Tobacco TGA factors differ with respect to interaction with NPR1, activation potential and DNA-binding properties
In higher plants, as-1-like cis elements mediate auxin- and salicylic acid-inducible transcription. Originally found in viral and T-DNA promoters, they are also functional elements of plant promoters activated during the defence response against pathogens. Tobacco bZIP transcription factor TGA1a was the first recombinant protein shown to bind to as-1. cDNAs for two novel tobacco as-1-binding bZIP proteins (TGA2.1 and TGA2.2) were isolated. Revealing a high degree of amino acid identity in the bZIP domain (89%) and the C-terminus (79%), the two TGA2 factors differ remarkably with respect to the length of the N-terminus (170 amino acids in TGA2.1 versus 43 amino acids in TGA2.2). TGA2.1 and TGA2.2, but not TGA1a, interacted with ankyrin repeat protein NPR1, a central activator of the plant defence response. In contrast, TGA2.1 and TGA1a, but not TGA2.2, functioned as transcriptional activators in yeast. Apart from conferring transcriptional activation, the N-terminal domain of TGA2.1 led to reduced in vitro as-1-binding activity and almost completely abolished binding to one half site of this bifunctional element. When being part of a heterodimer with TGA2.2, TGA2.1 was efficiently recruited to a single half site, though double occupancy of the element was still preferred. In contrast, TGA1a preferred to bind to only one palindrome, a feature that was also maintained in heterodimers between TGA1a and TGA2.1 or TGA2.2
Interaction of NIMIN1 with NPR1 modulates PR gene expression in Arabidopsis
The Arabidopsis thaliana NONEXPRESSER OF PR GENES1 (NPR1, also known as NIM1) protein is an essential positive regulator of salicylic acid (SA)-induced PATHOGENESIS-RELATED (PR) gene expression and systemic acquired resistance (SAR). PR gene activity is regulated at the level of redox-dependent nuclear transport of NPR1. NPR1 interacts with members of the TGA family of transcription factors that are known to bind to SA-responsive elements in the PR-1 promoter. In an attempt to identify proteins involved in SA-mediated signal transduction, we previously described the isolation of three novel genes encoding distinct albeit structurally related proteins designated NIMIN1 (for NIM11-INTERACTING1), NIMIN2, and NIMIN3 that interact with NPR1 in the yeast two-hybrid system. Here,we show that NIMIN1 and NPR1 can be copurified from plant extracts, providing biochemical evidence for their interaction. We provide functional evidence for this interaction by describing transgenic plants constitutively expressing high amounts of NIMIN1. These plants show reduced SA-mediated PR gene induction and a compromised SAR, thus mimicking the described phenotype conferred by npr1. Moreover, they showed reduced RESISTANCE gene-mediated protection. These effects were dependent on the ability of NIMIN1 to interact with NPR1. Mutant plants with a T-DNA insertion in NIMIN1 as well as transgenic plants with reduced NIMIN1 mRNA levels showed hyperactivation of PR-1 gene expression after SA treatment but no effect on the disease resistance phenotype. Our results strongly suggest that NIMIN1 negatively regulates distinct functions of NPR1, providing a mechanism to modulate specific features of SAR

Three Eichendorff lieder of Hans Pfitzner : study of musical and poetic affinity
"Joseph von Eichendorff relied upon the sounds of words to create a poetic atmosphere; this is metaphorical rather than literal. Hans Pfitzner rarely used overt text-painting (madrigalism), yet created highly evocative music. This, like the poetry of Eichendorff, is metaphorical interpretation rather than literal depiction; therefore, Pfitzner's Eichendorff settings lend compound illumination to the poetry. "Three Eichendorff Lieder of Hans Pfitzner: A Study of Poetic and Musical Affinity" explores three such settings. This project is limited to the examination of Pfitzner's setting of three Eichendorff poems: "Der Versp tete Wanderer," "Das Alter," and "In Danzig." To the best of the author's knowledge, each of these poems has received only one other musical setting. Analysis of the music and poetry will be for the purpose of demonstrating the affinity of Pfitzner's and Eichendorff's compositional aims, rather than to formal theoretical ends. Eichendorff's general stylistic traits will be noted, though extensive poetic analysis will be limited to the three poems at hand. Likewise, Pfitzner's general compositional traits will be set forth, and in-depth analysis will be reserved for the three Lieder topical to this study. Drawing on information gathered from examination of the poetry and Pfitzner's settings thereof, the present study will relate the compositional goals of Eichendorff and Pfitzner to show an affinity between composer and poet. Eichendorff's poetic intentions as interpreted by Pfitzner's music will be elucidated."--Abstract from author supplied metadata
