleküle mit einer ähnlichen Funktion nahe. In dieser Studie wurde die Proteinexpression von Smac und XIAP in Tissue micro Arrays von 194 Astrozytomen der WHO Grade I-IV mittels Immunhistochemie analysiert. Es konnte gezeigt werden, dass die Expression von XIAP signifikant mit dem Grad der Malignität ansteigt, während die geringste Expression von Smac in Grad III Astrozytomen detektiert wurden. Auch für Smac wurde die höchste Expression in Glioblastomen gemessen. Das XIAP Protein konnte nicht in neuronalen Zellen nachgewiesen werden, während Smac eine kräftige Anfärbung von Neuronen des normalen Kortex und der Infiltrationszone von diffusen Gliomen zeigte. Darüber hinaus konnte eine signifikante Korrelation von Patientenalter und XIAP Expression innerhalb der Gruppe der Glioblastome festgestellt werden. Die beiden Proteine zeigten eine unterschiedliche Verteilung innerhalb der Tumoren. Während die Expression von XIAP eine deutliche Korrelation von Tumor Zentrum und Peripherie zeigte, war ein solcher Zusammenhang für Smac nicht nachweisbar. Eine Korrelation beider Proteine mit der Expression der aktiven Caspase-8 und -9 als auch der Apoptoserate (ermittelt durch den TUNEL Assay) konnte keine direkten Assoziationen dieser Parameter aufzeigen. Das antiaoptotisch wirksame Smac demonstrierte eine signifikante Assoziation mit dem Überleben im WHO Grad II mit einer Verknüpfung einer hohen Expression mit einer schlechteren Prognose. Für XIAP konnten keine Assoziationen mit dem Überleben nachgewiesen werden. Zusammenfassend demonstriert diese Arbeit eine signifikante Hochregulation von XIAP in malignen Gliomen und bekräftigt die Möglichkeit einer anti-XIAP Therapie. Das Expressionmuster von SMAC gibt Hinweise darauf, dass die Apoptose in Grad III und IV Tumoren unterschiedlich reguliert sein könnte

    Sir John Ross Bt: the last lord Chancellor of Ireland 1921-1922

    Sir John Ross was appointed Lord Chancellor of Ireland in 1921, being the last to hold that office with its abolition in 1922. Ross was born and raised in Londonderry, before proceeding to Trinity College, Dublin. Briefly an MP in the 1890s, Ross was chiefly interested in the law. Called to the Irish Bar in 1880, he took silk in 1891. His career as a Chancery barrister, and later a judge, led him to the Irish woolsack. As a result of the Government of Ireland Act 1920, his role as Lord Chancellor was very different to that of his predecessors. However, as Lord Chancellor he took no back seat role, hearing cases in the newly-established High Court of Appeal for Ireland up until his office was abolished. Ross also served as Speaker of the short-lived Senate of Southern Ireland. He retired to Northern Ireland where he died in 1935.<br/

    Coincidence of semilobar holoprosencephaly and Chiari II malformation: correlation of prenatal diagnostics and neuropathologic findings

    As holoprosencephaly and Chiari II malformation differ considerably, both in pathogenesis and in phenotypic localization, the coincidence of both entities is extremely rare. The case presented is, to our knowledge, the first published report comprising a combination of a semilobar holoprosencephaly associated with a Chiari II malformation and a myelomeningocele diagnosed prenatally and confirmed by postmortem neuropathologic evaluation. These findings indicate that in the case of pre- and postnatal detection of a myelomeningocele and/or Chiari II malformation, possible additional intracranial malformation, such as a semilobar holoprosencephaly, should also be taken into account and vice versa
