    Chiens chez Modiano: análisis interdisciplinar de la representación animal en su obra

    Más que por un animalismo strictu sensu, la obra de Patrick Modiano se caracteriza por un profundo animalismo latente, en consonancia con una escritura minimalista marcada por la atenuación y por la poética de la ambigüedad. La investigación desvela cómo los cuentos de Marcel Aymé aportan a su imaginario de lector infantil un mundo en el que los animales viven y dialogan con una pareja de hermanas con las que el pequeño Modiano se identifica junto a Rudy, su hermano pequeño. Los accidentes que sufrieron dos perros con los que Modiano tuvo relación de niño le hicieron percibir una experiencia de la muerte, que precedió al trauma de la temprana desaparición de Rudy. A la influencia de Aymé se une también la de un Raymond Queneau, gran “animalier” poética y vitalmente, que actuó como mentor del escritor en ciernes. La investigación pone también el foco en dos animales de juguete que actúan como talismanes, u objetos transicionales, con los que el Modiano niño alivia su soledad en los internados en los que ha sido confinado. Asimismo destaca el rol de compensación de las carencias afectivas familiares que desempeñaron dos perros con los que Modiano vagabundeó en su juventud. El análisis de Lacombe Lucien pone de manifiesto la relevancia de la representación animal a lo largo del film en el que aparece la figura de un gran perro melancólico que se prolongará en Villa Triste, donde se convierte en un espejo del narrador y en un “voyeur d’ombres”. A partir de Livret de famille, los animales se incorporan a la compleja combinación de oposiciones binarias que recorre el conjunto de su obra, especialmente al binomio identidad/alteridad. Y el tema de la caza aparece por primera vez como una metáfora de la persecución de los judíos, en la que además incrusta la figura de un padre representado como víctima, a pesar de haber sido un colaborador. En su narrativa, la figura del perro es a veces un explorador anímico en el deambular del protagonista de sus novelas por las calles de Paris, a veces un psicopompo en la búsqueda fantasmagórica de personajes desaparecidos, en ocasiones un doble simbólico del narrador cuando no del hermano muerto e, incluso de ambos, en un sujeto desdoblado que los aúna: le chien et moi / mon frère et moi. De forma que la muerte del hermano, y el temor a ser también él víctima mortal del abandono familiar, se expresan en la autoficción mediante el recuerdo encubridor de la muerte del perro de su infancia. La figura del perro se convierte así en una máscara con la que el autor puede “mentir vrai” y ajustar cuentas con sus progenitores, especialmente con la madre. Además, la decodificación que hacemos del criptograma de la lista de los garajes de Rémise de peine vendría a corroborar la hipótesis del profesor Lecarme según la cual los padres dejaron morir a Rudy a causa de una negligencia En Dieu prend-il soin de bœufs?, por un lado se condensan todas las metáforas animales de su obra y por otro, el sujeto desdoblado se unifica en un definitorio sobrenombre: le chien noir du chagrin. El estudio comparado de este cuento con La femme pauvre de Léon Bloy nos ha permitido descubrir la profunda influencia de su animalismo en Modiano. Asimismo, la investigación demuestra que en su obra la idea poética de la figura del perro encarnando almas errantes, tiene su origen en la leyenda sobre Charles Cros –que creía que su perro llevaba el alma de Baudelaire– y en la influencia de algunos poemas del propio Cros y de Tristan Corbière. La tesis demuestra finalmente la influencia en Modiano de la simbología animal de Los apuntes de Malte Laurids Brigge y de la octava de las Elegías de Duino, de Rainer María Rilke.More than a strictu sensu animalism, the work of Patrick Modiano is characterized by a deep latent animalism, in consonance with a minimalist writing marked by the attenuation and by the poetics of ambiguity. The investigation reveals how the tales of Marcel Aymé bring to his childhood reader’ imaginary a world in which animals live and dialogue with a pair of sisters with whom the child Modiano, together with his little brother Rudy, identifies with. The accidents suffered by two dogs with which Modiano had a childhood relationship made him perceive an experience of death, which preceded the trauma of Rudy’s early disappearance. To the influence of Aymé is also united the one of Raymond Queneau, great "animalier" poetically and vitally, that acted like mentor of the budding writer. The research also focuses on two toy animals that act as talismans, or transitional objects, with which the child Modiano alleviates his loneliness in the boarding schools in which he has been confined. It highlights as well the role of compensation for the family affective deficiencies that played two dogs with which Modiano wandered in his youth. Lacombe Lucien’s analysis reveals the relevance of animal representation throughout the film in which appears the figure of a great melancholic dog that will be prolonged in Villa Triste, where it becomes a mirror of the narrator and a "voyeur d’ombres ". From Livret de famille, animals are incorporated into the complex combination of binary oppositions that runs throughout his work, especially the identity/alterity binomial. And the subject of the hunt appears for the first time as a metaphor of the persecution of the Jews, in which also it inlays the figure of a father represented like a victim, in spite of having been a collaborator. In his narrative, the figure of the dog is sometimes a psychic explorer in the wandering of the main character of his novels in the streets of Paris, sometimes a psychopompo in a phantasmagoric search for missing characters, sometimes a symbolic double of the narrator or of the dead brother, and even of both, in a split subject that unites them: le chien et moi / mon frère et moi. So that the death of the brother, and the fear of being himself also the mortal victim of family abandonment, are expressed in autofiction by the screen memory of the death of the dog of his childhood. The figure of the dog thus becomes a mask with which the author can "mentir vrai" and settle a score with his parents, especially with the mother. In addition, the decoding we do of the cryptogram of the list of the garages of Rémise de peine would corroborate the hypothesis of Professor Lecarme, according to which the parents let Rudy die because of negligence. In Dieu prend-il soin de bœufs, on the one hand all the animal metaphors of his work are condensed and, on the other, the splitted subject is unified in a definitory nickname: le chien noir du chagrin. The comparative study of this tale with La femme pauvre by Léon Bloy has allowed us to discover the profound influence of his animalism in Modiano. Research also shows that in his work the poetic idea of the figure of the dog incarnating wandering souls has its origin in the legend about Charles Cros –who believed that his dog carried the soul of Baudelaire– and in the influence of some poems of Cros himself and Tristan Corbière. The thesis finally shows the influence in Modiano of the animal symbology of The Notebooks of Malte Laurids Brigge and of the eighth of Duino Elegies, by Rainer Maria Rilke

