    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     La modification des profils spécifiques en acides gras (AG) dans des huiles végétales peut servir à l’amélioration de leur qualité nutritionnelle, mais également à l’émergence d’une industrie biosourcée où les AG peuvent représenter une alternative aux ressources carbonées fossiles. Cependant des difficultés à modifier le profil en AG des huiles végétales, partagées par l’ensemble des équipes de recherche impliquées subsistent. Bien que la biosynthèse des AG est aujourd’hui bien caractérisée chez les plantes, des incertitudes persistent concernant les voies de transfert des AG du pool d’acyl-CoA vers les précurseurs lipidiques, et les formes de transport entre les différents compartiments impliqués dans les synthèses lipidiques. La carnitine est un acteur essentiel du trafic intracellulaire des AG chez l’animal et la levure. La présence avérée chez la plante d’acyl-Carnitines au côté de la carnitine libre, et la mise en évidence d’activités enzymatiques associées, prouvent également le lien entre la carnitine et le métabolisme des AG. Le travail de cette thèse a donc porté sur un approfondissement des connaissances sur le rôle de la carnitine dans le métabolisme lipidique chez Arabidopsis thaliana. A ce titre, nous avons pu montrer par LC-QQQ-MS/MS dans nos extraits végétaux, que l’oléoyl-, la palmitoyl- et la stéaroyl-Carnitine sont les espèces majoritaires et correspondent aux 3 AG (C18:1, C16:0 et C18:0) principalement synthétisés et exportés des plastes, pour alimenter la synthèse lipidique de la voie eucaryotique. De plus, une augmentation significative des teneurs en acyl-Carnitines a été mesurée dans des processus développementaux nécessitant une forte synthèse de glycérolipides, tels que le développement post-Germinatif (2 j post-Imbibition) ou lors de l’organogenèse racinaire, en concomitance avec le niveau d’expression des gènes marqueurs de la synthèse lipidique eucaryotique. D’autre part, la comparaison des teneurs en acyl-Carnitines et en carnitine estérifiée a révélée un résultat inédit montrant qu’une grande fraction de la carnitine est estérifiée à des AG inhabituels ou des acides organiques que nous cherchons actuellement à identifier par LC-QTOF-MS/MS. Ainsi, l’ensemble de nos résultats suggèrent que la carnitine pourrait être impliquée dans le trafic intracellulaire d’AG pour la synthèse de lipides eucaryotiques et de lipides particuliers et/ou dans la prise en charge d’acides organiques encore non identifiés dans les plantes.Specific modification of fatty acid profiles (FA) in vegetable oils can help to improve their nutritional quality but also to the development of a bio-Based industry where FA may represent an alternative to fossil fuel sources. However problems to modify the FA profile of vegetable oils still remain and are shared by all the teams involved. Although the biosynthesis of AG is now well characterized in plants, uncertainties remain about the tranfert of AG of acyl-CoA pool to lipid precursors and the forms of transport between different compartments involved in lipid synthesis. Carnitine plays an essential role in intracellular trafficking of FA in animals and yeast. The known presence of acyl-Carnitines in plant alongside the free carnitine, and measurement of associatied-Enzymatic activities also show the link between FA metabolism and carnitine. During this thesis we tried to improve our understanding of the role of carnitine in lipid metabolism in Arabidopsis thaliana. We have shown by LC-QQQ-MS / MS in our plant extracts that oleoyl-, palmitoyl- and stearoyl-Carnitine are the predominant species and correspond to the three main AG (C18:1, C16:0 and C18:0) synthesized and exported from the plastids to supply the lipid synthesis of the eukaryotic pathway. Moreover, a significant increase of the levels of acyl-Carnitines were measured in developmental processes requiring high glycerolipid synthesis, such as the post-Germinative growth (2 days post-Imbibition) during root organogenesis, concomitantly with the level of expression of the eukaryotic lipid synthesis marker genes. Furthermore, comparison of the levels of acyl-Carnitine and carnitine ester revealed an unpublished results showing that a large fraction of carnitine is esterified to unusual FA or organic acids that we are currently identifing by LC QTOF-MS/MS. Thus, all of our results suggest that carnitine could be involved in the intracellular traffic of FA for the synthesis of lipids and particular eukaryotic lipids and/or in the transport of yet unidentified organic acids in plants

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used

    Role of ALDH10s enzymes in cartinine biosynthesis and plants physiologie in Arabidopsis

