Method: This study employs qualitative research and an abductive approach to achieve its objectives. Data collection is categorized into secondary research, involving a review of existing literature, and primary research, which includes a case study conducted through interviews and observations. Both semi-structured and unstructured interviews were employed with the case company and the benchmarking organization. The use of benchmarking allows for the comparison of organizational practices, challenges, and outcomes, aiding in identifying areas for improvement and gaining insights into effective strategies. Conclusion: Addressing the first research question, the material flow can be improved by transitioning to a pull-based system and implementing concepts such as supermarkets and point-of-use storage. Furthermore, integrating automated systems for tracking cart availability and establishing clear protocols for standardized work procedures are crucial.     ABSTRACT   Water is one of the primary needs in human life, starting from daily activities such as drinking, bathing, cooking, to the field of industrial processing on a large scale, therefore good quality water is needed. Depot Windy is a refill drinking water business produced by PT. Lautan Bening which has been operating since 2016, with daily sales of around 200-300 gallons issued from 07.00 AM - 19.00 PM. However, there are problems related to the distribution of information between administrators, delivery staff and customers, so unwanted things often occur, such as gallons being stolen, gallons not being picked up, and payment transactions being delayed. In this journal, the author describes the development of an android-based gallon water ordering application using the SDLC (Software Development Life Cycles) method and the Waterfall development method which is connected via the user's cellphone. By creating good communication facilities, it can make it easier for sellers and customers to make transactions online and in real time, make it easier for customers to know the availability of gallons of water, and achieve the company's long-term goal of increasing sales

    This research describe the forms of resistance by female characters in novel "Bako" by Darman Moenir through the lens of critical discourse analysis. The study is grounded Sara Mills\u27 theory to analyze the injustices faced by women. The primary issue of this research is how women, either individually or as representatives of a group, articulate their rights, and how their struggle is portrayed through the selection of words, sentences, and narrative forms presented to audience. The analytical technique in this research is Sara Mills\u27 discourse analysis.The research delves into portrayal of women in the novel, highlighting the resistance movements undertaken by the character Ummi, who serves as the protagonist challenging the norms of the matrilineal culture prevalent in Minangkabau society. As both the object and reader, audience is led by the author to become aware of the errors that often marginalize women in terms of tradition, social norms, and culture.     ABSTRAK Penelitian ini bertujuan untuk merancang sebuah kurikulum khusus untuk pengajaran Bahasa Arab di Pondok Pesantren Ma’had Islamy berdasarkan kurikulum multidimensi. Tidak adanya kurikulum terpadu sebagai pedoman bagi guru dan siswa dalam melaksanakan aktifitas belajar mengajar yang efektif di pesantren adalah dasar pijakan penelitian ini. Untuk itu, peneliti melihat bahwa kurikulum multidimensi sangat cocok dengan tujuan utama pembelajaran Bahasa Arab dan motivasi siswa untuk belajar Bahasa Arab di sana. Penelitian ini adalah penelitian pengembangan dengan menggunakan pendekatan kualitatif, serta memakai langkah-langkah penelitian pengembangan yang dikemukakan oleh Sugiono dan langkah-langkah pengembangan kurikulum yang dikemukakan oleh Jack C. Richard dengan modifikafi. Adapun subjek coba dari penelitian ini terdiri dari 45 orang siswa dan 10 orang guru di Pondok Pesantren Ma’had Islamy. Teknik pengumpulan data menggunakan observasi, wawancara, dan angket. Sedangkan pengolahan datanya menggunakan teknik analisis isi, analisis deskriptif dan analisis statistik deskriptif. Hasil penelitian ini adalah: (1) Kurikulum berbasis multidimensi untuk pengajaran Bahasa Arab di Ma’had Islamy yang berisi: gambaran umum, profil Ma’had Islamy, kalender pendidikan, kurikulum multidimensi, tujuan pembelajaran Bahasa Arab, isi (materi pelajaran), aktifitas pembelajaran, metode- metode pembelajaran, media yang digunakan, evaluasi dan ujian, buku sumber pengajaran dan silabus. (2) Hasil dari validasi yang telah dilakukan oleh para validator kurikulum pembelajaran Bahasa Arab terhadap kurikulum ini secara umum mencapai 92,7% “baik sekali”. Menurut para validator, isi kurikulum yang didesain ini jelas, memenuhi syarat yang ditentukan, sesuai dengan tingkatan siswa dan mengandung unsur-unsur kurikulum multidimensi serta menjaga karakteristiknya. Dan hasil analisis data yang didapat melalui angket ataupun wawancara dengan para guru menunjukkan bahwa kecocokan kurikulum ini untuk diterapkan di Ma’had Islami mencapai tingkat 95,24%. Ketua yayasan, pimpinan pondok dan para guru di pondok pesantren ini telah sepakat untuk memakai dan menerapkan kurikulum ini sebagai pedoman bagi mereka dalam pelaksanaan aktivitas belajar mengajar setelah dilakukan perbaikan seperlunya. ABSTRACT This research was intentionally performed to design a specific curriculum for teaching process of Arabic language at the Ma’had Islamy Islamic Boarding School based on multidimensional curriculum. The reason of choosing this topic was the absence of an integrated curriculum which can be used as a guide for teachers and students to perform an effective learning and teaching process in the Ma’had Islamy. The author assumed that the multidimensional curriculum is relevant with the main goals of Arabic teaching in this school and useful to motivate the students to learn Arabic language in the Ma’had Islamy. This development research with qualitative approach, used a model proposed by Sugiono and the curriculum development model proposed by Jack C. Richard with modification. The subjects of the research were 45 students and 10 teachers from the Ma’had Islamy Islamic Boarding School. The data was collected by using observation, interview, and questionnaire. The data was then analysed by using a content analysis technique, a descriptive analysis and a descriptive statistic analysis. The result of this study says that: (1) the proposed multidimensional curriculum for the Arabic language teaching at the Ma’had Islamy consists of: general overview, profile of the Ma’had Islamy, academic calendar, multidimensional curriculum, goals of Arabic language learning, contents (subject material), learning activities, learning methods, media, evaluation and examinations, teaching resources and sillaby . (2) The final assessment of this designed curriculum from the validators resulted the score of 92,7%, which means “very good”. The validators stated that the content of this curriculum is clear and has fulfilled the requisite requirement, relevant with the student levels, contains of multidimensional curriculum elements, and keep it’s characteristics. The result of data analysis obtained from the questionnaires and interview with the teachers shows that the suitability of the designed curriculum to be applied at the Ma’had Islamy has a score of 95,24 %. The head of the organisation, the head of Islamic boarding school, and teachers at the school have agreed to use and apply the designed curriculum as a guide for them during the teaching and learning process of Arabic language after necessary revision

