    GNCnn: A QuPath extension for glomerulosclerosis and glomerulonephritis characterization based on deep learning

    The digitalization of traditional glass slide microscopy into whole slide images has opened up new opportunities for pathology, such as the application of artificial intelligence techniques. Specialized software is necessary to visualize and analyze these images. One of these applications is QuPath, a popular bioimage analysis tool. This study proposes GNCnn, the first open-source QuPath extension specifically designed for nephropathology. It integrates deep learning models to provide nephropathologists with an accessible, automatic detector and classifier of glomeruli, the basic filtering units of the kidneys. The aim is to offer nephropathologists a freely available application to measure and analyze glomeruli to identify conditions such as glomerulosclerosis and glomerulonephritis. GNCnn offers a user-friendly interface that enables nephropathologists to detect glomeruli with high accuracy (Dice coefficient of 0.807) and categorize them as either sclerotic or non-sclerotic, achieving a balanced accuracy of 98.46%. Furthermore, it facilitates the classification of non-sclerotic glomeruli into 12 commonly diagnosed types of glomerulonephritis, with a top-3 balanced accuracy of 84.41%. GNCnn provides real-time updates of results, which are available at both the glomerulus and slide levels. This allows users to complete a typical analysis task without leaving the main application, QuPath. This tool is the first to integrate the entire workflow for the assessment of glomerulonephritis directly into the nephropathologists' workspace, accelerating and supporting their diagnosis

    C-rel deficiency and its effect on proliferation and survival in immunodeficient lymphoblastic B cells

    The analysis of lymphocytes from immunodeficient patients has provided valuable insights into understanding the development and function of the immune system. In particular, studies of patients with Hyper IgM syndrome, a rare immunodeficiency resulting from defects in both CD40L in activated CD4+ T cells and components of the CD40 signaling pathway in B cells have been crucial for the elucidation of molecular pathways involved in the activation, proliferation and differentiation of B lymphocytes. Previously our lab generated EBV transformed lymphoblastic cells with inducible LMP1 from a female patient diagnosed with non-X-linked HIGM (Pt1-LCLtet cells, Pt1). Analysis of these cells revealed a CD40-specific activation defect associated with low c-Rel levels. To broaden our understanding of the Pt1 phenotype we analyzed c-Rel expression and found that low c-Rel levels were the outcome of depressed transcriptional activation of the c-rel promoter. In-vivo analysis revealed that low transcription was coincident with increased p65 activity at the c-rel promoter although this was insufficient to induce normal c-Rel expression. Further investigations of the Pt1-LCLtet cells uncovered a proliferation and survival defect characterized by reduced c-Myc transcription and increased levels of caspase-4. This defect was complemented after c-Rel overexpression, supporting a causative role for c-Rel deficiency in the observed phenotype of the Pt1-LCLtet cells. The LCLtet cells were further utilized as a model system for the independent analysis of LMP1 and CD40 signaling. These studies revealed several outcomes of LMP1 signaling in the biology and function of CD40. Specifically, LMP1 signaling induced a unique pattern of CD40 phosphorylation as well as the expression of several novel CD40 isoforms. Moreover, when CD40 and LMP1 were activated simultaneously a synergistic effect was observed towards p65 and p38 phosphorylation suggesting a functional interaction that might occur during early stages of EBV infection or malignant transformation where both LMP1 and CD40 are co-expressed in the infected cell. Collectively, our results demonstrate that a majority of the defects displayed in the Pt1-LCLtet cells are a result of reduced c-Rel expression. Importantly, these results ascribe c-Rel a novel function as a negative regulator of caspase-4 expression and a putative key player in the control of cell survival in EBV-immortalized B cells.Ph.D.Includes bibliographical referencesby Anibal J.     Regulation of yeast choline kinase by zinc depletion and the yeast phosphatidate phosphatase by zinc depletion and growth

    In the yeast Saccharomyces cerevisiae, the CKI1-encoded choline kinase catalyzes the committed step in the synthesis of phosphatidylcholine via the CDP-choline branch of the Kennedy pathway. Analysis of a PCKI1-lacZ reporter gene revealed that CKI1 expression was regulated by intracellular levels of the essential mineral zinc. Zinc depletion resulted in a concentration-dependent induction of CKI1 expression. This regulation was mediated by the zinc-sensing and zinc-inducible transcriptional activator Zap1p. A purified Zap1p probe interacted with two putative UASZRE sequences (ZRE1 and ZRE2) in the CKI1 promoter. Mutations of ZRE1 and ZRE2 to a nonconsensus UASZRE attenuated the induction of CKI1 expression in response to zinc depletion. A UASINO element in the CKI1 promoter was responsible for stimulating CKI1 expression, but this element was not involved in the regulation by zinc depletion. The induction of CKI1 expression in zinc-depleted cells translated into increased choline kinase activity in vitro and in vivo, and an increase in phosphatidylcholine synthesis via the Kennedy pathway. The yeast PAH1-encoded PA phosphatase (PAP) catalyzes the dephosphorylation of PA to yield DAG and Pi. The DAG produced by the PAP reaction is utilized for the synthesis of TAG and the synthesis of PE and PC via the Kennedy pathway. Analysis of the PPAH1-lacZ reporter gene demonstrated that PAH1 expression was regulated by intracellular levels of zinc. The induction of the PAH1 gene in response to zinc depletion was dependent on the concentration of zinc. PAP regulation by zinc depletion was mediated by the zinc responsive transcription factor Zap1p. Purified Zap1p interacted with three putative UASZRE sequences located in the PAH1 promoter. The Zap1p mediated induction in PAH1 expression in response to zinc depletion resulted in an increase in PAP activity in vitro. The induction in PAP activity in zinc-depleted cells translated into an increase in DAG and TAG levels. The pah1Δ mutation resulted in the decrease of PC synthesis via the Kennedy pathway in response to zinc depletion. The regulation of the yeast PAH1-encoded PAP by growth was examined. The DAG produced in the PAP reaction is converted into the storage lipid TAG throughout growth. As cells progress from the exponential to the stationary phase of growth the PAP activity is induced, resulting in an increased in DAG and TAG levels. The induction in PAP activity coincided with a decrease in PAP protein levels.     The ambassador of Spain to the Russian Empire Anibal Morillo y Pérez del Villar and his “Memories”

