Isolasi, Identifikasi, dan Karakterisasi Cendawan Blas Pyricularia oryzae Hasil Rejuvenasi
<p>Isolation, Identificaton, and Charaterization of Blast Fungus Pyricularia oryzae Derived from Rejuvenation. Puji Lestari, Wawan, Tri P. Priyatno, Wening Enggarini, Reflinur, and Yadi Suryadi. Blast disease caused by Pyricularia oryzae (Po) is one of the important diseases on rice crop in Indonesia. This study was aimed at obtaining blast fungus pathogen rejuvenated from preserved seed origin, agar medium and rice leaf tissue. In addition, we identified blast isolates for long-term storage collection, and characterized isolates towards their specific locus of genes virulence. The results revealed that 22.7% isolates stored in agar media produced a typical mycelium of Po. The specificity of total genomic DNA banding pattern of isolates which were analyzed using specific primers encoding virulence genes of Cut1, Erg2, and Pwl2, showed six haplotypes consist of B-001 (1 isolate), C-011 (1 isolate), D-111 (8 isolate), F-110 (1 isolate), G-100 (3 isolate), and H-101 (2 isolate). None of haplotypes A-000 and E-010 were found among the isolates studied. In regard to its pathogenicity, the majority of Po fungal isolates had Cut1, Pwl2, and Erg2 genes. Among the total isolates of the Po fungus, the greatest proportion of genes were Pwl2 (87.5%) followed by Cut1 (75%) and Erg2 (62.4%) genes.</p><p> </p><p><strong>Abstrak</strong></p><p>Penyakit blas yang disebabkan oleh cendawan Pyricularia oryzae (Po) merupakan salah satu penyakit penting pada pertanaman padi di Indonesia. Penelitian ini bertujuan untuk mendapatkan cendawan patogen blas yang berasal dari hasil rejuvenasi simpanan benih, media agar, dan jaringan daun padi. Selain itu, identifikasi koleksi isolat blas yang ditujukan untuk penyimpanan jangka panjang dan karakterisasi kespesifikan tiap isolat terhadap lokus spesifik gen virulensi. Hasil pengujian menunjukkan 22,7% isolat yang tersimpan dalam media agar masih menunjukkan tipikal miselium Po. Kespesifikan pola pita DNA genomik total isolat yang dianalisis berdasarkan primer penyandi gen spesifik virulensi Cut1, Erg2, dan Pwl2, diperoleh sebanyak enam haplotipe meliputi B-001 (1 isolat), C-011 (1 isolat), D-111 (8 isolat), F-110 (1 isolat), G-100 (3 isolat), dan H-101 (2 isolat). Tidak ditemukan haplotipe A-000 dan E-010 pada isolat Po yang dianalisis. Dalam hubungannnya dengan patogenisitas, mayoritas cendawan Po mempunyai gen Cut1, Pwl2, dan Erg2. Di antara total isolat cendawan Po, gen yang paling besar ditemukan proporsinya adalah gen Pwl2 (87,5%) diikuti Cut1 (75%) dan Erg2 (62,4%).</p></jats:p
Creation of Tri-Planar Incisions in Vitrectomy: An Explorative Design-Study into Trocar Insertion Mechanisms
Background: In vitrectomy, a type of eye surgery, surgery instruments must pass through an incision in the sclera. The architecture of the incision is of great im-portance to the closure of the wound after the procedure and through that the risk for complications like hypotony and endophthalmitis. Current studies suggest that tri-planar incisions provide better sealing, however for a surgeon it is challenging to pro-duce the desired architecture easily and consistently. The objective of this study is to: “Design and verify a new, reliable and easy to use method to create tri-planar inci-sions for scleral cannulas to limit leakage after extraction of the cannula.” Method: Analysis of the factors affecting the tri-planar incision method in terms of reliability and ease of use was performed, leading to a set of design requirements. Based on the requirements, different incision methods were explored and compared. The selected method was transformed into a functional prototype, which was subse-quently evaluated on consistency and leakage. The evaluation was done by insertion on silicone sheet, eye phantoms and ex-vivo porcine eyes. The tri-planar incision was compared in leakage to straight and oblique incisions, two other common type of ar-chitectures.Results: The method performed using a designed device was able to create the de-sired tri-planar consistently. These incisions provided a better seal than a straight in-cision. However, for tri-planar compared to an oblique incision the test was inconclu-sive and further research is needed to distinguish between the two.Conclusion: As tri-planar incisions can now consistently be made with the use of the new method, further research can be performed to link the number and type of com-plications, like hypotony and endophthalmitis, to the incision statistics.Mechanical Engineerin
Identifikasi Entomopatogen Bakteri Merah pada Wereng Batang Coklat (Nilaparvata lugens Stål.)
