    Purpose of review The aim of the review was to provide an overview of safety and efficacy of the available treatments including information from both clinical trials and real-world data. Additional information form ongoing studies using other approaches than increasing SMN protein are also reported. Recent findings In the last 3years, there have been over 24 studies reporting safety and the impact of the available drugs on different aspects of function, including respiratory and bulbar function. These findings, obtained in a real-world setting, are extremely important to define the spectrum of responses in individuals with different age, weight, SMN2 copies, and other variables and will be of help to the families and the clinicians to set up the right expectations at the time of starting a new treatment. Summary The large number of studies that became available in the last few years support and expand the information on safety and efficacy provided by the clinical trials

Increasing amounts in µM concentrations of each protein were used as indicated above each individual gel. Free DNA and protein-DNA complexes were resolved in 6% polyacrylamide gel electrophoresis at 120 V in 0.25X TBE gels and stained with ethidium bromide. For presentation purposes the color of all images was inverted from the original image. A similar result was obtained for the <i>perA</i> promoter region with mutants PerA<sub>Y29A</sub>, PerA<sub>S194A</sub> and PerA<sub>Y255A</sub> (data not shown).</p

    On the efficient numerical simulation of heterogenous anisotropic diffusion models of tumor invasion using GPUs

    Mathematical models for tumor invasion are often used to predict the behaviour of cancer evolution and can produce strikingly nontrivial patterns. Therefore, their numerical solution often requires high spatial resolution to capture detailed biophysical phenomena. As a consequence, long computational times are required when using a serial implementation of numerical schemes. Parallel computing can improve dramatically the time efficiency of some numerical methods such as finite differences algo- rithms, which are relatively simple to implement and apply to tumor invasion models. For clinical operators and applied scientists involved in setting up realistic experiments, the possibility of running fast comparative simulations using simple algorithms implemented into affordable processors is of primary interest, and that is where Graphical Processing Units (GPUs) excel. In this work we focus on a mathematical model of anisotropic and heterogeneous diffusion of tu- mor cells, a set of time-evolution parabolic equations. The model includes chemotaxis and haptotaxis effects, that appear in other biophysical phenomena [1]. We compute numerical solutions consider- ing spatial discretization by centered finite differences and time integration through an explicit Euler method. The choice of time-explicit algorithms is due to their greater ease of implementation, and performance, on GPU devices, despite the limitations related to their reduced stability properties. The codes used in this work are designed using CUDA. The CUDA platform (Compute Unified De- vice Architecture NVIDIA 2007), was designed to support GPU execution of programs and focuses on data parallelism. With CUDA, graphics cards can be programmed with a medium-level language, that can be seen as an extension to C/C++/Fortran, without requiring a great deal of hardware exper- tise.     The ‘margin of appreciation’ in echr case-law as a boundary line to legal transplants

    This study investigates the influence of the European Human Rights Court on State 'margin of appreciation' on the diffusion of accepted or rejected cultural and political choices through the jurisprudence of the case-law. The margin of appreciation is aninterpretative argument, a criterion, a parameter, which allows the Court to preserve rather than censure national State policies and choices on difficult issues which, involving moral and ethical questions, and which do not receive common answers in the ,European legal systems. The work considers some decisions of the Court on abortion, ovum donation, pre-implant embryo analysis and artificial procreation techniques, to find out if, in such subjects, which are all strongly connected to ethical, moral and social values, the case law of the Court is diachronically coherent in its various decisions or if there are considerable and relevant incoherences and interpretative mismatches among them. In this latter hypothesis, the explanation for these contradictions lies in the different use of the margin of appreciation argument and in the single rules and models being scrutinized by the Court. An examination of these suggests that sometimes the ECHR intends to preserve a certain national model through the margin of appreciation and at other times prefers to substitute it with another one, using different arguments, such as the general consensus of contracting States. Indeed, it seems that in the decision-making process the Court's approach is firstly, to consider the choice of the model, and ;particularly that of the rule or the value that has to be promoted or discouraged among the contracting States and secondly, the choice of the hermeneutic argument, that can be adopted in order to achieve this goal, according to an anti-formalist hermeneutic approach.
