    Assessment of the potential utility of different regions of Streptococcus uberis adhesion molecule (SUAM) for mastitis subunit vaccine development

    Streptococcus uberis is one of the most prevalent pathogens causing clinical and subclinical mastitis worldwide. Among bacterial factors involved in intramammary infections caused by this organism, S. uberis adhesion molecule (SUAM) is one of the main virulence factors identified. This molecule is involved in S. uberis internalization to mammary epithelial cells through lactoferrin (Lf) binding. The objective of this study was to evaluate SUAM properties as a potential subunit vaccine component for prevention of S. uberis mastitis. B epitope prediction analysis of SUAM sequence was used to identify potentially immunogenic regions. Since these regions were detected all along the gene, this criterion did not allow selecting a specific region as a potential immunogen. Hence, four fractions of SUAM (-1fr, 2fr, 3fr and 4fr), comprising most of the protein, were cloned and expressed. Every fraction elicited a humoral immune response in mice as predicted by bioinformatics analysis. SUAM-1fr generated antibodies with the highest recognition ability towards SUAM native protein. Moreover, antibodies against SUAM-1fr produced the highest proportion of internalization inhibition of S. uberis to mammary epithelial cells. In conclusion, SUAM immunogenic and functionally relevant regions were identified and allowed to propose SUAM-1fr as a potential candidate for a subunit vaccine for S. uberis mastitis prevention.Fil: Perrig, Melina Soledad.     One-pot sonocatalyzed synthesis of sol–gel graphite electrodes containing gold nanoparticles for application in amperometric sensing

    We propose here a one-pot synthetic approach to prepare sol–gel graphite electrodes containing gold nanoparticles (AuNPs). At variance with the traditional synthesis, in which AuNPs are prepared in advance with respect to the silica matrix, they were here obtained directly inside the sol–gel, during its formation. Two reduction methods, namely chemical and thermal reduction, were used to achieve AuNPs starting from a suitable gold precursor, either consisting of NaAuCl 4 or HAuCl 4 . Different experimental parameters were tested in order to direct the synthesis of the material to the characteristics sought, namely Si/Au molar ratio, graphite (g): silane precursor (mL) ratio, chemical nature of the gold precursor and of the reductant, duration and temperature of thermal treatment. Sol–gel was prepared by means of sonocatalysis, in order to reduce the amount of solvent and time necessary for the hydrolysis step. Composition of the material, as well as shape, size and distribution of AuNPs inside the silica matrix was evaluated by spectroscopic and microscopic techniques. Furthermore, electrochemical tests allowed us to ascertain the good conductivity of the composite material and the electrocatalytic activity of AuNPs with respect to glucose and ascorbic acid oxidation. These tests demonstrated that the electrodes obtained by thermal reduction show the best performance in terms of sensitivity for the detection of these analytes, suggesting the possible application of this composite in the field of amperometric sensin

    Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Filosofia e Ciências Humanas, Programa de Pós-Graduação em História, Florianópolis, 2015.Este esboço de biografia procura citar algumas imagens de Otto Maria Carpeaux: construções biográficas de naturezas múltiplas, elaboradas em contextos, por atores e sob condições igualmente díspares. Está constituído a partir de uma visão crítica da História, o que permite que ?outras imagens?, fragmentárias e não monumentais, também tenham espaço. Em diálogo com o princípio da montagem, este esboço apresenta-se em duas partes. Na primeira, Imagens possíveis, estão citadas as imagens elaboradas em vida e post mortem acerca do austríaco-brasileiro que nasceu em Viena em 1900, se exilou no Brasil em 1939 e morreu no Rio de Janeiro, em 1978. Na segunda, Montagens possíveis, apresentam-se duas possibilidades de exercício biográfico: pela leitura alegórica do documentário O velho e o Novo (Otto Maria Carpeaux), entendido como instrumento de intervenção no contexto ditatorial brasileiro e de uma reelaboração biográfica concernentes às suas experiências europeias; e pelo Caderno de imagens críticas, registro dos encontros em Carpeaux pelo meio de imagens críticas produzidas a partir da cesura do presente.Abstract : This biographical sketch attempts to quote some images of Otto Maria Carpeaux: various types of biographical constructions, carried out in different contexts by disparate authors under conditions just as distinct. It stems from a critical view of history, allowing for ?other images? fragmented and non-monumental ? to share the space.In dialogue with the montage principle, this sketch has two parts. The first, Possible Images, quotes the images produced during and after the life of the Austrian-Brazilian, who was born in Vienna in 1900, went to Brazil in exile in 1939 and died in Rio de Janeiro in 1978. The second part, Possible Montages, presents two possibilities of a biographical exercise: through the allegorical reading of documentary O Velho e o Novo (Otto Maria Carpeaux), understood as an instrument of intervention in the Brazilian dictatorship context and as a biographical retelling of the author?s European experiences; and through my Scrapbook of Critical Images, a record of the encounters in Carpeaux through critical images produced from the caesura of the present

