    Reorientational Dynamics of Amyloid-β from NMR Spin Relaxation and Molecular Simulation

    Amyloid-β (Aβ) aggregation is a hallmark of Alzheimer's disease. As an intrinsically disordered protein, Aβ undergoes extensive dynamics on multiple length and time scales. Access to a comprehensive picture of the reorientational dynamics in Aβ requires therefore the combination of complementary techniques. Here, we integrate 15N spin relaxation rates at three magnetic fields with microseconds-long molecular dynamics simulation, ensemble-based hydrodynamic calculations, and previously published nanosecond fluorescence correlation spectroscopy to investigate the reorientational dynamics of Aβ1-40 (Aβ40) at single-residue resolution. The integrative analysis shows that librational and dihedral angle fluctuations occurring at fast and intermediate time scales are not sufficient to decorrelate orientational memory in Aβ40. Instead, slow segmental motions occurring at ∼5 ns are detected throughout the Aβ40 sequence and reach up to ∼10 ns for selected residues. We propose that the modulation of time scales of reorientational dynamics with respect to intra- and intermolecular diffusion plays an important role in disease-related Aβ aggregation

    Mediastinal sympathetic tumours in children

    Eleven children suffering from mediastinal tumours of sympathetic origin (4 neuroblastomas, 2 ganglioneuroblastomas, and 5 ganglioneuromas) are presented. In almost all instances the tumours were discovered by chance following radiological studies of the chest undertaken for various reasons; in 1 case the tumour was unexpectedly discovered during surgery for a retroperitoneal mass. VMA urinary levels were in all cases within the normal range. Surgery was performed as soon as possible. In 5 cases the mass was completely removed, in 5 others incompletely and in 1 case only the extrathoracic portion of the neoplasm was partially removed. In 5 of the 6 malignant cases surgery was followed by complementary radio- and chemotherapy. All the children are in good health 2 mo. to 15 yrs. from the date of surgery. The outstanding results obtained in the histologically unfavourable cases (including the 5 in which surgical removal was incomplete) are further confirmation of how the adequately treated mediastinal neuroblastoma has greater possibilities for being cured than the retroperitoneal one. Age (5 children were under two years) and possible spontaneous maturation of the neoplasm may have played an important role in the favourable outcome as well. Furthermore, in those cases in which the residual neoplasm was subjected to electrosurgical fulguration, a role in the healing process can probably be attributed to immunological factors

    WHY DEMAND UNCERTAINTY CURBS INVESTMENT

    From a theoretical point of view, uncertainty over the demand for a firm’s product may have unclear effects on investments, as it depends on a number of factors, such as the production technology and the degree of competition in the product market. Most of the empirical research has been based on cross-section analysis, which has prevented a deeper investigation of the interplay of different factors in the temporal dimension. The aim of this paper is to extend the results of the empirical literature by using a panel of Italian firms over a fairly long period of time (1996-2004), covering a complete business cycle. A key finding of our paper concerns the role played by the degree of the competition faced by Italian firms over the 1996-2004 period. More specifically, the gradual loss of market power experienced by Italian manufacturing firms along with the increasing flexibility of the labour input may have contributed to weaken the negative effect of uncertainty on investment decisions

    Forecasting industrial production in the Euro area

    The creation of the Euro area has increased the importance of obtaining timely information about short-term changes in the area's real activity. In this paper we propose a number of alternative short term forecasting models, ranging from simple ARIMA models to more complex cointegrated VAR and conditional models, to forecast the index of industrial production in the euro area. A conditional error-correction model in which the aggregate index of industrial production for the area is explained by the US industrial production index and the business confidence index from the European Commission harmonised survey on manufacturing firms achieves the best score in terms of forecasting capacity

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
