Das phänotypische Spektrum bei Patienten mit Sequenzveränderungen im RP1L1-Gen
Durch das RP1L1-Gen wird ein netzhautspezifisches Protein kodiert. Sequenzveränderungen in diesem Gen verursachen Veränderungen in den lichtempfindlichen Photorezeptoren und daraus resultierende Retinopathien. Mit dem RP1L1-Gen ist die seltene okkulte Makuladystrophie (OMD) assoziiert. Trotz funktioneller Beeinträchtigungen wurde der Name dieser autosomal dominanten Netzhauterkrankung vom unauffällig erscheinenden Augenhintergrund abgeleitet.
In der vorliegenden Studie wurden Daten von 22 Patienten mit einer nachgewiesenen Sequenzänderung im RP1L1-Gen retrospektiv analysiert. Das Ziel der Arbeit war die Charakterisierung des phänotypischen Spektrums. Mit der Absicht, die unterschiedlichen Phänotypen zu vergleichen, erfolgte zunächst eine Einteilung anhand der RP1L1-Veränderung in drei Gruppen anhand der Genotypen (Gruppe I: Veränderung der 45. Aminosäureposition, Gruppe II: Veränderung der 110. Aminosäureposition, Gruppe III: Veränderung unterschiedlicher Aminosäurepositionen im RP1L1-Protein). Darauffolgend wurden die Untersuchungsergebnisse der Visusprüfung, der Farbsinnprüfung, der Perimetrie (Goldmann-Perimetrie und Mikroperimetrie), der elektrophysiologischen Untersuchungen (Ganzfeld-ERG und mf-ERG), der optischen Kohärenztomographie, der Fundusautofluoreszenz (FAF) und der Fundusphotographie analysiert.
Im Kontrast zur Definition der okkulten Makuladystrophie mit klinisch blandem Augenhintergrund zeigten die Ergebnisse der Fundus- und FAF-Untersuchung pathologische Veränderungen. Weitere morphologische Auffälligkeiten konnten im Rahmen der OCT-Volumenscan-Analyse beschrieben werden. Ein Frühstadium der OMD mit vier betroffenen Schichten (Schichten: ELM, PIS, ISe und POS) und eine Progredienz der Veränderungen in späteren Stadien (Schichten: RNFL, GCL+IPL, INL, ONL und RPE) wurde nachgewiesen. Außerdem war die Reduktion der Netzhautdicke – ein typisches Zeichen der OMD – erkennbar. So stehen die morphologischen Veränderungen im Einklang mit Funktionsstörungen wie einer Visusreduktion, Verwechslungen im Farbsinntest, relativen und absoluten Zentralskotomen, einer unruhigen und exzentrischen Fixation, einer Empfindlichkeitsminderung bis zum Empfindlichkeitsverlust oder einer Reduktion der korrelierten Potenziale für die zentralen Hexagone im mf-ERG.
Insgesamt zeigte sich innerhalb der Gruppen eine klinische Heterogenität. Allerdings konnten keine gruppenbezogenen Unterschiede erkannt werden und die Befunde waren in allen drei Gruppen unabhängig von der Sequenzveränderung vergleichbar. Schlussfolgernd stellt sich ein einheitlicher Phänotyp dar. Dieser ist nicht spezifisch für eine spezielle genetische Variante des RP1L1-Gens und drückt sich als variable Expressivität und unvollständige Penetranz der Variationen aus. Zusätzlich zu den häufig beschriebenen RP1L1-Varianten (p.R45W, p.P110L) sind wahrscheinlich weitere Varianten wie p.P110S, p.S144del, p.A2016S und p.T350K mit der OMD assoziiert.
Resümierend eröffnen die Kenntnisse über die phänotypische Heterogenität bei Patienten mit nachgewiesenen RP1L1-Varianten einen neuen Blickwinkel auf Patienten mit reduziertem Visus und unauffälligen morphologischen Veränderungen der Netzhaut. Im Rahmen der Genotyp-Phänotyp-Korrelation sind diese Informationen für die Diagnostik, Abschätzung der Krankheitsprogression und für die Entwicklung der Therapie hilfreich
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Zum genotypischen und phänotypischen Spektrum der North-Carolina-Macular-Dystrophy (NCMD) bei Sequenzvariationen in PRDM13 und IRX1
Bei der North-Carolina-Macular-Dystrophy (NCMD) handelt es sich um eine seltene autosomal-vererbte Erkrankung der Netzhaut. Die betroffenen Patienten zeigen im Bereich der Makula degenerative bis hin zu vernarbenden Veränderungen, die sich hinsichtlich ihrer Ausprägung auch innerhalb einer Familie unterscheiden.
Bisherige Untersuchungen konnten eine Korrelation der North-Carolina-Macular- Dystrophy (NCMD) mit den Genen PRDM13 und IRX1 zeigen. Es wurde von drei verschiedenen Sequenzvarianten V1 – V3 im Promotor-Kontroll-Bereich (PKR) von PRDM13 berichtet. Des Weiteren wurden Copy-Number-Variations (CNV) in Form von Tandemduplikationen in PRDM13 und IRX1 entdeckt.
In Bezug auf die North-Carolina-Macular-Dystrophy (NCMD) wurde die Patientendatenbank des Labors für Molekulare Ophthalmologie der Klinik und Poliklinik für Augenheilkunde der Justus-Liebig-Universität Gießen phänotypisch und genotypisch ausgewertet. Im Rahmen dieser Arbeit wurden fünf verschiedene Sequenzvarianten (M1 – M5) innerhalb der Promotor-Kontroll-Region von dem PRDM13-Gen ermittelt. Darunter wurde in zwei Indexpatienten die Sequenzvariante V2 (≡M3) vorgefunden. Eine Tandemduplikation in PRDM13 oder IRX1 konnte nach Etablierung entsprechender Positivkontrollen in den untersuchten Patienten nicht festgestellt werden.
Um die detektierten Sequenzvarianten in PRDM13 zu beurteilen, wurde in der vorliegenden Arbeit ein Luciferase-Reporterassay entwickelt. Mehrere Messreihen zeigten auf, dass die Sequenzvarianten eine veränderte Genexpression in PRDM13 bewirkten, indem sie die Luciferase-Aktivität der Versuchsreihe beeinflussen. Basierend auf den Ergebnissen des Luciferase-Assays wird in dieser Arbeit angenommen, dass es sich bei der Promotor-Kontroll-Region von PRDM13 um einen Silencer handelt.
Im Rahmen der Phänotypisierung wurden deutliche Unterschiede der makulären Vernarbungen vorgefunden. Die Ergebnisse der Fundusautofluoreszenzen ermöglichen ein weiteres Grading zu der bisherigen Klassifizierung über Fundusphotographien. Durch eine analytische Untersuchung der optischen Kohärenztomographie (OCT) mithilfe der DiOCTA Software konnten die makulären Läsionen quantifiziert werden und den einzelnen Netzhautschichten zugeordnet werden. Hierbei zeigt sich vor allem das retinale Pigmentepithel (RPE) und die innere Körnerschicht (INL) betroffen.
Die Ergebnisse dieser Arbeit unterstützen insgesamt die Berichte bisheriger Studien, dass eine Überexpression des PRDM13-Gens durch Unterdrückung der Silencer- Funktion der Promotor-Kontroll-Region als Ursache einer North-Carolina-Macular- Dystrophy (NCMD) anzunehmen ist
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