    Relative expression of genes associated with adhesion to bovine mammary epithelial cells by Streptococcus uberis

    Streptococcus uberis is one of the most prevalent environmental pathogens of bovine mastitis. Biofilm growth ability by S. uberis looks to depend first upon the adherence of cells to a surface. The S. uberis ability to adhere to mammary gland epithelia might provide an advantage to colonize the lactating mammary gland. The objectives of this study were (a) to select S.uberis strains according to their ability to form biofilm, (b) to determine adherence to and internalization into MAC-T cells and (c) to investigate the expression profile adherence genes in these S. uberis strains. For the assays, the MAC-T bovine mammary epithelial cell line was used. Relative expression of genes acdA, lmb, scpA, sua, fbp and lbp was quantified by RT-qPCR. We observed that the RC38 strain from clinical bovine mastitis showed in the six genes higher values than control in both conditions. While the strain with greater ability to adhere, from clinical mastitis and biofilm producer (RC29) evidenced higher values in group 1 (G1) (bacteria after the initial contact with MAC-T cells) and decrease in group 2 (G2) (both adhered and internalized bacteria) than control. Strains with a moderate or strong capacity for biofilm production showed significantly lower relative expression values in the G2. In all adherence associated genes, strain RC19 showed relative expression values incremented in G1, while in G2 decreased expression. In conclusion, we did not find a single profile of relative expression because the relative expression levels of each gene differed depending on the strain and the co-culture stage of S. uberis cells from which RNA was obtained.Fil: Fessia, Alumine Soledad.     The 21 years of a state of exception experienced by Brazil broke out on March 31, 1964, when the military deposed then President João Goulart, accused of defending communist ideals. In Tropical sol da liberdade, Ana Maria Machado weaves, from different perspectives (the memory of the protagonist Lena, the testimony of her mother, letters and reports) images of the horrors that marked the state of exception imposed by the coup. In this article, we analyze the literary construction of the work and the way in which the author, situated in the characteristics of contemporary literature, reflects on the fabric of the literary text while making literature a space of memory.Os 21 anos de estado de exceção vividos pelo Brasil eclodiram no dia 31 de março de 1964, quando os militares depuseram o então presidente João Goulart, acusado de defender ideais comunistas. Ana Maria Machado, em Tropical sol da liberdade, tece, a partir de diferentes perspectivas (a memória da protagonista Lena, o testemunho da mãe, cartas e reportagens) imagens dos horrores que marcaram o estado de exceção imposto pelo golpe.     Immune Neuroendocrine Interactions during a Fungal Infection in Immunocompetent or Immunosuppressed Hosts

