    A Novel Cryptic Clostridial Peptide That Kills Bacteria by a Cell Membrane Permeabilization Mechanism

    This work reports detailed characteristics of the antimicrobial peptide Intestinalin (P30), which is derived from the LysC enzyme of Clostridium intestinale strain URNW. The peptide shows a broader antibacterial spectrum than the parental enzyme, showing potent antimicrobial activity against clinical strains of Gram-positive staphylococci and Gram-negative pathogens and causing between 3.04 6 0.12 log kill for Pseudomonas aeruginosa PAO1 and 7.10 6 0.05 log kill for multidrug-resistant Acinetobacter baumannii KPD 581 at a 5 mM concentration. Moreover, Intestinalin (P30) prevents biofilm formation and destroys 24-h and 72-h biofilms formed by Acinetobacter baumannii CRAB KPD 205(reduction levels of 4.28 and 2.62 log CFU/mL, respectively). The activity of Intestinalin is combined with both no cytotoxicity and little hemolytic effect against mammalian cells. The nuclear magnetic resonance and molecular dynamics (MD) data show a high tendency of Intestinalin to interact with the bacterial phospholipid cell membrane. Although positively charged, Intestinalin resides in the membrane and aggregates into small oligomers. Negatively charged phospholipids stabilize peptide oligomers to form water- and ion-permeable pores, disrupting the integrity of bacterial cell membranes. Experimental data showed that Intestinalin interacts with negatively charged lipoteichoic acid (logK based on isothermal titration calorimetry, 7.45 6 0.44), causes membrane depolarization, and affects membrane integrity by forming large pores, all of which result in loss of bacterial viability.</p

    Comparison of the Antibacterial Activity of Selected Deep Eutectic Solvents (DESs) and Deep Eutectic Solvents Comprising Organic Acids (OA-DESs) Toward Gram-Positive and Gram-Negative Species

    Deep eutectic solvents (DESs) have been intensively investigated in recent years for their antibacterial properties, with DESs that comprise organic acids (OA-DESs) showing promising antibacterial action. However a majority of the reports focused only on a limited number strains and techniques, which is not enough to determine the antibacterial potential of a substance. To bridge this gap, the antibacterial activity of classical DESs and OA-DESs is assessed on twelve Gram-negative and Gram-positive bacteria strains, with some of them exhibiting specific resistance toward antibiotics. The investigated formulations of OA-DESs comprise glycolic, malic, malonic, and oxalic acids as representatives of this group. Using a range of microbiological assays as well as physicochemical characterization methods, a major difference of the effectiveness between the two groups is demonstrated, with OA-DESs exhibiting, as expected, greater antibacterial effectiveness than classical DESs. Most interestingly, slight differences in the minimum inhibitory and bactericidal concentration values as well as time-kill kinetics profiles are observed between Gram-positive and Gram-negative strains. Transmission electron microscopy analysis reveals the effect of the treatment of the bacteria with the representatives of both groups of DESs, which allows us to better understand the possible mechanism-of-action of these novel materials.</p

    Improvement of the dialogue between the elder preschool children in the fairy tale production process

Pētījuma mērķis: pētīt teorētiski un praktiski vecākā pirmsskolas vecuma bērnu dialogrunas pilnveides iespējas pasaku iestudēšanas procesā. Pētījuma teorētiskajā daļā tiek raksturota dialogrunas būtība, analizētas vecākā pirmsskolas vecuma bērnu attīstības likumsakarības pētāmās problēmas aspektā, kā arī literāro darbu izmantošanas iespējas dialogrunas pilnveidei vecākā pirmsskolas vecuma bērniem. Pētījuma empīriskajā daļā raksturotas pētījuma izmantotās metodes – pedagoģiskais vērojums, izmēģinājumdarbība. Izstrādāti bērnu vērošanas kritēriji, veikta sākotnējā bērnu dialogprasmju vērošana, attīstītas bērnu dialogrunas prasmes tautas un literāro pasaku iestudējumos, veikta atkārtota bērnu prasmju vērošana un rezultātu salīdzināšana. Realizētie iestudējumi izmēģinājumdarbības laikā pierādīja, ka ar dažādu pasaku iestudējumiem ir iespējams uzlabot bērnu runu, tieši dialogrunu, jo bērni vairāk sāk lietot vienkāršus paplašinātus un saliktus teikumus, ievērot runas tempu, regulēt balss skaļumu, atdarināt dažādas balss intonācijas, nepārtraukt citus, ieklausīties sarunu biedrā. Aim of the research: to research posibilities of perfection of elder preschool children’s tales theoretically and practically. The theoretical part of the research consists of dialogue speech description, analysis of connections between the subject of the research and the development of elder preschool children and possibilities of using literary work in the development of the elder preschool children. The empirical part contains the description of research methods used- pedagogial observation and trialing. Observation of children criteria has been developed, observation of basic children’s dialogue skills has been done, children’s skills in dialogue speech during native and literary plays have been improved, repeated observation of children’s skills and comparison of the results has been made. The trailing of plays has proven that with different plays it is possible to improve children’s speech, especially dialogue speech, because children tend to use more simple expanded and perfect sentences, control the temp of their speech, regulate the volume of their voice, imitate different voice intonations, not interrupt others.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Plotka - mimo że jest stale obecna w różnych sferach funkcjonowania mass mediów - nie stała się w Macedonii przedmiotem badań. Tematyka szerzenia nieprawdziwych informacji, pomówień, przekazywania wiadomości w określonym celu była w mediach szczególnie aktualna w 2015 roku jako informacja "bomba". Materiał badawczy służący definiowaniu tego typu wypowiedzi został zaczerpnięty z tabloidów i stanowi część zjawiska opisywanego jako "mowa nienawiści", która umożliwia szerzenie na masową skalę informacji w określonym celu. W artykule została przedstawiona analiza zjawiska plotki występującej w macedońskich środkach masowego przekazu. Szczególną uwagę poświęcono językowym środkom wykorzystywanym w tego typu informacji.Rumour communication, although always present in different areas of mass media in the Republic of Macedonia, has not yet been researched. Topics devoted to various rumours, unverified information spread, "gossiping" circulation of information of specific purpose were particularly popular in 2015 through a part of this kind of information transmitted in the mass media as information "bomb". The scientific approach to define such information was especially present in the yellow press and in researches related to "hate speech" as an opportunity for mass dissemination of specific information of specific purpose. Here is an overview and analysis of rumour communication and the ways it is expressed in the mass media in Republic of Macedonia. Special emphasis is given to the linguistic devices used in the dissemination of such kind of information

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