<p>Indentification of Entomopathogenic Red Bacterial from<br />Brown Planthopper (Nilaparvata lugens Stål.). Tri P.<br />Priyatno, Yohana A. Dahliani, Yadi Suryadi, I Made<br />Samudra, Dwi N. Susilowati, Iman Rusmana, Baskoro S.<br />Wibowo, and Cahyadi Irwan. Red bacteria isolated from<br />brown planthopper (BPH) has been proven pathogenic<br />against BPH and others insects. Application of 106 to 107<br />cells/ml of red bacteria caused 65.6-78.2% mortality of BPH.<br />The 50% effective concentration (EC50) and lethal time of red<br />bacteria against BPH is 2.8 x 105 cells/ml and 6.8 days,<br />respectively. Based on phenotypic characters tested on GN<br />MicroPlateTM Biolog kit and 16S rRNA sequneces analysis,<br />red bacteria was identified as Serratia marcescens with 99%<br />similarity. Red pigmen produced by S. marcescens strain<br />BPH is secondary metabolite determined as prodigiosin<br />showing bactericidal activities against Xanthomonas oryzae<br />pv. oryzae. We concluded that S. marcescens did not only<br />potent as biocontrol agent to BPH, but also it can be used to<br />control plant pathogenic bacteria.</p></jats:p

Performance of Three Genera of Entomopathogenic Fungi as Potential Microbial Control Agents Against the Flea Beetle Phyllotreta Striolata F. (Coleoptera : Chrysomelidae)
The striped flea beetle (FB), Phyllotreta striolata F. (Coleoptera: Chrysomelidae), is not
only a serious pest of canoia and mustard but also feed on a wide range of other
brassicas. Entomopathogenic fungi (EF) are promising agent for biological control of
FB and are gaining increasing attention worldwide as mycoinsecticide. The potential of
three genera of EF, Metarhizium anisopliae, Beauveria bassiana and Paecilomyces
fumosoroseus, has been studied in the laboratory and the field against the striped FB,
Phyllotreta strio/ata F.
Surveys for FB naturally infected with EF indicated that M anisopliae v. manus and B.
bassiana were the potential EF in the populations of FB sampled from vegetable area at
UPM's Research Park, Serdang. However, the incidence of infection was very low.
Therefore, introduction of virulent isolates into a temporary habitat must be done. Test for pathogenicity of 16 isolates ofEF against adult FB found only one isolate of M
anisopliae (MPs) causing mortality in excess of 50%. Four isolates were tested for
pathogenicity against the eggs and larvae of the FB. Two isolates of M anisopliae (MPs
and Cy3), one B. bassiana (WIs) and one P. fumosoroseus (Pt) were found to be highly
pathogenic against the FB larvae while both isolates of M anisopliae were infective
against the FB eggs.
The resistance of FB adults against EF was caused by the existence of fungistatic
compounds on the integument. Five straight chain fatty acids (C4, C6, C7, C8, and C9)\ud
suspected as fungistatic compounds based on analysis using Gas Chromatography were
proven to inhibit conidial germination.
Two media, rice flour and sponge-rice flour medium, examined for conidial massproduction
of M anisopliae v. majus and P. fumosoroseus indicated that the spongerice
flour medium was shown to be potentially efficient for mass-production of fungal
spores
Pathogenicity of Paecilomyces fumosaroseus (Wise) Brown & Smith, Beauveria bassiana (Bals.) Vuill. and Metarhizium anisopliae (Metsch.) sorokin on the striped flea beetle Phyllotreta striolata F. (Coleoptera: Chrysomelidae)
The ability of three species of entomopathogenic fungi, Paecilomyces fumosoroseus (Wise) Brown & Smith, Beauveria bassiana (Bals.) Vuill. and Metarhizium anisopliae (Metsch.) Sorokin to cause infection on the adults, eggs and larvae of the striped flea beetle Phyllotreta striolata F. was tested. Only one isolate of M. anisopliae and none of the B. bassiana and P. fumosoroseus caused adult mortality in excess of 50% at a concentration of 2 x 107 conidia mL1. One isolate of?, fumosoroseus (Pf), one B. bassiana (Wls) and two of M. anisopliae (MPs and Cy3) were pathogenic to the first instar larvae of P. striolata causing more than 50% mortality at a concentration of 2 x 106 conidia mL1. The rate of larval mortality increased with increase in conidia concentration. The respective estimated LT50 values for Pf, Wls, MPs and Cy3 for the larvae at 2 x 106 conidia mL1 were2.9, 3.5, 3.0 and 3.0 days. The two isolates of M. anisopliae were also highly pathogenic to the eggs causing significant inhibition of hatching, while B. bassiana and P. fumosoroseus were less pathogenic. Estimates of the median lethal concentration for Cy3 and MPs were 13.0 x 105 and 5.03 x 105 conidia mL1 respectively
Seleksi In Vitro dan Pengujian Mutan Tanaman Pisang Ambon Kuning untuk Ketahanan terhadap Penyakit Layu Fusarium