    The yeast Candida albicans belongs to the microflora of healthy individuals, although it can infect a variety of tissues ensuing changes in the host's immune status. To evaluate the effect of neuroendocrine input on the early immune response during the fungal infection, we use a 3-day paradigm of chronic varied stress in Wistar rats infected with C. albicans. We find that stress mediators contribute to the spread of the fungus and downregulate critical functions of phagocytic cells at the infection site. Phenotypic and functional alterations of effector cells account for the decreased resistance to candidiasis and condition the development of the adaptive response. Stressed hosts exhibit a higher fungal burden in kidneys and livers associated with hyphal forms. The hepatic inflammatory reaction is compromised with severe steatosis, increment of functional enzymes, marked lipid peroxidation and hepatocyte apoptosis. Moreover, infection-related sickness symptoms are significantly increased by exposure to stress with anorexia, weight loss, lack of leptin and depletion of glycogen depots. Food deprivation exacerbates the liver injury.     10 pages, English and Spanish translation availableAlfonso X was king of “Castilla, León, Sevilla, Córdoba, Murcia, Jaén, and el Algarbe.” As evidenced by his title, he came to have possession of various kingdoms in Iberia. He was born in Toledo in 1221 and died in Seville in 1284, at 63 years of age. He is called the Learned King because he was an author, poet, musician, and historian, and because he supported artists and translators. Furthermore, he employed Christians, Jews, and Muslims in his translation workshop in Toledo. His Cantigas de Santa Maria is a repertoire of songs compiled in four manuscripts between 1257 and 1283. The Cantigas consist of 420 narrative songs that focus on the Virgen Mary and the miracles that she performs across Christendom. Originally, the text was written in Galician-Portuguese, from the northeast region of Iberia. Galicia is the site of Santiago de Compostela, the final destination of the pilgrimage route known as the Camino de Santiago (Saint James’s Way). Each story is comprised of text, images, and music. The combination of texts and images portrays day-to-day life and, often, the less-documented common peoples of the Middle Ages in Europe. This unit, edited by Diane Burke Moneypenny and Allison Carberry Gottlieb, contains the original text in Galician-Portuguese of Cantiga 5, “How the Virgin Mary helped the Empress of Rome.” The Spanish version (for use in contexts where the language of instruction is Spanish) has an introduction, notes, and translation into modern Spanish (by Sol Miguel-Prendes).     Genistein loaded in self-assembled bovine serum albumin nanovehicles and their effects on mouse mammary adenocarcinoma cells

    Antitumor activity of plant-derived flavonoids has been researched during recent decades. Among them, genistein (Gen) stands out for showing cytotoxic activity against breast cancer cells. However, its low water solubility, limited bioavailability, and fast metabolism hinder its administration in chemopreventive therapies. To overcome these obstacles, bovine serum albumin nanovehicles (BSAnp) were obtained by a heat-induced self-assembly process at 70 °C and two aqueous medium pH (9.0 and 11.0) and assayed for the Gen loading. Thus, in this work, Gen loading in BSAnp was studied by spectroscopic techniques and compared with the one obtained for its stereoisomer, chrysin (Chrys). Results revealed that Gen binds to BSAnp via fluorescence quenching mechanism forming inclusion complexes. Compared to Chrys, Gen binding to BSAnp involved more molecules, whereas the association constant was similar for both flavonoids. In general, flavonoid loading in protein systems was strongly affected by the combined effects of BSA conformational state (native vs. aggregated), nanovehicle size, and flavonoid chemical structure. To evaluate the antitumor properties freeze-dried powders were obtained, and they were assayed in vitro after reconstitution by XTT technique and Annexin V-FITC flow cytometry against mouse mammary adenocarcinoma F3II cells. Gen-loaded BSAnp produced a significant decrease in cell viability compared with unloaded BSAnp systems, being the highest cytotoxic effects found for the lowest sized Gen-loaded BSAnp. The leading cytotoxicity mechanism for Gen-loaded systems was apoptosis. Summarizing, it can be concluded that BSAnp constitute versatile nanovehicles for potential flavonoid incorporation in pharmaceutical and nutraceutical matrices.Fil: Ferrado, Joana Belén.     In vitro gastrointestinal digestion and cytotoxic effect of ovalbumin-conjugated linoleic acid nanocomplexes

    The aim of this work was to examine the behavior of conjugated linoleic acid (CLA) delivery systems based on ovalbumin (OVA) and their derived nanoparticles (OVAn1 and OVAn2), under static in vitro gastrointestinal digestion model. In addition, potential cytotoxic effect of these inclusion complexes on a human colon cancer cell line (HT-29) was evaluated. OVA was resistant to gastric and intestinal digestion, while OVA nanoparticles were very susceptible to digestive enzymes hydrolysis. Particle size distribution (PDS) and sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE) for OVA evidenced the presence of a protein fragment of similar size after simulated digestive process. Conversely, for nanoparticles, partial and total hydrolysis in gastric and intestinal phases, respectively, was evidenced. After in vitro gastrointestinal digestion, released CLA (RCLA) was assayed. In case of OVA, as CLA carrier, RCLA was 37%, while for OVA nanoparticles, lower RCLA values (~10–20%) were obtained. From cytotoxic assays, it was observed that CLA molecule was responsible for cell death, whereas OVA or their derived nanoparticles were not cytotoxic on HT-29 cells. On the other hand, flow cytometry analysis revealed that main death mechanism for CLA, and their inclusion complexes was apoptosis. OVA-CLA and OVAn1-CLA inclusion complexes displayed the highest potential cytotoxic activity and apoptotic index. Information derived from this work could be relevant for the design of CLA delivery systems as promising nanosupplements for production of new functional and excipient foods for both prevention and control of colon cancer.Fil: Visentini, Flavia Fátima.     Caminhos Cruzados e Um Lugar ao Sol: o projeto literário de Erico Verissimo

