    Obiettivo del congresso è l’approfondimento, nel contesto internazionale, delle conoscenze e delle strategie di intervento relative alla conservazione e valorizzazione de patrimonio architettonico, urbano e paesaggistico. Il congresso vuole riunire la comunità accademica internazionale interessata alla divulgazione scientifica e offrire uno spazio per la discussione dei temi relativi alla documentazione, conservazione e riuso del patrimonio architettonico, ma anche garantire un’opportunità di diffusione dei risultati dei progetti di ricerca e la condivisione dell’esperienza didattica. ReUSO 2016 mira ad un’analisi delle procedure pratiche per la valorizzazione del patrimonio architettonico storico nei diversi contesti culturali per promuovere considerazioni sulla musealizzazione e sulla flessibilità delle nuove funzioni previste per i beni architettonici in relazione alle variazioni di mercato. Mira ad un’analisi delle strategie urbane e degli aspetti tecnologici connessi al recupero del costruito con l’obiettivo di diffondere le conoscenze teoriche e pratiche rispetto al miglioramento delle prassi costruttive per riflettere sulla compatibilità degli interventi in relazione al valore culturale e materiale del patrimonio. Il carattere interdisciplinare di questo convegno intende riconnettere una serie di tematiche che fino ad ora hanno viaggiato da un punto di vista scientifico parallelamente. Il congresso consolida e rinnova le tematiche sviluppate nelle precedenti edizioni, approfondendo la specificità del riuso nei diversi settori scientifici e nelle diverse discipline che ruotano attorno all’architettura

    Ciona robusta hemocyte populational dynamics and PO-dependent cytotoxic activity

    Hemocyte populations from the ascidian Ciona robusta, separated through a Percoll discontinuous density gradient, are further characterized by May-Grünwald-Giemsa staining and a cytochemical reaction for phenoloxidase. Variability in cell density, acidophilic property and phenoloxidase activity suggest multiple hemocyte type populations, cell lineages and morphotypes that may be involved in distinct cellular responses. Therefore, unilocular refractile granulocytes, typical of this ascidian species, enriched in a fraction separated from the hemolymph show in vitro phenoloxidase-dependent cytotoxic activity against mammalian erythrocytes and a tumor cell lineage, in addition the properties listed above indicate relationships with vacuolated signet ring cells. Finally, bromo-deoxyuridine with, diamino-phenylindole fluorescent reaction and May-Grünwald-Giemsa staining show that in the hemolymph there are hyaline amoebocytes and granulocytes with potential proliferating activity. Present findings and reviewed images of previously reported inflammatory hemocytes in the tunic and pharynx allow us to speculate on theoretical outlines of hemocyte differentiation pathways

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    The inflammatory Response of Urochordata : The basic process of the Ascidians' innate immunity

    Ascidians form a widespread marine invertebrate group and are heterogeneous in terms of the taxonomic groups’ evolutionary lineages. The ascidian genomes lack significant homologies for rearranging genes of the vertebrate adoptive immunity. Genome analysis, gene sequencing, and transcriptional profiling have allowed us to disclose upregulation of innate immunity genes and cell labeling with riboprobes and antibodies has identified hemocyte types in tunic and pharynx inflammatory responses. Lymphocyte-like cells are stem cells and their immunocompetence has been proposed. Granulocyte types (compartment/morula cells) and hemocytes with large granules/vacuoles (compartment/morula cells) are mature cells expressing and releasing inflammatory components. LPS stimulates gene families of innate immune receptor homologs of the mammalian counterparts, as well as immune regulatory genes, during inflammatory responses. Proinflammatory components are involved in allogeneic reactions, and nonself and missing-self recognitions may be proposed. The findings on Ig-like domains contained in chitin-binding proteins (VCBPs) indicate the ancestral origin of vertebrate adaptive immunity and show that relevant genetic circuitry was already in place in the common ancestor of the protochordates and vertebrates. On the other hand, ascidians share with the other invertebrates the prophenoloxidase system that produces melanin and is involved in the inflammatory cytotoxic mechanism. The peroxinectin gene is also upregulated. Damage signals could be proinflammatory, but there are difficulties in assessing this that presumably could be examined during larva metamorphosis. Findings indicate that genetic circuitries relevant for vertebrate innate immunity were already in place in the common ancestor of the protochordates and vertebrates

    The article presents the research activities conducted on several rooms included into the extension project of the Grandi Uffizi Museum. The research work addresses to how define accurate survey methods for monitoring the static performances and plastic deformations of the structures. The research, strongly supported by the heads of the construction works, has helped the technical reconnaissance operations on these rooms, subjected to structural reinforcements and safety measures. To achieve compliant results in line with the project requirements of the construction site it has been necessary to identify the most appropriate graphic representation methods. The main 3D laser scanner survey activities have been accompanied by specific experiences of photomodeling and intensive photographic documentation campaigns. The post production and data processing steps have produced a substantial documentation of graphic materials as floor plans, detailed sections, with the elaboration of wire frame vector drawings and ortho-photo reconstructions, 3D models for the simulation of the static performances and plastic deformations with the overlapping and comparison of the different investigation steps by implementing the reverse engineering processes. The opportunity to support the construction site from 2010 to the present, improving these methods has contributed to the deepening of detection and representation techniques for the definition of operational methods characterized by a high metrical reliability with a detailed data post production results, extremely recommended for supporting sophisticated construction sites as the Grandi Uffizi represents

    Hemocytes of Rhynchophorus ferrugineus (Olivier)(Coleoptera: Curculionidae) and their response to Saccharomyces cerevisiae and Bacillus thuringiensis

    Originally from tropical Asia, the Red Palm Weevil (RPW Rhynchophorus ferrugineus (Olivier) is the most dangerous and deadly pest of many palm trees, and there have been reports of its recent detection in France, Greece and Italy. At present, emphasis is on the development of integrated pest management based on biological control rather than on chemical insecticides, however the success of both systems is often insufficient. In this regard, RPW appears to be one pest that is very difficult to control. Thus inves- tigations into the natural defences of this curculionid are advisable. RPW hemocytes, the main immuno- competent cells in the insect, are described for the first time. We identified five hemocyte cell types from the hemolymph of R. ferrugineus: plasmatocytes (50%), granulocytes (35%), prohemocytes (8%), oenocytes (4%) and spherulocytes (3%). SEM observations were also carried out. Some aspects of RPW interaction with non-self organisms, such as Saccharomyces cerevisiae and the entomopathogen bac- terium, Bacillus thuringiensis (Bt), are discussed. Plasmatocytes and granulocytes were involved in nodules and capsule formation as well as in the phagocytosis of yeast. The hemocyte response of RPW larvae to sub-lethal doses of commercial products containing Bt was examined. In vivo assays were carried out and Bt in vegetative form was found in the hemolymph. After a diet containing Bt, the number of total hemo- cytes, mainly plasmatocytes, in the RPW larva hemolymph declined sharply (12%) and then remained at a low level, while the number of other circulating cells was almost unchanged
