    IL-1 induces IL-1. IV. IFN-gamma suppresses IL-1 but not lipopolysaccharide-induced transcription of IL-1

    : IL-1 induces its own gene expression in human PBMC, in cultured smooth muscle and in endothelial cells. IL-1-induced IL-1 may be part of a self-amplification or an autocrine growth factor in a variety of responses, and thus the endogenous regulation of IL-1 production likely contributes to the outcome of immunologic or inflammatory responses. In the present study, IFN-gamma consistently increased LPS-induced IL-1, but reduced the total amount of IL-1-induced IL-1 from PBMC. This reduction was observed in populations of adherent cells and cells selected with anti-Leu M3 antibody. On a molar basis, IFN-gamma and IFN-alpha 2 were equally effective in reducing IL-1-induced IL-1 synthesis. In PBMC of 24 human subjects, IFN-gamma also reduced PMA-induced IL-1 synthesis (67% decrease, p less than 0.001). The augmentation of LPS-stimulated IL-1 by IFN-gamma was observed only when added at the same time as LPS, but IFN-gamma could be added 6 h after stimulation with IL-1 and still suppress IL-1 production. LPS-induced mRNA for IL-1 beta was modestly enhanced by IFN-gamma whereas mRNA levels for TNF were markedly increased. In contrast, IFN-gamma suppressed mRNA accumulation for IL-beta after stimulation with IL-1 alpha by 60 to 95%. The addition of IFN-gamma 30 min before stimulation of PBMC with IL-1 suppressed IL-1 beta mRNA for up to 30 h. The half-life of IL-1 beta mRNA of approximately 2 h was not influenced by IFN-gamma. Thus, IFN-gamma reduces IL-1-induced IL-1 synthesis by suppressing IL-1-induced transcription, whereas the same concentrations of IFN-gamma augment LPS-induced IL-1 production by increasing transcription

    Behavioral assessment of high-dose amphetamine withdrawal: Importance of training and testing conditions

    Chronic d-amphetamine-treated rats were given twice daily injections at a dose of 7.5 mg/kg for 2 weeks. Acute amphetamine and saline groups of rats were given saline treatments during this time, except that for the acute group the final injection was 7.5 mg/kg d-amphetamine. Acute and chronic amphetamine groups habituated to the locomotor activity testing apparatus showed increases in both distance traveled and repetitive movement time that lasted up to 6 h following the final injection. When animals were not habituated to the activity test apparatus, however, a significant decrease in repetitive movement time was noted for the chronic amphetamine group 24-54 h following the final amphetamine injection; no differences were observed for distance traveled when the locomotor activity apparatus was novel. Swim test immobility time was assessed twice following the last injection, with the second test following the first by approximately 24 h. During the first test, decreases in immobility were observed for both chronic and acute amphetamine groups, 6-12 h following the last injection. However, during the second test, decreases in immobility time were observed only for the chronic amphetamine groups 36-72 h following the final injection. These results indicate that 24 to 72 h after the end of the chronic amphetamine regimen a withdrawal effect was observed for both repetitive movement time in the locomotor activity test and immobility time in the swim test. The withdrawal effect was observed only for the locomotor activity groups for whom the test apparatus was novel, and only during the second test of immobility time for the swim test groups. Thus, the method of behavioral assessment can be critical for the demonstration of a high-dose amphetamine withdrawal effect

    Polyaniline, poly(ethylene oxide), poly(propylene glycol), poly(vinylpyrrolidinone), methyl cellulose, polyethylenimine, polyethylene, and Nylon-6, were encapsulated into MoS2. Electrical conductivity and thermoelectric power measurements of pressed pellets of (PEO)(0.92)MoS2 and (Polyaniline)(0.35)MoS2 show p-type metallic behavior. Below 14 K and 9 K respectively both show a metal to insulator transition.     Relationship between HDL Cholesterol Efflux Capacity, Calcium Coronary Artery Content, and Antibodies against ApolipoproteinA-1 in Obese and Healthy Subjects

