ABSTRACT
First Person is a series of interviews with the first authors of a selection of papers published in Journal of Cell Science, helping early-career researchers promote themselves alongside their papers. Chaitali Khan is the first author on ‘The initiator caspase Dronc plays a non-apoptotic role in promoting DNA damage signalling in D. melanogaster’, published in Journal of Cell Science. Chaitali is a PhD student in the lab of Basuthkar J. Rao at the Tata Institute of Fundamental Research, Mumbai, India, investigating the principles of tissue homeostasis in the context of DNA damage, cell death and cellular signalling.</jats:p
Sustainable competitive advantage of service sector organisations in India through environmental regulations compliance
This research began with the objective of developing an understanding of the implications of environmental regulations on businesses. This study is significantly motivated by the academically famed and much debated \u93Porter Hypothesis\u94, which proposes that businesses can gain considerable competitive advantage if they comply with the most stringent environmental regulations. The author wanted to explore the applicability of this proposition in relation to the service sector organizations in India. Firstly, the globally adopted generic environmental management strategies were identified through literature review to prepare a questionnaire in order to gather viewpoints on what constitutes the present scenario in India in terms of climate change related modifications being introduced by service sector firms into their management strategies. Restoration of autophagy decreases neuronal cell death after traumatic brain injury
Traumatic brain injury (TBI) is the leading cause of death and disability in children and young adults in United States. Around 5.3 million people are estimated to be living TBI-related disability in United States alone (https://www.ninds.nih.gov). TBI is often associated with accumulation of aggregated proteins and neuronal cell death, underscoring importance of degradation pathway such as autophagy. In our study, we attempted to understand the role of autophagy in TBI-related neuronal cell death. Using lateral fluid percussion injury model in rats, we found that autophagy flux was inhibited after TBI. Importantly, we discovered that treatment with 18 amino acid TAT-Beclin1 peptide increased autophagy flux and significantly reduced neuronal cell death after TBI. Further investigation revealed the role of integrated stress response (ISR) mediated by ER stress markers ATF4, CHOP and GADD34, as well as transcription factor TFEB as mediators of neuronal cell death after TBI. TAT-Beclin1 peptide induced autophagy decreased ISR in neurons and inhibited TFEB signaling, likely due to increased AKT activity. Using LFP injury model in rats as well as cortical neuron stretch injury in vitro, we also found that impaired autophagy in TBI leads to sequestration of Wnt signal mediator Dishevelled-1 (DVL-1) in autophagosomes. Sequestration of Dvl-1 likely contributes to decreased Wnt signaling, increased GSK3 activity, and phospho-Tau buildup. Furthermore, we report that TAT-Beclin1 induced autophagy prevented Dvl-1 sequestration, decreased pTau buildup and reduced neuronal cell death. Based on these findings, we postulate that autophagy flux is inhibited after TBI, and induction of autophagy by TAT-Beclin1 is beneficial in decreasing TBI-related neuronal cell death.Ph.D.Includes bibliographical referencesby Chaitali M. Considering ethics and power relations in a qualitative study exploring experiences of divorce among British-Indian adult children
"This paper outlines the need for the critical scrutiny of ethics and power relations embedded in
the research process, particularly when researching minority ethnic communities. Research with
minority groups within institutional and structural contexts is challenging. It demands close engagement
for recruitment, participation of minority community members for research. in addition,
critical interpretation and reflexivity on part of researchers is vital to ensure that knowledge
generated is not biased, or harmful that pathologises minority groups.
This paper systematically considers issues of recruitment, participation, and interpretation
throughout the research process through a qualitative research study carried out with British-
Indian adult children of divorce. In doing so, it considers the strategies used and critical discusses
their outcomes. It does not present findings but considers the experiences of conducting
this research within the larger contexts of inquiry focussing on minority ethnic groups. The research
processes are reflexively considered by the author and emphasises the need to consider
power as dynamic relationship by engaging with the positions of the researcher and the researched,
differing agendas, gender, cultural and linguistic influences within the interview that
have shaped the data obtained.