    La carnitine est un acide aminé non protéinogène présent dans l’ensemble du vivant. Son rôle dans le transport intracellulaire des acides gras est bien caractérisé chez les animaux. Chez les plantes aussi, la carnitine intervient dans le métabolisme lipidique, mais son rôle précis demeure peu documenté. Chez les animaux et les levures, la carnitine est synthétisée à partir de la triméthyllysine (TML) via 4 réactions enzymatiques. L’identification chez les plantes de la TML et de la γ-butyrobétaïne, le précurseur direct de la carnitine, suggère que la voie de biosynthèse de la carnitine chez les plantes est similaire à celle des animaux. Une recherche d’homologues a permis d’identifier les deux aldéhyde déshydrogénases 10 (ALDH10) d’Arabidopsis comme potentiellement impliquées dans la synthèse de γ-butyrobétaïne. Dans le cadre de ce travail de thèse, l’implication des AtALDH10 dans la biosynthèse de carnitine et dans la physiologie de la plante a été étudiée. Dans un premier temps, les AtALDH10 recombinantes ont été caractérisées in vitro. Cette étude indique qu’elles peuvent synthétiser la γ-butyrobétaïne, ainsi que des osmoprotectants. Ensuite, un mutant perte de fonction pour les deux AtALDH10 a été généré et étudié. Il présente une sensibilité accrue au stress salin, une hampe florale plus haute, un nombre plus élevé de hampes florales secondaires, et des siliques de taille réduite contenant moins de graines viables. Les graines parvenant à maturité sont enrichies en protéines et en lipides, et le développement de la plante est plus rapide. Ces résultats suggèrent une intervention des AtALDH10 dans la réponse au stress hydrique, l’architecture de la plante, le développement précoce. La teneur en γ-butyrobétaïne dans les tissus est significativement réduite chez le mutant, confirmant le rôle des AtALDH10 dans la synthèse de γ-butyrobétaïne in planta. La teneur résiduelle en γ-butyrobétaïne semble suffisante pour maintenir une teneur en carnitine inchangée. Cela pose la question de la régulation de la voie de biosynthèse de la carnitine chez les plantes, et suggère un lien entre la γ-butyrobétaïne et la réponse au stress, les voies de signalisation et l’architecture de la plante. Outre ce travail, une recherche bioinformatique a permis d’identifier une protéine d’Arabidopsis dont la structure est proche de celle de l’enzyme humaine responsable de la conversion de la γ-butyrobétaïne en carnitine. L’étude de cette protéine n’a pas confirmé son implication dans la synthèse de carnitine. L’identification et la caractérisation de mutants affectés dans la synthèse de carnitine ou son intervention dans le transport des acides gras serait précieuse pour une meilleure compréhension du rôle de la carnitine chez les plantes.Carnitine is a non-proteinogenic amino acid present in all living organisms. Its role in theintracellular transport of fatty acids is well characterized in animals. In plants also, carnitine islinked to lipid metabolism, but its precise role remains poorly understood. In animals andyeasts, carnitine is synthetized from trimethyllysine (TML) through 4 enzymatic steps. The identification in plant tissues of TML and γ-butyrobetaine, the direct precursor of carnitine,suggests that its biosynthesis follows the same steps. A bioinformatics research of candidatesidentified the two ALDH10 enzymes as possibly involved in the synthesis of γ-butyrobetaine.The present study deals with the characterization of the AtALDH10s, to elucidate their role incarnitine biosynthesis and plant physiology. Firstly, the two AtALDH10s were characterized in vitro. This study demonstrates their capability to synthetize γ-butyrobetaine, as well as osmoprotectants. Secondly, an Arabidopsis loss-of-function for the two AtALDH10 has beenobtained and characterized. This mutant is more sensitive to salt stress, bears a longer main shoot and more secondary shoots, and smaller siliques containing less seeds. Its viable seeds are enriched in proteins and fatty acids, and the seedling development is faster. These resultssuggest an intervention of AtALDH10s in the resistance to osmotic stress, the control of plant architecture and the early development. The γ-butyrobetaine content is reduced in the mutant, confirming the involvement of AtALDH10s in the synthesis of γ-butyrobetaine in planta. The residual content of γ-butyrobetaine seems to be sufficient to maintain an unchanged level of carnitine in the mutant. These results raise the question of the regulation of the carnitine biosynthesis pathway and suggest that γ-butyrobetaine could be involved in response to osmotic stress, signalisation pathways and plant architecture. Besides this work, a bioinformatic research lead to the identification of a protein with a structure close to that of the enzyme involved in the conversion of γ-butyrobetaine into carnitine in humans. However, the characterization of this protein did not confirm its involvement in carnitine biosynthesis. The identification and characterization of mutants impaired in enzymatic activities linked to carnitine, such as its biosynthesis or its intervention in the transport of fatty acids, would be valuable to pave the way of a better understanding of the role of carnitine in plants