<p>Fusarium wilt of banana (Musa spp.) caused by<br />Fusarium oxysporum f. sp. cubense (Foc) is the most serious<br />problem faced in banana cultivation in terms of plant<br />productivity and fruit quality. Mutation breeding is one of the<br />alternative method that can be applied in producing new<br />banana cultivar. Mutants can be induced by chemical<br />mutagen such as ethyl methane sulfonate (EMS) followed by<br />in vitro selection and then evaluation of the mutants to<br />fusarium wilt disease in glasshouse and Foc infected field.<br />The aim of this research was obtained EMS induced and in<br />vitro selected mutants of banana var. Ambon Kuning and<br />evaluated Foc disease resistant clones in glasshouse and<br />Foc infected field. The first step to obtain the explants for<br />this research was initiation and formation of multiple bud<br />clumps (MBC) using MS basal media supplemented with 5,<br />10, and 20 mg/l of benzyladenin. Plant regeneration of MBC<br />was also studied by using MS media containing 0, 0.2, and 1<br />mg/l of benzyladenin. To induce mutagenesis, MBC was<br />soaked in 0.1, 0.3, and 0.5% (v/v) EMS for 1, 2, and 3 hours.<br />The assesment of resistant MBC mutants to Fusarium<br />phytotoxin was conducted by using fusaric acid (FA) as<br />selection agent in concentration of 30, 45, and 60 ppm.<br />Putative mutant plants produced by in vitro selection were<br />further tested using spore solution of Foc race 4 in<br />glasshouse. Meanwhile, Foc resistance assesment in the<br />infected field was conducted in Pasirkuda Experimental<br />Station, Bogor Agricultural University. The results showed<br />that MBC can be formed in MS basal media supplemented<br />with 10 or 20 mg/l benzyladenin. The EMS played a role in<br />obtaining mutants by producing 68 MBC putative mutants<br />tolerant to Foc based on FA selection. Further evaluation in<br />the glasshouse was obtained 64 Foc resistant plants from<br />391 putative mutants produced by in vitro selection.<br />Evaluation in the Foc infected field showed six clones<br />survived until generative phase (12 month of age).</p></jats:p

Konstribusi PKH Terhadap Kesejahteraan Keluarga Penerima Manfaat (KPM)

Abstract
Penelitian ini bertujuan mendeskripsikan konstribusi PKH, factor pendukung dan penghambatnya terhadap kesejahteraan Keluarga Penerima Manfaat (KPM) di Kecamatan Candimulyo. Penelitian menggunakan pendekatan kualitatif deskriptif. Wawancara, observasi, dan telaah dokumen digunakan sebagai teknik pengumpulan data. Analisis data menggunakan siklus interaktif. Hasil penelitian menunjukkan, bahwa PKH berkonstribusi kepada peningkatan kesejahteraan keluarga, khususnya mereduksi kekurangan kualitas dan kuantitas pangan, kesehatan, dan pendidikan, sehingga KPM cenderung lebih sejahtera. Factor pendukung antara lain KPM memiliki motivasi/antusiasme tinggi dalam memenuhi komitmen di bidang Pendidikan, kesehatan, dan kesejahteraan social. Faktor penghambat antara lain masih ditemukan eklusif eror dan inklusif eror dalam penetapan sasaran garap (KPM). Rekomendasi yang diajukan diantaranya dalam verifikasi dan validasi (Vefivali) data KPM perlu perluasan pelibatan semua unsur masyarakat mulai dari RT, RW, kelurahan/Desa, kecamatan, kebupaten/kota hingga tokoh masyarakat setempat
Resistance and Phenotypic Character of Chili M2 Mutant Lines Against Chilli Veinal Mottle Virus
<p>Chilli veinal mottle virus infection (ChiVMV) could reduce the quality and 60–100% of yield losses of chili. Among the chili<br />varieties released, no one has been resistant to ChiVMV, mainly due to a high variation of ChiVMV strains and not well mapped.<br />Therefore, finding a new source of ChiVMV resistant genes is pivotal role in order to assembly new varieties. Approach through<br />in vitro mutation induction using mutagen ethyl methane sulfonate (EMS) is one of the efforts to increase genetic diversity.<br />Previous studies has successfully acquired 800 M2 lines through callus induction of Gelora variety with EMS. This study aimed to<br />obtain M2 lines resistant to ChiVMV and having a good agronomical characters. A total of 800 chili M2 lines that derived from chili<br />M2 mutations using mutagen EMS has been tested in greenhouse to ChiVMV resistance and studied character phenotype. The<br />results showed that of the 800 lines, there were 28 strains obtained showed a response tolerant and resistant to ChiVMV. Eight<br />mutant lines of which have good agronomic characters. The mutant lines are M2.100, M2.108, M2.200, M2. 122, M2.238, M2.353,<br />M2.420, and M2.517. Eight lines will be selected and further observed to obtain chili promising lines that are resistant to ChiVMV<br />and high yielding.</p></jats:p
Defect (p,q) five-branes
AbstractWe study a local description of composite five-branes of codimension two. The formulation is constructed by virtue of SL(2,Z)×SL(2,Z) monodromy associated with two-torus. Applying conjugate monodromy transformations to the complex structures of the two-torus, we obtain a field configuration of a defect (p,q) five-brane. This is a composite state of p defect NS5-branes and q exotic 522-branes. We also obtain a new example of hyper-Kähler geometry. This is an ALG space, a generalization of an ALF space which asymptotically has a tri-holomorphic two-torus action. This geometry appears in the conjugate configuration of a single defect KK5-brane
