Polymerization of misfolded Z alpha-1 antitrypsin protein lowers CX3CR1 expression in human PBMCs
The CX3CR1 (chemokine (C-X3-C motif) receptor 1) expression levels on immune cells have significant importance in maintaining tissue homeostasis under physiological and pathological conditions. The factors implicated in the regulation of CX3CR1 and its specific ligand CX3CL1 (fractalkine) expression remain largely unknown. Recent studies provide evidence that host`s misfolded proteins occurring in the forms of polymers or amyloid fibrils can regulate CX3CR1 expression. Herein, a novel example demonstrates that polymers of human ZZ alpha-1 antitrypsin (Z-AAT) protein, resulting from its conformational misfolding due to the Z (Glu342Lys) mutation in SERPINA1 gene, strongly lower CX3CR1 mRNA expression in human PBMCs. This parallels with increase of intracellular levels of CX3CR1 and Z-AAT proteins. Presented data indicate the involvement of the CX3CR1 pathway in the Z-AAT-related disorders and further support the role of misfolded proteins in CX3CR1 regulation.Funder: Deutsche Forschungsge-meinschaft, Grant number: STR 1095/6-1, Author: Pavel Strnad. Walking through Paper Dreams Pavla Z.
This diploma work is mainly a monography of a Czech author Pavel Zajicek. The base of this study is the analysis and the interpretation of his lifelong official and samizdat literary work. In the analysis of some of his prerevolutionary books as the main source I used mainly the critiques of the contemporary literary magazines. I also present the life of the author itself, especially in the context of the period he lived in. I will briefly mention two musical bands that are closely related to this personality {--} DG 307 and PPU. In my studies I use my personal e-mail with Pavel Z. and I do not forget to observe all accesible dialogues with this poet. This diploma work is mainly a monography of a Czech author Pavel Zajicek. Description of theoretical ap- proaches to modeling of localized surface plasmons on nanoparticles and nanostruc- tures is given. These techniques are used to study the process of optical excitation of localized surface plasmons on individual metallic nanoparticles and to detemine main performance characteristics of LSP-based sensors. Experimental study of LSP on spherical nanoparticles is reported. It is demonstrated that metallic nanoparticles supporting LSP can be used to detect changes in the refractive index or molecular binding events. The Practical part is the research part, the research was done using questionnaire surveys, classrom visits and interview with the starting teachers and teachers with long experience. Title: Scientific Literacy of 15-years old pupils in the Czech Republic in the International Survey PISA Author: Pavel Petrášek Department: Department of Physics Education Supervisor: PhDr. Martin Chvál, Ph.D., Department of Physics Education Abstract: My thesis focuses on the results of 15-year old pupils in selected tasks of the international survey PISA. It contains a brief introduction to the methodology of a survey and its realization in our conditions. The thesis presents relations between pupils` success and other variables found out in additional questionnaires, such as sex, parents` education, relation to mathematics or pupils` social and economical backgrounds. Besides the thesis contains the comparison of the importance of individual variables and results from the years 2000 and 2003. The thesis can be used as a theme for further research of mentioned relations and their comparison in a longer time horizon with other results. Electronic form of my thesis is on enclosed CD. Next part of this work is devoted to undesrtanding man in work of both thinkers. This work deals also contemporary evolutionism, based especially on statistical methods of work with genes and their models.