    En Modiano la figura del gos és a vegades un explorador anímic en el deambular del protagonista pels carrers de París, a vegades un guia en la cerca fantasmagòrica de personatges desapareguts, a vegades un doble simbòlic del narrador, quan no del germà mort, i fins i tot de tots dos, en un subjecte desdoblegat que els conjumina: el gos i jo / el meu germà i jo; de manera que la mort del germà i el temor a ser també ell víctima de l’abandó familiar s’expressen en la seua narrativa mitjançant el record encobridor de la mort d’un gos durant la seua infància. La figura del gos es converteix així en una màscara amb la qual l’autor d’“Un pedigrí” utilitza la veritat de les mentides literàries per a ajustar comptes amb els seus progenitors, especialment amb la mare. La descodificació d’una misteriosa llista de garatges modianescos revela la soterrada acusació de l’escriptor als seus pares per la seua negligència en la mort del fill xicotet, a qui l’escriptor ha rendit un homenatge secret en el Nobel Museum. Aquestes pàgines constitueixen una aproximació a la vida i a l’obra de Patrick Modiano a partir d’una original anàlisi de la figura del gos en la seua narrativa. El llibre mostra com els contes de Marcel Aymé aporten al seu imaginari de lector infantil un món en el qual els animals viuen i dialoguen amb una parella de germanes amb les quals el xicotet Modiano s’identifica al costat del seu germà Rudy.     The combination of transcriptomics and informatics identifies pathways targeted by miR-204 during neurogenesis and axon guidance

    Vertebrate organogenesis is critically sensitive to gene dosage and even subtle variations in the expression levels of key genes may result in a variety of tissue anomalies. MicroRNAs (miRNAs) are fundamental regulators of gene expression and their role in vertebrate tissue patterning is just beginning to be elucidated. To gain further insight into this issue, we analysed the transcriptomic consequences of manipulating the expression of miR-204 in the Medaka fish model system. We used RNA-Seq and an innovative bioinformatics approach, which combines conventional differential expression analysis with the behavior expected by miR-204 targets after its overexpression and knockdown. With this approach combined with a correlative analysis of the putative targets, we identified a wider set of miR-204 target genes belonging to different pathways. Together, these approaches confirmed that miR-204 has a key role in eye development and further highlighted its putative function in neural differentiation processes, including axon guidance as supported by in vivo functional studies. Together, our results demonstrate the advantage of integrating next-generation sequencing and bioinformatics approaches to investigate miRNA biology and provide new important information on the role of miRNAs in the control of axon guidance and more broadly in nervous system development. © The Author(s) 2014.     Increased serum miR-193a-5p during non-alcoholic fatty liver disease progression: Diagnostic and mechanistic relevance