    Abstract 1185: H3B-8800, a novel orally available SF3b modulator, shows preclinical efficacy across spliceosome mutant cancers

    Abstract Genomic characterization of hematologic and solid cancers has revealed recurrent somatic mutations affecting genes encoding the RNA splicing factors SF3B1, U2AF1, SRSF2 and ZRSR2. Recent data reveal that these mutations confer an alteration of function inducing aberrant splicing and rendering spliceosome mutant cells preferentially sensitive to splicing modulation compared with wildtype (WT) cells. Here we describe a novel orally bioavailable small molecule SF3B1 modulator identified through a medicinal chemistry effort aimed at optimizing compounds for preferential lethality in spliceosome mutant cells. H3B-8800 potently binds to WT or mutant SF3b complexes and modulates splicing in in vitro biochemical splicing assays and cellular pharmacodynamic assays. The selectivity of H3B-8800 was confirmed by observing lack of activity in cells expressing SF3B1R1074H, the SF3B1 mutation previously shown to confer resistance to other splicing modulators. Although H3B-8800 binds both WT and mutant SF3B1, it results in preferential lethality of cancer cells expressing SF3B1K700E, SRSF2P95H, or U2AF1S34F mutations compared to WT cells. In animals xenografted with SF3B1K700E knock-in leukemia K562 cells or mice transplanted with Srsf2P95H/MLL-AF9 mouse AML cells, oral H3B-8800 treatment demonstrated splicing modulation and inhibited tumor growth, while no therapeutic impact was seen in WT controls. These data were also evident in patient-derived xenografts (PDX) from patients with CMML where H3B-8800 resulted in a substantial reduction of leukemic burden only in SRSF2-mutant but not in WT CMML PDX models. Additionally, due to the high frequency of U2AF1 mutations in non-small cell lung cancer, H3B-8800 was tested in U2AF1S34F-mutant H441 lung cancer cells. Similar to the results from leukemia models, H3B-8800 demonstrated preferential lethality of U2AF1-mutant cells in vitro and in in vivo orthotopic xenografts at well tolerated doses. RNA-seq of isogenic K562 cells treated with H3B-8800 revealed dose-dependent inhibition of splicing. Although global inhibition of RNA splicing was not observed; H3B-8800 treatment led to preferential intron retention of transcripts with shorter and more GC-rich regions compared to those unaffected by drug. Interestingly, H3B-8800-retained introns commonly disrupted the expression of spliceosomal genes, suggesting that the preferential effect of H3B-8800 on spliceosome mutant cells is due to the dependency of these cells on expression of WT spliceosomal genes. These data identify a novel therapeutic approach with selective lethality in leukemias and lung cancers bearing a spliceosome mutation. Despite the essential nature of splicing, cancer cells without a spliceosome mutation were less sensitive to H3B-8800 compared with potent eradication of mutant counterparts. H3B-8800 is currently undergoing clinical evaluation in patients with MDS, AML, and CMML.
