Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Bestimmung der Hämoxygenase Aktivität in BMC1 Melanoma Zellen
Diplomarbeit - Veterinärmedizinische Universität Wien - 2023Das maligne Melanom ist eine gefährliche Form von Hautkrebs, die aus den pigmentproduzierenden Melanozyten entsteht und Tumore sowie aggressive Metastasen bildet. Beim Menschen weisen die meisten Melanome Mutationen im BRAF-Gen (BRAF V600) auf, die zu einer dauerhaften Aktivierung von Zellteilungssignalen führt. In experimentellen Studien wurde gezeigt, dass die konstitutive BRAF-Aktivierung mit einer erhöhten Expression von HO-1 assoziiert ist. Es ist daher anzunehmen, dass Melanomazellen von einer aktiven HO profitieren. Metastasierende Melanome, die eine BRAF V600 Mutation aufweisen, können mit BRAFi, wie Vemurafenib, behandelt werden, zeigen jedoch oft nach einiger Zeit eine Resistenz gegen das Medikament. Da bekannt ist, dass HO in tumorösem Gewebe vermehrt exprimiert wird und außerdem zur Metastasierung und Angiogenese von Melanomen beitragen kann, ist es möglich, dass die HO auch eine Rolle auch in der Resistenzentwicklung gegen BRAFi spielt. In anderen Tumorzellen konnte bereits gezeigt werden, dass eine HO-1 Erhöhung mit der Ausbildung der Chemoresistenz in Verbindung steht. Dabei wird vermutet, dass die Produkte, die in der HO-Reaktion gebildet werden, die Zellen gegen den oxidativen Stress und die Induktion der Apoptose schützen. Ziel der vorliegenden Diplomarbeit war es daher zu untersuchen, ob die HO-Aktivität auf die Gabe von Vemurafenib anspricht, und ob sie sich unter der Behandlung mit der Zeit verändert. Daher sollte die HO-Aktivität in Zellen der Linie BMC1-M1, die mit und ohne Vemurafenib behandelt wurden, in einer zeitlichen Kinetik erfasst werden. Es wurde vermutet, dass die HO- Aktivität der Zellen unter Einfluss von Vemurafenib zunächst gehemmt wird, sich aber im Anschluss wieder normalisiert, oder sogar ansteigt. BMC1-M1 Zellen wurden in drei Behandlungsgruppen (Kontrolle (Vehikel), Vemurafenib (10μM) und Hämin (1mM) eingeteilt, jeweils für 6h, 24h und 48h inkubiert. Die Behandlung mit Hämin wurde als Positivkontrolle mitgeführt. Für die Bestimmung der HO-Aktivität wurde ein gekoppelter biochemischer in vitro Assay eingesetzt. Dabei wurde als Maß für die HO-Aktivität die Menge an BR herangezogen, die vom Zellhomogenat in 30‘ je mg Protein gebildet wird. Zunächst wurden die experimentellen Rahmenbedingungen kontrolliert und sichergestellt, dass die im Assay entstandene BR-Menge einen direkten Rückschluss auf die zugrundeliegende HO- Aktivität erlaubt, da das durch die HO umgewandelte Hämin vollständig in BR überführt werden konnte. Dazu wurde auch das der HO nachgeschaltete Enzym, Biliverdinreduktase, auf eine ausreichend hohe Aktivität überprüft. Die BR-Menge wurde mittels Photometrie quantifiziert und die Proteinmenge in jedem Inkubations-Ansatz mit der Methode nach Bradford bestimmt. Im Vergleich zur Kontrollgruppe führte die Hämin- Behandlung in allen Zeitpunkten zu einer signifikant höheren HO-Aktivität. Dies zeigt, dass HO in BMC1-M1 induziert werden kann. Die Vemurafenib-Behandlung führte demgegenüber bereits nach 6h zu einer moderaten Hemmung der HO-Aktivität (etwa 25%), welche nach 24h signifikant war. Nach 48 hatte sich die HO-Aktivität jedoch wieder normalisiert, was nahelegt, dass die Hemmung zu diesem Zeitpunkt bereits überkommen wurde. Die erhobenen Daten bestätigen somit die Hypothese, dass die Behandlung mit Vemurafenib die HO-Aktivität nur transient hemmt, und die Hemmung im Späteren überkommen werden kann. Dies könnte an einer kompensatorischen Induktion der HO1 durch den Vemurafenib-induzierten Zellstress liegen. Um diese Vermutung zu bestätigen, müssten weitere Untersuchungen z.B. zur Genexpression durchgeführt werden. Jedenfalls zeigen die erhobenen Daten, dass Vemurafenib nur kurzzeitig die Bildung der zellschützenden HO-Produkte unterdrücken kann, und diese nach länger-andauernder Behandlung mit BRAFi wieder zum Zellschutz zur Verfügung stehen. Diese Ergebnisse stellen somit eine wichtige Grundlage zur Frage der Rolle der HO-Aktivität in der Krebstherapie dar.The malignant melanoma is a dangerous form of skin cancer that arises from pigmentproducing melanocytes and forms tumours as well as aggressive metastases. In humans, most melanomas exhibit mutations in the BRAF gene (BRAF V600), resulting in a sustained activation of cell division signals. Experimental studies have shown that constitutive BRAF activation is associated with increased expression of HO-1. It is therefore assumed that melanoma cells benefit from active HO. Metastatic melanomas harbouring a BRAF V600 mutation can be treated with BRAF inhibitors (BRAFi) such as Vemurafenib, but they often develop resistance to the medication over time. Since HO is known to be overexpressed in tumorous tissue and can contribute to the metastasis and angiogenesis of melanomas, it is possible that HO also plays a role in the development of resistance to BRAFi. In other tumour cells, an increase in HO-1 has been shown to be associated with the development of chemoresistance, with the products