    Background & Aims: Serum microRNA (miRNA) levels are known to change in non-alcoholic fatty liver disease (NAFLD) and may serve as useful biomarkers. This study aimed to profile miRNAs comprehensively at all NAFLD stages.Methods: We profiled 2,083 serum miRNAs in a discovery cohort (183 cases with NAFLD representing the complete NAFLD spectrum and 10 population controls). miRNA libraries generated by HTG EdgeSeq were sequenced by Illumina NextSeq. Selected serum miRNAs were profiled in 372 additional cases with NAFLD and 15 population controls by quantitative reverse transcriptase PCR.Results: Levels of 275 miRNAs differed between cases and population controls. Fewer differences were seen within individual NAFLD stages, but miR-193a-5p consistently showed increased levels in all comparisons. Relative to NAFL/non-alcoholic steatohepatitis (NASH) with mild fibrosis (stage 0/1), 3 miRNAs (miR-193a-5p, miR-378d, and miR378d) were increased in cases with NASH and clinically significant fibrosis (stages 2-4), 7 (miR193a-5p, miR-378d, miR-378e, miR-320b, miR-320c, miR-320d, and miR-320e) increased in cases with NAFLD activity score (NAS) 5-8 compared with lower NAS, and 3 (miR-193a-5p, miR-378d, and miR-378e) increased but 1 (miR-19b-3p) decreased in steatosis, activity, and fibrosis (SAF) activity score 2-4 compared with lower SAF activity. The significant findings for miR-193a-5p were replicated in the additional cohort with NAFLD. Studies in Hep G2 cells showed that following palmitic acid treatment, miR-193a-5p expression decreased significantly. Gene targets for miR-193a-5p were investigated in liver RNAseq data for a case subgroup (n = 80); liver GPX8 levels correlated positively with serum miR-193a-5p.Conclusions: Serum miR-193a-5p levels correlate strongly with NAFLD activity grade and fibrosis stage. MiR-193a-5p may have a role in the hepatic response to oxidative stress and is a potential clinically tractable circulating biomarker for progressive NAFLD.Lay summary: MicroRNAs (miRNAs) are small pieces of nucleic acid that may turn expression of genes on or off. These molecules can be detected in the blood circulation, and their levels in blood may change in liver disease including non-alcoholic fatty liver disease (NAFLD). To see if we could detect specific miRNA associated with advanced stages of NAFLD, we carried out miRNA sequencing in a group of 183 patients with NAFLD of varying severity together with 10 population controls. We found that a number of miRNAs showed changes, mainly increases, in serum levels but that 1 particular miRNA miR-193a-5p consistently increased. We confirmed this increase in a second group of cases with NAFLD. Measuring this miRNA in a blood sample may be a useful way to determine whether a patient has advanced NAFLD without an invasive liver biopsy. (C) 2021 The Author(s).     Analysis of miRNA signatures in CSF identifies upregulation of miR-21 and miR-146a/b in patients with multiple sclerosis and active lesions

