Regulation of UDP-GlcNAc:Galβ1-3GalNAc-R β 1-6-N-acetylglucosaminyltransferase (GlcNAc to GalNAc) in Chinese hamster ovary cells
UDP-GlcNAc:Galbeta3GalNAc-R (GlcNAc to GalNAc) beta1-6-N-acetylglucosaminyltransferase (i.e. core 2 GlcNAc-T) of the O-linked oligosaccharide pathway is developmentally regulated in human T cells, and changes in its activity have been associated with malignancies and the Wiskott-Aldrich immunodeficiency syndrome. Chinese hamster ovary (CHO) cells normally express low levels of core 2 GlcNAc-T activity (8-12 pmol/mg/h) which can be accurately measured with a two-step assay employing purified bovine
beta1-4Gal-T and high specific activity UDP-[3H]Gal to radiolabel the core 2 reaction product. CHO cells treated with 2 mM sodium butyrate for 24 h exhibited a 16-fold increase in core 2 GlcNAc-T activity, whereas several other differentiating agents including dimethyl
sulfoxide, retinoic acid, phorbol ester, and cholera toxin had no effect on activity. The addition of butyrate, cholera toxin, or dimethyl sulfoxide to CHO cells slowed cell proliferation and induced changes in cell morphology characteristic of cell differentiation. Induction of core 2 GlcNAc-T by butyrate was blocked by actinomycin D and cycloheximide. Butyrate treatment also elevated cytosolic cAMP levels with a time course which paralleled, but preceded, induction of core 2 GlcNAc-T activity by approximately 8 h. The protein kinase inhibitors H-7 and H-8 blocked butyrate-dependent induction of enzyme activity, whereas the inactive analogue H1004 had no effect. Core 2
GlcNAc-T showed a change in Km for UDP-GlcNAc, from 0.50 mM in untreated cells to 4.54 mM in butyrate + cholera toxin treated CHO cells, but no changes in Km for the synthetic acceptor, Galbeta1-3GalNAcalpha-paranitrophenyl. Despite the 9-fold increase in Km for
sugar nucleotide, Vmax/Km was 8.8-fold greater in treated compared with untreated cells. These observations suggest that in CHO cells induction of core 2 GlcNAc-T by butyrate treatment requires de novo gene transcription/translation, activation of protein kinase(s), and is associated with changes in the kinetic properties of the enzyme
A HOMOGENEOUS CELL-BASED ASSAY TO IDENTIFY N-LINKED CARBOHYDRATE PROCESSING INHIBITORS
Malignant transformation is accompanied by altered cell surface glycosylation. N-Linked oligosaccharides carrying β1–6GlcNAc branches are associated with tumor invasion and metastasis. Therefore, compounds that can enter cells and block biosynthesis of β1–6GlcNAc-branched glycans without overt cytotoxicity are potential anticancer agents. We have developed a homogeneous cell-based assay for detection of such compounds. The method enables identification of agents that block β1–6GlcNAc-branched glycan expression after incubation for 16–20 h with MDAY-D2 tumor cells, thereby protecting the cells from the subsequent addition of leukoagglutinin, a cytotoxic plant lectin. We observed that MDAY-D2 cell number is directly proportional to the level of endogenous alkaline phosphatase activity measured spectrophotometrically in cultures after the addition of substrate. The alkaline phosphatase assay was capable of detecting as few as 1500 cells. The assay was readily adapted for high-throughput screening as reagent costs are low and no cell harvesting and washing steps are required. Under high-throughput operating conditions, the coefficient of variation for controls was found to be 4.2%. The results suggest that measurement of alkaline phosphatase in this cell assay format may be adapted for wider applications in high-throughput screenings for compounds that relieve cells from other growth inhibitors

A Fluorescent Assay for the Determination of UDP-GlcNAc: Galβ1,3GalNAc-R (GlcNAc to GalNAc) β-1,6 N-Acetylglucosaminyltransferase Activity
A fluorescent assay for UDP-GlcNAc:Galbeta1,3GalNAc-R beta1,6-N-acetylglucosaminyltransferase (core 2 GlcNAc-T) activity has been developed involving dansylation of the enzyme reaction product. Core 2 GlcNAc-T detection was performed using unlabeled UDP-GlcNAc as the donor and Galbeta1,3GalNAcalpha-pAp as the acceptor. The product, Galbeta1,3(GlcNAcbeta1,6)-GalNAcalpha-pAp, was quantitatively derivatized with dansyl chloride at the NH2 moiety of the pAp group and the resultant fluorescent trisaccharide was separated on a Spherisorb ODS2 HPLC column. THis method, rapid and economical, was found to be sensitive enough for the detection of 1 pmol of reaction product and therefore represents a reliable alternative to assays which use radiolabeled substrates. Additionally, the approach described here can be adapted for the assay of other glycosyltransferases, where the acceptor substrate has a pAp group as a hydrophobic aglycon linker

Penelitian ini bertujuan untuk mendeskripsikan Kiprah Ibu Kasur dengan
sebagai Tokoh Pendidikan Anak di Indonesia pada tahun 1962 - 2002. Penelitian
ini menjelaskan mengenai kehidupan Ibu Kasur tentunya dengan mengenai
riwayat keluarga dan awal memasuki Dunia Anak di Indonesia. Keikutsertaan Ibu
Kasur dengan kegiatan Pak Kasur mengantarkan Ibu Kasur saat 1947
menggantikan Pak Kasur menjadi Pembawa juga Pengasuh Anak di media Radio
Republik Indonesia di Garut. Konsep dan metode pelajaran yang diserap Ibu
Kasur dari Pak Kasur dengan selalu mengedepankan motto belajar sambil
bermain serta pembelajaran melalui kesenian. Dengan kecintaannya terhadap
anak, Ibu Kasur menciptakan karya - karyanya melalui lagu dan dongeng yang
berisikan pesan moral dan nasihat untuk diterapkan dalam kehidupan anak.
Penelitian ini menggunakan metode deskriptif-naratif dengan sumber primer yang
digunakan berupa surat kabar kompas dan sumber sekundernya wawancara
dengan kedua anaknya di TK Mini Pak Kasur buku, jurnal, dan artikel.
Pada penelitian ini, penulis menjelaskan bahwa pengabdian Ibu Kasur
terhadap Pak Kasur tersalurkan ke dalam pendidikan anak usia dini. Penulis
menjelaskan bahwa Ibu Kasur memiliki karakter moral yang tinggi sedari menjadi
anak dengan pedoman ayahnya yang selalu menjadikan Ibu Kasur seorang kaka
yang dapat menjadi suri tauladan adik – adiknya. Selain itu, konsep yang
diajarkan serta dibuat suaminya dapat mengajarkan Ibu Kasur ke dalam Dunia
Pendidikan Anak yang dapat dijadikan sebagai bahan pembelajaran untuk
menumbuhkan ilmu pengetahuan melalui nilai moral, nilai keagamaan, dan nilai
sosial dengan metode melalui kesenian. Hal tersebut menimbulkan Ibu Kasur
dikenal sebagai Pengabdi Pendidikan Anak Usia Dini, hingga mendapat berbagai
penghargaan salah satunya dari Bapak Presiden Kedua, Ir. Soeharto yaitu Bintang
Budaya Parama Dharma pada tahun 1992.