formed in the HO reaction believed to protect cells against oxidative stress and apoptosis induction. Therefore, the aim of this thesis was to investigate whether HO activity responds to the administration of Vemurafenib and whether it changes over time during treatment. The HO activity in BMC1-M1 cells, treated with and without Vemurafenib, was measured over a temporal kinetic period. It was hypothesized that under the influence of Vemurafenib, the HO activity of the cells would initially be inhibited but would subsequently normalize or even increase. BMC1-M1 cells were divided into three treatment groups (control (vehicle), Vemurafenib (10μM), and hemin (1mM), incubated for 6h, 24h, and 48h, and used for determining HO activity using a coupled biochemical in vitro assay. The amount of bilirubin (BR) formed by the cell homogenate per mg of protein in 30 minutes was used as a measure of HO activity. First, the experimental conditions were controlled to ensure that the amount of BR generated in the assay allowed for a direct inference of the underlying HO activity since the Hemin converted by HO is completely converted to BR. Additionally, the activity of biliverdin reductase, the enzyme downstream of HO, was checked to be sufficiently high. The amount of BR was quantified by photometry and the protein amount in each incubation assay was determined using the Bradford method. Compared to the control group, hemin treatment resulted in significantly higher HO activity at all time points, indicating that HO can be induced in BMC1-M1 cells. In contrast, Vemurafenib treatment led to a moderate inhibition of HO activity (approximately 25%) after 6 hours, which became significant after 24 hours. However, after 48 hours, HO activity normalized again, suggesting that the inhibition had been overcome by that time. Thus, the collected data confirm the hypothesis that Vemurafenib treatment only transiently inhibits HO activity, and the inhibition can be overcome later. This could be due to compensatory induction of HO1 by the Vemurafenib-induced cellular stress. Further investigations, such as gene expression studies, would be necessary to confirm this assumption. Nevertheless, the data obtained demonstrate that Vemurafenib can only temporarily suppress the formation of cell-protective HO products, which become available again for cellular protection after prolonged treatment with BRAFi. These results thus provide an important basis for the question of the role of HO activity in cancer therapy
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Effekte von "OMNI BIOTIC® PANDA" auf die Proliferation von regulatorischen T-Zellen im Schwein
Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Hintergrund: Antibiotikaresistenzen sind eine wachsende Bedrohung für unser Gesundheitssystem, gleichzeitig war bzw. ist die Anwendung von antibakteriellen Substanzen für die Nutztierwirtschaft unumgänglich. Alternativen wie Probiotika sind deshalb notwendiger denn je. OMNI BIOTIC ® PANDA (OBP) ist ein Probiotikum aus der Humanmedizin, was unter anderem zu einer vermehrten Proliferation von regulatorischen T-Zellen führen soll.
Methoden: Es wurden isolierte Zellen aus peripher entnommenen Schweineblut (n=4) in mit OBP inkubiert. Insgesamt gab es drei Ansätze: (1) PBMC (Peripheral Blood Mononuclear Cells), entspricht 500 μl PBMC + 500 μl Medium; (2) Leerwert, entspricht 100 μl Standard + 50 μl Standartlösung; (3) 1:2, entspricht 500 μl PBMC + 500 μl OBP. Die Zellen wurden für die Fluoreszenzmikroskopie mit Antikörpern für die Erkennung T-Zell-spezifischer Oberflächenproteine CD4 und CD25 versetzt und per Programm ImageJ ® ausgezählt. Die Genexpression für CD25 und FOX P3 wurde mittels qPCR gemessen.
Ergebnisse: Fluoreszenzmikroskopie: Im Durchschnitt aller Schweine lässt sich ein deutlicher prozentueller Anstieg von CD4+-Zellen sehen und ein geringer prozentueller Abstieg von CD25+-Zellen der behandelten Zellen im Vergleich zum Kontrolldurchgang. Es gibt individuelle Unterschiede. Bei Schwein 1 gibt es einen geringeren Anteil von CD4+- und CD25+-Zellen, bei Schwein 2 und 4 steigen die Anteile beider Zellkategorien der behandelten Zellen. Bei Schwein 3 steigt der Anteil der CD4+- Zellen, aber der Anteil der CD25+-Zellen sinkt. Die Unterteilung der CD25 Oberflächenproteine in CD25 high und dim lässt keine Regel erkennen.
qPCR: Die behandelten Zellen zeigen im Durchschnitt im Vergleich mit den unbehandelten eine höhere Genexpression für FOX P3 und CD25. Auffällig ist, dass die behandelten Zellen von Schwein 1 in der Fluoreszenzmikroskopie und in der qPCR einer Verringerung in allen Markern zeigt.
Conclusio: Die erhobenen Ergebnisse deuten auf eine geänderte Antwort des T-Zell basierenden Immunsystems durch eine OBP-Behandlung hin. Vollendes kann jedoch nicht behauptet werden, dass OBP® zu einer vermehrten Proliferation von porzinen regulativen T-Zellen führt. Dafür bedarf es Folgeversuche mit einer größeren Stichprobenanzahl und optimiertem Versuchsdesign. Die Ergebnisse der vorliegenden Arbeit liefern dafür eine wichtige Basis
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
