Heterologous Matrix Metalloproteinase Gene Promoter Activity Allows In Vivo Real-time Imaging of Bleomycin-Induced Lung Fibrosis in Transiently Transgenized Mice
Idiopathic pulmonary fibrosis is a very common interstitial lung disease derived from chronic inflammatory insults, characterized by massive scar tissue deposition that causes the progressive loss of lung function and subsequent death for respiratory failure.Bleomycin is used as the standard agent to induce experimental pulmonary fibrosis in animal models for the study of its pathogenesis. However, to visualize the establishment of lung fibrosis after treatment, the animal sacrifice is necessary. Thus, the aim of this study was to avoid this limitation by using an innovative approach based on a double bleomycin treatment protocol, along with the in vivo images analysis of bleomycintreated mice. A reporter gene construct, containing the luciferase open reading frame under the matrix metalloproteinase-1 promoter control region, was tested on doublebleomycin-treated mice to investigate, in real time, the correlation between bleomycin treatment, inflammation, tissue remodeling and fibrosis. Bioluminescence emitted by the lungs of bleomycin-treated mice, corroborated by fluorescent molecular tomography, successfully allowed real time monitoring of fibrosis establishment. The reporter gene technology experienced in this work could represent an advanced functional approach for real time non-invasive assessment of disease evolution during therapy, in a reliable and translational living animal model.Fil: Stellari, Fabio Franco. Petrology and geochemistry of submarine volcanism in the Sicily Channel Rift
Submarine magmatism in the Sicily Channel Rift began in the early Pliocene and lasted until almost 200 yr ago. We present here petrological and geochemical data on volcanic rocks dredged from Graham and Nameless banks and Pantelleria seamounts in the Pelagian sector of the Sicily Channel Rift. Petrological evidence suggests that the ascent of magmas to the surface was relatively rapid, probably through channels superimposed over the major tectonic discontinuities of the Rift. Major and trace element data indicate an ocean island basalt affinity for Graham and Nameless bank alkaline lavas and a depleted tholeiitic signature for one Pantelleria seamount, which had a shallower mantle source. Sr-Nd-Pb isotopic compositions suggest a heterogeneous mantle source involving both focus zone ( FOZO) and high- mu( HIMU) components. Lead isotopic compositions are slightly less radiogenic in the tholeiitic basalt and more radiogenic ( closer to HIMU) in the alkaline lavas. We hypothesize that a decompressional melting mechanism affected the mantle beneath the Sicily Channel at different levels. Our data fit in a regional scenario characterized by the presence of HIMU or FOZO components in alkaline and calc-alkaline volcanics all around Sicily. The explanation for the common isotopic composition could lie in a ( fossil) plume head that variably contributed to magmatogenesis in response to variable lithospheric stretching

A DH-Leavened Musicological Toolbox
Graduate-level training in music research methodologies tends to ignore digital humanities work and overlook the use of digital tools created in support of new forms of reading. Training instead focuses on source material in the student’s area of interest. This material includes secondary and primary (archival) resources, as well as information resources, such as: monuments of music and critical editions; indexes; bibliographies and thematic catalogs; dictionaries and encyclopedias; digital libraries of scores or editions; and databases of period-specific newspapers or journals. Graduate students taking research methods courses already have a toolbox built from their experiences as musicians and students of music, including the ability to read and interpret music notation, to understand theoretical and analytical concepts in music, as well as a command of music history, including the canon of musical works.
Digital humanities has become a major area of academic endeavor at the “interface of technological development, epistemological change and methodological concerns." An important characteristic of digital humanities research has been its interdisciplinarity. We argue that graduate training in musicology needs to include coverage of methodologies applied by digital humanists in support of new forms of reading, not only to broaden the canon of research topics in musicology, but also to build common ground with researchers of other disciplines. We propose that librarians are well positioned to provide this expertise and training
A Twitter Case Study for Assessing Digital Sound
Academic and cultural heritage institutions around the world have made measurable strides in the development of digital sound archives oriented towards research and access, but their impact on scholarship and society has been little studied. Traditionally, impact has been measured by citations; yet these are problematic metrics for non-traditional outputs like sound recordings. Social media data provide a promising avenue of investigation for measuring scholarly as well as societal impact. Twitter in particular has been shown to provide a high number of references for cultural and research outputs in all disciplines. This study analyzes Twitter references pertaining to the collections of five digital sound archives: British Library Sounds, Europeana Sounds, the Internet Archive Audio Archive, PennSound and UbuWeb. Using text analysis methods to identify high frequency events and trends, and labeling them with a rubric designed for measuring the impact of digital heritage resources, this study provides preliminary insights on user values as they relate to digital sound collections. Despite the limitations of using social media data, the evidence gathered in this case study characterizes aspects of the use of digital sound collections, and may point to future priorities for the digital preservation of sound.Peer reviewe
Against the Grain: Reading for the Challenges of Collaborative DH Pedagogy
This article provides a critical review of the past five years of literature in digital humanities pedagogy and faculty-librarian collaboration, commingled with reflections on personal practice, which extend findings from the literature. Faculty-librarian partnerships in DH pedagogy reflect a rapidly evolving area of engagement calling for expertise in teaching, subject knowledge, scholarly communication, digital technologies, and DH research methodologies. Although there is a rapidly expanding body of literature on these partnerships, the challenges of the work tend to be minimized. This article expands upon commonly encountered difficulties, and it points to potential solutions and best practices.Peer reviewe
Modeling pulmonary fibrosis through Bleomycin delivered by osmotic minipump: a new histomorphometric method of evaluation
The systemic delivery of bleomycin (BLM) to mice through subcutaneously implanted osmotic minipumps may be used to experimentally mimic the typical features of systemic sclerosis and related interstitial lung diseases. The published studies on this model principally focused on induced dermal modifications, probably because lung lesions are typically mild, subpleurally localized and difficult to analyze. The use of high BLM doses to increase their severity has been proposed, but is ethically questionable because of compromising animal welfare. We propose a tailored histomorphometric method suitable to detect and quantify this type of mild lung lesions. Using a two steps automated image analysis, a peripheral region of interest with a depth of 250 μm from the pleural edge was defined on whole slide images and the fibrotic foci were histomorphometrically characterized. The effects of different BLM doses on lung alterations were evaluated in C57BL/6 mice and 60 U/kg resulted in a fair compromise between fibrotic lesions and animal welfare. This dose was also tested in time-course experiments. The analysis revealed a peak of histologic fibrotic-like alterations, cytokine expression, metalloprotease and macrophagic activation, between the 21th and 28th day post pump implant. The induced dermal fibrosis was characterized by the progressive loss of the white dermal adipose layer, an increase of dermal thickness, dermal hyperplasia and more compacted collagen fibers. Despite the trend to spontaneous resolution, our model allowed a double organ readout of the BLM effect and the identification of a therapeutic window for testing pharmacological compounds without using life threatening doses