    The article examines the  memoirs of a Spanish diplomat Anibal Morillo and Perez del  Villar, the   Count of  Cartagena. He  held  the   post of the  Spanish ambassador in the  Russian  Empire in 1914-1916 when World War  One  broke out.  “Memories  of my Embassy in Russia”  by Morillo is a specific source that shows the  life of the  zarist court and diplomatic circles of St. Petersburg in that period. The Count of Cartagena’s activity has not been considered much  in Russian  historiography.  Based on the analysis of his memoirs, the author of the article suggests that Morillo considered the  Russian revolution to be brought in from outside. A study of the  memoirs showed that the  Spanish ambassador at St. Petersburg preferred  German diplomacy and had a peculiar notion of  the  role of Russia  in unleashing the  war.  The  author of this  article concludes that Morillo’s ideas were partly shaped by the  internal problems and the international situation of his own country at the beginning of the 20th century and that the  Spanish ambassador  was one of  the  Spanish Germanophiles. Spain maintained strict  neutrality throughout the  war. The  Spanish embassy  in Russia  carried out  important humanitarian mission  and active mediation activities, supporting Russian  citizens on enemy territory and trying to improve the  situation of Russian  prisoners of war and facilitate their return. The issue of the  personal participation of Anibal Morillo in mediation is also  touched upon in this article

    An application of the scale-adapted simulation to the unsteady flow across a tube bundle

    In the last decades, one of the main objectives pursued in the field of computational fluid dynamics has been the development of turbulent flow models and simulations techniques capable of generating predictions of flow patterns accurate enough to many industrial applications in a reasonable wall-clock time and at an acceptable cost. Very often, a trade-off has to be sought between the engineer's expectations regarding the accuracy of the model and the limited computational resources available. In that framework, the present contribution aims at demonstrating the capabilities of the SST-SAS approach which is considered as an intermediate model with respect to accuracy and computational requirements. To that end, the turbulent flow through a tube bundle was selected as test case. Preliminary sensitivity analyses were carried out to properly choose the residuals tolerance level, duration of the physical time integration, mesh size and time step value. The SST-SAS results are compared with available experimental data as well as with different simulations results taken from the literature. A quantitative scoring criterion was defined to sort out the different models results: the SST-SAS was at the top, sharing the first place with a URANS-RSM approach.Fil: Grioni, Mauro.     Uterine adenocarcinoma with generalised metastasis in a bottlenose dolphin Tursiops truncatus from northern Patagonia, Argentina

    An endometrial adenocarcinoma with areas of squamous differentiation and generalised metastasis was observed in a bottlenose dolphin Tursiops truncatus stranded in northern Patagonia in July 1997. This is the second report of a uterine adenocarcinoma in a free-living cetacean and the first in a Delphinidae. This neoplasm likely compromised reproduction for several years. In addition, the dolphin presented tattoo-like skin lesions and its digestive tract was infested by Anisakis simplex, Pseudoterranova sp., Braunina cordiformis and Corynosoma australe.Fil: Sanchez, Juana Elisabet.     Skepticism and Clandestine Literature: Doutes des Pyrrhoniens

    The latest studies on clandestine philosophical literature reveal the importance of this corpus to understand how skeptical tropes and lexicon were modified during the Modern Age. We will try to show it by means of an anonymous text written surely after 1711 and entitled Doutes des pyrrhoniens. First, we will present the manuscript in its most important arguments; second, we will concentrate in the eighth chapter and specially in the relationship between religion and politics; finally, we will take into account a differentiation that the author establishes between "moderate pyrrhonism" and "furious pyrrhonism" to consider its importance. Our main argument will be that the difference between these two forms of pyrrhonism is political, not epistemological. Indeed, both forms coincide in that religion is a fact of power that has nothing to do with an alleged divine revelation. The question is no longer about the truth of religion, but about whether or not it is convenient to abandon it as a regulatory component of society.Fil: Bahr, Fernando Anibal.