    Esta dissertação trata da relação entre autor-criador e texto literário em Caminhos Cruzados (1935) e Um Lugar ao Sol (1936) de Erico Verissimo, sob as perspectivas do desdobramento de uma narrativa em outra e a de personagens-escritoras partilhando a função autoral com Verissimo-criador, posicionado na fissura entre o escritural e o biográfico. Questionávamos sobre o significado dessa multiplicação de vozes autorais no projeto verissiano e os vínculos que estabelecem com sua posição de escritor no contexto do Neorrealismo dos anos 30. Elaboramos as hipóteses a partir de dois pressupostos: o de personagens-escritoras revelando-se como dobras do próprio autor em uma dinâmica de encaixe entre narrativas e o da saga como núcleo estruturante de um projeto literário em processo de (re)escrita. Os fundamentos teóricos partiram dos estudos de Foucault, Bakhtin, Barthes e Giorgio Agamben sobre a função-autor, do conceito de encaixe de Todorov e do de saga de Johnni Langer, além de estudos críticos como os de Antonio Candido, Flávio Loureiro Chaves e Maria da Glória Bordini. Na análise, foram selecionados pares de personagens-escritoras, Noel-Fernanda e Vasco- Clarissa, que apontavam para o confronto entre projetos literários numa relação de afastamento-aproximação do engajamento social, refletindo o conflito de Verissimo no contexto neorrealista de 1930. Observou-se, também, o encaixe entre narrativas e entre personagens-autoras, ampliando o referencial de análise que abrangeu as demais obras do autor. Isto revelou uma arquitetônica cuja raiz é a saga, vista sob nova faceta: aquela que se tece entre famílias de narrativas. Concluímos que Caminhos Cruzados e Um Lugar ao Sol instauram a reflexão entre autor e texto literário por meio da migração de personagens e da tessitura de um romance no outro, fazendo da saga o princípio estético composicional do projeto verissiano, comprometido com o fazer artístico e, ao mesmo tempo, com o homem e a sociedadeThis dissertation deals with the relationship between author-creator and literary text Caminhos Cruzados (1935) and Um Lugar ao Sol (1936) written by Erico Verissimo, from the perspectives of the unfolding of a narrative in another, and the characters-writers sharing authorial function with Verissimo-creator, positioned in the crack between the book-biographical. We developed the hypotheses from two assumptions: the characters-writers appearing as the author's own folds into a dynamic fit between narratives and the "saga" as a structural core of a literary project in (re)writing process. The theorical foundation left of Foucault, Bakhtin, Barthes and Giorgio Agamben on the author-function studies, the concept of plug-in by Todorov and the saga of Johnni Langer, and critical studies such as Antonio Candido, Flávio Loureiro Chaves and Maria Glory Bordini. In the analysis, we selected pairs of characters-writers, Noel-Fernanda and Vasco-Clarissa, who pointed to the confrontation between literary projects a ratio of removal-approach of social engagement, reflecting the Verissimo s conflict in the context of the neorealist in the 1930s. There was also the fit between narratives and characters betweenauthors, expanding the analytical framework covering the other works of the author. This revealed an architectural whose root is the saga, seen under a new facet: that which is woven between families of narratives. We conclude that Caminhos Cruzados and Um Lugar ao Sol established a reflection between author and literary text through migration of characters and the web of a novel in the other, making the "saga" the aesthetic compositional principle of Verissimo s project, committed to making art and at the same time with the individual and societyCoordenação de Aperfeiçoamento de Pessoal de Nível Superio