    L'huile de lin est considérée comme une des plus riches en acide linolénique (oméga- 3), avec certaines variétés de lin oléagineux produisant jusqu'à 65% d'acide a-linolénique. Cette huile utilisée depuis longtemps en industrie chimique pour la confection de peintures, vernis, agents tensioactifs, présente aussi des bénéfices pour la santé humaine avec un apport journalier d'environ 2 g.f 1 (diminution de la pression artérielle, prévention de thrombose...). Il parait donc raisonnable aujourd'hui d'augmenter la production de l'huile de lin riche en oméga-3 pour répondre à ces différents besoins. La sélection de cultivars producteurs d'huile de la qualité demandée devient impérative. Pour cela, des connaissances sur les mécanismes de synthèse et d'accumulation AG polyinsaturés (PUFA) dans les graines de lin sont à acquérir. Dans ce contexte, nous avons évalué le rôle des enzymes de type LPAAT dans l'accumulation des PUFA dans le but de sélectionner des isoformes compétitives dans leur accumulation au niveau des TAG. Les gènes Lpaat ont été isolés à partir des graines de lin âgées de 20 JAF, phase active de remplissage en huile. La fonctionnalité des protéines correspondantes a été testée par complémentation dans une souche JC201 d’E. coli déficiente en activité LPAAT. Une caractérisation de ces enzymes a été effectuée par des tests in vitro et in vivo. Les résultats ont montré la présence d'une isoforme, nommée LPAAT2A, dont l'activité LPAAT présente une spécificité et sélectivité élevées vis-à-vis des PUFA. Dans un 2ème temps, nous avons mesuré in vitro et in vivo le potentiel des enzymes LPAAT provenant de litchi et de lin vis-à-vis des AG de type cyclopropanique. Ces AG possèdent des propriétés physico-chimiques intéressantes pour l'industrie des lubrifiants et des cosmétiques et sont produits par une enzyme spécifique « la CFA synthase ». Nous avons généré des lignées d'Arabidopsis exprimant la CFA synthase d’E.coli puis co-exprimé le gène Lpaat de litchi. Une augmentation de la teneur en AGC (AG cycliques) dans les lignées transgéniques obtenues reflète la spécificité de la LPAAT de litchi vis-à-vis des AGC. Nous nous sommes aussi intéressés aux enzymes de types sPLA2a (phospholipase A2). L'isolement du gène correspondant a été effectué à partir des graines de lin pendant la phase active de remplissage en huile. La caractérisation de son activité a été effectuée par des tests in vitro et in vivo. Les résultats ont montré que l'expression du gène sP/a2a de lin dans les graines d' Arabidopsis provoque une augmentation du poids de la graine ainsi qu'une augmentation du contenu en acide linolénique dans les TAG. L'ensemble de ces résultats montre l'existence d'un système enzymatique endogène chez le lin efficace vis-à­vis des PUFA et qui semble stimuler le métabolisme lipidique une fois exprimé chez Arabidopsis.Flaxseed oil contain high amount of oméga-3 and present different industrials applications and human health benefits. ln this work, we aimed to identify enzymes allowing the accumulation of high level of omega-3 in linseed plants. ln this context, we evaluated the role of Lysophophatidic acid acyltransferases (LPAAT) and Phospholipases A2 (PLA2) enzymes in the accumulation of omega-3 by in vitro and in vivo approaches. Results showed the presence of a LPAAT2A isoform into flax genome having a high specificity and selectivity toward omega-3. Expression of Lpaat2A and sPia2a gene in Arabidopsis seeds increase seed weight, oil production and omega-3 content (up to 10% and 11%) in transformant seeds respectively. These results showed the presence in linseed plant of an efficient enzymatic system toward omega-3 accumulation. Furthermore, we have evaluated in vitro and in vivo the potential of Litchi and flax LPAAT enzymes toward the production of cyclopropane fatty acids (CFA). These fatty acids are naturally produced by a specific enzyme «CFA synthase» with physico-chemical properties interesting to lubricant and cosmetics industries. ln this context, we have generated Arabidopsis lines expressing E. coli Cfa synthase with or not co-expression of litchi Lpaat gene. Result showed an increase of 25% in the content of CFA in transgenic line co-expressing Cfa synthase and litchi Lpaat comparing to the transgenic lines expressing the CFA synthase. This increase in the level of CFA in transgenic seeds reflects the specificity of Litchi LPAAT toward CFA