    Aims: To explore the associations between cholesterol efflux capacity (CEC), coronary artery calcium (CAC) score, Framingham risk score (FRS), and antibodies against apolipoproteinA-1 (anti-apoA-1 IgG) in healthy and obese subjects (OS). Methods and Results: ABCA1-, ABCG1-, passive diffusion (PD)-CEC and anti-apoA-1 IgG were measured in sera from 34 controls and 35 OS who underwent CAC score determination by chest computed tomography. Anti-apoA-1 IgG ability to modulate CEC and macrophage cholesterol content (MCC) was tested in vitro. Controls and OS displayed similar ABCG1-, ABCA1-, PD-CEC, CAC and FRS scores. Logistic regression analyses indicated that FRS was the only significant predictor of CAC lesion. Overall, anti-apoA-1 IgG were significantly correlated with ABCA1-CEC (r = 0.48, p < 0.0001), PD-CEC (r = −0.33, p = 0.004), and the CAC score (r = 0.37, p = 0.03). ABCA1-CEC was correlated with CAC score (r = 0.47, p = 0.004) and FRS (r = 0.18, p = 0.29), while PD-CEC was inversely associated with the same parameters (CAC: r = −0.46, p = 0.006; FRS: score r = −0.40, p = 0.01). None of these associations was replicated in healthy controls or after excluding anti-apoA-1 IgG seropositive subjects. In vitro, anti-apoA-1 IgG inhibited PD-CEC (p < 0.0001), increased ABCA1-CEC (p < 0.0001), and increased MCC (p < 0.0001). Conclusions: We report a paradoxical positive association between ABCA1-CEC and the CAC score, with the latter being inversely associated with PD in OS. Corroborating our clinical observations, anti-apoA-1 IgG enhanced ABCA1 while repressing PD-CEC, leading to MCC increase in vitro. These results indicate that anti-apoA-1 IgG have the potential to interfere with CEC and macrophage lipid metabolism, and may underpin paradoxical associations between ABCA1-CEC and cardiovascular risk

    A Genetic Polymorphism in the Pannexin1 Gene Predisposes for The Development of Endothelial Dysfunction with Increasing BMI

    Endothelial dysfunction worsens when body mass index (BMI) increases. Pannexin1 (Panx1) ATP release channels regulate endothelial function and lipid homeostasis in mice. We investigated whether the Panx1-400A>C single nucleotide polymorphism (SNP), encoding for a gain-of-function channel, associates with endothelial dysfunction in non-obese and obese individuals. Myocardial blood flow (MBF) was measured by 13N-ammonia positron emission/computed tomography at rest, during cold pressor test (CPT) or dipyridamole-induced hyperemia. Myocardial flow reserve (MFR) and endothelial function were compared in 43 non-obese (BMI < 30 kg/m2) vs. 29 obese (BMI ≥ 30 kg/m2) participants and genotyping for the Panx1-400A>C SNP was performed. Groups comprised subjects homozygous for the C allele (n = 40) vs. subjects with at least one A allele (n = 32). MBF (during CPT or hyperemia), MFR and endothelial function correlated negatively with BMI in the full cohort. BMI correlated negatively with MFR and endothelial function in non-obese Panx1-400C subjects, but not in Panx1-400A individuals nor in obese groups. BMI correlated positively with serum triglycerides, insulin or HOMA. MFR correlated negatively with these factors in non-obese Panx1-400C but not in Panx1-400A individuals. Here, we demonstrated that Panx1-400C SNP predisposes to BMI-dependent endothelial dysfunction in non-obese subjects. This effect may be masked by excessive dysregulation of metabolic factors in obese individuals

    SRLx: A Personalized Learner Interface for MOOCs

    Past research in large-scale learning environments has found one of the most inhibiting factors to learners’ success to be their inability to effectively self-regulate their learning efforts. In traditional small-scale learning environments, personalized feedback (on progress, content, behavior, etc.) has been found to be an effective solution to this issue, but it has not yet widely been evaluated at scale. In this paper we present the Personalized SRL Support System (SRLx), an interactive widget that we designed and open-sourced to improve learners’ self-regulated learning behavior in the Massive Open Online Course platform edX. SRLx enables learners to plan their learning on a weekly basis and view real-time feedback on the realization of those plans.
