    Understanding the role of membrane cholesterol upon Epstein Barr virus infection in astroglial cells.

    Background: EBV infection has long been postulated to trigger multiple sclerosis (MS) and anti-EBV antibodies showed a consistent presence in MS patients. Previous reports from our group have shown that the EBV infects different brain cells. Entry of the virus in neuronal cells is assisted by several host factors including membrane cholesterol. By using an inhibitor, methyl-β-cyclodextrin (MβCD), we evaluated the role of membrane cholesterol in EBV infection and pathogenesis. Methodology: The membrane cholesterol depleted cells were infected with EBV and its latent genes expression were assessed. Further, EBV-mediated downstream signalling molecules namely STAT3, RIP, NF-kB and TNF-α levels was checked at protein level along with spatial (periphery and nucleus) and temporal changes in biomolecular fingerprints with Raman microspectroscopy (RS). Results: Upon treatment with MβCD, lmp1 and lmp2a suggested significant downregulation compared to EBV infection. Downstream molecules like STAT3 and RIP, exhibited a decrease in protein levels temporally upon exposure to MβCD while NF-kB levels were found to be increased. Further, the intensity of the Raman spectra exhibited an increase in triglycerides and fatty acids in the cytoplasm of EBV-infected LN-229 cells compared to MβCD+EBV. Likewise, the Raman peak width of cholesterol, lipid and fatty acids were found to be reduced in EBV-infected samples indicates elevation in the cholesterol specific moieties. In contrast, an opposite pattern was observed in the nucleus. Moreover, the ingenuity pathway analysis revealed protein molecules such as VLDLR, MBP and APP that are associated with altered profile of cholesterol, fatty acids and triglycerides with infection-related CNS disorders. Conclusion: Taken together, our results underline the important role of membrane cholesterol over EBV entry/pathogenesis in astroglia cells which further trigger/exacerbate virus-associated neuropathologies. These results likely to aid into the prognosis of neurological disease like MS

This has some practical consequences for the clinical encounter, as discussed throughout the book.     RESUMENEn la correspondencia que, entre 1773 y 1778, mantuvo Antonio Valcárcel Pío de Saboya, conde de Lumiares, con el ilustrado Gregorio Mayans y con el anticuario gaditano Antonio Mosti, se contienen ciertas manifestaciones relativas a una poesía manuscrita inédita que Valcárcel tenía intención de publicar de forma anónima pese a saber que su autor era el jesuita Martín Marín.     This paper discusses three songs in the rani work ceremony at GBKP Runggun Tiga Juhar, STM Hulu sub-district, Deli Serdang Regency. Kerja rani is one of the ceremonies held on cultural week at GBKP Runggun Tiga Juhar. The rani work ceremony is one of the sources of church finances by collecting offerings and then conducting an auction. Some of the songs used in the rani work ceremony are Sendah kerja rani, Mbuah page, and Mejuah-juah. Sendah kerja rani is a song that must be sung because it is directly related to agriculture in the Karo tribe. While the other two songs, namely the songs Mbuah page and Mejuah-juah are taken from folk songs created by Djaga depari. This research aims to analyze the meaning of the text and the musical function of the three rani work ceremony songs. In answering the research objectives, the author uses the method of field work and desk work proposed by Bruno Netll. The theory used to answer the main problem in this paper is the semiotic theory pioneered by Roland Barthes, especially the meaning of denotation and connotation. The results of the study revealed that the rani work ceremony at GBKP Runggun Tiga Juhar is one way for the church to maintain culture and liven up the atmosphere of the ceremony. The three songs in the rani work ceremony have the process of the ceremony, the meaning of the text and its functions.     Human genetic factors associated with pneumonia risk, a cue for COVID-19 susceptibility

    Pneumonia, an acute respiratory tract infection, is one of the major causes of mortality worldwide. Depending on the site of acquisition, pneumonia can be community acquired pneumonia (CAP) or nosocomial pneumonia (NP). The risk of pneumonia, is partially driven by host genetics. CYP1A1 is a widely studied pulmonary CYP family gene primarily expressed in peripheral airway epithelium. The CYP1A1 genetic variants, included in this study, alter the gene activity and are known to contribute in lung inflammation, which may cause pneumonia pathogenesis. In this study, we performed a meta-analysis to establish the possible contribution of CYP1A1 gene, and its three variants (rs2606345, rs1048943 and rs4646903) towards the genetic etiology of pneumonia risk. Using PRISMA guidelines, we systematically reviewed and meta-analysed case-control studies, evaluating risk of pneumonia in patients carrying the risk alleles of CYP1A1 variants. Heterogeneity across the studies was evaluated using I2 statistics. Based on heterogeneity, a random-effect (using maximum likelihood) or fixed-effect (using inverse variance) model was applied to estimate the effect size. Pooled odds ratio (OR) was calculated to estimate the overall effect of the risk allele association with pneumonia susceptibility. Egger's regression test and funnel plot were used to assess publication bias. Subgroup analysis was performed based on pneumonia type (CAP and NP), population, as well as age group. A total of ten articles were identified as eligible studies, which included 3049 cases and 2249 healthy controls. The meta-analysis findings revealed CYP1A1 variants, rs2606345 [T vs G; OR = 1.12 (0.75–1.50); p = 0.02; I2 = 84.89%], and rs1048943 [G vs T; OR = 1.19 (0.76–1.61); p = 0.02; I2 = 0.00%] as risk markers whereas rs4646903 showed no statistical significance for susceptibility to pneumonia. On subgroup analysis, both the genetic variants showed significant association with CAP but not with NP. We additionally performed a spatial analysis to identify the key factors possibly explaining the variability across countries in the prevalence of the coronavirus disease 2019 (COVID-19), a viral pneumonia. We observed a significant association between the risk allele of rs2606345 and rs1048943, with a higher COVID-19 prevalence worldwide, providing us important links in understanding the variability in COVID-19 prevalence

    This volume (The Early Modern Period) is the third part of The History of Science and Natural Philosophy among the Croats (with particular reference to the exact sciences) by academician Žarko Dadić. The first two volumes were The Middle Ages (2015) and The Renaissance (2016). Volume I covered the Middle Ages up to 1526, and Volume II the period from 1526 to the end of the sixteenth century. The third part covers events from 1600 to 1699. According to the author, “the 17th century, though considered part of late medieval times, can also be identified as the Early Modern Age. In philosophical and scientific terms, it was a particular age which differed from the Renaissance and in which the foundations of completely new philosophical and scientific attitudes were laid”.
