Trauerrede für Peter Borowsky
Am 13. Oktober 2000 starb der Historiker Peter Borowsky, ein "Schüler" Fritz Fischers, im Alter von 62 Jahren. Nahezu vier Jahrzehnte hat er an der Universität Hamburg gewirkt: als Student, Wissenschaftlicher Assistent, Wissenschaftlicher Oberrat und Privatdozent für Neuere und Neueste Geschichte. Im Jahre 1996 wurde ihm der Titel "Professor" verliehen; eine Professorenstelle jedoch blieb ihm verwehrt. Als Autor auflagenstarker Geschichtswerke erreichte Peter Borowsky ein großes Publikum. Die gemeinsam mit Barbara Vogel und Heide Wunder verfasste, 1975 erstmals erschienene Einführung in die Geschichtswissenschaft ist bis heute ein wissenschaftlicher "Bestseller"; Borowskys Hitler-Biographie für Jugendliche und seine Überblicksdarstellungen zur Deutschen Geschichte nach 1945 erlebten ebenfalls zahlreiche Auflagen. Der Schwerpunkt seines Wirkens aber lag in der akademischen Lehre, in der er Herausragendes leistete. Generationen von Geschichtsstudierenden wurden von ihm unterrichtet, betreut und geprägt – an der Universität Hamburg wie am Smith College, Northampton/Massachusetts, wo der Gelehrte seit 1974 regelmäßig eine Gastprofessur wahrnahm. Binding of STEVOR protein to various receptor on erythrocytes
Malaria is currently one of the most life-threatening infectious disease that affects any individual located along the equator of the earth. With so many new cases and high mortality rate, studies have been carried out to find out the reason behind the severity of malaria. Results from these studies have showed that expression of different variant surface antigens does contributes to severity of the disease. The pathogenesis of malaria is due to the expression of variant surface antigen by the parasite known as Plasmodium falciparum. 3 families of variant surface antigen, expressed by Plasmodium falciparum, are PfEMP1, RIFINs and STEVORs. With many of the variant surface antigen not fully understood, there could be novel functions yet to be discovered. In this project, recombinant STEVOR protein are expressed and tested with protease-treated red blood cells so to characterize possible receptors which may have interactions with STEVORs.
The results have showed that different IPTG concentration as well as incubation conditions can affect the expression of recombinant STEVOR proteins in transformed cells. Expressed STEVORs may not be folded correctly which could hinder the protein purification process. Results of the western blot analysis confirm that STEVORs interact with glycophorin C on the red blood cells. One STEVOR manages to produce a unique result that is different from other STEVORs that were used for erythrocytes binding assays.
Glycophorin C are sensitive to trypsin and neuraminidase treatment while resistant to chymotrypsin treatment Semi-conserved region of STEVOR protein binds to glycophorin C to facilitate adhesion to the vasculature or mediate PfEMP-1-independent rosetting which are important for the pathogenesis of malaria. However, one of the STEVOR protein may interactions with receptors other than glycophorin C on the red blood cells. Binding to different receptors may lead to the activation of other chemical reactions that could contribute to the pathogenicity of the infectious disease.Bachelor of Science in Biological Science
Insights into the role of the Plasmodium falciparum reticulocyte binding protein RH1 in merozoite invasion.
The most serious form of malaria in humans is caused by the parasite, Plasmodium falciparum. Studies have shown that this parasite recognizes erythrocytes for invasion by binding to receptors on the erythrocyte surface. Other than the availability of receptors, ATP has been shown to be an important factor in determining whether an erythrocyte is suitable for invasion. Recent findings that ATP and ADP can bind to a nucleotide binding region (PfRH1-NBR) within Plasmodium falciparum reticulocyte-binding-like protein homologue 1(PfRH1) suggested that PfRH1 might serve as an ATP sensor during invasion. In this project, the biological relevance of ATP in the invasion process was investigated. My results showed that there was a reduction in extracellular ATP during early invasion thereby suggesting that ATP was used in this process. In addition, to further characterize the role of PfRH1-NBR, I had purified monoclonal antibodies that were raised against PfRH1-NBR and found that these antibodies can inhibit invasion. These findings suggested that ATP, through its interaction with PfRH1 is an important factor during parasite invasion.Bachelor of Science in Biological Science
Identification of gene features important for post-transcriptional regulation of Plasmodium falciparum invasion genes
Plasmodium falciparum which causes the most virulent form of human malaria is able to alter a repertoire of proteins steady state levels in order to switch from a sialic acid dependent to a sialic acid independent invasion pathway. The molecular basis of this switching mechanism is still unknown. Recent evidences from transcriptomic and proteomic approaches had postulated the post-transcriptional regulation (PTR) as the possible mechanism of regulating protein expression during invasion pathway switching. Here, it is showed that both the 5’UTR and 3’UTR of several genes contains the regulatory elements that are responsible for the differential regulation of protein expression during invasion pathway switching of P. falciparum. Identification of this provides experimental support to the postulation that UTRs are crucial regulators of PTR. Understanding the altered protein expression during host-parasite interaction would inspire the development of truly novel therapeutic approaches for the control of this human pathogen.Bachelor of Science in Biological Science
Identification of gene features important for post-transcriptional regulation of Plasmodium falciparum invasion genes.
Plasmodium falciparum, the major cause of human malaria, is able to up-regulate various ligands to invade erythrocytes possessing different receptors in a process known as invasion-pathway switching. Recent evidence from the W2mef P. falciparum clone has shown that this switch involves the post-transcriptional regulation (PTR) of many invasion-related genes in addition to previously characterized mechanisms. To identify gene features responsible for this PTR, the 5’ un-translated regions (UTR) of 7 genes up-regulated at the protein level were cloned into luciferase-reporter plasmids and then transfected into both normal W2mef and W2mef clones that had switched invasion pathways. No difference in luciferase activity was observed between transfected switched and un-switched clones, suggesting that motifs within the 5’ UTR of invasion genes are either not responsible for, or insufficient to drive PTR in P. falciparum.Bachelor of Science in Biological Science
