No full textThe objective of this work is to situate the writer Graciliano Ramos in the context of a study where we compare his adult production, more specifically the novel São Bernardo, and A terra dos meninos pelados, one of his works which is aimed at children. When considering the dynamic relationship established between author, text and reader, and knowing that the writer, in building work, is undergoing a creative intent and purpose of an aesthetic, ideological and social basis, and conscious of the need to seek effects of meaning, convey his thoughts and encourage interpretations, we try to answer a question that seems fundamental: are the two branches of Graciliano’s literature (adult and child) equally significant?A nossa intenção neste trabalho é situar o escritor Graciliano Ramos em um contexto de estudo onde se coteje a sua produção adulta, mais especificamente o livro São Bernardo, com A terra dos meninos pelados, um de seus trabalhos destinado às crianças. Ao considerarmos as relações dinâmicas que se estabelecem entre autor, texto e leitor, e sabendo que o escritor, ao elaborar o seu trabalho, o faz submetido a uma intenção criativa e a um propósito estético, ideológico e social, cônscio da necessidade de buscar efeitos de sentido, transmitir o seu pensamento e incitar interpretações, tentamos responder a uma questão que nos parece fundamental: podemos considerar os dois ramos literários de Graciliano (o adulto e o infantil) igualmente significativos
El desempeño de la RT-PCR con diferentes nucleótidos análogos y diferentes electrolitos
No full textLa transcripción reversa seguida de la reacción en cadena de la polimerasa (RT-PCR por sus siglas en inglés) se ha convertido en una de las técnicas más usadas en biología molecular para el análisis de secuencias de ARN. Esta técnica no es confiable cuando las enzimas involucradas, tanto la transcriptasa reversa de MMLV como la polimerasa Taq, se ven estorbadas en su avance de extensión cuando se encuentran con secuencias que se plegaron en forma de G-tetraplex (estructura G4) dentro de la secuencia molde de interés. Estas estructuras se estabilizan fuertemente en presencia de cationes monovalentes, siendo el más efectivo en este sentido el K+, ión rutinariamente usado en los procedimientos de RT y PCR. Esto resulta problemático pues, ante la estructura G4, las enzimas no pueden completar el copiado correcto de la secuencia molde. Por otra parte se ha encontrado que iones de litio o cesio ejercen el efecto de estabilización en menor medida. En este trabajo de tesis, se evaluó el uso de cationes monovalentes diferentes al potasio, como litio y cesio, para su aplicación en reacciones de RT y de PCR, así mismo se usó el nucleótido análogo c7dGTP (7-desaza-2´-desoxiguanosina 5’-trifosfato), y se determinó su efecto en la PCR. Se evaluaron dos enzimas, la transcriptasa reversa de MMLV y la ADN polimerasa Taq, en cuanto a su capacidad para realizar su trabajo de extensión en presencia de sales diferentes al cloruro de potasio, tanto en regiones que no pueden formar estructuras G4 como en regiones que sí forman dichas estructuras. Inicialmente se evaluó a la polimerasa Taq en PCR con un ADN molde de fácil amplificación y se demostró que realiza su trabajo de extensión en presencia de CsCl, LiCl y NaCl con rendimientos de amplificación similares a los obtenidos con KCl. Posteriormente se evaluó el desempeño en ADNs molde de difícil amplificación, donde la polimerasa Taq mostró el mejor desempeño en presencia de CsCl, mientras que con LiCl el rendimiento fue muy bajo y para KCl el rendimiento fue nulo. En los experimentos de transcripción reversa con un ARN molde incapaz de formar estructuras G4 se mostró un desempeño similar tanto para KCl como para CsCl; en cambio, en transcripción reversa de un ARN molde sintético apto para la formación de una estructura G4, la transcriptasa reversa de MMLV realizó una conversión casi total de ARN molde a ADN complementario en presencia de CsCl, mientras que con KCl la conversión fue prácticamente nula, y en presencia de LiCl o en ausencia de cloruro metálico añadido la transcriptasa reversa realizó un copiado parcial de ARN molde a ADN complementario . En las reacciones de PCR con el análogo c7dGTP la polimerasa Taq mostró que puede incorporar parcialmente o totalmente a este análogo, pero esto no demostró ningún beneficio para la amplificación de un ADN molde apto para formar la estructura G4. El CsCl es una buena opción para reemplazar al KCl en reacciones de RT-PCR en el caso de secuencias que tienen problemas de formación de estructuras G4, favoreciendo a la polimerasa Taq y a la transcriptasa reversa de MMLV en su trabajo de extensión, mientras que con KCl dichas enzimas se pueden ver afectadas originando de esta manera amplificaciones incompletas o nulas. La técnica de RT-PCR puede ser modificada, mejorando su reproducibilidad en determinadas situaciones problemáticas, obteniéndose resultados confiables de fácil interpretación en biología molecular

