Functional characterization of the HuR:CD83 mRNA interaction.
No full textMaturation of dendritic cells (DC) is characterized by expression of CD83, a surface protein that appears to be necessary for the effective activation of naïve T-cells and T-helper cells by DC. Lately it was shown that CD83 expression is regulated on the posttranscriptional level by interaction of the shuttle protein HuR with a novel posttranscriptional regulatory RNA element (PRE), which is located in the coding region of the CD83 transcript. Interestingly, this interaction commits the CD83 mRNA to efficient nuclear export via the CRM1 pathway. To date, however, the structural basis of this interaction, which potentially involves three distinct RNA recognition motifs (RRM1-3) in HuR and a complex three-pronged RNA stem-loop element in CD83 mRNA, has not been investigated in detail. In the present work we analyzed this interaction in vitro and in vivo using various HuR- and CD83 mRNA mutants. We are able to demonstrate that both, RRM1 and RRM2 are crucial for binding, whereas RRM3 as well as the HuR hinge region contributed only marginally to this protein:RNA interaction. Furthermore, mutation of uridine rich patches within the PRE did not disturb HuR:CD83 mRNA complex formation while, in contrast, the deletion of specific PRE subfragments from the CD83 mRNA prevented HuR binding in vitro and in vivo. Interestingly, the observed inhibition of HuR binding to CD83 mRNA does not lead to a nuclear trapping of the transcript but rather redirected this transcript from the CRM1- towards the NXF1/TAP-specific nuclear export pathway. Thus, the presence of a functional PRE permits nucleocytoplasmic trafficking of the CD83 transcript via the CRM1 pathway

No full textSilent films in Ireland from 1914 to 1935 focused on Irish rural life, the long struggle for independence from Great Britain, and the civil war that raged from 1922-1923 over the formation of the Irish Free State. Films like those from producers Ellen O’Mara and James Mark Sullivan’s indigenous Film Company of Ireland (1916-1920) specialized in romantic comedies/dramas of Irish life and historical melodramas on the Protestant Ascendency and its exploitation of Irish Catholic tenants. Screenwriter Mary Manning’s Guests of the Nation (1935) explored the Anglo/Irish War (1919-1921) and its devastating impact on the Irish countryside and its people. English-Irish author Dorothea Donn-Byrne worked in and outside this nationalist tradition. As Dolly Byrne, she provided the source material for Enter Madame (US 1922), a drawing room comedy about a self-absorbed opera singer and her troubled marriage, and, as Dorothea Donn Byrne, she wrote the original story for Land of Her Fathers (IE/US 1925), a romantic drama set against the backdrop of the Anglo/Irish War. In her prolific career, Donn-Byrne wrote for the stage and screen, and penned essays and short stories on a variety of topics that reflected the political and popular culture of the Anglo/Irish/American societies in which she lived. Her work in cinema points to the early dependence on source authors and story writers—particularly women—the nature of their contribution as cinematic authors, and the uncertainty surrounding their involvement in production

The thesis writer is interested in finding the factors that cause their marriage to fail. She assumes that the failure is caused by both Dorothea and Mr. Casaubon. Therefore, the thesis writer needs to divide the factors into two groups; the factors that cause the failure of the marriage from Dorothea?s side as well as from Mr. Casaubon?s side. From the author?s characterization, which reveals Dorothea and Mr. Casaubon?s attitudes, the thesis writer finds out the causes of the failure. Besides, the description of Victorian women and marriage helps the thesis writer understand the causes of the failure. The thesis writer concludes that Dorothea and Mr. Casaubon fail in their marriage because of some factors such their character traits, the absence of love and the difference of age. The fact that Dorothea has been disillusioned about her husband and marriage makes her unhappy with her marriage