The paper concludes that research with minority ethnic communities is important and critical but
needs to be conducted in ways that are cultural sensitivity, involve communities, provides opportunities
for participation. Due to their minority status, research with this groups presents a
greater need for analytical transparency-validation and critical researcher reflexivity." [author's abstract
Within and cross-corpus speech emotion recognition using latent topic model-based features
abstract: Owing to the suprasegmental behavior of emotional speech, turn-level features have demonstrated a better success than frame-level features for recognition-related tasks. Conventionally, such features are obtained via a brute-force collection of statistics over frames, thereby losing important local information in the process which affects the performance. To overcome these limitations, a novel feature extraction approach using latent topic models (LTMs) is presented in this study. Speech is assumed to comprise of a mixture of emotion-specific topics, where the latter capture emotionally salient information from the co-occurrences of frame-level acoustic features and yield better descriptors. Specifically, a supervised replicated softmax model (sRSM), based on restricted Boltzmann machines and distributed representations, is proposed to learn naturally discriminative topics. The proposed features are evaluated for the recognition of categorical or continuous emotional attributes via within and cross-corpus experiments conducted over acted and spontaneous expressions. In a within-corpus scenario, sRSM outperforms competing LTMs, while obtaining a significant improvement of 16.75% over popular statistics-based turn-level features for valence-based classification, which is considered to be a difficult task using only speech. Further analyses with respect to the turn duration show that the improvement is even more significant, 35%, on longer turns (>6 s), which is highly desirable for current turn-based practices. In a cross-corpus scenario, two novel adaptation-based approaches, instance selection, and weight regularization are proposed to reduce the inherent bias due to varying annotation procedures and cultural perceptions across databases. Experimental results indicate a natural, yet less severe, deterioration in performance - only 2.6% and 2.7%, thereby highlighting the generalization ability of the proposed features
Strategies for critical reflection in international contexts for social work students
International contexts provide social work students with the opportunity to develop knowledge of international social work, global citizenship and cultural competency. While these contexts are powerful sites of learning, there is a need to ensure that this occurs within a critical framework. The paradigm of critical reflection is used to facilitate this and has been popular in international programs. In this article, we develop this further by describing critically-reflective techniques and providing examples used in a pilot exchange program between a social work school in the UK and in India. The potential implications of these strategies for social work education are discussed. © The Author(s) 201
Low Complexity Optical Flow Using Neighbor-Guided Semi-Global Matching
abstract: Many real-time vision applications require accurate estimation of optical flow. This problem is quite challenging due to extremely high computation and memory requirements. This thesis focuses on designing low complexity dense optical flow algorithms.
First, a new method for optical flow that is based on Semi-Global Matching (SGM), a popular dynamic programming algorithm for stereo vision, is presented. In SGM, the disparity of each pixel is calculated by aggregating local matching costs over the entire image to resolve local ambiguity in texture-less and occluded regions. The proposed method, Neighbor-Guided Semi-Global Matching (NG-fSGM) achieves significantly less complexity compared to SGM, by 1) operating on a subset of the search space that has been aggressively pruned based on neighboring pixels’ information, 2) using a simple cost aggregation function, 3) approximating aggregated cost array and embedding pixel-wise matching cost computation and flow computation in aggregation. Evaluation on the Middlebury benchmark suite showed that, compared to a prior SGM extension for optical flow, the proposed basic NG-fSGM provides robust optical flow with 0.53% accuracy improvement, 40x reduction in number of operations and 6x reduction in memory size. To further reduce the complexity, sparse-to-dense flow estimation method is proposed. The number of operations and memory size are reduced by 68% and 47%, respectively, with only 0.42% accuracy degradation, compared to the basic NG-fSGM.
A parallel block-based version of NG-fSGM is also proposed. The image is divided into overlapping blocks and the blocks are processed in parallel to improve throughput, latency and power efficiency. To minimize the amount of overlap among blocks with minimal effect on the accuracy, temporal information is used to estimate a flow map that guides flow vector selections for pixels along block boundaries. The proposed block-based NG-fSGM achieves significant reduction in complexity with only 0.51% accuracy degradation compared to the basic NG-fSGM.Dissertation/ThesisMasters Thesis Computer Science 201