No full textAbstract. With model-driven development being on the verge of becoming an industrial standard, systematic development strategies based on safe model transformations is a demand. However, the lack of a formal semantics makes the application of such transformations inadequate, especially transformations that must take into account changes in both behavioural and structural diagrams. As part of the Master thesis briefly summarised in this paper [Ramos, 2005], we have defined a semantics for UML-RT through a mapping into a formal notation, called Circus. Based on the semantics, we have proposed a set of transformation laws that aims to systematise the evolution of UML-RT models during development, with preservation of both static and dynamic aspects. Soundness and completeness of the laws are also addressed, and a case study has been developed to illustrate the overall approach

Customs foreign trade manual writing and implementation for interns Case Company: Cedillo Ramos
Get PDFThe purpose of the thesis was to create a feasible and suitable alternative for Cedillo Ramos to determine the opportunity areas that the interns have to improve during the training process. One objective was to study the company's current situation by analysing it with qualitative and quantitative methods. Another goal relied on the design of a foreign trade training manual so it could be implemented to increase customer satisfaction and to facilitate the training process.
The theoretical framework was based on literature regarding customs legislation, supply chain management, service design, and six sigma philosophy. Most of the used authors are leaders on the field, such as Richard Gerson, Elizabeth Cudney and Omer Artun. However, some material retrieved to write the service design title was taken from lectures provided by Georg Berger, lecturer in the International Business degree programme.
The study was executed in an electronic survey sent to the most important and loyal clients. Moreover, by gathering data directly from the company's email, the author analysed the operational knowledge that the interns have in Cedillo Ramos. Eleven different clients were studied in-depth with their answers to the surveys and their previous foreign trade operations to interpret the results. Nonetheless, the analytical approach methodology was a mix of qualitative and quantitative data collection through long-distance interviews, surveys, and meetings to analyse the results with the commissioning company.
The findings revealed that the training process that the company was implementing ten years ago is no longer helpful and that the implementation of the manual revealed positive numbers regarding the leading time of the operations, the operational knowledge of the interns and customer satisfaction.
To sum up, there might be some room to pay close attention to the interns' process and perform their tasks when they are hired. By combining several activities, the company's culture will become a safer place for students who can be part of these foreign trade consultancy organisations and retain the loyal and vital customers that the company has.The Operational Manual delivered to Cedillo Ramos is in Spanish due to the company's requirements regarding the training process of the interns

No full textThe dataset provides the h-index and Author Impact Factor (AIF) from 2015-2019 and 5-Year (AIF) of 300 neurosurgical authors. The h-index was retrieved from Scopus in September 2020. The data required to calculate the AIF was retrieved from Scopus from September 2020 to April 2021. There are three groups of neurosurgical authors: (1) the 100 authors with the greatest number of articles of all time within general neurosurgical journals (“Experienced”); (2) the 100 authors with the greatest number of articles during the 2015-2019 period within general neurosurgical journals (“Trending ”); and (3) the 100 Post Graduation Year 7 (PGY 7) neurosurgical residents with the highest h-index (“Amateur”).
