Monetary Green Accounting and Ecosystem Services
The point of departure in this paper is that monetary green accounting aims at serving as indicator of wealth changes, sustainable use of natural capital, and performance of environmental policy. It is then investigated how wealth changes and sustainable development of natural capital can be estimated by means of ecosystem services. These services are defined as outputs from natural capital. The value of changes in natural capital, or wealth change, is thus measured as the value of impacts on current and future production of ecosystem services. It is then shown how this measure can be used as an indicator of sustainable use of the aggregate natural capital, and also how it can be applied efficient environmental policies. An empirical demonstration is made to the calculation of wealth changes to Swedish forests, agricultural landscape, wetlands, air quality, and coastal and marine ecosystems. The demonstration shows that the net welfare contribution from these natural capital assets during the period under study is positive, but that the use of the assets is unsustainable.

Darbā izmantotas D.Dzintares, I.Stangaines, M.Montesori, Ē.Eriksona, Ž.Piažē, Ļ. Vigotska, u.c. autoru atziņas. Darbs sastāv no 46 lapaspusēm, tajā skaitā - 9 attēli, 8 tabulas, 3 pielikumi un 17 literatūras un citi avoti. Atslēgas vārdi: adaptācija, rotaļa, empīriskais pētījums, rezultātu analīze.The author of qualification work is Oksana Gren. Work theme is “Use of play elements to promote adaptation to younger preschool children (3 - 4 years old)”. Work main aim is to identify the role of the play and its possibilities for a successful adaptation of 3 - 4 years old children, both in the family, and in the pre-school education institution. The work consists of two parts – theoretical and empirical. In the theoretical part 1. chapter is being analysed the nature and explanation of the concept of adaptation, characteristics and course, but in 2. chapter based on the findings and studies of educators and psychologists is being analysed use of play for pre-school children. In the empirical part is being explored parents’ attitude towards children’s play and analysed controlled plays. The main conclusions are, that parents should be more interested in play and after several playouts, the adaptation of children is improving, it stays easier. The qualification work hypothesis was approved, and the goal was achieved. In the work are used D. Dzintares, I. Stangaines, M. Montesori, Ē. Eriksona, Ž. Piažē, Ļ. Vigotska, etc. authors reviews. The work consists of 46 pages, including 9 pictures, 8 tables, 3 annexes. And the list of 17 literature and other references. Key words: adaptation, play, empirical research, analysis of results
Development and application of genetic engineering methods for Actinoplanes sp. SE50/110
Gren T. Development and application of genetic engineering methods for Actinoplanes sp. SE50/110. Bielefeld: Universität Bielefeld; 2017.The alpha-glucosidase inhibitor acarbose is used for treatment of diabetes mellitus type 2, and is manufactured industrially with overproducing derivatives of _Actinoplanes_ sp. SE50/110. This strain was reportedly optimized through step-by-step conventional mutagenesis procedures in the past, however this strategy seems to reach its limits by now. Despite of high industrial significance, only limited information exists regarding acarbose metabolism, function and regulation of these processes, due to the absence of proper genetic engineering methods and tools developed for this strain. In this work, a full toolkit and set of methods for genetic engineering of _Actinoplanes_ sp. SE50/110 were developed. A standardized protocol for a DNA transfer through _E. coli_ - _Actinoplanes_ conjugation was adjusted and applied for the transfer of phiC31, phiBT1 and VWB actinophage-based integrative vectors and pSG5-based replicative vector. Integration sites, occurring once per genome for all integrative vectors, were sequenced and characterized for the first time in _{Actinoplanes_ sp. SE50/110. Notably, the studied plasmids were proven to be stable and neutral with respect to strain morphology and acarbose production, enabling future use for genetic manipulations of _Actinoplanes_ sp. SE50/110. To further broaden the spectrum of available tools, a GUS reporter system, was established in _Actinoplanes_ sp. SE50/110. The set of different methods for gene knockouts was tested, which included integrative and replicative vector based knockouts, ReDirect system based knockouts and CRISPR-Cas9 genetic engineering. ReDirect system was further used to create a library of _Actinoplanes_ single knockout strains. Two of the strains, _Actinoplanes_ _acbD_ and _Actinoplanes_ _cadC_ knockout mutants were further characterized in detail regarding their phenotype.
The developed gene cloning system offers multiple possibilities to solve fundamental questions regarding acarbose production, in particular, formulation and verification of the complete acarbose metabolism model, as well as the rational design of acarbose overproducing strains
A pulsed TV holography system for the study of transients in experimental mechanics
An all-electronic system for pulsed holographic interferometry called pulsed TV holography is developed. This is a whole-field non-contacting optical measurement method suitable for studies of transient events like wave propagation in solids and fluids. Chemical wet processing of holographic film and optical reconstruction of holograms are no longer needed. The technique was first developed using a double pulsed ruby laser as light source. The holograms are recorded directly on a CCD-detector. Quantitative data of changes in optical path length, caused either by a deformation of a solid object or a change in refractive index in a fluid, are calculated directly in a computer. The system for pulsed TV holography has recently been further developed by the purchase of a new pulsed laser (twin cavity, injection seeded pulsed Nd:YAG) and a CCD camera (PCO Sensicam) with higher spatial resolution and dynamic range. In the survey of this thesis the increased versatility compared to a ruby laser based system is discussed. During the development of the pulsed TV holography system a number of experiments in mechanics and acoustics have been accomplished. Bending waves in impacted plates propagating at a speed of about 2000 m/s are easily “frozen” due to the short duration laser pulses (<30 ns). These waves act as supersonic travelling acoustic sources and generate sound waves in the surrounding air. For the first time, transient sound fields from impacted plates have been visualised and measured using pulsed holographic interferometry. In another experiment, we have demonstrated that the pulsed TV holography system is feasible in combination with tomography. By recording a three-dimensional acoustic pressure field from a number of viewing directions followed by a tomographic reconstruction, the pressure in any point can be calculated. Finally, a method to restore fringes lost by large bulk motions is proposed. This technique may become very attractive in the study of vibrations (preferable transient) on moving or rotating objects. In conclusion, pulsed TV holography is proved to be a fast and reliable method to quantitatively study transients in mechanics and acoustics. The technique has a great potential in experimental mechanics in the future.Godkänd; 2000; 20061116 (haneit