    Background MicroRNAs (miRNAs) have been reported as deregulated in active brain lesions derived from patients with multiple sclerosis (MS). In there, these post-transcriptional regulators may elicit very important effects but proper identification of miRNA candidates as potential biomarkers and/or therapeutic targets is scarcely available. Objective The aim of the study was to detect the presence of a set of candidate miRNAs in cell-free cerebrospinal fluid (CSF) and to determine their association with gadolinium-enhancing (Gd+) lesions in order to assess their value as biomarkers of MS activity. Methods Assessment of 28 miRNA candidates in cell-free CSF collected from 46 patients with MS (26 Gd+ and 20 Gd− patients) was performed by TaqMan assays and qPCR. Variations in their relative abundance were analyzed by the Mann-Whitney U test and further evaluated by receiver operating characteristic (ROC) analysis. Signaling pathways and biological functions of miRNAs were analyzed using bioinformatic tools (miRTarBase, Enrichr, REVIGO, and Cytoscape softwares). Results Seven out of 28 miRNA candidates were detected in at least 75% of CSF samples. Consistent increase of miR-21 and miR-146a/b was found in Gd+ MS patients. This increase was in parallel to the number of Gd+ lesions and neurofilament light chain (NF-L) levels. Gene Ontology enrichment analysis revealed that the target genes of these miRNAs are involved in biological processes of key relevance such as apoptosis, cell migration and proliferation, and in cytokine-mediated signaling pathways.     Em 1955, a pedido do diretor do TBC, Zbigniew Ziembinski, Manuel Bandeira traduz o drama histórico Maria Stuart (1801), de Friedrich Schiller. O poeta pernambucano, que já traduzira poemas de Goethe, Heine, Hölderlin e Rilke, enfrenta então sua primeira tradução de longo fôlego da língua alemã, e sua primeira tradução para o teatro. Partindo das formulações do pensamento alemão dos séculos XVIII e XIX, além do ensaio \"A tarefa do tradutor\", de Walter Benjamin, a respeito da tradução como forma, do tradutor como autor e da tradução como exercício integrante do processo de formação do poeta, a análise busca os traços ínsitos da poética e da versificação bandeirianas que se caracterizam como marcas d\'água com que o tradutor registra sua presença no texto schilleriano. Recursos recorrentes na poesia de Bandeira, bem como questões levantadas em seus trabalhos críticos, presentes em conferências, crônicas e antologias, são mobilizados de modo a tornar o texto traduzido como indissociável da obra de seu tradutor. Desse modo, a tradução de Maria Stuart pode ser vista como uma obra a quatro mãos, em que Bandeira, a cada escolha feita, manifesta seu próprio entendimento da peça e estabelece um diálogo com o poeta de Marbach.In 1955, at the request of the theater director Zbigniew Ziembinski, Manuel Bandeira translates the historical drama Maria Stuart (1801), by Friedrich Schiller. The poet who had already translated poems by Goethe, Heine, Hölderlin and Rilke, then faces his first long-term translation of the German language, and his first translation of a stage play. Starting from the formulations of 18th and 19th centuries German philosophy, as well as those from the essay \"The translator\'s task\", by Walter Benjamin, about translation as a form, the translator as author and translation as an integral exercise in the poet\'s formation process, the analysis seeks the embedded traits of Bandeiras poetics and versification that are characterized as watermarks with which the translator registers his presence in the Schillerian text. Recurring proceeds in Bandeira\'s poetry, as well as questions raised in his critical works, present in conferences, chronicles and anthologies, are mobilized in order to make the translated text as inseparable from the work of its translator. Therefore, Maria Stuart\'s translation can be seen as a four-handed work, in which Bandeira manifests through his choices his own understanding of the play and establishes a dialogue with the poet from Marbach

    Expanded CTG repeats trigger miRNA alterations in Drosophila that are conserved in myotonic dystrophy type 1 patients

    Myotonic dystrophy type 1 (DM1) is caused by the expansion of CTG repeats in the 3' untranslated region of the DMPK gene. Several missplicing events and transcriptional alterations have been described in DM1 patients. A large number of these defects have been reproduced in animal models expressing CTG repeats alone. Recent studies have also reported miRNA dysregulation in DM1 patients. In this work, a Drosophila model was used to investigate miRNA transcriptome alterations in the muscle, specifically triggered by CTG expansions. Twenty miRNAs were differentially expressed in CTG-expressing flies. Of these, 19 were down-regulated, whereas 1 was up-regulated. This trend was confirmed for those miRNAs conserved between Drosophila and humans (miR-1, miR-7 and miR-10) in muscle biopsies from DM1 patients. Consistently, at least seven target transcripts of these miRNAs were up-regulated in DM1 skeletal muscles. The mechanisms involved in dysregulation of miR-7 included a reduction of its primary precursor both in CTG-expressing flies and in DM1 patients. Additionally, a regulatory role for Muscleblind (Mbl) was also suggested for miR-1 and miR-7, as these miRNAs were down-regulated in flies where Mbl had been silenced. Finally, the physiological relevance of miRNA dysregulation was demonstrated for miR-10, since over-expression of this miRNA in Drosophila extended the lifespan of CTG-expressing flies.     Training-induced plasma miR-29a-3p is secreted by skeletal muscle and contributes to metabolic adaptations to resistance exercise in mice

    Objective: The adaptive response to different models of regular exercise involves complex tissue crosstalk. Our aim was to explore the involvement of extracellular vesicle (EV) microRNAs (miRNAs) in this process, the secretory role of skeletal muscle and its functional metabolic interaction with the liver. Methods: Plasma EV miRNAs obtained from mice after 4-weeks of endurance or resistance training were sequenced. Subsequent experiments using trained genetically modified mouse models and in vitro approaches involving knock-down and electrostimulated cells, were conducted. Results: Resistance training increased the expression of a group of 11 miRNAs functionally divided into two clusters. Among them, miR-29a-3p emerges as a molecular mediator released in EVs by skeletal muscle, with a relevant role in adaptation to endurance training, by contributing to modulate the expression and secretion of other miRNAs associated with training and regulating processes related to substrate availability, transport, and metabolic use in skeletal muscle and liver. Conclusions: Our study suggests that miR-29a-3p is a training-induced molecular mediator in the response and adaptation to resistance training, possibly due to its regulatory role in energy metabolism in skeletal muscle in response to exercise.
