Dissecting pathways involved in manganese homeostasis and stress in higher plant cells
Manganese (Mn) is an essential heavy metal micronutrient in plants having a range of cellular functions. Mn homeostasis involves the coordinated operation of transporters mediating cellular import and export and distribution between cell organelles. Significant progress has been made in identifying transport mechanisms in plants responsible for Mn uptake from the soil, distribution around the plant and compartmentalisation within the cell. These processes are required to ensure that Mn reaches its correct target proteins and also for cellular Mn detoxification. Transporters that have been implicated in Mn transport include members of the P2A-type ATPase, ZIP, CAX, CDF, NRAMP and OPT/YSL families. Here we discuss their roles in the acquisition, distribution and homeostasis of Mn and aspects of their regulation
Two additional type IIA Ca2+-ATPases are expressed in Arabidopsis thaliana: evidence that type IIA sub-groups exist
High affinity Ca2+-ATPases play a central role in calcium homeostasis by catalysing the active efflux of calcium from the cytoplasm. This study reports the identification of two additional type IIA (SERCA-type) Ca2+-ATPases from Arabidopsis (AtECA2 and AtECA3), and describes the detailed sequence analysis of these genes in comparison with AtECA1 and other plant and animal Ca2+-ATPases. Southern analysis suggests that each of these genes is present as a single copy and also that there may be a small family of moderately related genes that encode type IIA Ca2+-ATPases in Arabidopsis. Evidence is also provided from RT-PCR that these genes are expressed in Arabidopsis. Hydropathy analysis predicts that the topology of the Arabidopsis type IIA proteins is similar to the animal SERCA proteins. Emerging mechanisms for heavy metal transport in plants
Heavy metal ions such as Cu2+, Zn2+, Mn2+, Fe2+, Ni2+ and Co2+ are essential micronutrients for plant metabolism but when present in excess, these, and non-essential metals such as Cd2+, Hg2+ and Pb2+, can become extremely toxic. Thus mechanisms must exist to satisfy the requirements of cellular metabolism but also to protect cells from toxic effects. The mechanisms deployed in the acquisition of essential heavy metal micronutrients have not been clearly defined although a number of genes have now been identified which encode potential transporters. This review concentrates on three classes of membrane transporters that have been implicated in the transport of heavy metals in a variety of organisms and could serve such a role in plants: the heavy metal (CPx-type) ATPases, the natural resistance-associated macrophage protein (Nramp) family and members of the cation diffusion facilitator (CDF) family. We aim to give an overview of the main features of these transporters in plants in terms of structure, function and regulation drawing on information from studies in a wide variety of organisms. Increased metal tolerance and bioaccumulation of zinc and cadmium in Chlamydomonas reinhardtii expressing a AtHMA4 C-terminal domain protein
The use of microalgal biomass for metal pollutant bioremediation might be improved by genetic engineering to modify the selectivity or capacity of metal biosorption. A plant cadmium (Cd) and zinc (Zn) transporter (AtHMA4) was used as a transgene to increase the ability of Chlamydomonas reinhardtii to tolerate 0.2 mM Cd and 0.3 mM Zn exposure. The transgenic cells showed increased accumulation and internalization of both metals compared to wild-type. AtHMA4 was expressed either as the full-length (FL) protein or just the C-terminal (CT) tail, which is known to have metal-binding sites. Similar Cd and Zn tolerance and accumulation was observed with expression of either the FL protein or CT domain, suggesting that enhanced metal tolerance was mainly due to increased metal binding rather than metal transport. The effectiveness of the transgenic cells was further examined by immobilization in calcium alginate to generate microalgal beads that could be added to a metal contaminated solution. Immobilization maintained metal tolerance, while AtHMA4-expressing cells in alginate showed a concentration-dependent increase in metal biosorption that was significantly greater than alginate beads composed of wild-type cells. This demonstrates that expressing AtHMA4 FL or CT has great potential as a strategy for bioremediation using microalgal biomass.</p
